The mechanical properties of filled natural rubber latex vulcanizates were found to be improved by the addition of polyethylene glycols of different molecular weight and glycerol. There is a slight reduction in the optimum cure times of the compounds containing PEG/Glycerol. The morphology study shows that the filler distribution is more uniform in the compounds containing PEG/Glycerol.

ABSTRACT: p-Phenylenediamine was chemically attached to low molecular weight chlorinated paraffin wax. The polymer-bound p-phenylenediamine was characterized by vapor-phase osmometry (VPO), proton magnetic resonance spectroscopy ('H-NMR), infrared spectroscopy (IR), and thermogravimetric analysis (TGA). The efficiency and permanence of the polymer-bound p-phenylenediamine as an antioxidant was compared with a conventional amine-type antioxidant in natural rubber vulcanizates. The vulcanizates showed improved aging resistance in comparison to vulcanizates containing a conventional antioxidant. The presence of liquid polymer-bound p-phenylenediamine also reduces the amount of the plasticizer required for compounding.

ABSTRACT: Phenol was chemically attached to low molecular weight chlorinated polyisobutylene and stearic acid respectively. These phenolic antioxidants were characterised by IR, 1H NMR and TGA. The efficiency and permanence of these bound antioxidants were compared with conventional antioxidants in natural rubber vulcanisates. The vulcanisates showed comparable ageing resistance in comparison to vulcanisates containing conventional antioxidants. The presence of liquid polymer bound phenol reduce the amount of plasticiser required for compounding.

Low-protein content natural rubber latex was produced by using a nonionic surfactant-polyethylene glycol (PEG). Extractable protein content of natural rubber latex was found to decrease with PEG treatment and reduction increased with increase in the molecular weight of PEG. The low-protein latex samples were characterized by tensile testing, Fourier transform infrared and thermogravimetric analysis. The results have shown 35% reduction in the extractable protein content, without any compromise on the mechanical properties of the latex; however, thermal stability of low-protein latex was found to be reduced marginally with PEG treatment.

The thesis describes utilisation of reclaimed rubber, Whole Tyre Reclaim (WTR) produced from bio non- degradable solid pollutant scrap and used tyres. In this study an attempt has made to optimize the substitution of virgin rubber with WTR in both natural and synthetic rubber compounds without seriously compromising the important mechanical properties. The WTR is used as potent source of rubber hydrocarbon and carbon black filler. Apart from natural rubber (NR), Butadiene rubber (BR), Styrene butadiene rubber (SBR), Acrylonitrile butadiene rubber (NBR) and Chloroprene rubber (CR) were selected for study, being the most widely used general purpose and specialty rubbers. The compatibility problem was addressed by functionalisation of WTR with maleic anhydride and by using a coupling agent Si69.The blends were systematically evaluated with respect to various mechanical properties. The thermogravimetric analyses were also carried out to evaluate the thermal stability of the blends.Mechanical properties of the blends were property and matrix dependant. Presence of reinforcing carbon black filler and curatives in the reclaimed rubber improved the mechanical properties with the exception of some of the elastic properties like heat build up, resilience, compression set. When WTR was blended with natural rubber and synthetic rubbers, as the concentration of the low molecular weight, depolymerised WfR was increased above 46-weight percent, the properties deteriorates.When WTR was blended with crystallizing rubbers such as natural rubber and chloroprene rubber, properties like tensile strength, ultimate elongation were decreased in presence of WTR. Where as in the case of blends of WTR with non-crystallizing rubbers reinforcement effect was more prominent.The effect of functionalisation and coupling agent was studied in three matrices having different levels of polarity(NBR, CR and SBR).The grafting of maleic anhydride on to WTR definitely improved the properties of its blends with NBR, CR and SBR, the effect being prominent in Chloroprene rubber.Improvement in properties of these blends could also achieved by using a coupling agent Si69. With this there is apparent plasticizing effect at higher loading of the coupling agent. The optimum concentration of Si69 was 1 phr for improved properties, though the improvements are not as significant as in the case of maleic anhydride grafting.Thermal stability of the blend was increased by using silane-coupling agent.

This thesis has discussed the development of a new metal ion doped panchromatic photopolymer for various holographic applications. High-quality panchromatic holographic recording material with high diffraction efficiency, high photosensitivity and high spatial resolution is one of the key factors for the successful recording of true colour holograms. The capability of the developed material for multicolour holography can be investigated.In the present work, multiplexing studies were carried out using He-Ne laser (632.8 nm). Multiplexing can be done using low wavelength lasers like Ar+ ion (488 nm) and frequency doubled Nd: YAG (532 nm) lasers, so as to increase the storage capacity. The photopolymer film studied had a thickness of only 130 Cm. Films with high thickness (~500 Cm) is highly essential for competitive holographic memories . Hence films with high thickness can be fabricated and efforts can be made to record more holograms or gratings in the material.In the present study, attempts were made to record data page in silver doped MBPVA/AA photopolymer film. Image of a checkerboard pattern was recorded in the film, which could be reconstructed with good image fidelity. Efforts can be made to determine the bit error rate (BER) which provides a quantitative measure of the image quality of the reconstructed image . Multiple holographic data pages can also be recorded in the material making use of different multiplexing techniques.Holographic optical elements (HOEs) are widely used in optical sensors, optical information processing, fibre optics, optical scanners and solar concentrators . The suitability of the developed film for recording holographic optical elements like lenses, beam splitters and filters can be studied.The suitability of a reflection hologram recorded in acrylamide based photopolymer for visual indication of environmental humidity is reported . Studies can be done to optimize the film composition for recording of reflection holograms.An improvement in the spatial resolution of PVA/acrylamide based photopolymer by using a low molecular-weight poly (vinyl alcohol) binder was recently reported . Effect of the molecular weight of the binder matrix on the holographic properties of the developed photopolymer system can be investigated.Incorporation of nanoparticles into photopolymer system is reported to enhance the resolution and improve the dimensional stability of the system . Hence efforts can be made to incorporate silver nanoparticles into the photopolymer and its influence on the holographic properties can be studied.This thesis was a small venture towards the realization of a big goal, a competent holographic recording material with excellent properties for practical holographic applications. As a result of the present research, we could successfully develop an efficient panchromatic photopolymer system and could demonstrate its suitability for recording transmission holograms and holographic data page. The developed photopolymer system is expected to have significant applications in the fields of true-color display holography, wavelength multiplexing holographic storage, and holographic optical elements. Highly concentrated and determined effort has yet to be put forth for this expectation to become a reality.

In this paper, we present a laser-induced photoacoustic study on the photostability of laser dye Coumarin 540 doped in PMMA matrix and modified by the incorporation of low-molecular weight additives. The dependence of photostability of the dye on various experimental conditions, such as nature of solvents, incident optical power and dye concentration, is investigated in detail. The activation rates for the bleaching process are calculated for different concentrations and they suggest the possibility of two distinct mechanisms responsible for photodegradation. Further, analysis of the data confirms the linear dependence of photodegradation on the intensity of incident radiation. The role of different externally influencing parameters, such as wavelength and modulation frequency of incident radiation, is also discussed.

The present study aimed at the utlisation of microbial organisms for the production of good quality chitin and chitosan. The three strains used for the study were Lactobacillus plantarum, Lactobacililus brevis and Bacillus subtilis. These strains were selected on the basis of their acid producing ability to reduce the pH of the fermenting substrates to prevent spoilage and thus caused demineralisation of the shell. Besides, the proteolytic enzymes in these strains acted on proteinaceous covering of shrimp and thus caused deprotenisation of shrimp shell waste. Thus the two processes involved in chitin production can be affected to certain extent using bacterial fermentation of shrimp shell.Optimization parameters like fermentation period, quantity of inoculum, type of sugar, concentration of sugar etc. for fermentation with three different strains were studied. For these, parameters like pH, Total titrable acidity (TTA), changes in sugar concentration, changes in microbial count, sensory changes etc. were studied.Fermentation study with Lactobacillus plantarum was continued with 20% w/v jaggery broth for 15 days. The inoculum prepared yislded a cell concentration of approximately 108 CFU/ml. In the present study, lactic acid and dilute hydrochloric acid were used for initial pH adjustment because; without adjusting the initial pH, it took more than 5 hours for the lactic acid bacteria to convert glucose to lactic acid and during this delay spoilage occurred due to putrefying enzymes active at neutral or higher pH. During the fermentation study, pH first decreased in correspondence with increase in TTA values. This showed a clear indication of acid production by the strain. This trend continued till their proteolytic activity showed an increasing trend. When the available sugar source started depleting, proteolytic activity also decreased and pH increased. This was clearly reflected in the sensory evaluation results. Lactic acid treated samples showed greater extent of demineralization and deprotenisation at the end of fermentation study than hydrochloric acid treated samples. It can be due to the effect of strong hydrochloric acid on the initial microbial count, which directly affects the fermentation process. At the end of fermentation, about 76.5% of ash was removed in lactic acid treated samples and 71.8% in hydrochloric acid treated samples; 72.8% of proteins in lactic acid treated samples and 70.6% in hydrochloric acid treated samples.The residual protein and ash in the fermented residue were reduced to permissible limit by treatment with 0.8N HCI and 1M NaOH. Characteristics of chitin like chitin content, ash content, protein content, % of N- acetylation etc. were studied. Quality characteristics like viscosity, degree of deacetylation and molecular weight of chitosan prepared were also compared. The chitosan samples prepared from lactic acid treated showed high viscosity than HCI treated samples. But degree of deacetylation is more in HCI treated samples than lactic acid treated ones. Characteristics of protein liquor obtained like its biogenic composition, amino acid composition, total volatile base nitrogen, alpha amino nitrogen etc. also were studied to find out its suitability as animal feed supplement.Optimization of fermentation parameters for Lactobacillus brevis fermentation study was also conducted and parameters were standardized. Then detailed fermentation study was done in 20%wlv jaggery broth for 17 days. Also the effect of two different acid treatments (mild HCI and lactic acid) used for initial pH adjustment on chitin production were also studied. In this study also trend of changes in pH. changes in sugar concentration ,microbial count changes were similar to Lactobacillus plantarum studies. At the end of fermentation, residual protein in the samples were only 32.48% in HCI treated samples and 31.85% in lactic acid treated samples. The residual ash content was about 33.68% in HCI treated ones and 32.52% in lactic acid treated ones. The fermented residue was converted to chitin with good characteristics by treatment with 1.2MNaOH and 1NHCI.Characteristics of chitin samples prepared were studied and extent of Nacetylation was about 84% in HCI treated chitin and 85%in lactic acid treated ones assessed from FTIR spectrum. Chitosan was prepared from these samples by usual chemical method and its extent of solubility, degree of deacetylation, viscosity and molecular weight etc were studied. The values of viscosity and molecular weight of the samples prepared were comparatively less than the chitosan prepared by Lactobacillus plantarum fermentation. Characteristics of protein liquor obtained were analyzed to determine its quality and is suitability as animal feed supplement.Another strain used for the study was Bacillus subtilis and fermentation was carried out in 20%w/v jaggery broth for 15 days. It was found that Bacillus subtilis was more efficient than other Lactobacillus species for deprotenisation and demineralization. This was mainly due to the difference in the proteolytic nature of the strains. About 84% of protein and 72% of ash were removed at the end of fermentation. Xylanases with hydrolytic activity on xylan, one of the hemicellulosic materials present in plant cell walls, have been identified long back and the applicability of this enzyme is constantly growing. All these applications especially the pulp and paper industries require novel enzymes. There has been lot of documentation on microbial xylanases, however, none meeting all the required characteristics. The characters being sought are: higher production, higher pH and temperature optima, good stabilities under these conditions and finally the low associated cellulase and protease production. The present study analyses various facets of xylanase biotechnology giving emphasis on bacterial xylanases. Fungal xylanases are having problems like low pH values for both enzyme activity and growth. Moreover, the associated production of cellulases at significant levels make fungal xylanases less suitable for application in paper and pulp industries.Bacillus SSP-34 selected from 200 isolates was clearly having xylan catabolizing nature distinct from earlier reports. The stabilities at higher temperatures and pH values along with the optimum conditions for pH and temperature is rendering Bacillus SSP-34 xylanase more suitable than many of the previous reports for application in pulp and paper industries.Bacillus SSP-34 is an alkalophilic thertmotolerant bacteria which under optimal cultural conditions as mentioned earlier, can produce 2.5 times more xylanase than the basal medium.The 0.5% xylan concentration in the medium was found to the best carbon source resulting in 366 IU/ml of xylanase activity. This induction was subjected to catabolite repression by glucose. Xylose was a good inducer for xylanase production. The combination of yeast extract and peptone selected from several nitrogen sources resulted in the highest enzyme production (379+-0.2 IU/ml) at the optimum final concentration of 0.5%. All the cultural and nutritional parameters were compiled and comparative study showed that the modified medium resulted in xylanase activity of 506 IU/ml, 5 folds higher than the basal medium.The novel combination of purification techniques like ultrafiltraton, ammonium sulphate fractionation, DEAE Sepharose anion exchange chromatography, CM Sephadex cation exchange chromatography and Gel permeation chromatography resulted in the purified xylanase having a specific activity of 1723 U/mg protein with 33.3% yield. The enzyme was having a molecular weight of 20-22 kDa. The Km of the purified xylanase was 6.5 mg of oat spelts xylan per ml and Vmax 1233 µ mol/min/mg protein.Bacillus SSP-34 xylanase resulted in the ISO brightness increase from 41.1% to 48.5%. The hydrolytic nature of the xylanase was in the endo-form.Thus the organism Bacillus SSP-34 was having interesting biotechnological and physiological aspects. The SSP-34 xylanase having desired characters seems to be suited for application in paper and pulp industries.

The thesis mainly discussed the isolation and identification of a probiotic Lactobacillus plantarum, fermentative production of exopolysaccharide by the strain, its purification, structural characterisation and possible applications in food industry and therapeutics. The studies on the probiotic characterization explored the tolerance of the isolated LAB cultures to acid, bile, phenol, salt and mucin binding. These are some of the key factors that could satisfy the criteria for probiotic strains . The important factors required for a high EPS production in submerged fermentation was investigated with a collection of statistical and mathematical approach. Chapter 5 of the thesis explains the structural elucidation of EPS employing spectroscopic and chromatographic techniques. The studies helped in the exploration of the hetero-polysaccharide sequence from L. plantarum MTCC 9510. The thesis also explored the bioactivities of EPS from L. plantarum. As majority of chemical compounds identified as anti-cancerous are toxic to normal cells, the discovery and identification of new safe drugs has become an important goal of research in the biomedical sciences. The thesis has explored the anti-oxidant, anti-tumour and immunomodulating properties of EPS purified from Lactobacillus plantarum. The presence of (1, 3) linkages and its molecular weight presented the EPS with anti-oxidant, anti-tumour and immunomodulating properties under in vitro conditions.

The continually growing worldwide hazardous waste problem is receiving much attention lately. The development of cost effective, yet efficient methods of decontamination are vital to our success in solving this problem.Bioremediation using white rot fungi, a group of basidiomycetes characterized by their ability to degrade lignin by producing extracellular LiP, MnP and laccase have come to be recognized globally which is described in detail in Chapter 1.These features provide them with tremendous advantages over other micro-organisms.Chapter 2 deals with the isolation and screening of lignin degrading enzyme producing micoro-organisms from mangrove area. Marine microbes of mangrove area has great capacity to tolerate wide fluctuations of salinitie.Primary and secondary screening for lignin degrading enzyme producing halophilic microbes from mangrove area resulted in the selection of two fungal strains from among 75 bacteria and 26 fungi. The two fungi, SIP 10 and SIP ll, were identified as penicillium sp and Aspergillus sp respectively belonging to the class Ascomycetes .Specific activity of the purified LiP was 7923 U/mg protein. The purification fold was 24.07 while the yield was 18.7%. SDS PAGE of LiP showed that it was a low molecular weight protein of 29 kDa.Zymogram analysis using crystal violet dye as substrate confirmed the peroxidase nature of the purified LiP.The studies on the ability of purified LiP to decolorize different synthetic dyes was done. Among the dyes studied, crystal violet, a triphenyl methane dye was decolorized to the greatest extent.

The primary aim of this work has been to prepare efficient and cost effective polymer bound antioxidants by direct’ attachment of conventional antioxidants to a modified polymer. Due to the importance and easy availability of natural rubber in Kerala, it is proposed to make use of low molecular weight natural rubber as the polymer substrate for binding the antioxidant in most cases. The molecular weight of such low molecular weight natural rubber can be easily manipulated by varying the time of mastication, UV—irradiation etc. Further, the bound antioxidant may also get vulcanized during the vulcanization of the elastomer to which it is added, making the antioxidant non—volatile and non extractable. Several methods are proposed to be investigated for attaching the antioxidant to the low molecular weight natural rubber such as modified Friedel-Craft's alkylation reaction, binding during UV—irradiation, binding during aggressive mastication etc. The efficiency of such rubber bound antioxidants is proposed to be compared with that of conventional antioxidants in terms of volatility, extractability in solvents, ageing resistance etc. Naturally occuring antioxidants such as cardanol, are also proposed to be modified by binding them to low molecular weight natural rubber. The study is undertaken with the intention of generating a class of bound antioxidants which can be used in elastomers for aggressive and long term application.

Lipids constitute a significant portion of the biomass of earth and lipolytic enzymes play a very important role in lipid turn over. Apart from their biological significance, lipolytic enzymes are also very important in the fields of nutrition, food technology, medicine and preparative and analytical lipid biochemistry. Recent developments in the study of proteins and enzymes have largely benefited the study of lipolytic enzymes, that some of these enzymes were isolated in pure form. Even today there is a continuous search for new and potent sources of these lipolytic enzymes. The zest for elucidating the structure and mechanism of action of the enzymes obtained in pure form for biochemist still remains unabated. The literature shows no record of such an effort for the study of lipases from marine sources. The fact that many fishes like oil sardine, mackerel, cat fish, seer etc. contains large amounts of lipid shows the possibility of the existence of lipases in significant amounts necessitating their exhaustive study. Such a study will, not only provide alternate sources for lipase but also will provide methods to curb lipolysis and the resultant rancidity and off flavor development in fish and fishery products.

The microorganisms are recognized as important sources of protease inhibitors which are valuable in the fields of medicine, agriculture and biotechnology. The protease inhibitors of microbial origin are found to be versatile in their structure and mode of inhibition that vary from those of other sources. Although surplus of low molecular weight non-protein protease inhibitors from microorganisms have been reported, there is a dearth of reports on proteinaceous protease inhibitors. The search for new metabolites from marine organisms has resulted in the isolation of more or less 10,000 metabolites (Fuesetani and Fuesetani, 2000) many of which are gifted with pharmacodynamic properties. The existence of marine microorganisms was reported earlier, and they were found to be metabolically and physiologically dissimilar from terrestrial microorganisms. Marine microorganisms have potential as important new sources of enzyme inhibitors and consequently a detailed study of new marine microbial inhibitors will provide the basis for future research (Imada, 2004).

Engyodontium album isolated from marine sediment produced protease, which was active at pH 11. Process parameters influencing the production of alkaline protease by marine E. album was optimized. Particle size of <425 mm, 60% initial moisture content and incubation at 25 8C for 120 h were optimal for protease production under solid state fermentation (SSF) using wheat bran. The organism has two optimal pH (5 and 10) for maximal enzyme production. Sucrose as carbon source, ammonium hydrogen carbonate as additional inorganic nitrogen source and amino acid leucine enhanced enzyme production during SSF. The protease was purified and partially characterized. A 16-fold purified enzyme was obtained after ammonium sulphate precipitation and ion-exchange chromatography. Molecular weight of the purified enzyme protein was recorded approximately 38 kDa by SDS-PAGE. The enzyme showed maximum activity at pH 11 and 60 8C. Activity at high temperature and high alkaline pH suggests suitability of the enzyme for its application in detergent industry
