[EN]We present a new strategy, based on the idea of the meccano method and a novel T-mesh optimization procedure, to construct a T-spline parameterization of 2D geometries for the application of isogeometric analysis. The proposed method only demands a boundary representation of the geometry as input data. The algorithm obtains, as a result, high quality parametric transformation between 2D objects and the parametric domain, the unit square. First, we define a parametric mapping between the input boundary of the object and the boundary of the parametric domain. [EN]We present a new method to construct a trivariate T-spline representation of complex solids for the application of isogeometric analysis. The proposed technique only demands the surface of the solid as input data. The key of this method lies in obtaining a volumetric parameterization between the solid and a simple parametric domain. To do that, an adaptive tetrahedral mesh of the parametric domain is isomorphically transformed onto the solid by applying the meccano method. [EN]In this paper we review the novel meccano method. We summarize the main stages (subdivision, mapping, optimization) of this automatic tetrahedral mesh generation technique and we concentrate the study to complex genus-zero solids. In this case, our procedure only requires a surface triangulation of the solid. A crucial consequence of our method is the volume parametrization of the solid to a cube. We construct volume T-meshes for isogeometric analysis by using this result. The efficiency of the proposed technique is shown with several examples. A comparison between the meccano method and standard mesh generation techniques is introduced.-1…

[ES]El método de diseño aerodinámico de ventiladores centrífugos, utilizando la velocidad específica de rotación y las ecuaciones de las curvas características de una familia de ventiladores centrífugos de alabes rectos atrasados, permite implementar un programa informático donde la interface de usuario admite ingresar valores de caudal y presión total, para determinar la potencia del ventilador, velocidad del rotor, eficiencia y los parámetros geométricos, diseñándolo como una turbomáquina hidráulica. Las curvas de rendimiento que se emplearon fueron obtenidas de ensayos en laboratorios rusos, por analistas especializados, que muestran principalmente las relaciones existentes entre el caudal y la velocidad específica, presión total, potencia y eficiencia. Se seleccionó un ventilador centrífugo de doce alabes rectos atrasados, por su mayor porcentaje de eficiencia y construcción económica pudiendo este ser replicado a otras escalas mediante la ley de semejanza geométrica. El algoritmo matemático puede ser utilizado para el diseño de otras familias de ventiladores centrífugos.

[ES]El presente trabajo pretende estudiar el efecto mediador que tiene la adicción a la utilización del móvil entre la tríada oscura, que consiste en la confluencia en un mismo individuo de psicopatía, maquiavelismo y narcicismo, y el acoso a través del móvil a compañeros estudiantes en la universidad. En la investigación se aplican ecuaciones estructurales basadas en la varianza, Partial Least Squares (PLS), a una muestra de 402 estudiantes de último curso de los grados en Derecho y Administración y Dirección de Empresas de la Universidad de la de Las Palmas de Gran Canaria. Como principal resultado cabe mencionar el que soporta la hipótesis de mediación de la adicción al móvil.

[EN]We present advances of the meccano method for T-spline modelling and analysis of complex geometries. We consider a planar domain composed by several irregular sub-domains. These sub-regions are defined by their boundaries and can represent different materials. The bivariate T-spline representation of the whole physical domain is constructed from a square. [EN]Isogeometric analysis (IGA) has arisen as an attempt to unify the fields of CAD and classical finite element methods. The main idea of IGA consists in using for analysis the same functions (splines) that are used in CAD representation of the geometry. The main advantage with respect to the traditional finite element method is a higher smoothness of the numerical solution and more accurate representation of the geometry. IGA seems to be a promising tool with wide range of applications in engineering. However, this relatively new technique have some open problems that require a solution. Ein discoidales Lipoprotein aus dem Polychaeten Nereis virens (Annelida) wurde eingehend charakterisiert. Im Vordergrund standen dabei die transportierten Lipide, sowie die Ultrastruktur des Partikels. Das Nereis-Lipoprotein besitzt eine für Invertebraten atypische Lipidzusammensetzung: Außer den Phospholipiden gibt es keine klar dominierende Lipidklasse. Die Charakterisierung der Apolipoproteine zeigt Gemeinsamkeiten mit den Apolipophorinen der Insekten: Wie diese besitzt das Nereis-Lipoprotein zwei Apolipoproteine, die in einer 1:1-Stöchiometrie angeordnet sind. Das größere Protein (ApoNvLp I) ist dabei stärker zum wässrigen Medium exponiert ist als das kleinere (ApoNvLp II). Beide Proteinuntereinheiten sind N-glycosyliert. ApoNvLp II ist zusätzlich noch O-glycosyliert. Bei den Sekundärstrukturen dominieren β-Strukturen (35%) gegenüber α-Helices (14%); 28% waren ungeordnete Strukturen. Die Masse wurde mit verschiedenen Methoden bestimmt: sie liegt zwischen ~800 kDa (Gelfiltration) und ~860 kDa (Analytische Ultrazentrifugation). Der Sedimentationskoeffizient beträgt 9,7 S. Der zelluläre Lipoproteinrezeptor wurde aus einer großen Anzahl von Zellen und Geweben isoliert. Die biochemische Charakterisierung des Rezeptormoleküls zeigte es als ein monomeres, integrales, N- und O-glycosyliertes Membranprotein mit einer Masse von ~114 kDa. Die Bindungscharakteristika (Abhängigkeit von Ca2+, Disulfidbrücken) weisen es als Mitglied der LDLR-Superfamilie aus. In vitro-Inkubationsversuche mit fluoreszenzmarkierten Lipoproteinen zeigten die Aufnahme sowohl in Oocyten als auch in freie Coelomzellen (Elaeocyten) sowie in Spermatogonien- und Tetradenstadien. Auffällig war, dass die Lipide zusammen mit den Apolipoproteinen in die Dottergranula der Eizellen eingelagert wurden und nicht direkt in die Lipidtropfen. Auch bei den Elaeocyten wurden die Lipide nicht direkt in den Lipidtropfen eingelagert. Intakte Lipoproteine konnten per Dichtegradienten-Ultrazentrifugation nur aus Spermatogonien isoliert werden. Die isolierten Lipoproteine hatten die gleiche ‚Morphologie’ wie die aus der Coelomflüssigkeit isolierten, zeigten jedoch sehr viele Peptidfragmente im SDS-Gel, was auf eine beginnende Degradation hinweist. Es wird ein Modell für den Lipidtransport in Nereis virens vorgeschlagen, bei dem den Elaeocyten eine entscheidende Rolle im Lipidstoffwechsel zufällt.

The amyloid precursor protein (APP) is a type I transmembrane glycoprotein, which resembles a cell surface receptor, comprising a large ectodomain, a single spanning transmembrane part and a short C-terminal, cytoplasmic domain. It belongs to a conserved gene family, with over 17 members, including also the two mammalian APP homologues proteins APLP1 and APLP2 („amyloid precursor like proteins“). APP is encoded by 19 exons, of which exons 7, 8, and 15 can be alternatively spliced to produce three major protein isoforms APP770, APP751 and APP695, reflecting the number of amino acids. The neuronal APP695 is the only isoform that lacks a Kunitz Protease Inhibitor (KPI) domain in its extracellular portion whereas the two larger, peripheral APP isoforms, contain the 57-amino-acid KPI insert. rnRecently, research effort has suggested that APP metabolism and function is thought to be influenced by homodimerization and that the oligomerization state of APP could also play a role in the pathology of Alzheimer's disease (AD), by regulating its processing and amyloid beta production. Several independent studies have shown that APP can form homodimers within the cell, driven by motifs present in the extracellular domain, as well as in the juxtamembrane (JM) and transmembrane (TM) regions of the molecule, whereby the exact molecular mechanism and the origin of dimer formation remains elusive. Therefore, we focused in our study on the actual subcellular origin of APP homodimerization within the cell, an underlying mechanism, and a possible impact on dimerization properties of its homologue APLP1. Furthermore, we analyzed homodimerization of various APP isoforms, in particular APP695, APP751 and APP770, which differ in the presence of a Kunitz-type protease inhibitor domain (KPI) in the extracellular region. In order to assess the cellular origin of dimerization under different cellular conditions, we established a mammalian cell culture model-system in CHO-K1 (chinese hamster ovary) cells, stably overexpressing human APP, harboring dilysine based organelle sorting motifs at the very C-terminus [KKAA-Endoplasmic Reticulum (ER); KKFF-Golgi]. In this study we show that APP exists as disulfide-bound, SDS-stable dimers, when it was retained in the ER, unlike when it progressed further to the cis-Golgi, due to the KKFF ER exit determinant. These stable APP complexes were isolated from cells, and analyzed by SDS–polyacrylamide gel electrophoresis under non-reducing conditions, whereas strong denaturing and reducing conditions completely converted those dimers to monomers. Our findings suggested that APP homodimer formation starts early in the secretory pathway and that the unique oxidizing environment of the ER likely promotes intermolecular disulfide bond formation between APP molecules. We particularly visualized APP dimerization employing a variety of biochemical experiments and investigated the origin of its generation by using a Bimolecular Fluorescence Complementation (BiFC) approach with split GFP-APP chimeras. Moreover, using N-terminal deletion constructs, we demonstrate that intermolecular disulfide linkage between cysteine residues, exclusively located in the extracellular E1 domain, represents another mechanism of how an APP sub-fraction can dimerize within the cell. Additionally, mutational studies revealed that cysteines at positions 98 and 105, embedded in the conserved loop region within the E1 domain, are critical for interchain disulfide bond formation. Using a pharmacological treatment approach, we show that once generated in the oxidative environment of the ER, APP dimers remain stably associated during transport, reaching the plasma membrane. In addition, we demonstrate that APP isoforms, encompassing the KPI domain, exhibit a strongly reduced ability to form cis-directed dimers in the ER, whereas trans-directed cell aggregation of Drosophila Schneider (S2)-cells was isoform independent, mediating cell-cell contacts. Thus, suggesting that steric properties of KPI-APP might be the cause for weaker cis-interaction in the ER, compared to APP695. Finally, we provide evidence that APP/APLP1 heterointeractions are likewise initiated in the ER, suggesting a similar mechanism for heterodimerization. Therefore, dynamic alterations of APP between monomeric, homodimeric, and possibly heterodimeric status could at least partially explain some of the variety in the physiological functions of APP.rn

Although employees are encouraged to take exercise after work to keep physically fit, they should not suffer injury. Some sports injuries that occur after work appear to be work-related and preventable. This study investigated whether cognitive failure mediates the influence of mental work demands and conscientiousness on risk-taking and risky and unaware behaviour during after-work sports activities. Participants were 129 employees (36% female) who regularly took part in team sports after work. A structural equation model showed that work-related cognitive failure significantly mediated the influence of mental work demands on risky behaviour during sports (p < .05) and also mediated the directional link between conscientiousness and risky behaviour during sports (p < .05). A path from risky behaviour during sports to sports injuries in the last four weeks was also significant (p < .05). Performance constraints, time pressure, and task uncertainty are likely to increase cognitive load and thereby boost cognitive failures both during work and sports activities after work. Some sports injuries after work could be prevented by addressing the issue of work redesign.

Die Bedrohungssituation für Menschenrechte hat sich in den letzten Jahren gewandelt. Stellte früher meist ein zu mächtiger Staat die Hauptgefahr für Menschenrechte dar, resultieren Probleme heute oft aus dessen Schwäche, z. B. in Bürgerkriegen. Dies hat zwei Auswirkungen: Zum einen erlebt das humanitäre Völkerrecht eine Renaissance, zum anderen sind es zunehmend nichtstaatliche Gebilde, welche für Rechtsverletzungen verantwortlich sind. Dogmatisch wirft diese Konstellation verschiedenste Probleme auf: Menschenrechtliche Verpflichtungen mit unterschiedlichem Geltungsgrad überlappen sich mit solchen des humanitären Völkerrechts, staatliche Verpflichtungen verlangen vermehrt auch aktives Handeln, und in Folge der unterschiedlichen beteiligten Akteure lassen sich Lösungen oft nur unter Berücksichtigung der Regeln der Staatenverantwortlichkeit finden. Ziel des Autors ist es, aus diesem Gewirr anwendbarer Normen ein integrales Modell des Geltungsgrades, d. h. der Verpflichtungen aus Menschenrechten, zu entwickeln.

Este libro busca que el lector identifique e interprete conocimientos referidos a números reales y a sus operaciones, reconociendo los algoritmos y los procedimientos relacionados vinculándolos al cálculo de distintas medidas; que comprenda y sepa resolver problemas, seleccionando el tipo de razonamiento o argumentación que requiera la situación; que emplee sistemas de ecuaciones e inecuaciones para modelizar y resolver situaciones reales del entorno cotidiano; y que identifique, defina, grafique, describe e interprete distintos tipos de funciones asociándolas a situaciones reales. Se editó como material de aprendizaje destinado al personal de seguridad pública de la Provincia de Mendoza en el marco del proyecto pedagógico con modalidad a distancia para la terminalidad de estudios de EGB3 y Educación Polimodal –EDITEP–, implementado a partir de la firma del Convenio entre la Universidad Nacional de Cuyo y el Gobierno de la Provincia de Mendoza, en octubre de 2003.
