O experimento foi conduzido na fazenda Pedrões, no município de Uberaba-MG, para avaliar a qualidade do feno de Brachiaria decumbens enfardado após a queda de sementes. Os fenos foram submetidos aos seguintes tratamentos: feno não tratado, feno tratado com amônia anidra (3,0% NH3 na MS), feno tratado com uréia (5,0% na MS). A composição química, a digestibilidade dos fenos e o desempenho de novilhos foram avaliados usando um delineamento em blocos completos casualisados com três tratamentos e seis repetições. A amonização utilizando NH3 ou uréia aumentou o conteúdo de proteína bruta e a DIVMS. O tratamento com NH3 reduziu os conteúdos de FDN e FDA, e a aplicação de uréia reduziu os conteúdos de hemicelulose e lignina. A amonização não afetou os valores de nitrogênio insolúvel em detergente neutro e nitrogênio insolúvel em detergente ácido. Novilhos recebendo feno de brachiaria e farelo de soja (1,08 kg/dia de MS), feno tratado com amônia e milho grão (1,14 kg/dia de MS) e feno tratado com uréia e milho grão (1,14 kg/dia de MS) apresentaram consumo de MS de 1,97; 2,23 e 1,90% do peso vivo e ganhos de peso de 0,60; 0,53 e 0,37 kg/dia, e conversão alimentar 10,8; 12,8 e 16,9 kg MS/kg de ganho de peso, respectivamente.

O objetivo deste trabalho foi verificar a influência do armazenamento de botões florais masculinos por diferentes períodos, na produção e qualidade de sementes de abóbora cultivar Piramoita. O experimento foi desenvolvido no período de agosto a dezembro de 2005. O delineamento experimental utilizado foi em blocos ao acaso, com quatro repetições e cinco tratamentos: polinização manual com flor aberta no dia (zero dia de armazenamento) e polinização manual com botões armazenados em geladeira (7 ºC) por 1, 2, 3 e 4 dias. Quanto maior o período de armazenamento dos botões florais masculinos, menor a produção (número e massa) de sementes por fruto e a massa média do fruto. Também há uma perda de vigor e germinação das sementes quando se aumenta o período de armazenamento dos botões florais, reduzindo sua qualidade.

O presente trabalho constou de sete experimentos, que foram conduzidos na Faculdade de Ciências Agronômicas, campus de Botucatu, UNESP, São Paulo, Brasil, com o objetivo de avaliar os efeitos alelopáticos de Brachiaria decumbens, colhida em diferentes épocas, sobre o crescimento inicial de milho, arroz, trigo, soja, feijão, algodão e capim-braquiária (B. decumbens), bem como a dinâmica do nitrogênio no solo. A parte aérea de B. decumbens foi coletada durante as estações seca (outono-inverno) e chuvosa (primavera-verão), em área de pastagem localizada no município de Botucatu-SP. A matéria seca triturada de B. decumbens foi incorporada ao solo dos vasos na proporção de 3% p/p. O delineamento experimental foi inteiramente casualizado, com cinco repetições. Contando-se a partir do transplante, o experimento foi conduzido por 21 dias para milho e feijão, 24 dias para soja e trigo, 28 dias para arroz e 30 dias para algodão e capim-braquiária. O crescimento dessas plantas foi reduzido com a adição de capim-braquiária para as duas épocas, sendo o próprio capim-braquiária o mais afetado. Os efeitos inibitórios foram mais intensos para capim-braquiária coletado na estação chuvosa. A incorporação da matéria seca da parte aérea de B. decumbens reduziu, significativamente, os teores de nitrato no solo, em todos os estudos realizados.

Physical exercise and statins, recommended interventions to dyslipidaemia treatment, are independently related to cardiomyocytes alterations, characterized by miocardic hypertrophy and apoptosis, respectively. Thus, the objective of the present study was to analyze the effects of statin and aerobic physical exercise association in the morphometric parameters of cardiac cell nucleus. 40 male rats adults were divided into four groups: exercised (DE); sedentary (DS), exercised and statin use (DES); sedentary and statin use (DSS). The animals received during the whole experimental period a hiperlipidic diet added 20% of coconut oil and 1.25% of cholesterol; after 30 days of its ingestion, a blood collection was made to verify the dyslipidaemia. Simvastatin (20 mg) was taken five days a week, during eight weeks. During this period, the animals exercised 60 minutes daily in the treadmill. After the last day of the protocol, the cardiac muscle was collected and maintained in liquid nitrogen (-180 degrees C); the cuts were stained by Hematoxilin-Eosin method, and the cardiac fibers were submitted to the nuclear morphometric analyses. The data were analyzed using descriptive analyses, paired T test, Kruskal-Wallis test and Dunn post hoc test; for all analyses, it was adopted p<0.05. It was verified that the group receiving statin presented values statistically significant in comparison to the other groups, in the tridimensional and linear variables. The exercised and statin group, the values obtained in the morphometric analyses were similar to the control group. It is suggested that statins alone can cause alterations in the nucleus of cardiac cells that can be related to apoptosis occurrence and, when exercise is practiced associated to statin administration, the effects of statin can be reduced, what can be related to beneficial adaptations of cardiac mitochondrial in response to physical exercise, turning them more resistant to apoptotic stimuli.

An essential key to pathogenicity in Yersinia is the presence of a 70 kb plasmid (pYV) which encodes a type-III secretion system and several virulence outer proteins whose main function is to enable the bacteria to survive in the host. Thus, a specific immune response is needed in which cytokines are engaged. The aim of this study was to assess the influence of Yersinia outer proteins (Yops) released by Yersinia pseudotuberculosis on the production of the proinflammatory cytokines, interleukin-12 (IL-12), and tumor necrosis factor alpha (TNF-alpha), and nitric oxide (NO) by murine peritoneal macrophages. To this end, female Swiss mice were infected intravenously with wild-type Y pseudotuberculosis or with mutant strains unable to secrete specific Yops (YopE, YopH, YopJ, YopM, and YpkA). on the 7th, 14th, 21st, and 28th days after infection, the animals were sacrificed and the cytokines and NO were assayed in the peritoneal macrophages culture supernatants. A fall in NO production was observed during the course of infection with all the strains tested, though during the infection with the strains that did not secrete YopE and YopH, the suppression occurred later. There was, in general, an unchanged or sometimes increased production of TNF-alpha between the 7th and the 21st day after infection, compared to the control group, followed by an abrupt decrease on the last day of infection. The IL-12 production was also suppressed during the infection, with most of the strains tested, except with those that did not secrete YopJ and YopE. The results suggest that Yops may suppress IL-12, TNF-alpha, and NO production and that the most important proteins involved in this suppression are YopE and YopH. (C) 2004 Elsevier B.V. The fruit of Indian Eugenia jambolana have been shown to have therapeutic properties, but because the therapeutic potential of a plant is related to the geographic region in which the plant was grown and to the part of the plant used, we investigated Brazilian Eugenia jambolana fruit using the same preparation and experimental methods as have been used in India. The well-established metabolic cage model was used to evaluate the physiological and metabolic parameters associated with streptozotocin-induced diabetes in rats (n = 10) which had been administered, by gavage, 50 mg per day of lyophilised Eugenia jambolana fruit-pulp extract for 41 days. We found that, compared to untreated controls, rats treated with the lyophilised fruit-pulp showed no observable difference in body weight, food or water intake, urine volume, glycaemia, urinary urea and glucose, hepatic glycogen, or on serum levels of total cholesterol, HDL cholesterol or triglycerides. No change was observed in the masses of epididymal or retroperitoneal adipose tissue or of soleus or extensor digitorum longus muscles. This lack of any apparent effect on the diabetes may be attributable to the regional ecosystem where the fruit was collected and/or to the severity of the induced diabetes. (C) 2004 Elsevier B.V.. A germinação das sementes de grão-de-bico foi acompanhada por um período de 6 dias, no qual pequenas variações nos teores de nitrogênio e globulina total foram registradas. A globulina majoritária (tipo 11 S) apresentou maiores variações após o quarto dia de germinação. A natureza e distribuição da fração globulina majoritária isolada na cromatografia em Sepharose CL-6B mostrou pequenas modificações ao final do período de germinação. A eletroforese em gel de poliacrilamida com dodecilssulfato de sódio do pico eluído na cromatografia em Sepharose CL-6B demonstra modificações nas bandas de proteínas entre os pesos moleculares de 20 e 30 kDa e acima de 60 kDa, indicando degradação protéica durante o período. Atividade proteolítica foi detectada na fração albumina da semente que aumentou até o quarto dia, seguido de queda até o sexto dia de germinação, quando da utilização de globulina total isolada da semente e caseína como substratos. Farinha de grão-de-bico, frações albumina e globulina total isoladas não apresentaram aumento na digestibilidade in vitro; entretanto, a fração globulina majoritária isolada foi mais suscetível à hidrólise após germinação.

The addition of a hydroxymethyl group to the antimicrobial drug nitrofurazone generated hydroxymethylnitrofurazone (NFOH), which had reduced toxicity when its activity against Trypanosoma cruzi was tested in a murine model of Chagas' disease. Four groups of 12 Swiss female mice each received 150 mg of body weight/kg/day of NFOH, 150 mg/kg/day of nitrofurazone (parental compound), 60 mg/kg/day of benznidazole (BZL), or the solvent as a placebo. Treatments were administered orally once a day 6 days a week until the completion of 60 doses. NFOH was as effective as BZL in keeping direct parasitemia at undetectable levels, and PCR results were negative. No histopathological lesions were seen 180 days after completion of the treatments, a time when the levels of anti-T. cruzi antibodies were very low in mice treated with either NFOH or BZL. Nitrofurazone was highly toxic, which led to an overall rate of mortality of 75% and necessitated interruption of the treatment. In contrast, the group treated with its hydroxymethyl derivative, NFOH, displayed the lowest mortality (16%), followed by the BZL (33%) and placebo (66%) groups. The findings of histopathological studies were consistent with these results, with the placebo group showing the most severe parasite infiltrates in skeletal muscle and heart tissue and the NFOH group showing the lowest. The present evidence suggests that NFOH is a promising anti-T. cruzi agent.

Toluene and verapamil are subject to extensive oxidative metabolism mediated by CYP enzymes, and their interaction can be stereoselective. In the present study we investigated the influence of toluene inhalation on the enantioselective kinetic disposition of verapamil and its metabolite, norverapamil, in rats. Male Wistar rats (n = 6 per group) received a single dose of racemic verapamil (10 mg/kg) orally at the fifth day of nose-only toluene or air (control group) inhalation for 6 h/day (25, 50, and 100 ppm). Serial blood samples were collected from the tail up to 6 h after verapamil administration. The plasma concentrations of verapamil and norverapamil enantiomers were analyzed by LC-MS/MS by using a Chiralpak AD column. Toluene inhalation did not influence the kinetic disposition of verapamil or norverapamil enantiomers (p > 0.05, Kruskal-Wallis test) in rats. The pharmacokinetics of verapamil was enantioselective in the control group, with a higher plasma proportion of the S-verapamil (AUC 250.8 versus 120.4 ng.h.mL(-1); p <= 0.05, Wilcoxon test) and S-norverapamil (AUC 72.3 versus 52.3 ng.h.mL(-1); p <= 0.05, Wilcoxon test). Nose-only exposure to toluene at 25, 50, or 100 ppm resulted in a lack of enantioselectivity for both verapamil and norverapamil. The study demonstrates the importance of the application of enantioselective methods in studies on the interaction between solvents and chiral drugs.
