The increased incidence of infections caused by the opportunistic pathogen Cryptococcus neoformans, which mainly affects immunocompromised patients but can also infect immunocompetent individuals, has needed additional studies on this micro-organism`s pathogenicity and factors related to virulence, such as enzyme production, for a better understanding of the aetiology of cryptococcosis. The aim of this study was to verify the applicability of non-denaturing PAGE for analysis of laccases by quantification of the amount of melanin pigment produced by clinical and environmental strains of C. neoformans. After incubation of the gel with the substrate L-dopa, strains produced melanin spots of a bright brown to black colour. Quantification of these spots was performed by densitometry analysis and the amount of melanin produced was calculated and compared among the strains. All strains showed laccase activity. Serotype B strains showed a higher melanin intensity than serotype A strains. Over half of the clinical strains (56.2%) showed the lowest melanin intensities, suggesting that melanin production may not be the main virulence factor against host defence. The clinical strain ICB 88 revealed two melanin spots on the gel, indicating the presence of two laccase isoforms. The environmental strains showed the highest values of melanin intensity, which may be related to previous exposure to environmental stress conditions.

The surface of midgut cells in Hemiptera is ensheathed by a lipoprotein membrane (the perimicrovillar membrane), which delimits a closed compartment with the microvillar membrane, the so-called perimicrovillar space. In Dysdercus peruvianus midgut perimicrovillar space a soluble aminopeptidase maybe involved in the digestion of oligopeptides and proteins ingested in the diet. This D. peruvianus aminopeptidase was purified to homogeneity by ion-exchange chromatography on an Econo-Q column, hydrophobic interaction chromatography on phenyl-agarose column and preparative polyacrylamide gel electrophoresis. The results suggested that there is a single molecular species of aminopeptidase in D. peruvianus midgut. Molecular mass values for the aminopeptidase were estimated to be 106 kDa (gel filtration) and 55 kDa (SDS-PAGE), suggesting that the enzyme occurs as a dimer under native conditions. Kinetic data showed that D. peruvianus aminopeptidase hydrolyzes the synthetic substrates LpNA, RpNA, A beta NA and AsnMCA (K(m)s 0.65, 0.14, 0.68 and 0.74 mM, respectively). The aminopeptidase activity upon LpNA was inhibited by EDTA and 1,10-phenanthroline, indicating the importance of metal ions in enzyme catalysis. One partial sequence of BLAST-identified aminopeptidase was found by random sequencing of the D. peruvianus midgut cDNA library. Semi-quantitative RT-PCR analysis showed that the aminopeptidase genes were expressed throughout the midgut epithelium, in the epithelia of V1, V2 and V3. Malphigian tubules and fat body, but it was not expressed in the salivary glands. These results are important in furthering our understanding of the digestive process in this pest species. Adults of Quesada gigas (Hemiptera: Cicadidae) have a major alpha-glucosidase bound to the perimicrovillar membranes, which are lipoprotein membranes that surround the midgut cell microvilli in Hemiptera and Thysanoptera. Determination of the spatial distribution of alpha-glucosidases in Q. gigas midgut showed that this activity is not equally distributed between soluble and membrane-bound isoforms. The major membrane-bound enzyme was solubilized in the detergent Triton X-100 and purified to homogeneity by means of gel filtration on Sephacryl S-100, and ion-exchange on High Q and Mono Q columns. The purified alpha-glucosidase is a protein with a pH optimum of 6.0 against the synthetic substrate p-nitrophenyl alpha-D-glucoside and M(r) of 61,000 (SDS-PAGE). Taking into account V(Max)/K(M) ratios, the enzyme is more active on maltose than sucrose and prefers oligomaltodextrins up to maltopentaose, with lower efficiency for longer chain maltodextrins. The Q gigas alpha-glucosidase was immunolocalized in perimicrovillar membranes by using a monospecific polyclonal antibody raised against the purified enzyme from Dysdercus peruvianus. The role of this enzyme in xylem fluid digestion and its possible involvement in osmoregulation is discussed. (C) 2009 Elsevier Inc. All rights reserved.

Denna uppsats har till syfte att få en överblick av hälsoläget inom Nås provinsialläkaredistrikt under hösten 1918 och våren 1919, då spanska sjukan grasserade. Uppsatsen undersöker vilken belastning spanska sjukan blev för epidemisjukvården och hur kommunen sörjde för den under epidemin. Mortaliteten under spanska sjukan visade ett liknande mönster som i andra delar av världen där ett stort antal unga människor avled. I undersökningen belyses även könstillhörigheten samt yrkeskategorisering av de döda. Under epidemins inledning var männen i majoritet men senare under epidemin blir det flest kvinnor, som avled i sviterna av spanskan. Mortalitetens yrkeskategorisering visar en klar majoritet för kroppsarbetare av olika slag samt hustrur till hemmansägare eller skogsarbetare och industriarbetare. Dödligheten var störst inom Vansbro municipalsamhälle under epidemin, beroende på att järnvägen och många resande passerade där. Pressens rapportering visar att Vansbro / Dala – Järna drabbades hårt av spanska sjukan.Som källmaterial användes provincialläkarrapporter från medicinalstyrelsens arkiv, kommunalnämndens och hälsovårdsnämndens protokoll, dödböcker och tidningartiklar ur tre lokala dagstidningar.

Uppsatsens syfte har varit att undersöka hur Kungliga Gotlands infanteriregemente drabbades av spanska sjukan under 1918-1920. Det har även jämförts hur detta regemente drabba¬des i jämförelse med Dalarnas och Västmanlands regementen under 1918. Frågeställning¬arna som besvarats handlar om hur många som insjuknade och dog, hur regementets ansva¬riga agerade samt vilka åtgärder som vidtogs när epidemin hade nått regementet. Det huvudsakliga materialet jag har använt mig av är förste provinsialläkarens årsberättelser och regementets inkomna och utgående skrivelser. Studiens resultat visar att Kungliga Gotlands infanteriregemente med sina cirka 500 sjuk¬domsfall och fyra dödsfall under epidemins första period, 1918 drabbades i hög grad i jämfö¬relse med Dalregementet, men i liten grad i jämförelse med Västmanlands rege¬mente. Anledningen till att regementet på Gotland klarade sig relativt lindrigt från spanska sjukan kan bero på att en fungerande sjukvård snabbt kunde sättas in samt att man vidtog andra åt¬gärder såsom inskränkningar på permissioner, striktare personlig hygien och renlighet i loka¬lerna samt skyddsympning.

Spanska sjukan var en världsomfattande epidemi, en pandemi, som drog över världen i flera influensavågor mellan 1918-1920. Syftet har varit att ta reda på hur Säters hospital drabbades av och hanterade influensautbrottet 1918. Med hjälp av överläkarens årsberättelse för 1918 har jag lyckats få fram att 40,7 % av alla patienter blev smittade av spanska sjukan varav 15,3 % av dem avled till följd av sjukdomen. 529 personer smittades av spanska sjukan på Säters sjukhusområde varav 57 personer avled, vilket motsvarar 10,8 %. Av de 529 smittade männen och kvinnorna avled 12,6 % kvinnor (34 stycken) och 8,9 % män (23 stycken) till följd av in-fluensan. Överläkaren beskrev i årsberättelsen att Säters hospital klarade sig förhållandevis bra, tack var det paviljongsystemet som möjliggjorde att influensan kunde isoleras.

Single-page applications have historically been subject to strong market forces driving fast development and deployment in lieu of quality control and changeable code, which are important factors for maintainability. In this report we develop two functionally equivalent applications using AngularJS and React and compare their maintainability as defined by ISO/IEC 9126. AngularJS and React represent two distinct approaches to web development, with AngularJS being a general framework providing rich base functionality and React a small specialized library for efficient view rendering. The quality comparison was accomplished by calculating Maintainability Index for each application. Version control analysis was used to determine quality indicators during development and subsequent maintenance where new functionality was added in two steps. The results show no major differences in maintainability in the initial applications. As more functionality is added the Maintainability Index decreases faster in the AngularJS application, indicating a steeper increase in complexity compared to the React application. Source code analysis reveals that changes in data flow requires significantly larger modifications of the AngularJS application due to its inherent architecture for data flow. We conclude that frameworks are useful when they facilitate development of known requirements but less so when applications and systems grow in size.
