Spermatogenesis is a complex process reliant upon interactions between germ cells (GC) and supporting somatic cells. Testicular Sertoli cells (SC) support GCs during maturation through physical attachment, the provision of nutrients, and protection from immunological attack. This role is facilitated by an active cytoskeleton of parallel microtubule arrays that permit transport of nutrients to GCs, as well as translocation of spermatids through the seminiferous epithelium during maturation. It is well established that chemical perturbation of SC microtubule remodelling leads to premature GC exfoliation demonstrating that microtubule remodelling is an essential component of male fertility, yet the genes responsible for this process remain unknown. Using a random ENU mutagenesis approach, we have identified a novel mouse line displaying male-specific infertility, due to a point mutation in the highly conserved ATPase domain of the novel KATANIN p60-related microtubule severing protein Katanin p60 subunit A-like1 (KATNAL1). We demonstrate that Katnal1 is expressed in testicular Sertoli cells (SC) from 15.5 days post-coitum (dpc) and that, consistent with chemical disruption models, loss of function of KATNAL1 leads to male-specific infertility through disruption of SC microtubule dynamics and premature exfoliation of spermatids from the seminiferous epithelium. The identification of KATNAL1 as an essential regulator of male fertility provides a significant novel entry point into advancing our understanding of how SC microtubule dynamics promotes male fertility. Such information will have resonance both for future treatment of male fertility and the development of non-hormonal male contraceptives.

Seafloor massive sulfide (SMS) mining will likely occur at hydrothermal systems in the near future. Alongside their mineral wealth, SMS deposits also have considerable biological value. Active SMS deposits host endemic hydrothermal vent communities, whilst inactive deposits support communities of deep water corals and other suspension feeders. Mining activities are expected to remove all large organisms and suitable habitat in the immediate area, making vent endemic organisms particularly at risk from habitat loss and localised extinction. As part of environmental management strategies designed to mitigate the effects of mining, areas of seabed need to be protected to preserve biodiversity that is lost at the mine site and to preserve communities that support connectivity among populations of vent animals in the surrounding region. These "set-aside" areas need to be biologically similar to the mine site and be suitably connected, mostly by transport of larvae, to neighbouring sites to ensure exchange of genetic material among remaining populations. Establishing suitable set-asides can be a formidable task for environmental managers, however the application of genetic approaches can aid set-aside identification, suitability assessment and monitoring. There are many genetic tools available, including analysis of mitochondrial DNA (mtDNA) sequences (e.g. COI or other suitable mtDNA genes) and appropriate nuclear DNA markers (e.g. microsatellites, single nucleotide polymorphisms), environmental DNA (eDNA) techniques and microbial metagenomics. When used in concert with traditional biological survey techniques, these tools can help to identify species, assess the genetic connectivity among populations and assess the diversity of communities. How these techniques can be applied to set-aside decision making is discussed and recommendations are made for the genetic characteristics of set-aside sites. A checklist for environmental regulators forms a guide to aid decision making on the suitability of set-aside design and assessment using genetic tools. This non-technical primer document represents the views of participants in the VentBase 2014 workshop.

1. We analysed 41 years of data (1968–2008) from Blelham Tarn, U.K., to determine the consequences of eutrophication and climate warming on hypolimnetic dissolved oxygen (DO).
2. The establishment of thermal stratification was strongly related to the onset of DO depletion in the lower hypolimnion. As a result of a progressively earlier onset of stratification and later overturn, the duration of stratification increased by 38 ± 8 days over the 41 years.
3. The observed rate of volumetric hypolimnetic oxygen depletion (VHODobs) ranged from 0.131 to 0.252 g O2 m−3 per day and decreased significantly over the study period, despite the increase in the mean chlorophyll a (Chl a) concentration in the growing season. The vertical transport of DO represented from 0 to 30% of VHODobs, while adjustments for interannual differences in hypolimnetic temperature were less important, ranging from −11 to 9% of VHODobs.
4. The mean wind speed during May made the strongest significant contribution to the variation in VHODobs. VHODobs adjusted for the vertical transport of DO and hypolimnetic temperature differences, VHODadj, was significantly related to the upper mixed layer Chl a concentration during spring.
5. Hypolimnetic anoxia (HA) ranged from 27 to 168 days per year and increased significantly over time, which undoubtedly had negative ecological consequences for the lake.
6. In similar small temperate lakes, the negative effects of eutrophication on hypolimnetic DO are likely to be exacerbated by changes in lake thermal structure brought about by a warming climate, which may undermine management efforts to alleviate the effects of anthropogenic eutrophication.

Collimated transport of fast electron beam through solid density matter is one of the key issues behind the success of the fast ignition scheme by means of which the required amount of ignition energy can be delivered to the hot spot region of the compressed fuel. Here we report on a hot electron beam collimation scheme in solids by tactfully using the strong magnetic fields generated by an electrical resistivity gradient according to Faraday's law. This was accomplished by appropriately fabricating the targets in such a way that the electron beam is directed to flow in a metal which is embedded in a much lower resistivity and atomic number metal. Experimental results showed guided transport of hot electron beam over hundreds of microns length inside solid density plasma, which were obtained from two experiments examining the scheme for petawatt laser driven hot electron beam while employing various target configurations.

O sucesso do recrutamento em espécies com ciclos de vida complexos, i.e. com duas ou mais fases de desenvolvimento, depende do fornecimento de larvas aos locais apropriados do assentamento (mecanismos de pré-assentamento) e na consequente sobrevivência destas larvas até aos estágios bentónicos iniciais (mecanismos de pós-assentamento). O estudo dos principais processos físicos que controlam a variabilidade intra-anual do recrutamento do caranguejo Carcinus maenas foi possível com à obtenção em séries diárias da abundância de megalopas no plâncton e do seu assentamento em substratos artificiais de assentamento, e de parâmetros físicos relacionados com a amplitude de maré e o upwelling, durante os meses da época reprodutiva deste caranguejo. Os nossos resultados demonstraram um padrão cíclico de fornecimento de megalopas de C. maenas aos estuários na costa ocidental de Portugal, relacionado com a amplitude de maré e favorecido pelos ventos de sul. O assentamento de megalopas nos substratos artificiais de assentamento mostrou-se desacoplado do fornecimento nos dois estuários. Os dados obtidos sugerem que as megalopas são transportadas para a costa após a acção de ventos de downwelling, e o fornecimento para os estuários ocorre por transporte selectivo por corrente de maré. A utilização de substratos artificiais para medir densidades de juvenis de Carcinus maenas deve ser ponderada, uma vez que se verificou que em habitats estruturalmente menos complexos os juvenis utilizam os colectores como refúgio; em habitats mais complexos, os juvenis preferiram refugiar-se nas ervas marinhas existentes. No entanto, a estimativa de abundâncias diárias medidas nos colectores permitiu a descrição das abundâncias dos vários estádios de desenvolvimento juvenis, bem como as respectivas taxas de crescimento. As capacidades locomotoras de diferentes classes de tamanho de juvenis de C. maenas foram estimadas mediante o cálculo de um índice de locomoção. Os nossos resultados sugerem que os juvenis mais pequenos estão relativamente protegidos em habitats com vegetação densa, provavelmente porque este tipo de habitats inibe os movimentos dos juvenis de maiores dimensões. Foram também encontradas evidências da existência de segregação temporal na actividade locomotora das diferentes classes de juvenis, que funcionará como um mecanismo que permite reduzir o canibalismo e, consequentemente, aumentar a capacidade de suporte dos habitats juvenis. Foi realizado um estudo sobre o canibalismo nos juvenis de C. maenas na Ria de Aveiro, in situ e utilizando densidades reais. Verificou-se que as presas mais pequenas eram as mais vulneráveis, sendo os juvenis de maiores dimensões os predadores mais eficazes. Por outro lado, as taxas de canibalismo foram menores em habitats com elevadas densidades de refúgio (Zostera noltii). A estimativa das taxas diárias de mortalidade devidas ao canibalismo são aproximadamente metade das taxas diárias de mortalidade devidas aos processos de pré-assentamento, indicando que esta população de C. maenas será regulada pelo fornecimento larvar, i.e., será uma população regulada essencialmente por mecanismos préassentamento.

Environmental transport of pollutants comprises distinct processes such as volatilization, leaching and surface runoff. Sorption is one of the most important phenomena that affects leaching, and thus the fate of hydrophobic organic pollutants in soils and also control their distribution in the soil/water environment. The work developed focuses the optimization of analytical techniques for monitoring the sorption behaviour of organic pollutants, 17α- ethinylestradiol (EE2) and atrazine, and their fate in aqueous environment. Initially, the development of several analytical techniques, such as micellar electrokinetic chromatography, spectral deconvolution, using UV-Vis and fluorescence spectroscopy, and also enzyme linked immunosorbent assay was performed. Optimization, method performance and recovery tests are described and results discussed. Moreover, in order to evaluate the applicability of the previously optimized method, atrazine and EE2 sorption to soil samples was performed. The work developed provide several options, in terms of methodology to follow sorption of atrazine onto soils, however the choice depends on the laboratory conditions and on the analyst preferences. The advantages and disadvantages of each methodology should be evaluated first. The second part of this work consisted in the sorption behaviour study of those two different hydrophobic organic pollutants onto different soil samples. Soil organic matter chemical characterization, being essential to understand the binding mechanism responsible for the interactions, was made. The results of atrazine binding to organic matter pointed out that carboxyl units and aromaticrich organic matter are the most efficient binding agents for atrazine. EE2 adsorbs strongly to soil organic matter and is mainly stabilized by hydrophobic interactions, through aromatic nuclei face to face with surface and/or another EE2 molecule association. Farmyard manure soil contains higher aromatic and carboxyl units, indicating that this type of manure can be effectively used to minimize the residual toxicity of EE2 and atrazine present in soils, increasing the sorption and reducing leaching onto water resources. Since the final destination of organic pollutants can be ground, surface and/or waste water, atrazine and 17α-ethinylestradiol were quantified in several water samples.

Over the years it was observed at the Ria de Aveiro lagoon inlet, near the head of the north breakwater, a depth increase that might threaten the stability of this structure. A trend of accretion in the navigation channel of this lagoon is observed, endangering the navigation in this region. In order to understand the origin of these and other trends observed, the knowledge of the sediment transport in the study area is imperative. The main aim of this work is understanding the dominant physical processes in the sediment transport of sediment at the Ria de Aveiro lagoon inlet and adjacent area, improving knowledge of this region morphodynamics. The methodology followed in this study consisted in the analyzes of the topohydrographic surveys performed by the Administration of the Aveiro Harbor, and in the numerical simulations results performed with the morphodynamic modeling system MORSYS2D. The analysis of the surveys was performed by studying the temporal evolution of the bathymetry. The numerical analysis was based on the implementation of the model at the study area, sensitivity analysis of the formulations used to compute the sediment transport to the variation of input parameters (e.g. depth, sediment size, tidal currents) and analysis of the sediment uxes and bathymetric changes predicted. The simulations considered as sediment transport forcing the tidal currents only and the coupled forcing of tides and waves. Considering the wave e ect as sediment transport forcing, both monochromatic waves and a wave regime were simulated. The results revealed that the observed residual sediment transport patterns are generated due to the channel con guration. Inside the lagoon the uxes are mainly induced by the tidal currents action, restricting the action of waves to the inlet and adjacent coast. In the navigation channel the residual sediment uxes predicted are directed o - shore with values between 7 and 40 m3=day generating accretions of approximately 10 m3=day for the shallower region and 35 m3=day for the region between the tidal gauge and the tri^angulo das mar es. At the inlet, the residual uxes are approximately 30 m3=day inducing trends of erosion of approximately 20 m3=day. At the North side of the nearshore accretion is predicted, while at the South side is predicted erosion, at the rates of 250 and 1500 m3=day, respectively. It was also concluded that the waves with higher contribution to the residual sediment uxes are those with heights between 4 and 5 m. However, the storm waves with heights bigger than 5 m, despite their 10% of frequency of occurrence are responsible for 25% of the observed sediment transport.

Exposure to allergens is pivotal in determining sensitization and allergic symptoms in individuals. Pollen grain counts in ambient air have traditionally been assessed to estimate airborne allergen exposure. However, the exact allergen content of ambient air is unknown. We therefore monitored atmospheric concentrations of birch pollen grain and the matched major birch pollen allergen Bet v 1 simultaneously across Europe within the EU-funded project HIALINE (Health Impacts of Airborne Allergen Information Network). Pollen count was assessed with Hirst type pollen traps at 10 l/min at sites in France, United Kingdom, Germany, Italy and Finland. Allergen concentrations in ambient air were sampled at 800l/min with a Chemvol high-volume cascade impactor equipped with stages PM>10μm, 10 μm>PM>2.5μm, and in Germany also 2.5 μm>PM>0.12μm. The major birch pollen allergen Bet v 1 was determined with an allergen specific ELISA. Bet v 1 isoform patterns were analyzed by 2D-SDS-PAGE blots and mass spectrometric identification. Basophil activation was tested in an FcεR1-humanized rat basophil cell line passively sensitized with serum of a birch pollen lmptomatic patient. Compared to 10 previous years, 2009 was a representative birch pollen season for all stations. About 90% of the allergen was found in the PM>10μm fraction at all stations. Bet v 1 isoforms pattern did not varied substantially neither during ripening of pollen nor between different geographical locations. The average European allergen release from birch pollen was 3.2 pg Bet v 1/pollen and did not vary much between the European countries. However, in all countries a >10-fold difference in daily allergen release per pollen was measured which could be explained by long range transport of pollen with a deviating allergen release. Basophil activation by ambient air extracts correlated better with airborne allergen than with pollen concentration. Although Bet v 1 is a mixture of different isoforms, its fingerprint is constant across Europe. Bet v 1 was also exclusively linked to pollen. Pollen from different days varied >10-fold in allergen release. Thus exposure to allergen is inaccurately monitored by only monitoring birch pollen grains. Indeed, a humanized basophil activation test correlated much better with allergen concentrations in ambient air than with pollen count. Monitoring the allergens themselves together with pollen in ambient air might be an improvement in allergen exposure assessment.

This study aims to determine the potential origin of Olea pollen recorded in Badajoz in the Southwest of the Iberian Peninsula during 2009–2011. This was achieved using a combination of daily average and diurnal (hourly) airborne Olea pollen counts recorded at Badajoz (south-western Spain) and Évora (south-eastern Portugal), an inventory of olive groves in the studied area and air mass trajectory calculations computed using the HYSPLIT model. Examining olive pollen episodes at Badajoz that had distinctly different diurnal cycles in olive pollen in relation to the mean, allowed us to identify three different scenarios where olive pollen can be transported to the city from either distant or nearby sources during conditions with slow air mass movements. Back trajectory analysis showed that olive pollen can be transported to Badajoz from the West on prevailing winds, either directly or on slow moving air masses, and from high densities of olive groves situated to the Southeast (e.g. Andalucía). Regional scale transport of olive pollen can result in increased nighttime concentrations of this important aeroallergen. This could be particularly important in Mediterranean countries where people can be outdoors during this time due to climate and lifestyle. Such studies that examine sources and the atmospheric transport of pollen are valuable for allergy sufferers and health care professionals because the information can be incorporated into forecasts, the outputs of which are used for avoiding exposure to aeroallergens and planning medication. The results of studies of this nature can also be used for examining gene flow in this important agricultural crop.

Airborne pollen transport at micro-, meso-gamma and meso-beta scales must be studied by atmospheric models, having special relevance in complex terrain. In these cases, the accuracy of these models is mainly determined by the spatial resolution of the underlying meteorological dataset. This work examines how meteorological datasets determine the results obtained from atmospheric transport models used to describe pollen transport in the atmosphere. We investigate the effect of the spatial resolution when computing backward trajectories with the HYSPLIT model. We have used meteorological datasets from the WRF model with 27, 9 and 3 km resolutions and from the GDAS files with 1 ° resolution. This work allows characterizing atmospheric transport of Olea pollen in a region with complex flows. The results show that the complex terrain affects the trajectories and this effect varies with the different meteorological datasets. Overall, the change from GDAS to WRF-ARW inputs improves the analyses with the HYSPLIT model, thereby increasing the understanding the pollen episode. The results indicate that a spatial resolution of at least 9 km is needed to simulate atmospheric flows that are considerable affected by the relief of the landscape. The results suggest that the appropriate meteorological files should be considered when atmospheric models are used to characterize the atmospheric transport of pollen on micro-, meso-gamma and meso-beta scales. Furthermore, at these scales, the results are believed to be generally applicable for related areas such as the description of atmospheric transport of radionuclides or in the definition of nuclear-radioactivity emergency preparedness.

BACKGROUND: Researchers have tested the beliefs of sportspeople and sports medicine specialists that cognitive strategies influence strength performance. Few investigators have synthesised the literature. OBJECTIVES: The specific objectives were to review evidence regarding (a) the cognitive strategy-strength performance relationship; (b) participant skill level as a moderator; and (c) cognitive, motivational, biomechanical/physiological, and emotional mediators. METHOD: Studies were sourced via electronic databases, reference lists of retrieved articles, and manual searches of relevant journals. Studies had to be randomised or counterbalanced experiments with a control group or condition, repeated measures, and a quality control score above 0.5 (out of 1). Cognitive strategies included goal setting, imagery, self-talk, preparatory arousal, and free choice. Dependent variables included maximal strength, local muscular endurance, or muscular power. RESULTS: Globally, cognitive strategies were reliability associated with increased strength performance (results ranged from 61 to 65 %). Results were mixed when examining the effects of specific strategies on particular dependent variables, although no intervention had an overall negative influence. Indeterminate relationships emerged regarding hypothesised mediators (except cognitive variables) and participant skill level as a moderator. CONCLUSIONS: Although cognitive strategies influence strength performance, there are knowledge gaps regarding specific types of strength, especially muscular power. Cognitive variables, such as concentration, show promise as possible mediators.

The continued growth in the volume of international trade poses considerable economic and sustainability challenges, particularly as transport routes become more congested and concern grows about the role of transport movements in accelerating climate change. Rail freight plays a major role in the inland transport of containers passing through the main British container ports, and potentially could play a more significant role in the future. However, there is little detailed understanding of the nature of this particular rail market, especially in terms its current operating efficiency. This paper examines container train service provision to/from the four main ports, based on analysis of a representative survey of more than 500 container trains between February and August 2007. The extent to which the existing capacity is utilised is presented, and scenarios by which the number of containers carried could be increased without requiring additional train service provision are modelled, to identify the theoretical potential for greater rail volumes. Finally, the paper identifies the challenges involved in achieving higher load factors, emphasising the importance both of wider supply chain considerations and government policy decision-making.

In the proposed model, the independent system operator (ISO) provides the opportunity for maintenance outage rescheduling of generating units before each short-term (ST) time interval. Long-term (LT) scheduling for 1 or 2 years in advance is essential for the ISO and the generation companies (GENCOs) to decide their LT strategies; however, it is not possible to be exactly followed and requires slight adjustments. The Cournot-Nash equilibrium is used to characterize the decision-making procedure of an individual GENCO for ST intervals considering the effective coordination with LT plans. Random inputs, such as parameters of the demand function of loads, hourly demand during the following ST time interval and the expected generation pattern of the rivals, are included as scenarios in the stochastic mixed integer program defined to model the payoff-maximizing objective of a GENCO. Scenario reduction algorithms are used to deal with the computational burden. Two reliability test systems were chosen to illustrate the effectiveness of the proposed model for the ST decision-making process for future planned outages from the point of view of a GENCO.
