Objectives: To evaluate the degree of tubular involvement in INS at various stage of the disease. Methods: 19 patients with INS were studied. 13 were steroid responders (group 1). 5 of them had biopsy which showed MCD. 6 patients were non responder to steroid or were steroid dependant with frequent relapses (group 2). Biopsies showed 3 FSGS and 3 MCD. They were treated with prednisone, ciclosporin and/ or mycofenolate mofetil. Protein, microalbumin (ALB), alpha-microglobulin (AMG), N-acetyl-beta-D-glucosaminidase (NAG) and creatinine (cr) were measured in each urine sample. Patients were considered in remission if prot/ cr ratio (g/mol) was < 20 (group 1a and 2a), and in relapse if the ratio was > 200 (group 1c and 2c). Some patients in group 1 had non nephrotic proteinuria (group 1b). Tubular dysfunction was defi ned by NAG/cr ratio (mg/mmol) > 0.86 or by AMG/cr ratio (mg/mmol) > 1.58. Conclusions: These data indicate that tubular dysfunction occurs in INS but only in patients in relapse. In this population, tubular dysfunction was independent of the severity of the nephrotic syndrome, the treatment protocol and the histopathology.

o interesse da República de Cabo Verde na utiliza9ao da energia renovável é basicamente económico. O país é muito pobre e todo o custo de energia pesa profundamente na sua economia nacional,visto nao existirem recursos de energia "tradicional". A utiliza~ao da energia renovável para bombagem de água é por isso, de grande importancia porque reduz a importa~ao de petróleo. Tanto a energia eólica como a energia solar oferecem grandes possibilidades em Cabo Verde. O país está sendo atingido pelo pior e mais langa período de seca jamais experimentado na sua história. Pode-se imaginar os problemas enfrentados para abastecimento de água a populac~o, abeberamento de animais e irriga- .., , ,,, , ~ao das poucas areas agrlcolas. O abastecimento de água potável é extremamente importante neste país, é apenas para a sobrevivencia da maior parte da sua popula9ao rural (190.000 habitantes), com pouco poder aquisitivo dado a diminuisao da área agrícola irrigada e a quantidade de gado, nos últimos 15 anos. Na agricultura irrigada, de momento totalizando apenas 2.000 ha, utilizase relativamente grande quantidade de combustível, porque 50% consiste em irrigasao por bombagem. Por esta razao o governo de Cabo Verde estimula fortemente a utilizasao das energias renováveis. As actividades neste campo come~aram em 1977 com a cria~ao da Divisao de Energias Renováveis (DER) dentro do Ministério do Desenvolvimento Rural (MUR). O governo de Cabo Verde e a PNUD (Projecto PNUD CVI/76/X05 - Demonstrasao de Energia Nao Convenciorial) puseram fundos a disposi~ao. Este projecto contribuiu com 3 aerogeradores e um volontário, Sr. Van Meel, que trabalhou anteriormente no Grupo de Energia Eólica de Eindhoven e que faz parte do CWD. Desde o seu início, a Divisao deu prioridade a bombagem de água para o abastecimento de água potável, o que foi e continua a ser urna necessidade urgente nas Ilhas de Cabo Verde. Os Servi~os da Igreja Mundial ofereceram 25 aerobombas Dempster com ventoinha de 8' de diametro e mais tarde a UNICEF distribuiu 10 Dempsters de ]4'. Gradualmente mais organiza90es financiadoras envolveram-se nas actividades da DER, como por exemplo a ClMADE, FAC, Embaixada An~ricana, Embaixada do Canadá, ICCO, etc. O Ministério do Desenvolvimento e Coopera9ao dos Países Baixos (DGIS) encarregou- se, em 1979, da norneasao do voluntárío da PNUD. Também em 1979 a USAID envolveu-se. Foi reconhecido e implementado um projecto de energia renovável (ref. 1). Este projecto comecou em Agosto de 1980. O papel prin cipal do projecto consistia na aquisiq~o de material e equipamento para urna nova oficina da DER. Urna das premissas do projecto era que o DGIS prestaria assistencia él Divisao. A pedido do DGIS a CWD realizou um estudo sobre a viabilidade de aplica9ao de aerobombas durante o primeiro semestre de 1980 (ref. 2). O estudo revelou que 75% das necessidades de água por bombagem em Cabo Verde, pode ria ser desempenhado pelos moinhos. - O projecto proposto diz respeito nao apenas a assistencia técnica, mas também ao financiamento da nova oficina e a aquisi9ao de cerca de 30 moi nhos. Este projecto bilateral entre Cabo Verde e os Países Baixos - Energias Renováveis", depois da aprova<;ao de Cabo Verde e do DGIS, veia a comecar no dia 1 de Julho de 1981 e estendeu-se por 3 anos. O período de 1977 a Julho de 1981 classifica-se agora como a primeira fase das actividades. Durante este período o CWD contribuiu com a assist€ncia técnica. O período de Julho de 1981 a Julho de 1984 é classificado como segunda fase e será relatado neste documento. O projecto foi executado pela DER. O Director da DER é, desde Fevereiro de 1983, o Sr. Daniel Livramento, licenciado em Física (Brasil 1982). Outros funcionários do corpo directivo sao o Sr. David Cardoso (desde Novem bro de 1983) e muito recentemente, desde Maio de 1984, o Sr. F. Ferreira7 A colaboracao dos Países Baixos para o projecto tem sido efectuada por En genheiros Consultores do DHV, em nome e como associados do CWD. O Sr. K.Versteegh, consultor do CWD, comecou a trabalhar para o projecto em 1 de Julho de 1981 e o Sr. N.Pieterse,~consultordo CWD, em 15 de Setembro de 1981. O Sr. Van Meel deixou o projecto a 1 de Novembro de 1981. O projecto foi avaliado em Novembro de 1983, por urna missao tripartida (Cabo Verde, USAID, DGIS) - Ref. 3. As conclusoes desta missao de avaliacao foram, em geral, positivas e o prolongamento do projecto foi fortemen te recomendado, com €nfase sobre aprodu~ao local. Entretanto, a DER e o CWD prepararam urna proposta para a extensao do projecto por mais 3 anos (Ref. 4). Esta extensao foi aprovada por Cabo Verde e pelo DGIS. O Sr. Versteegh foi substituído em Julho de 1984 e o Sr. Pieterse será substituido em Setembro de 1984 pelos Srs. J.Diepens e H. van der Spek, respectivamente. O conteúdo deste relatório é o seguinte: No capitulo 2 é apresentada urna exposi~ao do trabalho que incluiu um resumo das actividades realizadas na fase 1. Nos restantes capítulos descrevem-se as actividades durante a segunda fase. O relatório divide-se em tr€s partes: la. During the last 2 years, several novel genes that encode glucose transporter-like proteins have been identified and characterized. Because of their sequence similarity with GLUT1, these genes appear to belong to the family of solute carriers 2A (SLC2A, protein symbol GLUT). Sequence comparisons of all 13 family members allow the definition of characteristic sugar/polyol transporter signatures: (1) the presence of 12 membrane-spanning helices, (2) seven conserved glycine residues in the helices, (3) several basic and acidic residues at the intracellular surface of the proteins, (4) two conserved tryptophan residues, and (5) two conserved tyrosine residues. On the basis of sequence similarities and characteristic elements, the extended GLUT family can be divided into three subfamilies, namely class I (the previously known glucose transporters GLUT1-4), class II (the previously known fructose transporter GLUT5, the GLUT7, GLUT9 and GLUT11), and class III (GLUT6, 8, 10, 12, and the myo-inositol transporter HMIT1). Functional characteristics have been reported for some of the novel GLUTs. Like GLUT1-4, they exhibit a tissue/cell-specific expression (GLUT6, leukocytes, brain; GLUT8, testis, blastocysts, brain, muscle, adipocytes; GLUT9, liver, kidney; GLUT10, liver, pancreas; GLUT11, heart, skeletal muscle). GLUT6 and GLUT8 appear to be regulated by sub-cellular redistribution, because they are targeted to intra-cellular compartments by dileucine motifs in a dynamin dependent manner. Sugar transport has been reported for GLUT6, 8, and 11; HMIT1 has been shown to be a H+/myo-inositol co-transporter. Thus, the members of the extended GLUT family exhibit a surprisingly diverse substrate specificity, and the definition of sequence elements determining this substrate specificity will require a full functional characterization of all members.

High-molecular-weight (HMW) penicillin-binding proteins (PBPs) are divided into class A and class B PBPs, which are bifunctional transpeptidases/transglycosylases and monofunctional transpeptidases, respectively. We determined the sequences for the HMW PBP genes of Streptococcus gordonii, a gingivo-dental commensal related to Streptococcus pneumoniae. Five HMW PBPs were identified, including three class A (PBPs 1A, 1B, and 2A) and two class B (PBPs 2B and 2X) PBPs, by homology with those of S. pneumoniae and by radiolabeling with [3H]penicillin. Single and double deletions of each of them were achieved by allelic replacement. All could be deleted, except for PBP 2X, which was essential. Morphological alterations occurred after deletion of PBP 1A (lozenge shape), PBP 2A (separation defect and chaining), and PBP 2B (aberrant septation and premature lysis) but not PBP 1B. The muropeptide cross-link patterns remained similar in all strains, indicating that cross-linkage for one missing PBP could be replaced by others. However, PBP 1A mutants presented shorter glycan chains (by 30%) and a relative decrease (25%) in one monomer stem peptide. Growth rate and viability under aeration, hyperosmolarity, and penicillin exposure were affected primarily in PBP 2B-deleted mutants. In contrast, chain-forming PBP 2A-deleted mutants withstood better aeration, probably because they formed clusters that impaired oxygen diffusion. Double deletion could be generated with any PBP combination and resulted in more-altered mutants. Thus, single deletion of four of the five HMW genes had a detectable effect on the bacterial morphology and/or physiology, and only PBP 1B seemed redundant a priori.

We tested whether individuals can exert control over the expression of attitudinal ambivalence and if this control is exerted with self-presentational concerns. Using the self-presentation paradigm, participants reported more ambivalence about Genetically Modified Organisms ("GMO") in a standard and a self-enhancement (present yourself positively) conditions than in a self-depreciation (present yourself negatively) condition, on both felt (Experiments 1a and 2a) and potential ambivalence, in its cognitive (Experiments 1b and 2b) and affective components (Experiments 1b and 2c). The role of ambivalent attitudes in conveying a positive social value was confirmed by the fact that the above effect was found on a controversial attitude object (GMOs) but the opposite appeared on a non-controversial one (e.g. tooth brushing, a truism; Experiment 3). Such a reversal was obtained by directly manipulating the perception of controversy on GMOs (Experiment 4). Attitudinal ambivalence may thus serve an adaptive function, i.e. achieving a positive social value.

(3R)-hydroxyacyl-CoA dehydrogenase is part of multifunctional enzyme type 2 (MFE-2) of peroxisomal fatty acid beta-oxidation. The MFE-2 protein from yeasts contains in the same polypeptide chain two dehydrogenases (A and B), which possess difference in substrate specificity. The crystal structure of Candida tropicalis (3R)-hydroxyacyl-CoA dehydrogenase AB heterodimer, consisting of dehydrogenase A and B, determined at the resolution of 2.2A, shows overall similarity with the prototypic counterpart from rat, but also important differences that explain the substrate specificity differences observed. Docking studies suggest that dehydrogenase A binds the hydrophobic fatty acyl chain of a medium-chain-length ((3R)-OH-C10) substrate as bent into the binding pocket, whereas the short-chain substrates are dislocated by two mechanisms: (i) a short-chain-length 3-hydroxyacyl group ((3R)-OH-C4) does not reach the hydrophobic contacts needed for anchoring the substrate into the active site; and (ii) Leu44 in the loop above the NAD(+) cofactor attracts short-chain-length substrates away from the active site. Dehydrogenase B, which can use a (3R)-OH-C4 substrate, has a more shallow binding pocket and the substrate is correctly placed for catalysis. Based on the current structure, and together with the structure of the 2-enoyl-CoA hydratase 2 unit of yeast MFE-2 it becomes obvious that in yeast and mammalian MFE-2s, despite basically identical functional domains, the assembly of these domains into a mature, dimeric multifunctional enzyme is very different.

Background. Hepatitis C virus (HCV) nonstructural protein 5A (NS5A) has been shown to modulate multiple cellular processes, including apoptosis. The aim of this study was to assess the effects of HCV NS5A on apoptosis induced by Toll-like receptor (TLR) 4 ligand, lipopolysaccharide (LPS). Methods. Apoptotic responses to TLR4 ligands and the expression of molecules involved in TLR signaling pathways in human hepatocytes were examined with or without expression of HCV NS5A. Results. HCV NS5A protected HepG2 hepatocytes against LPS-induced apoptosis, an effect linked to reduced TLR4 expression. A similar downregulation of TLR4 expression was observed in Huh-7-expressing genotype 1b and 2a. In agreement with these findings, NS5A inhibited the expression of numerous genes encoding for molecules involved in TLR4 signaling, such as CD14, MD-2, myeloid differentiation primary response gene 88, interferon regulatory factor 3, and nuclear factor-κB2. Consistent with a conferred prosurvival advantage, NS5A diminished the poly(adenosine diphosphate-ribose) polymerase cleavage and the activation of caspases 3, 7, 8, and 9 and increased the expression of anti-apoptotic molecules Bcl-2 and c-FLIP. Conclusions. HCV NS5A downregulates TLR4 signaling and LPS-induced apoptotic pathways in human hepatocytes, suggesting that disruption of TLR4-mediated apoptosis may play a role in the pathogenesis of HCV infection.

The distribution of Sn4+ cations within the five crystallographic sites of the magnetoplumbite (M) ‐like compound BaFe12−2xCoxSnxO19 has been analyzed using single‐crystal x‐ray‐diffraction data. The species Fe3+ and Co2+ cannot be distinguished using x rays because of their very similar atomic numbers; however, the calculation of the apparent valencies for the different sites allows an insight into the Co2+ cation segregation. The use of previous data from neutron powder diffraction allows a precise picture of the cation distribution, which indicates a pronounced site selectivity for both Sn4+ and Co2+ cations. The Sn4+ cations prefer the 4f2 sites and to a much lower extent the 12k sites, while they do not enter the octahedral 2a sites at all. Co2+ cations are distributed among tetrahedral and octahedral sites displaying a clear preference for the tetrahedral 4f1 sites. Magnetic measurements indicate that the compound still exhibits uniaxial anisotropy with the easy direction parallel to the c axis. Nevertheless, the magnetic structure shows a considerable degree of noncolinearity. A strong reduction of the magnetic anisotropy regarding that of the undoped compound is also detected.

Microtubule-associated protein 1A (MAP1A) is essential during the late differentiation phase of neuronal development. Here, we demonstrated the presence of two MAP1A isoforms with a differential spatial distribution in the adult mouse barrel cortex. Antibody A stained MAP1A in pyramidal and stellate cells, including dendrites that crossed layer IV in the septa between barrels. The other antibody, BW6 recognized a MAP1A isoform that was mainly confined to the barrel hollow and identified smaller caliber dendrites. Previously, an interaction of MAP1A and the serotonin 5-hydroxytryptamine 2A (5-HT(2A)) receptor was shown in the rat cortex. Here, we identified, by double-immunofluorescent labeling, MAP1A isoform and serotonin 5-HT(2A) receptor distribution. MAP1A co-localized mainly with 5-HT(2A) receptor in larger apical dendrites situated in septa. This differential staining of MAP1A and a serotonin receptor in defined barrel compartments may be due to changes in the expression or processing of MAP1A during dendritic transport as a consequence of functional differences in processing of whisker-related sensory input.

Soil organic matter (SOM) plays a crucial role in soil quality and can act as an atmospheric C-CO2 sink under conservationist management systems. This study aimed to evaluate the long-term effects (19 years) of tillage (CT-conventional tillage and NT-no tillage) and crop rotations (R0-monoculture system, R1-winter crop rotation, and R2- intensive crop rotation) on total, particulate and mineral-associated organic carbon (C) stocks of an originally degraded Red Oxisol in Cruz Alta, RS, Southern Brazil. The climate is humid subtropical Cfa 2a (Köppen classification), the mean annual precipitation 1,774 mm and mean annual temperature 19.2 ºC. The plots were divided into four segments, of which each was sampled in the layers 0-0.05, 0.05-0.10, 0.10-0.20, and 0.20-0.30 m. Sampling was performed manually by opening small trenches. The SOM pools were determined by physical fractionation. Soil C stocks had a linear relationship with annual crop C inputs, regardless of the tillage systems. Thus, soil disturbance had a minor effect on SOM turnover. In the 0-0.30 m layer, soil C sequestration ranged from 0 to 0.51 Mg ha-1 yr-1, using the CT R0 treatment as base-line; crop rotation systems had more influence on soil stock C than tillage systems. The mean C sequestration rate of the cropping systems was 0.13 Mg ha-1 yr-1 higher in NT than CT. This result was associated to the higher C input by crops due to the improvement in soil quality under long-term no-tillage. The particulate C fraction was a sensitive indicator of soil management quality, while mineral-associated organic C was the main pool of atmospheric C fixed in this clayey Oxisol. The C retention in this stable SOM fraction accounts for 81 and 89 % of total C sequestration in the treatments NT R1 and NT R2, respectively, in relation to the same cropping systems under CT. The highest C management index was observed in NT R2, confirming the capacity of this soil management practice to improve the soil C stock qualitatively in relation to CT R0. The results highlighted the diversification of crop rotation with cover crops as a crucial strategy for atmospheric C-CO2 sequestration and SOM quality improvement in highly weathered subtropical Oxisols.
