The aim of the present study was to estimate the index and individual responses to selection for milk (MY), fat (FY) and protein (PY) yields for different breeding goals for two commercial buffalo milk production systems in São Paulo State characterized by: 1) all milk produced is sold to the industry (MILK) and 2) all milk produced is used in the mozzarella cheese-making process at the farm (MOZZARELLA). The current payment policy is based exclusively on milk volume. The mozzarella price refers to the wholesale selling price. Index responses to selection (IR) were calculated for three different breeding goals (BG): 1) MY exclusively (BG(1)); 2) FY + PY (BG(2)) and 3) MY + FY + PY (BG(3)). IR for the MILK system were US$ 41.79 (BG(1)), US$ 5.91 (BG(2)) and US$ 38.22 (BG(3)). For the MOZZARELLA system, IR were US$ 179.50 (BG(1)), US$ 262.85 (BG(2)) and US$ 402.41 (BG(3)). The results suggest that for the present circumstances, selection for milk components is not advantageous when milk is produced for sale to the industry. However, when mozzarella making is added to the system, the selection for components and milk volume is the most economically beneficial.

The objective of this study was to apply factor analysis to describe lactation curves in dairy buffaloes in order to estimate the phenotypic and genetic association between common latent factors and cumulative milk yield. A total of 31 257 monthly test-day milk yield records from buffaloes belonging to herds located in the state of São Paulo were used to estimate two common latent factors, which were then analysed in a multi-trait animal model for estimating genetic parameters. Estimates of (co)variance components for the two common latent factors and cumulated 270-d milk yield were obtained by Bayesian inference using a multiple trait animal model. Contemporary group, number of milkings per day (two levels) and age of buffalo cow at calving (linear and quadratic) as covariate were included in the model as fixed effects. The additive genetic, permanent environmental and residual effects were included as random effects. The first common latent factor (F1) was associated with persistency of lactation and the second common latent factor (F2) with the level of production in early lactation. Heritability estimates for Fl and F2 were 0.12 and 0.07, respectively. Genetic correlation estimates between El and F2 with cumulative milk yield were positive and moderate (0.63 and 0.52). Multivariate statistics employing factor analysis allowed the extraction of two variables (latent factors) that described the shape of the lactation curve. It is expected that the response to selection to increase lactation persistency is higher than the response obtained from selecting animals to increase lactation peak. Selection for higher total milk yield would result in a favourable correlated response to increase the level of production in early lactation and the lactation persistency.

Babesia spp. infections were investigated in Bos taurus x Bos indicus dairy cows and calves and in Boophilus microplus engorged female ticks and eggs. Blood samples and engorged female ticks were collected from 25 cows and 27 calves. Babesia spp. was detected in ticks by microscopic examination of hemolymph of engorged female and by squashes of egg samples. Cattle infection was investigated in blood thin smears and by DNA amplification methods (PCR and nested PCR), using specific primers for Babesia bovis and Babesia bigemina. Merozoites of B. bovis (3 animals) and B. bigemina (12 animals) were detected exclusively in blood smears of calves. DNA amplification methods revealed that the frequency of B. bigemina infection in calves (92.6%) and in cows (84%) and of B. bovis in calves (85.2%) and in cows (100%) did not differ significantly (P > 0.05). Babesia spp. infection was more frequent in female ticks and eggs collected from calves (P < 0.01) than from cows, especially in those which had patent parasitemia. Hatching rates of B. microplus larvae were assessed according to the origin of engorged females, parasiternia of the vertebrate host, frequency and intensity of infection in engorged female tick, and frequency of egg infection. Hatching rate was lower in samples collected from calves (P < 0.01) than from cows, and in those in which Babesia spp. was detected in egg samples (P < 0.01). Published by Elsevier B.V.

This investigation was carried out within the Parana sedimentary basin, Brazil, involved the sampling of effluents and groundwater from monitoring stations situated at different sites at São Paulo State, and was realized with the purpose of evaluating the presence of fats, oil and grease (FOG) in different matrices. Several tests were realized with very distinct materials (cooking oil, butter, margarine, pig fat, vacuum pump oil) in order to properly calibrate the spectrophotometric system. Each matrix was dissolved with 1,1,1-trichloroethane and from the stock solutions it was prepared working standards from different dilutions. The data obtained were plotted on absorbance vs. concentration graph that yielded a successful calibration curve when a mineral oil for vacuum pump was utilized in the experiments at a wavelength corresponding to 410 mn. The results obtained for the analyzed samples were compared with the limiting value established by the São Paulo State legislation on the prevention and pollution control of the environment that was established in 8(th) September 1976 by Rule No. 8468.

Sera collected from 447 dairy cattle on 14 dairy farms were tested for Neospora caninum antibodies by use of an immunofluorescent antibody technique. Positive reactions with titres greater than or equal to 1:200 were found in 63 (14.09%) of animals. Neospora positive sera were also tested for Toxoplasma gondii antibodies by using a commercial latex agglutination test. Antibodies to T. gondii were detected in 3 (4.76%) of 63 N. caninum positive sera. These results indicate that N. caninum infection is widespread among dairy cattle in Bahia state. A new outbreak of Vaccinia virus was observed in Southwest region of São Paulo State. Brazil. The disease was observed in four small dairy farms with manual milking. Lesions were detected in cattle and in humans previously vaccinated and not vaccinated against smallpox. Although several reports of Vaccinia virus outbreaks have been occuring in Brazil, it was not yet reported in this region. This Outbreak reinforces the Viral circulation in Our country. The disease in persons previously vaccinated and not vaccinated against smallpox reinforces the absence of immunity, the risk to the human health, and the need for more epidemiologic and immunologic studies.

The prevalence of Leptospira interrogans serovars in dairy cattle was determined by analyzing 464 serum samples from cows on 15 properties in Garanhuns municipal district, Pernambuco State, Brazil. A microscopic seroagglutination test including 12 serovars of Leptospira interrogans as antigens was used. Samples with titres 3 100 were considered positive. Two hundred and twenty-one (47.63%) of the samples were positive to one or more serovars. The prevalence of the serovars was hardjo (21.98%), bratislava (15.73%), castellonis(11.64%), tarassovi (10.56%), pyrogenes(1.72%), icterohaemorrhagiae (1.08%), pomona (0.86%), wolffi (0.86%), grippotyphosa (0.86%), djasiman (0.43%), canicola (0.21 %), and copenhageni (0.21 %).

Neospora caninum, bovine viral diarrhea virus (BVDV) and bovine herpes virus 1 (BHV-1) are worldwide spread pathogens associated with reproductive problems in cattle. The present work aimed to observe the infection pattern of these three pathogens in two dairy herds with distinct reproductive managements from Triângulo Mineiro, Minas Gerais State, Brazil. The herds were not vaccinated against either N. caninum, BVDV or BHV-1. Blood samples were collected and analyzed for presence of specific antibodies, and N. caninum IgG avidity was measured in N. caninum positive samples. In herd 1, 34 out of 174 sampled cows (20%) had antibodies to N. caninum and the seropositivity of BVDV and BHV-1 were 62% and 86%, respectively. Of 69 sampled cows in herd 2. 7 (10%) had antibodies to N. caninum, and 49% and 39% were seropositive to BVDV and BHV-1, respectively. The IgG avidity profiles indicated that N. caninum had been present in both herds for some years and that herd 1 had an ongoing horizontal spread of the parasite. The results indicate that the studied reproductive pathogens were present in the herds and partly may have contributed to their reproductive problems.

Anaplasma marginale is the most prevalent pathogen of cattle transmitted by ticks in the world. This study aimed to evaluate the risk factors for anaplasmosis in dairy cattle. Fifty dairy cattle from the herd of Empresa de Pesquisa Agropecuaria do Estado do Rio de Janeiro were selected by proportional stratified sampling. The risk factors evaluated were: physiological state, race pattern, number of lactations, milk production, infestation by Rhipicephalus microplus and animal density. Antibody activity against A. marginale was determined using the indirect enzyme-linked immunosorbent assay. The percentual values of seroprevalence for A. marginale were submitted to X2 test, and the level of minimum significance, to keep a factor in the model of logistic regression, was fixated in 5%. It was observed that pregnancy and lactation influenced significantly (p<0.05) in the seropositivity of the animals. Bos indicus animals had 5.21 times more chances of being seropositive than B. taurus animals. Primiparous female had 88% more chances of being seropositive than pluriparous female. Animals with high milk production were 63% more positive than low production animals. When infested by ticks the animals had 39% more chance of being seropositive to A. marginale. Bos indicus animals presented 5.21 times more chance of being seropositive than B. taurus animals. Primiparous females presented 88% more chance of being seropositive than the pluriparous ones. High milk production animals were 63% more positive than the low production ones. When infested by ticks the animals had 39% more chance of being seropositive to A. marginale. High density grazing provided for the animals 3.2 times more chances of being seropositive than low density ones. The herd was classified as erratic to A. marginale, even being placed in a steady enzootic area.

As the methodologies available for the detection of positive selection from genomic data vary in terms of assumptions and execution, weak correlations are expected among them. However, if there is any given signal that is consistently supported across different methodologies, it is strong evidence that the locus has been under past selection. In this paper, a straightforward frequentist approach based on the Stouffer Method to combine P-values across different tests for evidence of recent positive selection in common variations, as well as strategies for extracting biological information from the detected signals, were described and applied to high density single nucleotide polymorphism (SNP) data generated from dairy and beef cattle (taurine and indicine). The ancestral Bovinae allele state of over 440,000 SNP is also reported. Using this combination of methods, highly significant (P<3.17×10-7) population-specific sweeps pointing out to candidate genes and pathways that may be involved in beef and dairy production were identified. The most significant signal was found in the Cornichon homolog 3 gene (CNIH3) in Brown Swiss (P = 3.82×10-12), and may be involved in the regulation of pre-ovulatory luteinizing hormone surge. Other putative pathways under selection are the glucolysis/gluconeogenesis, transcription machinery and chemokine/cytokine activity in Angus; calpain-calpastatin system and ribosome biogenesis in Brown Swiss; and gangliosides deposition in milk fat globules in Gyr. The composite method, combined with the strategies applied to retrieve functional information, may be a useful tool for surveying genome-wide selective sweeps and providing insights in to the source of selection.

O objetivo do presente trabalho foi pesquisar a prevalência e a etiologia da mastite bovina na bacia leiteira do município de Rondon do Pará, bem como avaliar o perfil de sensibilidade e resistência dos agentes isolados frente aos antimicrobianos. Foram avaliadas 237 vacas mestiças de aptidão leiteira, pertencentes a nove propriedades, as quais utilizavam ordenha manual uma vez ao dia e sistema de criação extensivo em pastagens de Brachiaria brizantha, com fornecimento de sal mineral e água ad libitum. Realizou-se o exame clínico da glândula mamária, o teste da caneca telada e o California Mastitis Test. Dos 935 quartos mamários avaliados, 6,6% apresentaram mastite subclínica, 1,3% mastite clínica e 92,1% foram negativos. As bactérias isoladas na mastite clínica foram Staphylococcus spp. coagulase negativo (25%), Staphylococcus aureus (16,7%), Streptococcus spp. (8,3%) e Corynebacterium spp. (8,3%). Na mastite subclínica foram Staphylococcus spp. coagulase negativo (32,3%), Staphylococcus aureus (17,7%), Staphylococcus intermedius (1,6%), Streptococcus spp. (4,8%), Corynebacterium spp. (4,8%) e Staphylococcus spp. coagulase negativo/S. aureus (1,6%). Não houve crescimento microbiano em 41,7% das amostras com mastite clínica e 37,1% com mastite subclínica. No antibiograma, 100% dos isolados de Staphylococcus spp. coagulase negativo, S. aureus, S. intermedius, e Streptococcus spp. foram sensíveis ao sulfazotrim. Por outro lado Corynebacterium spp. foi 100% resistente ao mesmo antimicrobiano. A cefalotina, cefoxitina e gentamicina, apresentaram eficácia frente às bactérias isoladas do gênero Staphylococcus spp., as quais neste trabalho representam a grande maioria dos agentes causadores de mastite. A mastite foi diagnosticada em todos os rebanhos pesquisados, contudo o número de animais acometidos foi considerado baixo; isso provavelmente deve-se à baixa produção de leite dos animais e a permanência do bezerro ao pé após a ordenha, o que favorece o esvaziamento da glândula mamária. Diante disso, faz-se necessário que medidas higiênico-sanitárias e de manejo sejam adotadas.

O presente estudo descreve a ocorrência de intoxicação por chumbo em bovinos e galinhas no Pará, Brasil. Em um lote composto de 80 bezerros de um rebanho leiteiro, 10 animais ficaram doentes e nove morreram, e um animal se recuperou após ser removido do piquete. Após a inspeção deste piquete, foi observada a presença de baterias de caminhões usados para armazenar a energia captada por painéis solares. Os sinais clínicos observados nos bezerros incluíam dificuldade respiratória, corrimento nasal, salivação excessiva, opacidade da córnea, pressão da cabeça contra objetos e decúbito. As galinhas tinham diminuída oviposição e os ovos produzidos eram com cascas malformadas ou tinham a casca mais fina. Os achados de necropsia e as alterações histopatológicas observadas nos bovinos eram de pouco significado, com exceção de um animal que mostrou leve astrocitose no córtex cerebral. Em uma das galinhas, na histopatologia renal observou-se leve necrose tubular aguda multifocal. As concentrações de chumbo médios nos fígados e rins dos bovinos eram 93,91mg/kg e 209,76mg/kg, respectivamente, e a concentração média no fígado de galinhas foi 105,02mg/ kg. Concluiu-se que a fonte de contaminação por chumbo nesses bezerros e galinhas eram placas de bateria de caminhão, aos quais os animais tiveram acesso na pastagem.
