A series of quasi-static and dynamic tensile tests at varying temperatures were carried out to determine the mechanical behaviour of Ti-45Al-2Nb-2Mn+0.8vol.% TiB2 XD as-HIPed alloy. The temperature for the tests ranged from room temperature to 850  ∘C. The effect of the temperature on the ultimate tensile strength, as expected, was almost negligible within the selected temperature range. Nevertheless, the plastic flow suffered some softening because of the temperature. This alloy presents a relatively low ductility; thus, a low tensile strain to failure. The dynamic tests were performed in a Split Hopkinson Tension Bar, showing an increase of the ultimate tensile strength due to the strain rate hardening effect. Johnson-Cook constitutive relation was used to model the plastic flow. A post-testing microstructural of the specimens revealed an inhomogeneous structure, consisting of lamellar α2 + γ structure and γ phase equiaxed grains in the centre, and a fully lamellar structure on the rest. The assessment of the duplex-fully lamellar area ratio showed a clear relationship between the microstructure and the fracture behaviour.

A system for estimation of unknown rectangular room dimensions based on two radio transceivers, both capable of full duplex operations, is presented. The approach is based on CIR measurements taken at the same place where the signal is transmitted (generated), commonly known as self- to-self CIR. Another novelty is the receiver antenna design which consists of eight sectorized antennas with 45° aperture in the horizontal plane, whose total coverage corresponds to the isotropic one. The dimensions of a rectangular room are reconstructed directly from radio impulse responses by extracting the information regarding features like round trip time, received signal strength and reverberation time. Using radar approach the estimation of walls and corners positions are derived. Additionally, the analysis of the absorption coefficient of the test environment is conducted and a typical coefficient for office room with furniture is proposed. Its accuracy is confirmed through the results of volume estimation. Tests using measured data were performed, and the simulation results confirm the feasibility of the approach.

A system for simultaneous 2D estimation of rectangular room and transceiver localization is proposed. The system is based on two radio transceivers, both capable of full duplex operations (simultaneous transmission and reception). This property enables measurements of channel impulse response (CIR) at the same place the signal is transmitted (generated), commonly known as self-to-self CIR. Another novelty of the proposed system is the spatial CIR discrimination that is possible with the receiver antenna design which consists of eight sectorized antennas with 45° aperture in the horizontal plane and total coverage equal to the isotropic one. The dimensions of a rectangular room are reconstructed directly from spatial radio impulse responses by extracting the information regarding round trip time (RTT). Using radar approach estimation of walls and corners positions is derived. Tests using measured data were performed, and the simulation results confirm the feasibility of the approach.

This work aims to contribute to a further understanding of the fundamentals of crystallographic slip and grain boundary sliding in the γ-TiAl Ti–45Al–2Nb–2Mn (at%)–0.8 vol%TiB2 intermetallic alloy, by means of in situ high-temperature tensile testing combined with electron backscatter diffraction (EBSD). Several microstructures, containing different fractions and sizes of lamellar colonies and equiaxed γ-grains, were fabricated by either centrifugal casting or powder metallurgy, followed by heat treatment at 1300 °C and furnace cooling. in situ tensile and tensile-creep experiments were performed in a scanning electron microscope (SEM) at temperatures ranging from 580 °C to 700 °C. EBSD was carried out in selected regions before and after straining. Our results suggest that, during constant strain rate tests, true twin γ/γ interfaces are the weakest barriers to dislocations and, thus, that the relevant length scale might be influenced by the distance between non-true twin boundaries. Under creep conditions both grain/colony boundary sliding (G/CBS) and crystallographic slip are observed to contribute to deformation. The incidence of boundary sliding is particularly high in γ grains of duplex microstructures. The slip activity during creep deformation in different microstructures was evaluated by trace analysis. Special emphasis was placed in distinguishing the compliance of different slip events with the Schmid law with respect to the applied stress.

La utilización de barras corrugadas de acero inoxidable en estructuras de hormigón armado, se está mostrando como una alternativa con gran futuro en estructuras expuestas a ambientes muy agresivos o que requieran vidas en servicio muy elevadas. Estos aceros inoxidables cuentan con similares propiedades mecánicas que los aceros al carbono pero un comportamiento muy mejorado frente a la corrosión, especialmente frente a cloruros. Dentro de los aceros inoxidables, los del tipo dúplex tienen como ventaja una composición con una cantidad menor de níquel, reduciendo de esta manera el coste de estos y haciendo que su precio dependa menos de las fluctuaciones del precio del níquel. Este trabajo estudia la resistencia frente a la corrosión bajo tensión de estos aceros inoxidables del tipo dúplex (AISI 2001 y AISI 2205). The use of stainless steel reinforcing bars in concrete structures is proving to be an alternative with great future in structures exposed to aggressive environments or that are required to perform very long service lives. These steels have similar mechanical properties as carbon steels but very improved corrosion resistance, particularly against chlorides. Within stainless steels, duplex type ones have as an advantage their lower content of nickel in their composition, reducing this way their price and making it less related to nickel price fluctuations. This project will study stress corrosion cracking behavior for some of these duplex stainless steels (AISI 2001 and AISI 2205)

Históricamente la fractura ha sido considerada siempre como un efecto indeseado entre los materiales, dado que su aparición supone un cese del material en servicio, puesto que un material fracturado carece de importancia desde el punto de vista comercial. Consecuentemente, la Mecánica de Fractura ha experimentado un desarrollo importante en las últimas décadas como no lo hizo en toda la historia de los materiales. El desarrollo de nuevos campos a nivel científico y técnico han estado de la mano con el desarrollo de nuevos materiales que satisfagan las necesidades particulares de cada sector o aplicación. Este requerimiento se ve acentuado cuando se incorpora el aspecto económico, dado que, así como se necesitan materiales con mayor resistencia a la fractura, corrosión etc, también se necesita que su precio en el mercado sea accesible y que permita una aplicación rentable. En los últimos 70 años, desde los requerimientos de nuevos materiales resistentes a la fractura con los buques Liberty hasta el boom petrolero, pasando por las aplicaciones aeroespaciales se han desarrollado diversas teorías que explican el comportamiento de los materiales, en cuando a la tenacidad a la fractura en distintas temperaturas, composiciones químicas, materiales compuestos etc. Uno de los sectores que más ha demandado un desarrollo, por su amplitud en cuanto a requerimientos y consumo global, así como su impacto en la economía mundial, es el sector de gas, petróleo y petroquímica. Muchos de los proyectos que se intentaron desarrollar hasta hace menos de 25 años eran inviables por su elevado coste de ejecución y su bajo retorno de inversión debido a la caída de los precios del petróleo. Con una demanda creciente a nivel mundial y unos precios que apuntan hacia la estabilización o alza moderada, nuevos sistemas de trasporte por tuberías han sido necesarios desarrollar, desde el punto de vista de ingeniería, con el menos coste posible y de un modo seguro. Muchas de estas aplicaciones se vieron incrementadas cuando nuevos requerimientos en cuanto a resistencia a la corrosión fueron necesarios: demanda de materiales que no se corroan, con prestaciones seguras a nivel mecánico y un bajo coste. Esta nueva etapa se conoce como Aleaciones Resistentes a la Corrosión (CRA´s por sus siglas en inglés) en las cuales uno de los factores de diseño seguro recaían indiscutiblemente en la mecánica de fractura. Por estas razones era necesario entender como influía en la resistencia a la fractura las aportaciones que podrían hacerse sobre una superficie metálica. Al realizar el presente estudio se comenzó analizando la influencia que tenían modificaciones en el rango iónico sobre aceros al carbono. Estudios previos sobre láminas de acero ferrítico usadas en reactores de fisión nuclear demostraron que aportes de iones, en este particular el Helio, influían en el comportamiento de la tenacidad a la fractura en función de la temperatura. De este modo, un primer análisis fue hecho sobre la influencia de iones de nitrógeno aportados sobre superficies de acero al carbono y como modificaban su tenacidad a la fractura. Este primer análisis sirvió para comprobar el impacto que tenían pequeñas dosis de iones de nitrógeno en la tenacidad a la fractura. Otro desarrollo con una mayor aplicación industrial fue hecho sobre superficies de acero al carbono con aporte por soldadura de los materiales más usados para evitar la corrosión. El análisis se centró fundamentalmente en la influencia que tenían distintos materiales aportados como el MONEL 400, DUPLEX 928, INCONEL 625 y STAINLESS-STEEL 316 en referencia a características de diseño como la tensión elástica y la tensión a la rotura. Este análisis permitió conocer el impacto de los materiales aportados en los ensayos de tracción en probetas de acero al carbono. Una explicación acerca del comportamiento fue soportada por el análisis macrofractográfico de los perfiles fracturados y las macro deformaciones en la superficie de las probetas. Un posterior desarrollo teórico permitió modelar matemáticamente la fractura de las probetas aportadas por soldadura en la región elástica por medio de la Ley de Hooke, así como la teoría de Plasticidad de Hill para la región de deformación plástica. ABSTRACT Fracture mechanics has been extensively studied in the last 70 years by the constant requirements of new materials with low costs. These requirements have allowed surface modified welded materials in which it is necessary to know the influence of design fundamentals with the material surface welded. Several specimens have been studied for ductile fracture in longitudinal tensile tests for carbon steel surface-modified by weld overlay MONEL 400, DUPLEX 928, INCONEL 625 and STAINLESS-STEEL 316. Similarly of macro photographic analyzes to level the fractured surfaces that explain the behavior curves obtained in Tensile – displacement charts. The contribution of weld overlay material shows a significant impact on the yield and tensile stress of the specimens which was modeled according to Hooke's law for elastic area and Hill´s theory of plasticity to the plastic one.

Las características y capacidades de los aceros inoxidables sinterizados se han investigado en una doble vertiente. Por una parte con vista a sus capacidades de resistencia a la oxidación en caliente y por otra parte se ha investigado su capacidad para retener microorganismos que contribuyan a la descontaminación de un ambiente. Por ello, para cada una de estas funciones se han utilizado los aceros inoxidables sinterizados, que se han considerado más adecuados. Para estudiar sus capacidades de resistencia a la oxidación en caliente se ha utilizado un acero inoxidable austenítico AISI 304L, un acero inoxidable ferrítico AISI 430L y un acero inoxidable Fe-16Cr-3Al. Para estudiar sus capacidades para retener microorganismos se ha utilizado un acero inoxidable austenítico AISI 316L, un acero inoxidable ferrítico AISI 430L y un acero inoxidable dúplex 50%/50% de los anteriores. Para esta última finalidad los aceros se han compactado a tres diferentes presiones 300, 500 y 700 MPa, a las que corresponden diferentes porosidades. En relación con el comportamiento frente a la oxidación en caliente, se han cuantificado los incrementos positivos o negativos de volumen, masa y densidad en los diferentes tipos de sinterización y estados de tratamiento de oxidación. Como tónica general de comportamiento, puede decirse que los aceros sinterizados bajo vacío son más resistentes a la oxidación, que los sinterizados en atmósfera de N2-5H2 y que los aceros inoxidables austeníticos son algo más resistentes, que los Cr-Al y estos, a su vez, más que los aceros inoxidables ferríticos. Respecto a la retención de microorganismos, los tres aceros inoxidables sinterizados se han ensayado en diferentes medios de cultivo, utilizando cuatro especies de bacterias. Los mejores resultados se han obtenido con Staphylococcus aureus, muy favorable para su observación y recuento. Se han cuantificado, una vez sinterizados y colonizados por los microorganismos, para cada material y presión de compactación, las áreas de cada uno de los poros y el número de microorganismos situados en los poros y en la superficie sin poros. Se ha establecido en cada caso la densidad de microorganismos en las zonas de poros y en las zonas sin poros. Como tónica general puede decirse, que los aceros inoxidables austeníticos aparecen más favorables para estos estudios, que los aceros dúplex y estos más que los inoxidables ferríticos. Asimismo, se desprende que las áreas de los poros dependen de forma unívoca de la presión de compactación y que para áreas de poros decrecientes las densidades de microorganismos son crecientes. En consecuencia, podría deducirse, que a igualdad de área de poros en una superficie, aquella que tuviera los poros más pequeños, retendría mayor cantidad de bacterias. ABSTRACT The characteristics and capacities of sintered stainless steels have been researched from two perspectives: firstly, with a view to their resistance to hot oxidation, and secondly their capacity to retain microorganisms able to decontaminate the environment. For both these functions, sintered stainless steels were used, which are considered to be the most fit for purpose. To study their resistance to hot oxidation, we used austenitic stainless steel AISI 304L, ferritic stainless steel AISI 430L and stainless steel Fe-16Cr-3Al. To study their ability to retain microorganisms, we used austenitic stainless steel AISI 316L, ferritic stainless steel AISI 430L, and duplex stainless steel, being a 50/50 blend of the two former ones. For this last purpose, the steels were compacted at three different pressures (300, 500 and 700 MPa) corresponding to different porosities. With regard to the hot oxidation, we quantified the positive or negative increments in volume, mass and density in the different types of sintering and oxidation treatment states. As a general performance trend, we observed that vacuum sintered steels are more resistant to oxidation than those sintered in an atmosphere of N2-5H2, and that austenitic stainless steels are slightly more resistant than the Cr-Al steels which, in turn, are more resistant than the ferritic stainless steels. With regard to the retention of microorganisms, the three sintered stainless steels were tested in different culture media using four types of bacteria. The best results for observation and counting were obtained with Staphylococcus aureus bacteria. Once sintered and colonized by microorganisms, for each material and compacting pressure we quantified the areas of the pores and the number of microorganisms situated in the pores and on the pore-free surface. In each case, the density of microorganisms in the pores and in the pore-free areas was established. As a general rule, we can say that the austenitic stainless steels appear to be more favourable for this type of study than the duplex steels which, in turn, are more favourable than the ferritic stainless steels. It also emerged that the areas with the pores depend unequivocally on the compacting pressure, and that the smaller the area of the pore the higher the density of the microorganisms. Consequently, it can be deduced that comparing an equal area of pores on a surface, the one with the smaller pores would retain a larger number of bacteria.

Splicing of nuclear precursors of mRNA (pre-mRNA) involves dynamic interactions between the RNA constituents of the spliceosome. The rearrangement of RNA–RNA interactions, such as the unwinding of the U4/U6 duplex, is believed to be driven by ATP-dependent RNA helicases. We recently have shown that spliceosomal U5 small nuclear ribonucleoproteins (snRNPs) from HeLa cells contain two proteins, U5–200kD and U5–100kD, which share homology with the DEAD/DEXH-box families of RNA helicases. Here we demonstrate that purified U5 snRNPs exhibit ATP-dependent unwinding of U4/U6 RNA duplices in vitro. To identify the protein responsible for this activity, U5 snRNPs were depleted of a subset of proteins under high salt concentrations and assayed for RNA unwinding. The activity was retained in U5 snRNPs that contain the U5–200kD protein but lack U5–100kD, suggesting that the U5–200kD protein could mediate U4/U6 duplex unwinding. Finally, U5–200kD was purified to homogeneity by glycerol gradient centrifugation of U5 snRNP proteins in the presence of sodium thiocyanate, followed by ion exchange chromatography. The RNA unwinding activity was found to reside exclusively with the U5–200kD DEXH-box protein. Our data raise the interesting possibility that this RNA helicase catalyzes unwinding of the U4/U6 RNA duplex in the spliceosome.

Membrane bilayer fusion has been shown to be mediated by v- and t-SNAREs initially present in separate populations of liposomes and to occur with high efficiency at a physiologically meaningful rate. Lipid mixing was demonstrated to involve both the inner and the outer leaflets of the membrane bilayer. Here, we use a fusion assay that relies on duplex formation of oligonucleotides introduced in separate liposome populations and report that SNARE proteins suffice to mediate complete membrane fusion accompanied by mixing of luminal content. We also find that SNARE-mediated membrane fusion does not compromise the integrity of liposomes.

The spectrum of mutations induced by the naturally occurring DNA adduct pyrimido[1,2-α]purin-10(3H)-one (M1G) was determined by site-specific approaches using M13 vectors replicated in Escherichia coli. M1G was placed at position 6256 in the (−)-strand of M13MB102 by ligating the oligodeoxynucleotide 5′-GGT(M1G)TCCG-3′ into a gapped-duplex derivative of the vector. Unmodified and M1G-modified genomes containing either a cytosine or thymine at position 6256 of the (+)-strand were transformed into repair-proficient and repair-deficient E. coli strains, and base pair substitutions were quantitated by hybridization analysis. Modified genomes containing a cytosine opposite M1G resulted in roughly equal numbers of M1G→A and M1G→T mutations with few M1G→C mutations. The total mutation frequency was ≈1%, which represents a 500-fold increase in mutations compared with unmodified M13MB102. Transformation of modified genomes containing a thymine opposite M1G allowed an estimate to be made of the ability of M1G to block replication. The (−)-strand was replicated >80% of the time in the unadducted genome but only 20% of the time when M1G was present. Correction of the mutation frequency for the strand bias of replication indicated that the actual frequency of mutations induced by M1G was 18%. Experiments using E. coli with different genetic backgrounds indicated that the SOS response enhances the mutagenicity of M1G and that M1G is a substrate for repair by the nucleotide excision repair complex. These studies indicate that M1G, which is present endogenously in DNA of healthy human beings, is a strong block to replication and an efficient premutagenic lesion.

RecA is a 38-kDa protein from Escherichia coli that polymerizes on single-stranded DNA, forming a nucleoprotein filament that pairs with homologous duplex DNA and carries out strand exchange in vitro. To observe the effects of mismatches on the kinetics of the RecA-catalyzed recombination reaction, we used assays based upon fluorescence energy transfer that can differentiate between the pairing and strand displacement phases. Oligonucleotide sequences that produced 2–14% mismatches in the heteroduplex product of strand exchange were tested, as well as completely homologous and heterologous sequences. The equilibrium constant for pairing decreased as the number of mismatches increased, which appeared to result from both a decrease in the rate of formation and an increase in the rate of dissociation of the intermediates. In addition, the rate of strand displacement decreased with increasing numbers of mismatches, roughly in proportion to the number of mismatches. The equilibrium constant for pairing and the rate constant for strand displacement both decreased 6-fold as the heterology increased to 14%. These results suggest that discrimination of homology from heterology occurs during both pairing and strand exchange.

The crystal structure of the RNA dodecamer 5′-GGCC(GAAA)GGCC-3′ has been determined from x-ray diffraction data to 2.3-Å resolution. In the crystal, these oligomers form double helices around twofold symmetry axes. Four consecutive non-Watson–Crick base pairs make up an internal loop in the middle of the duplex, including sheared G·A pairs and novel asymmetric A·A pairs. This internal loop sequence produces a significant curvature and narrowing of the double helix. The helix is curved by 34° from end to end and the diameter is narrowed by 24% in the internal loop. A Mn2+ ion is bound directly to the N7 of the first guanine in the Watson–Crick region following the internal loop and the phosphate of the preceding residue. This Mn2+ location corresponds to a metal binding site observed in the hammerhead catalytic RNA.

LEF-1 (lymphoid enhancer-binding factor 1) is a cell type-specific member of the family of high mobility group (HMG) domain proteins that recognizes a specific nucleotide sequence in the T cell receptor (TCR) α enhancer. In this study, we extend the analysis of the DNA-binding properties of LEF-1 and examine their contributions to the regulation of gene expression. We find that LEF-1, like nonspecific HMG-domain proteins, can interact with irregular DNA structures such as four-way junctions, albeit with lower efficiency than with specific duplex DNA. We also show by a phasing analysis that the LEF-induced DNA bend is directed toward the major groove. In addition, we find that the interaction of LEF-1 with a specific binding site in circular DNA changes the linking number of DNA and unwinds the double helix. Finally, we identified two nucleotides in the LEF-1-binding site that are important for protein-induced DNA bending. Mutations of these nucleotides decrease both the extent of DNA bending and the transactivation of the TCRα enhancer by LEF-1, suggesting a contribution of protein-induced DNA bending to the function of TCRα enhancer.

Although three human genes encoding DNA ligases have been isolated, the molecular mechanisms by which these gene products specifically participate in different DNA transactions are not well understood. In this study, fractionation of a HeLa nuclear extract by DNA ligase I affinity chromatography resulted in the specific retention of a replication protein, proliferating cell nuclear antigen (PCNA), by the affinity resin. Subsequent experiments demonstrated that DNA ligase I and PCNA interact directly via the amino-terminal 118 aa of DNA ligase I, the same region of DNA ligase I that is required for localization of this enzyme at replication foci during S phase. PCNA, which forms a sliding clamp around duplex DNA, interacts with DNA pol δ and enables this enzyme to synthesize DNA processively. An interaction between DNA ligase I and PCNA that is topologically linked to DNA was detected. However, DNA ligase I inhibited PCNA-dependent DNA synthesis by DNA pol δ. These observations suggest that a ternary complex of DNA ligase I, PCNA and DNA pol δ does not form on a gapped DNA template. Consistent with this idea, the cell cycle inhibitor p21, which also interacts with PCNA and inhibits processive DNA synthesis by DNA pol δ, disrupts the DNA ligase I–PCNA complex. Thus, we propose that after Okazaki fragment DNA synthesis is completed by a PCNA–DNA pol δ complex, DNA pol δ is released, allowing DNA ligase I to bind to PCNA at the nick between adjacent Okazaki fragments and catalyze phosphodiester bond formation.

The replication of damaged nucleotides that have escaped DNA repair leads to the formation of mutations caused by misincorporation opposite the lesion. In Escherichia coli, this process is under tight regulation of the SOS stress response and is carried out by DNA polymerase III in a process that involves also the RecA, UmuD′ and UmuC proteins. We have shown that DNA polymerase III holoenzyme is able to replicate, unassisted, through a synthetic abasic site in a gapped duplex plasmid. Here, we show that DNA polymerase III*, a subassembly of DNA polymerase III holoenzyme lacking the β subunit, is blocked very effectively by the synthetic abasic site in the same DNA substrate. Addition of the β subunit caused a dramatic increase of at least 28-fold in the ability of the polymerase to perform translesion replication, reaching 52% bypass in 5 min. When the ssDNA region in the gapped plasmid was extended from 22 nucleotides to 350 nucleotides, translesion replication still depended on the β subunit, but it was reduced by 80%. DNA sequence analysis of translesion replication products revealed mostly −1 frameshifts. This mutation type is changed to base substitution by the addition of UmuD′, UmuC, and RecA, as demonstrated in a reconstituted SOS translesion replication reaction. These results indicate that the β subunit sliding DNA clamp is the major determinant in the ability of DNA polymerase III holoenzyme to perform unassisted translesion replication and that this unassisted bypass produces primarily frameshifts.
