Objective: In this study, we compared the frequency and intensity of childhood traumas in alcohol- or other drug-dependent patients, in patients with depression, and in a control group without psychiatric diagnoses. Methods: The study had a retrospective design of a clinical sample of men and women from the groups listed above. They were evaluated by the same standardized instrument: the Childhood Trauma Questionnaire.. Results: A higher frequency and intensity of emotional, physical, and sexual abuse were found in alcohol- and other drug-dependent patients than in patients with depression, who, in turn, presented significantly higher proportions than the control group. In all of the cases, the frequency was higher among women than men. Conclusion: Because of the high frequency and intensity of childhood traumas among alcohol- or other drug-dependent patients and depressed patients, the assessment of problems due to childhood traumas among these patients is essential to a better understanding of the etiology of those disorders and to their treatment. © 2010 Elsevier Ltd. Background. From shotgun libraries used for the genomic sequencing of the phytopathogenic bacterium Xanthomonas axonopodis pv. citri (XAC), clones that were representative of the largest possible number of coding sequences (CDSs) were selected to create a DNA microarray platform on glass slides (XACarray). The creation of the XACarray allowed for the establishment of a tool that is capable of providing data for the analysis of global genome expression in this organism. Findings. The inserts from the selected clones were amplified by PCR with the universal oligonucleotide primers M13R and M13F. The obtained products were purified and fixed in duplicate on glass slides specific for use in DNA microarrays. The number of spots on the microarray totaled 6,144 and included 768 positive controls and 624 negative controls per slide. Validation of the platform was performed through hybridization of total DNA probes from XAC labeled with different fluorophores, Cy3 and Cy5. In this validation assay, 86% of all PCR products fixed on the glass slides were confirmed to present a hybridization signal greater than twice the standard deviation of the deviation of the global median signal-to-noise ration. Conclusions. Our validation of the XACArray platform using DNA-DNA hybridization revealed that it can be used to evaluate the expression of 2,365 individual CDSs from all major functional categories, which corresponds to 52.7% of the annotated CDSs of the XAC genome. As a proof of concept, we used this platform in a previously work to verify the absence of genomic regions that could not be detected by sequencing in related strains of Xanthomonas. © 2010 Moreira et al; licensee BioMed Central Ltd.

Land use classification has been paramount in the last years, since we can identify illegal land use and also to monitor deforesting areas. Although one can find several research works in the literature that address this problem, we propose here the land use recognition by means of Optimum-Path Forest Clustering (OPF), which has never been applied to this context up to date. Experiments among Optimum-Path Forest, Mean Shift and K-Means demonstrated the robustness of OPF for automatic land use classification of images obtained by CBERS-2B and Ikonos-2 satellites. © 2011 IEEE.

The growing use of sensitive loads in the electric power system, especially in industrial applications, increases voltage sags related production losses considerably, stimulating a demand for power electronics' based solutions to mitigate the effects of such problems. This paper shows the implementation and some industrial certification tests of a power equipment prototype designed to correct sags and swells, a dynamic voltage restorer, which is one of the many possible solutions for voltage sags and swells problems Experimental results of a 75kVA prototype are shown both in laboratory and full load conditions, in a certification institution (IEE-USP). © 2011 IEEE.

In the Nilo Coelho irrigation scheme, Brazil, the natural vegetation has been replaced by irrigated agriculture, bringing importance for the quantification of the effects on the energy exchanges between the mixed vegetated surfaces and the lower atmosphere. Landsat satellite images and agro-meteorological stations from 1992 to 2011 were used together, for modelling these exchanges. Surface albedo (α0), NDVI and surface temperature (T0) were the basic remote sensing retrieving parameters necessary to calculate the latent heat flux (λE) and the surface resistance to evapotranspiration (rs) on a large scale. The daily net radiation (Rn) was obtained from α0, air temperature (Ta) and short-wave transmissivity (τsw) throughout the slob equation, allowing the quantification of the daily sensible heat flux (H) by residual in the energy balance equation. With a threshold value for rs, it was possible to separate the energy fluxes from crops and natural vegetation. The averaged fractions of Rn partitioned as H and λE, were in average 39 and 67%, respectively. It was observed an increase of the energy used for the evapotranspiration process inside irrigated areas from 51% in 1992 to 80% in 2011, with the ratio λE/Rn presenting an increase of 3 % per year. The tools and models applied in the current research, can subsidize the monitoring of the coupled climate and land use changes effects in irrigation perimeters, being valuable when aiming the sustainability of the irrigated agriculture in the future, avoiding conflicts among different water users. © 2012 SPIE.

An efficient cryopreservation protocol was developed for mature seeds of Oncidium flexuosum Sims. Seed morphology, protocorm formation, and early seedling development were also assessed. The effects of phloroglucinol and Supercool X-1000® as cryoprotectant additives in the vitrification solution were investigated. Dehydration using the plant vitrification solution 2 (PVS2) for 60 and 120 min prior to immersion in liquid nitrogen promoted the highest frequency of in vitro seed germination 6 weeks following culture on half-strength Murashige and Skoog (1/2 MS) medium. Mature seeds submitted to vitrification for 120 min in PVS2 and 1 % phloroglucinol at 0 °C enhanced germination by 68 %, whereas in PVS2 and 1 % Supercool X-1000® germination was just moderately enhanced (26 %). In vitro-germinating seedlings developed healthy shoots and roots without the use of plant growth regulators. After 6 months of growth, there were no differences between in vitro- and ex vitro-grown seedlings for various phenotypic characteristics, including shoot length, number of leaves, number and length of roots, and fresh and dry weight. Seedlings were transferred to greenhouse conditions and successfully acclimatized, further developing into normal plants with over 90 % survival. Comparative analysis of seedlings from control and vitrified seeds using flow cytometry indicated that no change in ploidy levels occurred as a result of cryopreservation, therefore maintaining seedlings genetic stability. In this study, vitrification with PVS2 for 120 min with the addition of 1 % phloroglucinol offers a simple, safe, and feasible protocol for cryopreservation of O. flexuosum mature seeds. © 2013 Springer Science+Business Media Dordrecht.

Metagenomics has been widely employed for discovery of new enzymes and pathways to conversion of lignocellulosic biomass to fuels and chemicals. In this context, the present study reports the isolation, recombinant expression, biochemical and structural characterization of a novel endoxylanase family GH10 (SCXyl) identified from sugarcane soil metagenome. The recombinant SCXyl was highly active against xylan from beechwood and showed optimal enzyme activity at pH 6,0 and 45°C. The crystal structure was solved at 2.75 Å resolution, revealing the classical (β/α)8-barrel fold with a conserved active-site pocket and an inherent flexibility of the Trp281-Arg291 loop that can adopt distinct conformational states depending on substrate binding. The capillary electrophoresis analysis of degradation products evidenced that the enzyme displays unusual capacity to degrade small xylooligosaccharides, such as xylotriose, which is consistent to the hydrophobic contacts at the +1 subsite and low-binding energies of subsites that are distant from the site of hydrolysis. The main reaction products from xylan polymers and phosphoric acid-pretreated sugarcane bagasse (PASB) were xylooligosaccharides, but, after a longer incubation time, xylobiose and xylose were also formed. Moreover, the use of SCXyl as pre-treatment step of PASB, prior to the addition of commercial cellulolytic cocktail, significantly enhanced the saccharification process. All these characteristics demonstrate the advantageous application of this enzyme in several biotechnological processes in food and feed industry and also in the enzymatic pretreatment of biomass for feedstock and ethanol production. © 2013 Alvarez et al.

This paper analyzes land use change in Rio Claro City and its surroundings, located in the southeastern state of Sao Paulo, in the period from 1988 to 1995, using air-borne digital imagery and a cellular automata model. The simulation experiment was carried out in the Dinamica EGO platform and the results revealed a constrained urban sprawl, resulting from both the densification of residential areas implemented in previous years and the economic recession that led to an internal financial crisis in Brazil during the early 1990s. The simulation outputs were validated using a multi-resolution procedure based on a fuzzy similarity index and showed a satisfactory fitness in relation to the historical reference data. © 2013 IEEE.

Breast cancer is a public health problem throughout the world. Moreover, breast cancer cells have a great affinity for hydroxyapatite, leading to a high occurrence of bone metastasis. In this work we developed a bio-nanocomposite (bio-NCP) in order to use such affinity in the diagnosis and treatment of breast cancer. The bio-NCP consists of magnetic nanoparticles of Mn and Zn ferrite inside a polymeric coating (chitosan) modified with nanocrystals of apatite. The materials were characterized with synchrotron X-ray Powder Diffraction (XPD), Time-of-Flight Neutron Powder Diffraction (NPD), Fourier Transformed Infra-red Spectroscopy (FTIR), Scanning Electron Microscopy (SEM) and magnetic measurement with a Physical Property Measurement System (PPMS). We obtained ferrite nanoparticles with a high inversion degree of the spinel structure regarding the Fe and Mn, but with all the Zn in the A site. The coating of such nanoparticles with chitosan had no notable effects to the ferrite microstructure. In addition, the polymeric surface can be easily modified with apatite nanocrystals since the hydration of the bio-NCP during synthesis can be controlled. The resulting bio-NCP presents a spherical shape with a narrow size distribution and high magnetic response at room temperature and is a very promising material for early diagnosis of breast cancer and its treatment. © 2013 Elsevier B.V.

The Economic Commission for Latin America and the Caribbean (ECLAC) Subregional Headquarters for the Caribbean, in collaboration with the Division of Production, Productivity and Management at ECLAC Headquarters in Chile, convened a one-day workshop on “Boosting SME Development and Competitiveness in the Caribbean”, at the Subregional Headquarters for the Caribbean in Port of Spain on 14 May 2009. The workshop was the culmination of country studies that were carried out under an Italian Government-funded project to assess the policies, institutions and instruments for dynamic Small and Medium Enterprises (SME) development and competitiveness in Jamaica, Suriname and Trinidad and Tobago. The aim was to use the lessons learned from the three country studies to inform policy and practice in the other member countries of the Caribbean Development and Cooperation Committee (CDCC). The main objectives of the workshop were to: (a) share and discuss the findings of the country studies and lessons learned; (b) provide a forum for high quality discussion of the policy environment, instruments, business development and support services required for successful SME development in the Caribbean; and (c) map out a strategy for moving from analysis and recommendation to policy implementation and business changes in order to promote a dynamic and competitive SME sector. The workshop aimed to arrive at practical solutions to major constraints and a weighting of key actions in order of priority of implementation, by adopting a problem-solving approach. Participants at the workshop included representatives of key SME support institutions in the region, actual SMEs and academic researchers. The list of participants and provisional programme are annexed to this report.
