Leptin acts as a potent inhibitory factor against obesity by regulating energy expenditure, food intake, and adiposity. The obese diabetic db/db mouse, which has defects in leptin receptor, displays enhanced neural responses and elevated behavioral preference to sweet stimuli. Here, we show the effects of leptin on the peripheral taste system. An administration of leptin into lean mice suppressed responses of peripheral taste nerves (chorda tympani and glossopharyngeal) to sweet substances (sucrose and saccharin) without affecting responses to sour, salty, and bitter substances. Whole-cell patch-clamp recordings of activities of taste receptor cells isolated from circumvallate papillae (innervated by the glossopharyngeal nerve) demonstrated that leptin activated outward K+ currents, which resulted in hyperpolarization of taste cells. The db/db mouse with impaired leptin receptors showed no such leptin suppression. Taste tissue (circumvallate papilla) of lean mice expressed leptin-receptor mRNA and some of the taste cells exhibited immunoreactivities to antibodies of the leptin receptor. Taken together, these observations suggest that the taste organ is a peripheral target for leptin, and that leptin may be a sweet-sensing modulator (suppressor) that may take part in regulation of food intake. Defects in this leptin suppression system in db/db mice may lead to their enhanced peripheral neural responses and enhanced behavioral preferences for sweet substances.

Two novel type I ribosome-inactivating proteins (RIPs) were found in the storage roots of Mirabilis expansa, an underutilized Andean root crop. The two RIPs, named ME1 and ME2, were purified to homogeneity by ammonium sulfate precipitation, cation-exchange perfusion chromatography, and C4 reverse-phase chromatography. The two proteins were found to be similar in size (27 and 27.5 kD) by sodium dodecyl sulfate-polyacrylamide gel electrophoresis, and their isoelectric points were determined to be greater than pH 10.0. Amino acid N-terminal sequencing revealed that both ME1 and ME2 had conserved residues characteristic of RIPs. Amino acid composition and western-blot analysis further suggested a structural similarity between ME1 and ME2. ME2 showed high similarity to the Mirabilis jalapa antiviral protein, a type I RIP. Depurination of yeast 26S rRNA by ME1 and ME2 demonstrated their ribosome-inactivating activity. Because these two proteins were isolated from roots, their antimicrobial activity was tested against root-rot microorganisms, among others. ME1 and ME2 were active against several fungi, including Pythium irregulare, Fusarium oxysporum solani, Alternaria solani, Trichoderma reesei, and Trichoderma harzianum, and an additive antifungal effect of ME1 and ME2 was observed. Antibacterial activity of both ME1 and ME2 was observed against Pseudomonas syringae, Agrobacterium tumefaciens, Agrobacterium radiobacter, and others.

The tobacco N and Arabidopsis RPS2 genes, among several recently cloned disease-resistance genes, share highly conserved structure, a nucleotide-binding site (NBS). Using degenerate oligonucleotide primers for the NBS region of N and RPS2, we have amplified and cloned the NBS sequences from soybean. Each of these PCR-derived NBS clones detected low-or moderate-copy soybean DNA sequences and belongs to 1 of 11 different classes. Sequence analysis showed that all PCR clones encode three motifs (P-loop, kinase-2, and kinase-3a) of NBS nearly identical to those in N and RPS2. The intervening region between P-loop and kinase-3a of the 11 classes has high (26% average) amino acid sequence similarity to the N gene although not as high (19% average) to RPS2. These 11 classes represent a superfamily of NBS-containing soybean genes that are homologous to N and RPS2. Each class or subfamily was assessed for its positional association with known soybean disease-resistance genes through near-isogenic line assays, followed by linkage analysis in F2 populations using restriction fragment length polymorphisms. Five of the 11 subfamilies have thus far been mapped to the vicinity of known soybean genes for resistance to potyviruses (Rsv1 and Rpv), Phytophthora root rot (Rps1, Rps2, and Rps3), and powdery mildew (rmd). The conserved N- or RPS2-homologous NBS sequences and their positional associations with mapped soybean-resistance genes suggest that a number of the soybean disease-resistance genes may belong to this superfamily. The candidate subfamilies of NBS-containing genes identified by genetic mapping should greatly facilitate the molecular cloning of disease-resistance genes.

Os fungos do gênero Metarhizium são entomopatogênicos e ainda apresentam relações endofíticas e podem viver saprofiticamente no solo. A associação desses microrganismos com insetos é bem conhecida, mas as interações diretas com as plantas ainda são incipientes. Objetivou-se com este estudo, determinar a capacidade de colonização endofítica em raízes de cana-de-açúcar (Saccharum spp.) de M. robertsii, M. anisopliae e três linhagens brasileiras recém-descobertas, bem como revelar o potencial destes fungos como antagonistas a dois fungos fitopatogênicos responsáveis pela podridão-vermelha (Fusarium moniliforme e Colletotrichum falcatum) e no controle de duas importantes pragas, a broca da cana-de-açúcar, Diatraea saccharalis e o nematóide-das-galhas, Meloidogyne javanica. Foram utilizados 24 isolados de Metarhizium spp. nos experimentos de promoção de crescimento de plantas após a inoculação em gemas de cana-de-açúcar em condições de casa-de-vegetação e campo. O efeito da inoculação do fungo em mudas de cana-de-açúcar foi investigado quanto a mortalidade e redução no desenvolvimento de D. saccharalis e redução nas populações de M. javanica. Experimentos de inibição in vitro em cultivo pareado foram conduzidos com os fungos F. moniliforme e C. falcatum e Trichoderma harzianum. Além disto, foram realizados testes qualitativos \"in vitro\" para avaliar a capacidade desses fungos em produzir β-1,3-Glucanase e Sideróforos. A inoculação de Metarhizium de todas as espécies testadas promoveu o crescimento de parte aérea, peso úmido e seco de raízes em relação ao controle (água). Enquanto o tratamento de gemas rotineiramente usado na usina onde se realizou a produção de mudas pré-brotadas com aplicação do fungicida Comet® + o fertilizante Biozyme TF resultou em pegamento de 32% e 59,6%, apenas com o uso de isolados de Metarhizium spp. estes valores foram de até 50% e 86,4%, nos dois experimentos em campo, respectivamente. As mortalidades observadas em lagartas de D. saccharalis que se alimentaram das plantas inoculadas com isolados de Metarhizium spp. atingiram valores de até 80%, sendo a mortalidade confirmada pela esporulação do fungo de até 55%. As lagartas sobreviventes apresentaram menor peso do que aquelas do controle não tratado. A inoculação de Metarhizium spp. em mudas de cana-de-açúcar conferiu proteção a M. javanica, resultando em uma densidade de massa de ovos por grama de raiz nas mudas inoculadas com o fungo até 59,8% menor no primeiro experimento e 64,1% menor no segundo em relação as mudas sem inoculação. Os ensaios de antagonismo mostraram que, todos os isolados de Metarhizium spp. apresentaram alguma forma de inibição tanto para F. moniliforme como para C. falcatum. Dos 24 isolados testados, 20,8% produziram a enzima hidrolítica, β-1,3-glucanase, o que pode estar associado a capacidade de inibição dos fungos fitopatogênicos. Constatou-se que 8 (33,3%) dos isolados produziram sideróforos e sugere que esses fungos apresentam mecanismos de disponibilização do ferro. O conhecimento gerado com este estudo poderá subsidiar novas estratégias de utilização de Metarhizium spp. em campo na produção de MPB de cana-de-açúcar.

The EU’s October summit was dominated by one issue; the migration and refugee crisis, with EU leaders intent on putting on a public display of unity after weeks of bitter arguments, and concentrating on fire-fighting and immediate measures to tackle the most pressing reasons for, and impacts of, the crisis. Longer-term measures to address some of the root causes of increased migratory flows, support for the integration of newly arrived refugees or the introduction of new channels of legal migration, were not discussed. The Summit also spent little time on two issues that had originally been expected to be a key part of the agenda: the forthcoming British referendum on EU membership, where irritation with the slow pace of talks and British vagueness about its demands were in evidence; and the governance of Economic and Monetary Union (EMU), where EU leaders missed another opportunity for a thorough debate about future perspectives on the basis of the ‘Five Presidents’ Report’.

After many meetings and long hours of negotiations, the overwhelming feeling when a deal between the EU and Turkey was struck, was one of “mission accomplished”! Faced with an unprecedented crisis and forced to appease increasingly hostile public opinions back home, EU leaders had only one objective in mind: reducing the number of migrants arriving in the EU so that order can return in the framework of EU rules. However, a closer look at the Summit Conclusions and the EU-Turkey statement leaves a bitter taste, according to Yves Pascouau. In this Commentary, he questions the feasibility of the final EU-Turkey deal, saying that it creates more problems than it solves: besides the obvious legal and practical issues, it is far from certain which member states will be willing to do their part, or whether or not the EU can come up with a strategic vision on human mobility for the future.

Die Diskussionen um die kantonalen Wahlrechtsreformen, den doppelten Pukelsheim und die Höhe von Wahlhürden halten an. - In sechs Kantonen fanden 2014 Gesamterneuerungswahlen für die Parlamente statt. Verliererin ist die BDP, die in Bern elf Sitze verliert; per Saldo legt die GLP um elf und die SVP um zehn Sitze zu. Die FDP verliert insgesamt acht und die CVP sechs Sitze. Die GP kann sich erholen und die SP stagniert. - Die in sechs Kantonen geführten Angriffe der SVP auf Regierungsmandate scheitern mit Ausnahme des Kantons Glarus; in Neuenburg verliert die Volkspartei ihren 2013 eroberten Sitz nach nur einem Jahr wieder an die FDP. - Im Kanton Glarus verliert die SP ihren seit 1942 gehaltenen Regierungssitz an die SVP. - In Bern wird die rot-grüne Regierungsmehrheit nur dank des Jurasitzes verteidigt. - In Nidwalden und in Glarus werden Amtierende abgewählt; in Nidwalden bleibt die Regierungszusammensetzung aber bestehen. - Im Kanton Glarus müssen beide verstorbenen Ständeräte ersetzt werden; die FDP und die SVP können ihre Sitze verteidigen.

A stable isotope record from the eastern Weddell Sea from 69°S is presented. For the first time, a 250,000-yr record from the Southern Ocean can be correlated in detail to the global isotope stratigraphy. Together with magnetostratigraphic, sedimentological and micropalaeontological data, the stratigraphic control of this record can be extended back to 910,000 yrs B.P. A time scale is constructed by linear interpolation between confirmed stratigraphic data points. The benthic d18O record (Epistominella exigua) reflects global continental ice volume changes during the Brunhes and late Matuyama chrons, whereas the planktonic isotopic record (Neogloboquadrina pachyderma) may be influenced by a meltwater lid caused by the nearby Antarctic ice shelf and icebergs. The worldwide climatic improvement during deglaciations is documented in the eastern Weddell Sea by an increase in production of siliceous plankton followed, with a time lag of approximately 10,000 yrs, by planktonic foraminifera production. Peak values in the difference between planktonic and benthic d13C records, which are 0.5 per mil higher during warm climatic periods than during times with expanded continental ice sheets, also suggest increased surface productivity during interglacials in the Southern Ocean.
