During vertebrate craniofacial development, neural crest cells (NCCs) contribute to most of the craniofacial pharyngeal skeleton. Defects in NCC specification, migration and differentiation resulting in malformations in the craniofacial complex are associated with human craniofacial disorders including Treacher-Collins Syndrome, caused by mutations in TCOF1. It has been hypothesized that perturbed ribosome biogenesis and resulting p53 mediated neuroepithelial apoptosis results in NCC hypoplasia in mouse Tcof1 mutants. However, the underlying mechanisms linking ribosome biogenesis and NCC development remain poorly understood. Here we report a new zebrafish mutant, fantome (fan), which harbors a point mutation and predicted premature stop codon in zebrafish wdr43, the ortholog to yeast UTP5. Although wdr43 mRNA is widely expressed during early zebrafish development, and its deficiency triggers early neural, eye, heart and pharyngeal arch defects, later defects appear fairly restricted to NCC derived craniofacial cartilages. Here we show that the C-terminus of Wdr43, which is absent in fan mutant protein, is both necessary and sufficient to mediate its nucleolar localization and protein interactions in metazoans. We demonstrate that Wdr43 functions in ribosome biogenesis, and that defects observed in fan mutants are mediated by a p53 dependent pathway. Finally, we show that proper localization of a variety of nucleolar proteins, including TCOF1, is dependent on that of WDR43. Together, our findings provide new insight into roles for Wdr43 in development, ribosome biogenesis, and also ribosomopathy-induced craniofacial phenotypes including Treacher-Collins Syndrome.

The employment effect from raising the minimum wage has long been studied but remains in dispute. Our meta-analysis of 236 estimated minimum wage elasticities and 710 partial correlation coefficients from 16 UK studies finds no overall practically significant adverse employment effect. Unlike US studies, there seems to be little, if any, overall reporting bias. Multivariate meta-regression analysis identifies several research dimensions that are associated with differential employment effects. In particular, the residential home care industry may exhibit a genuinely adverse employment effect.

This book will inspire academics, teachers and trainers to use film and television in their classrooms and to shows them how it might be done. It brings together respected international scholars who recount their experiences of how they have used moving images in their classrooms (defined widely to include distance-learning) with their explanations of why they chose this method of teaching and how they put their intentions into action. The book also illustrates how particular subjects might be taught using film and television as an inspiration to demonstrate the range of opportunities that these media offer. Finally, this book considers some of the practical issues in using film and television in the classroom such as copyright, technology, and the representation of reality and drama in films. This is a 'practical, how to' book that answers the questions of those people who have considered using film and television in their classroom but until now have shied away from doing so. The opportunity to see how others have used film effectively breaks down psychological barriers and makes it seem both realistic and worthwhile.

The Alaotran gentle lemur Hapalemur alaotrensis is a Critically Endangered lemur, which exclusively inhabits the marshes around Lac Alaotra in northeast Madagascar. In the past decades the population of H. alaotrensis has experienced a dramatic decline due to poaching, habitat destruction and degradation. Surveys have been carried out periodically to follow the status of the population. Here we present the results of a survey carried out between May and June 2008 in the southwestern part of the marshes around Alaotra and discuss the key findings derived from the analysis of the data collected. Our study indicates that the probability of detecting the species in an area where it is present is very low and depends on factors that vary in space and time. These results stress the need to account for imperfect detection when monitoring this species, an issue especially relevant when reporting population trends. Our analyses also show that habitat fragmentation is a key determinant of habitat suitability for H. alaotrensis, with fragmented areas of marsh showing low suitability. Finally, our observations and analysis suggest that the protection provided by the local community to H. alaotrensis in Andreba is contributing to the conservation of this Critically Endangered species. This highlights the need to continue working on engaging the local communities in the conservation of the marshes at Lac Alaotra as a critical element to secure the future of H. alaotrensis.

Technology effects, business process development, and productivity growth are considered in the context of a single company: Wal-Mart. The starting point is the 2001 McKinsey Global Institute report, which finds that over 1995–2000, a quarter of U.S. productivity growth is attributable to the retail industry, and almost a sixth of that is attributable to Wal-Mart. Wal-Mart is interesting as well because of its rapid growth in Canada. This is now Canada’s largest private sector employer. We also consider other evidence relevant to public policy formation concerning Wal-Mart and conclude with a discussion of options for partially filling important data gaps.

AimsThe association between central obesity and insulin resistance reflects the properties of visceral adipose tissue. Our aim was to gain further insight into this association by analysing the lipid composition of subcutaneous and omental adipose tissue in obese women with and without insulin resistance.MethodsSubcutaneous and omental adipose tissue and serum were obtained from 29 obese non-diabetic women, 13 of whom were hyperinsulinemic. Histology, and lipid and gene profiling were performed.ResultsIn omental adipose tissue of obese, insulin-resistant women, adipocyte hypertrophy and macrophage infiltration were accompanied by an increase in GM3 ganglioside and its synthesis enzyme ST3GAL5; in addition, phosphatidylethanolamine (PE) lipids were increased and their degradation enzyme, PEMT, decreased. ST3GAL5 was expressed predominantly in adipose stromovascular cells and PEMT in adipocytes. Insulin resistance was also associated with an increase in PE lipids in serum.InterpretationThe relevance of these findings to insulin resistance in humans is supported by published mouse studies in which adipocyte GM3 ganglioside, increased by the inflammatory cytokine tumour necrosis factor-α, impaired insulin action, and PEMT was required for adipocyte lipid storage. Thus, in visceral adipose tissue of obese humans, an increase in GM3 ganglioside secondary to inflammation may contribute to insulin resistance and a decrease in PEMT may be a compensatory response to adipocyte hypertrophy.International Journal of Obesity accepted article preview online, 26 October 2015. doi:10.1038/ijo.2015.223.

Coarse woody debris (CWD) is a common structural component of terrestrial ecosystems, and provides important habitat for biota. Fires modify the distribution of CWD, both spatially and temporally. Changes in fire regimes, such as those arising from prescribed burning and changing climatic conditions, make it critical to understand the response of this resource to fire. We created a conceptual model of the effects of fire on logs and dead trees in topographically diverse forests in which trees often survive severe fire. We then surveyed paired sites, in a damp gully and adjacent drier slope, ~3.5. years after a large wildfire in south-eastern Australia. Sites were stratified by fire severity (unburnt, understorey burnt and severely burnt), and fire history (burnt ≤3. years or ≥20. years prior to the wildfire). Both components of the fire regime influenced CWD availability in gullies. Severe wildfire and fire history ≤3. years reduced the volume of small logs (10-30. cm diameter) in gullies, while severe wildfire increased the number of large dead trees in gullies. CWD on slopes was not affected by fire severity or history at ~3.5. years post-fire. Log volumes on slopes may recover more quickly after wildfire through rapid collapse of branches and trees. Gullies generally supported more logs than slopes, but longer inter-fire intervals in gullies may allow fuel loads to accumulate and lead to comparatively larger fire impacts. Given that fire severity and fire interval are predicted to change in many fire-prone ecosystems in coming decades, this study highlights the importance of understanding the interacting effects of multiple components of the fire regime with landscape structure. In particular, variation in fire interval and fire severity in relation to topographic position will influence the pattern of accumulation of coarse woody debris across the landscape, and therefore the structure and quality of habitats for biota.

Research consistently reports higher rates of problematic drinking among lesbian, bisexual and queer women than among heterosexual women, but relatively little research has identified underlying factors. Within this context, the aim of the present study was to qualitatively explore the sociocultural influences on alcohol consumption among lesbian, bisexual and queer women in Australia. An ethnographic study including in-depth interviews and 10 sessions of participant observation was conducted with 25 Australian lesbian, bisexual and queer women. Analysis of transcripts and fieldnotes focused on lesbian, bisexual and queer-related influences on alcohol consumption. Three lesbian, bisexual and queer-related factors were identified that influenced alcohol use: (1) coping, (2) connection and (3) intersections with lesbian, bisexual and queer identity. Most participants reported consuming alcohol to cope with discrimination or to connect with like-minded others. Alcohol use had positive influences for some women through facilitating social connection and wellbeing. Women with a high lesbian, bisexual and queer identity salience were more likely to seek lesbian, bisexual and queer community connection involving alcohol, to publicly identify as lesbian, bisexual and queer and to experience discrimination. National policies need to address underlying causes of discrimination against lesbian, bisexual and queer women. Alcohol policies and clinical interventions should acknowledge the impact of discrimination on higher alcohol consumption amongst lesbian, bisexual and queer women compared with heterosexual women, and should utilise health promotion messages regarding safe drinking that facilitate lesbian, bisexual and queer social connection.

In fire-prone regions, wildfire influences spatial and temporal patterns of landscape heterogeneity. The likely impacts of climate change on the frequency and intensity of wildfire highlights the importance of understanding how fire-induced heterogeneity may affect different components of the biota. Here, we examine the influence of wildfire, as an agent of landscape heterogeneity, on the distribution of arboreal mammals in fire-prone forests in south-eastern Australia. First, we used a stratified design to examine the role of topography, and the relative influence of fire severity and fire history, on the occurrence of arboreal mammals 2-3 years after wildfire. Second, we investigated the influence of landscape context on the occurrence of arboreal mammals at severely burnt sites. Forested gullies supported a higher abundance of arboreal mammals than slopes. Fire severity was the strongest influence, with abundance lower at severely burnt than unburnt sites. The occurrence of mammals at severely burned sites was influenced by landscape context: abundance increased with increasing amount of unburnt and understorey-only burnt forest within a 1 km radius. These results support the hypothesis that unburnt forest and moist gullies can serve as refuges for fauna in the post-fire environment and assist recolonization of severely burned forest. They highlight the importance of spatial heterogeneity created by wildfire and the need to incorporate spatial aspects of fire regimes (e.g., creation and protection of refuges) for fire management in fire-prone landscapes.

Wildfires have major impacts on ecosystems globally. Fire regimes (including fire frequency, intensity, season and type of fire) influence the status of species by altering habitat suitability at the site scale, and by creating heterogeneity at the landscape scale. The relative effects of site and landscape-scale fire attributes on animal species are rarely examined together. Such knowledge is important, given that fire regimes are sensitive to changing land management practices; and that fires are predicted to become larger and more frequent in some regions as a result of climate change. Here, we tested the relative influence of elements of the fire regime (fire severity, fire history) at the site-scale, and the landscape context (extent of surrounding unburnt forest, fire heterogeneity) on the occurrence of native terrestrial mammals after severe wildfire in south-eastern Australia. We conducted surveys by using automatically triggered, infrared cameras at 80 sites in fire-prone eucalypt forests, 2-3. years post-wildfire. Thirteen native mammal species were recorded, eight of which were detected with sufficient frequency for analysis. Most species were widespread (35-90% of sites) and recorded in all fire severity classes. Fire effects at the site-level were more influential than landscape context effects arising from heterogeneity in the fire regime (e.g. extent of surrounding unburnt forest). Fire severity was the most influential of the fire-regime elements investigated, but it affected different species in different ways. This study highlights three main points relevant to conservation of terrestrial mammals after wildfire. First, spatial variation in fire severity associated with wildfire (ranging from unburned to severely burned stands) is an important contributor to the post-fire status of species. Second, post-fire environmental conditions are significant: here, rapid regeneration of vegetation following drought-breaking rains greatly influenced the suitability of post-fire habitats. Third, it is valuable to consider the effects of the fire regime at multiple scales, including both the site (forest stand) and its landscape context. Insights from short-term surveys, such as this, will be enhanced by complementary longitudinal studies, especially where they encompass environmental variation through the post-fire succession.

HydroShare is an online, collaborative system being developed for open sharing of hydrologic data and models. The goal of HydroShare is to enable scientists to easily discover and access hydrologic data and models, retrieve them to their desktop or perform analyses in a distributed computing environment that may include grid, cloud or high performance computing model instances as necessary. Scientists may also publish outcomes (data, results or models) into HydroShare, using the system as a collaboration platform for sharing data, models and analyses. HydroShare is expanding the data sharing capability of the CUAHSI Hydrologic Information System by broadening the classes of data accommodated, creating new capability to share models and model components, and taking advantage of emerging social media functionality to enhance information about and collaboration around hydrologic data and models. One of the fundamental concepts in HydroShare is that of a Resource. All content is represented using a Resource Data Model that separates system and science metadata and has elements common to all resources as well as elements specific to the types of resources HydroShare will support. These will include different data types used in the hydrology community and models and workflows that require metadata on execution functionality. The HydroShare web interface and social media functions are being developed using the Drupal content management system. A geospatial visualization and analysis component enables searching, visualizing, and analyzing geographic datasets. The integrated Rule-Oriented Data System (iRODS) is being used to manage federated data content and perform rule-based background actions on data and model resources, including parsing to generate metadata catalog information and the execution of models and workflows. This presentation will introduce the HydroShare functionality developed to date, describe key elements of the Resource Data Model and outline the roadmap for future development.

O objetivo deste artigo é o de analisar a organização rítmica de uma obra para piano do compositor brasileiro Cláudio Santoro. A análise da obra denominada Paulistana n.º 2 – Tempo de Catira – apoia-se na fundamentação t³rica postulada por Grosvenor Cooper & Leonard Meyer (1960). Este trabalho alia os pressupostos t³ricos fundamentados em estudos recentes sobre o ritmo e sua organização, em agrupamentos e em níveis hierárquicos com o propósito de delinear uma interpretação musical. Neste contexto, as considerações t³ricas são utilizadas para entender a partitura. Os dois pólos, o t³rico e o prático, são complementares e juntos contribuem para um entendimento musical equilibrado.
