Bread is one of the most widely consumed foods. Its impact on human health is currently of special interest for researchers. We aimed to identify biomarkers of bread consumption by applying a nutrimetabolomic approach to a free-living population. An untargeted HPLC q-TOF-MS and multivariate analysis was applied to human urine from 155 subjects stratified by habitual bread consumption in three groups: non-consumers of bread (n = 56), white-bread consumers (n = 48) and whole-grain bread consumers (n = 51). The most differential metabolites (variable importance for projection ≥1.5) included compounds originating from cereal plant phytochemicals such as benzoxazinoids and alkylresorcinol metabolites, and compounds produced by gut microbiota (such as enterolactones, hydroxybenzoic and dihydroferulic acid metabolites). Pyrraline, riboflavin, 3-indolecarboxylic acid glucuronide, 2,8-dihydroxyquinoline glucuronide and N-α-acetylcitrulline were also tentatively identified. In order to combine multiple metabolites in a model to predict bread consumption, a stepwise logistic regression analysis was used. Receiver operating curves were constructed to evaluate the global performance of individual metabolites and their combination. The area under the curve values [AUC (95 % CI)] of combined models ranged from 77.8 % (69.1 86.4 %) to 93.7 % (89.4 98.1 %), whereas the AUC for the metabolites included in the models had weak values when they were evaluated individually: from 58.1 % (46.6 69.7 %) to 78.4 % (69.8 87.1 %). Our study showed that a daily bread intake significantly impacted on the urinary metabolome, despite being examined under uncontrolled free-living conditions. We further concluded that a combination of several biomarkers of exposure is better than a single biomarker for the predictive ability of discriminative analysis.

Immaturity of the gut barrier system in the newborn has been seen to underlie a number of chronic diseases originating in infancy and manifesting later in life. The gut microbiota and breast milk provide the most important maturing signals for the gut-related immune system and reinforcement of the gut mucosal barrier function. Recently, the composition of the gut microbiota has been proposed to be instrumental in control of host body weight and metabolism as well as the inflammatory state characterizing overweight and obesity. On this basis, inflammatory Western lifestyle diseases, including overweight development, may represent a potential target for probiotic interventions beyond the well documented clinical applications. The purpose of the present undertaking was to study the efficacy and safety of perinatal probiotic intervention. The material comprised two ongoing, prospective, double-blind NAMI (Nutrition, Allergy, Mucosal immunology and Intestinal microbiota) probiotic interventions. In the mother-infant nutrition and probiotic study altogether 256 women were randomized at their first trimester of pregnancy into a dietary intervention and a control group. The intervention group received intensive dietary counselling provided by a nutritionist, and were further randomized at baseline, double-blind, to receive probiotics (Lactobacillus rhamnosus GG and Bifidobacterium lactis) or placebo. The intervention period extended from the first trimester of pregnancy to the end of exclusive breastfeeding. In the allergy prevention study altogether 159 women were randomized, double-blind, to receive probiotics (Lactobacillus rhamnosus GG) or placebo 4 weeks before expected delivery, the intervention extending for 6 months postnatally. Additionally, patient data on all premature infants with very low birth weight (VLBW) treated in the Department of Paediatrics, Turku University Hospital, during the years 1997 - 2008 were utilized. The perinatal probiotic intervention reduced the risk of gestational diabetes mellitus (GDM) in the mothers and perinatal dietary counselling reduced that of fetal overgrowth in GDM-affected pregnancies. Early gut microbiota modulation with probiotics modified the growth pattern of the child by restraining excessive weight gain during the first years of life. The colostrum adiponectin concentration was demonstrated to be dependent on maternal diet and nutritional status during pregnancy. It was also higher in the colostrum received by normal-weight compared to overweight children at the age of 10 years. The early perinatal probiotic intervention and the postnatal probiotic intervention in VLBW infants were shown to be safe. To conclude, the findings in this study provided clinical evidence supporting the involvement of the initial microbial and nutritional environment in metabolic programming of the child. The manipulation of early gut microbial communities with probiotics might offer an applicable strategy to impact individual energy homeostasis and thus to prevent excessive body-weight gain. The results add weight to the hypothesis that interventions aiming to prevent obesity and its metabolic consequences later in life should be initiated as early as during the perinatal period.

A microbiota do solo é de grande importância no desenvolvimento de culturas. Os métodos de controle, químico (brometo de metila) e físico (solarização), alteram essa microbiota. O presente trabalho objetivou estudar o comportamento da comunidade de fungos em solo solarizado e fumigado (brometo de metila). O delineamento experimental foi de blocos ao acaso, com três tratamentos (solarização, brometo de metila e testemunha) e sete repetições. A comunidade de fungos do solo foi avaliada de forma quantitativa e qualitativa, em três momentos (antes, durante e após a solarização) com amostras coletadas de três profundidades (0-5; 10-15 e 20-25 cm). Durante a solarização ocorreu uma redução na comunidade de fungos do solo, em termos quantitativos, em todas as camadas amostradas. No entanto, essa diminuição foi mais significativa na camada superficial (0-5cm). Em termos qualitativos, a solarização reduziu também o número de diferentes espécies de fungos do solo, mas na camada de 20-25 cm, essa diminuição foi a zero aos 56 dias de avaliação. A recolonização da microbiota do solo, em termos quantitativos, foi maior no tratamento com brometo de metila do que nos demais. Entretanto, esse aumento não foi o mesmo em termos qualitativos. Nos tratamentos solarizado e testemunha, o aumento na comunidade de fungos do solo foi acompanhado pela diversificação de espécies fúngicas.

The prevalence of inflammatory based diseases has increased in industrialized countries over the last decades. For allergic diseases, two primary hypotheses have been proposed to explain this phenomenon, namely the hygiene and dietary evolution based hypothesis. Particularly, the reduced early exposure to microbes and an increase in the amount of polyunsaturated fatty acids (especially n-6 PUFA) in the diet have been discussed. Often, these two factors have been studied independently, even though both factors have been shown to possess potential health benefits and their mode of action to share similar mechanisms. The hypothesis of the present study was that demonstrate that PUFA and probiotics are not separate entities as such but do interact with each other. In the present study, we investigated whether maternal diet and atopic status influence the PUFA composition of breast milk and serum fatty acids of infants, and whether the fatty acid absorption and utilization of infant formula fatty acids is affected by supplementation of infant formula with probiotic bacteria (Lactobacillus GG and Bifidobacterium lactis Bb-12). Moreover, we investigated the mechanisms by which different PUFA influence the physicochemical and functional properties of probiotics as well as functionality of epithelial cells in vitro. We demonstrated a carry-over effect of dietary fatty acids from maternal diet via breast milk into infants’ serum lipid fatty acids. Our data confirmed the previously shown allergy –related PUFA level imbalances, though it did not fully support the impaired desaturation and elongation capacity hypothesis. We also showed that PUFA incorporation into phospholipids of infants was influenced by probiotics in infant formula in a strain dependent manner. Especially,Bifidobacterium lactis Bb-12 in infant formula promoted the utilization of n-3 PUFA. Mechanistically, we demonstrated that probiotics (Lactobacillus GG, Lactobacillus casei Shirota and Lactobacillus bulgaricus) did incorporate and interconvert exogenous free PUFA in the growth medium into bacterial fatty acids strain and PUFA dependently. In general, high concentrations of free PUFA inhibited the growth and mucus adhesion of probiotics, whereas low concentrations of specific long chain PUFA were found to promote the growth and mucus adhesion of Lactobacillus casei Shirota. These effects were paralleled with only minor alterations in hydrophobicity and electron donor – electron acceptor properties of lactobacilli. Furthermore, free PUFA were also demonstrated to alter the adhesion capacity of the intestinal epithelial cells; n-6 PUFA tended to inhibit the Caco-2 adhesion of probiotics, whereas n-3 PUFA had either no or minor effects or even promote the bacterial adhesion (especially Lactobacillus casei Shirota) to PUFA treated Caco-2 cells. The results of this study demonstrate the close and bilateral interactions between dietary PUFA and probiotics. Probiotics were shown to influence the absorption and utilization of dietary PUFA, whereas PUFA were shown to alter the functional properties of both probiotics and mucosal epithelia. These findings suggest that a more thorough understanding of interactions between PUFA and intestinal microbiota is a prerequisite, when the beneficial effects of new functional foods containing probiotics are designed and planned for human intervention studies.

The endogenous microbiota, constituting the microbes that live inside and on humans, is estimated to outnumber human cells by a factor of ten. This commensal microbial population has an important role in many physiological functions, with the densest microbiota population found in the colon. The colonic microbiota is a highly complex and diverse bacterial ecosystem, and a delicate balance exists between the gut microbiota and its host. An imbalance in the microbial ecosystem may lead to severe symptoms in and also beyond the gastrointestinal tract. Due to the important role of the gut microbiota in human health, means of its modification have been introduced in the dietary concepts of pro-, pre- and synbiotics. Prebiotics, which are usually carbohydrates, strive to selectively influence beneficial microbes resident in the colon with the aim of modifying the composition and functionality of the commensal microbial population towards a purportedly healthier one. The study of prebiotic effects on colonic micro-organisms is typically done by using human faecal material, though this provides relatively little information on bacterial populations and metabolic events in different parts of the colon. For this reason, several in vitro models have been developed to investigate the gut microbiota. The aim of this doctoral thesis was to screen through some of the promising prebiotic candidates, characterize their effects on the microbiota through the use of two in vitro methods (pure microbial cultures and a colon simulator model) and to evaluate their potential as emerging prebiotics or synbiotics when combined with the probiotic Bifidobacterium lactis . As a result of the screening work and subsequent colon simulation studies, several compounds with promising features were identified. Xylo-oligosaccharides (XOS), which have previously already shown promise as prebiotic compounds, were well fermented by several probiotic Bifidobacterium lactis strains in pure culture studies and in the following simulation studies utilizing the complex microbiota by endogenous B. lactis Another promising compound was panose, a trisaccharide belonging to isomalto-oligosaccharides (IMO) that also was also able to modify the microbiota in vitro by increasing the number of beneficial microbes investigated. Panose has not been widely studied previously and therefore, this thesis work provided the first data on panose fermentation in mixed colonic microbiota. Galacto-oligosaccharide (GOS) is an established prebiotic, and it was studied here in conjunction with another potential polygosaccharide polydextrose (PDX) and probiotic B. lactis Bi-07. In this final study, the synbiotics including GOS were more effective than the constituting pro- or prebiotics alone in modulating the microbiota composition, thus indicating a synergy resulting from the combination. The results obtained in this in vitro work can be, and have already been, utilized in product development aimed at the nutritional modification of the human colonic microbiota. Some of the compounds have entered the human clinical intervention phase to nvestigate in more detail the prebiotic and synbiotic properties seen in these in vitro studies.

Rapid identification and resistance determination of pathogens in clinical specimens is vital for accurate treatment and monitoring of infectious diseases. Antimicrobial drug resistance is increasing globally and healthcare settings are facing this cost-intensive and even life-threatening problem. The incidence of resistant pathogens in Finland has remained relatively steady and manageable at least for the time being. DNA sequencing is the gold standard method for genotyping, mutation analysis, and identification of bacteria. Due to significant cost decrease in recent years, this technique is available to many research and clinical laboratories. Pyrosequencing technique, a rapid real-time DNA sequencing method especially suitable for analyzing fairly short stretches of DNA, was used in this study. Due to its robustness and versatility, pyrosequencing was applied in this study for identification of streptococci and detection of certain mutations causing antimicrobial resistance in different bacteria. Certain streptococcal species such as S. pneumoniae and S. pyogenes are significantly important clinical pathogens. S. pneumoniae causes e.g. pneumonia and otitis media and is one of the most important community-acquired pathogens. S. pyogenes, also known as group A streptococcus, causes e.g. angina and erysipelas. In contrast, the socalled alpha-haemolytic streptococci, such as S. mitis and S. oralis, belong to the normal microbiota, which are regarded to be non-pathogenic and are nearly impossible to identify by phenotypic methods. In this thesis, a pyrosequencing method was developed for identification of streptococcal species based on the 16S rRNA sequences. Almost all streptococcal species could be differentiated from one another by the developed method, including S. pneumoniae from its close relatives S. mitis and S. oralis . New resistance genes and their variants are constantly discovered and reported. In this study, new methods for detecting certain mutations causing macrolide resistance or extended spectrum beta-lactamase (ESBL) phenotype were developed. These resistance detection approaches are not only suitable for surveillance of mechanisms causing antimicrobial resistance but also for routine analysis of clinical samples particularly in epidemic settings. In conclusion, pyrosequencing was found to be an accurate, versatile, cost-effective, and rapid DNA sequencing method that is especially suitable for mutation analysis of short DNA fragments and identification of certain bacteria.

Dietary and microbial factors are thought to contribute to the rapidly increasing prevalence of T1D in many countries worldwide. The impact of these factors on immune regulation and diabetes development in non-obese diabetic (NOD) mice are investigated in this thesis. Diabetes can be prevented in NOD mice through dietary manipulation. Diet affects the composition of intestinal microbiota, which may subsequently influence intestinal immune homeostasis. However, the specific effects of anti-diabetogenic diets on gut immunity and the explicit associations between intestinal immune disruption and type 1 diabetes onset remain unclear. The research presented herein demonstrates that newly weaned NOD mice suffer from a mild level of colitis, which shifts the colonic immune cell balance towards a proinflammatory status. Several aberrations can also be observed in the peritoneal B cells of NOD mice; an increase in activation marker expression, increased trafficking to the pancreatic lymph nodes and significantly higher antigen presenting cell (APC) efficiency towards insulin-specific T cells. A shift towards inflammation is likewise observed in the colon of germ-free NOD mice, but signs of peritoneal B cell activation are lacking in these mice. Remarkably, most of the abnormalities in the colon, peritoneal macrophages and the peritoneal B cell APC activity of NOD mice are abrogated when NOD mice are maintained on a diabetes-preventive, soy-based diet (ProSobee) from the time of weaning. Dietary and microbial factors hence have a significant impact on colonic immune regulation and peritoneal B cell activation and it is suggested that these factors influence diabetes development in NOD mice.

Avaliou-se o desempenho de dois reatores anaeróbios de fluxo ascendente com manta de lodo (UASB) em escala-piloto com volumes de 908 L e 188 L, instalados em série, alimentados com águas residuárias de suinocultura com concentrações médias de sólidos suspensos totais (SST) variando de 2.216 mg L-1 a 7.131 mg L-1 e submetidos a tempos de detenção hidráulica (TDH) de 62,3 e 31,1 h, no primeiro reator, e de 12,9 e 6,5 h, no segundo reator. As eficiências médias de remoção de DQOtotal variaram de 74,0% a 89,6% no Reator 1 e de 34,3% a 45,1% no Reator 2, resultando em valores médios de 86,6% a 93,1% para o sistema de tratamento em dois estágios com carga orgânica volumétrica (COV) na faixa de 3,40 a 14,44 kg DQOtotal m-3 reator d-1 no Reator 1. As concentrações de metano no biogás foram acima de 75% para o Reator 1 e de 80% para o Reator 2. Os valores médios de pH variaram na faixa de 6,9 a 8,2 para o efluente do Reator 1 e de 7,0 a 8,3 para o efluente do Reator 2. Os ácidos voláteis totais mantiveram-se estáveis com concentrações médias abaixo de 200 mg L-1. Esses resultados indicaram que as condições de carga orgânica, em termos de DQO e SSV, impostas ao sistema de tratamento anaeróbio em dois estágios, não foram limitantes para que houvesse o desenvolvimento de lodo com microbiota adaptada e com alta atividade, propiciando altas eficiências médias de remoção de matéria orgânica (86,6 a 93,1% para DQOtotal e 85,6 a 88,2% para SSV) e taxas de produção de metano de 0,156 a 0,289 m³ CH4 kg-1 de DQO removida.

O mercado consumidor, atualmente, exige aumento gradativo do controle da qualidade dos produtos. Os métodos manuais de controle existentes, aplicáveis à produção animal, começam a se mostrar ineficientes para garantir percentual crescente dessa qualidade, pois essa garantia somente pode ser efetiva se houver rastreamento confiável do animal, desde o seu nascimento até o abate. A identificação individual apresenta grande importância nesse enfoque, pois possibilita a coleta de informações inerentes ao animal. A rastreabilidade eletrônica utiliza dispositivo eletrônico, que emite um sinal ativado por um leitor fixo, colocado onde for necessário registrar um determinado evento, ou um leitor manual que permite maior independência do operador. Sabendo-se da importância da identificação eletrônica como ferramenta para a rastreabilidade dos animais produzidos comercialmente, teve-se o objetivo de estudar o uso do identificador eletrônico (transponder), de forma a garantir tanto a leitura manual do indivíduo, como a leitura com antena fixa. Foram implantados nos seguintes locais: 1) fronte; 2) lóbulo externo; 3) parte posterior da base auricular, e 4) dentro de um brinco e implantado no lóbulo externo da orelha. Foram analisados os fatores seqüela e migração, posterior ao implante, assim como a eficiência de leitura na antena tanto fixa como manual. O melhor local de implante foi a parte posterior da base auricular.

O presente estudo teve o objetivo de avaliar a partida e a adaptação de um reator anaeróbio horizontal de leito fixo (RAHLF) no tratamento de águas residuárias do processamento primário dos frutos do cafeeiro (ARC). O reator foi construído com tubos de PVC de 0,2 m de diâmetro e 3,2 m de comprimento. O sistema foi preenchido com cubos de espuma de poliuretano para imobilização de biomassa ativa. O reator apresentou volume total de 0,1 m³ e volume útil equivalente a 0,04 m³. Em média, houve remoção de 49% da matéria orgânica, com o reator trabalhando sob carga orgânica volumétrica média de 2,66 kg m-3 d-1, medida como DQO. A suplementação de alcalinidade, somada à inoculação prévia de biomassa, proporcionou partida estável do RAHLF, confirmada pelo consumo de ácidos voláteis e adaptação da microbiota ao resíduo. O sistema apresentou resistência às variações de vazão e de carga orgânica observadas, e os teores de fenol e potássio monitorados não causaram inibição da atividade biológica no RAHLF. O maior controle sobre as variações de carga é fator importante na continuidade dos estudos.

In this study it was evaluated the start-up procedures of anaerobic treatment system with three horizontal anaerobic reactors (R1, R2 and R3), installed in series, with volume of 1.2 L each. R1 had sludge blanket, and R2 and R3 had half supporter of bamboo and coconut fiber, respectively. As an affluent, it was synthesized wastewater from mechanical pulping of the coffee fruit by wet method, with a mean value of total chemical oxygen demand (CODtotal) of 16,003 mg L-1. The hydraulic retention time (HRT) in each reactor was 30 h. The volumetric organic loading (VOL) applied in R1 varied from 8.9 to 25.0 g of CODtotal (L d)-1. The mean removal efficiencies of CODtotal varied from 43 to 97% in the treatment system (R1+R2+R3), stabilizing above 80% after 30 days of operation. The mean content of methane in the biogas were of 70 to 76%, the mean volumetric production was 1.7 L CH4 (L reactor d)-1 in the system, and the higher conversions were around at 0.20 L CH4 (g CODremoved)-1 in R1 and R2. The mean values of pH in the effluents ranged from 6.8 to 8.3 and the mean values of total volatile acids remained below 200 mg L-1 in the effluent of R3. The concentrations of total phenols of the affluent ranged from 45 to 278 mg L-1, and the mean removal efficiency was of 52%. The start-up of the anaerobic treatment system occurred after 30 days of operation as a result of inoculation with anaerobic sludge with active microbiota.

A administração oral e a aspersão com amitraz reproduziram experimentalmente em 17eqüinos e um asinino um quadro de intoxicação muito similar a outro que vinha ocorrendo em cavalos no Estado do Rio de Janeiro. O início dos sintomas após a administração oral variou entre 15min. e 2h05min., na aplicação por aspersão variou entre 6h28min. e 8h38min. A evolução nos casos de administração oral foi de 4 a 9 dias, nos de aspersão de 5 a 6 dias. Somente morreram animais que receberam a administração oral. Um animal aspergido com o amitraz foi sacrificado. Por via oral foram usadas dosagens de 5,5 mg/kg (uma administração), 5,8 mg/kg (duas administrações) e num terceiro animal, doses que variaram entre 7,2 e 36,4 mg/kg (cinco administrações). Nas aplicações por aspersão, a intoxicação foi reproduzida com soluções nas concentrações de 0,1 e 0,2%. Com relação ao sistema nervoso, os principais sinais observados foram apatia, sonolência, ptoses palpebral e auricular, dificuldade de apreensão, mastigação e deglutição do alimento, arrastar das pinças dos cascos no solo, exposição do pênis, sensibilidade cutânea diminuída/ausente, instabilidade em estação, abdução dos membros, cabeça baixa, incoordenação, bocejos, flacidez labial, exposição da língua, cruzamento dos membros ao caminhar, resposta postural diminuída após cruzar e abduzir os membros, reflexos do lábio superior, palatal, lingual, de deglutição e flexor diminuídos/ausentes, reflexos auricular, palpebral e de ameaça diminuídos e resposta ambulatória diminuída ao teste de girar em círculo de pequeno raio. No que se refere ao sistema digestivo, foram evidenciados, principalmente, hipomotilidade/atonia intestinal, edema dos lábios, distensão abdominal, deitar e levantar com freqüência, rolar no solo, olhar para o flanco, gemer e impactação do intestino grosso. Observaram-se ainda taquicardia, aumento do tempo de preenchimento capilar e mucosas congestas, estridor, taquipnéia, dispnéia, secreção nasal, bradipnéia e respiração abdominal. Todos os três casos naturais ocorreram após aspersão do amitraz. Os primeiros sintomas foram observados 2 e 3 dias após o banho. A evolução foi de 6, 7 e 17 dias. Um animal manifestou a maioria dos sinais referentes ao sistema nervoso observados nos experimentos, com exceção dos sinais de cruzamento dos membros ao caminhar, bocejos, lábios flácidos e exposição do pênis. Outro animal, intoxicado espontaneamente, manifestou somente sintomas digestivos como rolar, ''patear'', hipomotilidade/atonia intestinal e impactação do intestino grosso. Um terceiro animal, inicialmente manifestou sintomas digestivos caracterizados por patear, rolar, atonia intestinal e impactação do intestino grosso, com conseqüente desenvolvimento de laminite; na fase final exibiu acentuada sintomatologia nervosa mostrando compressão da cabeça contra obstáculos, incoordenação motora com cruzamento dos membros ao caminhar e relutância em se movimentar. Baseados no quadro clínico observado, são sugeridos possíveis locais de lesão no sistema nervoso.

Uma versão condensada em português de um artigo de revisão sobre a periodontite da "cara inchada" dos bovinos, publicado em inglês, está apresentada com algumas informações adicionais. A doença foi responsável por grandes perdas de bovinos jovens, principalmente nas décadas de 1970 e l980 no Brazil Central. Em face da periodontite progressiva e a perdas de dentes, os animais não podem se alimentar convenientemente, tornam-se emaciados e podem morrer. A doença foi tida como uma deficiência ou desequilíbrio mineral. Mas as pesquisas de campo e de laboratório, realizadas durante 30 anos, mostraram que trata-se de doença infecciosa multifatorial a ser definida como Periodontite Epizoótica Bovina. Chegou-se à conclusão que os fatores principais para o seu desenvolvimento são: (1) a idade dos bovinos na fase de erupção dos dentes premolares e molares; (2) a presença de bactérias do grupo Bacteroides spp nos espaços subgengivais; e (3) a ingestão com a forragem de concentrações subinibitórias de antibióticos, sobretudo de estreptomicina, produzidos por actinomicetos cujo número é aumentado em solos virgens recém-cultivados na formação de pastagens após a derrubada da mata ou da vegetação de Cerrado; isto leva a um aumento da aderência dos bacteróides ao epitélio gengival e à destruição dos tecidos peridentários. Hoje em dia, a doença perdeu a sua importância e praticamente desapareceu, porque a microbiota do solo entrou novamente em equilíbrio e a abertura de grandes áreas virgens para a pecuária cessou. Porém, novos surtos podem ocorrer em áreas anteriormente positivas para a doença quando, na reforma de pastagens ou capineiras, houver um novo desequilíbrio da microbiota do solo. Outros antibióticos, como a espiramicina e virginiamicina, administrados por via oral ou adicionado a misturas minerais, podem controlar a periodontite.

Este trabalho teve por objetivo estudar os efeitos da acidose láctica ruminal, induzida experimentalmente em caprinos, sobre o quadro clínico, as características físico-químicas e microbiológicas do fluido ruminal. Para tal, foram utilizados dez animais, sem raça definida (SRD), com peso médio de 25 kg, machos, com um a dois anos de idade, fistulados, clinicamente sadios e mantidos em baias. Após se estabelecer os padrões de normalidade para as variáveis estudadas, os animais foram induzidos experimentalmente a ter a acidose ruminal empregando-se a sacarose, na dose de 15 g/kg de peso corporal. As observações clínicas e laboratoriais foram realizadas nos intervalos de 4, 8, 12, 16, 24, 32, 48, 72, 96, 120 e 144 horas, após a indução (PI) onde se avaliou a intensidade do processo da acidose láctica ruminal. Os caprinos estudados apresentaram manifestações clínicas de acidose láctica ruminal já a partir das 4 horas PI, cujos sinais como apatia, anorexia, taquicardia, atonia ruminal, distensão abdominal e diarréia de intensidade variável foram observados. As características do suco ruminal sofreram alterações, ocorreu a redução do pH para valores inferiores a 5, a cor tornou-se leitosa, o odor ácido e a consistência aquosa. O tempo de atividade de sedimentação (TAS) reduziu seus valores, e a dinâmica da fauna e flora ficaram comprometidas, havendo um predomínio da microbiota Gram-positiva. Ao término do período de observação constatou-se que em alguns animais não ocorreu o restabelecimento pleno das variáveis analisadas.
