Since new technologies based on solid phase assays (SPA) have been routinely incorporated in the transplant immunology laboratory, the presence of pretransplantation donor-specific antibodies (DSA) against human leukocyte antigen (HLA) molecules has generally been considered as a risk factor for acute rejection (AR) and, in particular, for acute humoral rejection (AHR). We retrospectively studied 113 kidney transplant recipients who had negative prospective T-cell and B-cell complement-dependent cytotoxicity (CDC) crossmatches at the time of transplant. Pretransplantation sera were screened for the presence of circulating anti-HLA antibody and DSA by using highly sensitive and HLA-specific Luminex assay, and the results were correlated with AR and AHR posttransplantation. We found that approximately half of our patient population (55/113, 48.7%) had circulating anti-HLA antibody pretransplantation. Of 113 patients, 11 (9.7%) had HLA-DSA. Of 11 rejection episodes post-transplant, only two patients had pretransplantation DSA, of whom one had a severe AHR (C4d positive). One-year allograft survival was similar between the pretransplantation DSA-positive and -negative groups. Number, class, and intensity of pretransplantation DSA, as well as presensitizing events, could not predict AR. We conclude that, based on the presence of pretransplantation DSA, post-transplantation acute rejections episodes could not have been predicted. The only AHR episode occurred in a recipient with pretransplantation DSA. More work should be performed to better delineate the precise clinical significance of detecting low titers of DSA before transplantation.

To identify genetic loci influencing central obesity and fat distribution, we performed a meta-analysis of 16 genome-wide association studies (GWAS, N = 38,580) informative for adult waist circumference (WC) and waist-hip ratio (WHR). We selected 26 SNPs for follow-up, for which the evidence of association with measures of central adiposity (WC and/or WHR) was strong and disproportionate to that for overall adiposity or height. Follow-up studies in a maximum of 70,689 individuals identified two loci strongly associated with measures of central adiposity; these map near TFAP2B (WC, P = 1.9x10(-11)) and MSRA (WC, P = 8.9x10(-9)). A third locus, near LYPLAL1, was associated with WHR in women only (P = 2.6x10(-8)). The variants near TFAP2B appear to influence central adiposity through an effect on overall obesity/fat-mass, whereas LYPLAL1 displays a strong female-only association with fat distribution. By focusing on anthropometric measures of central obesity and fat distribution, we have identified three loci implicated in the regulation of human adiposity.

The northeastern portion of the Mont Blanc massif in western Switzerland is predominantly comprised of the granitic rocks of the Mont Blanc intrusive suit, and the Mont Blanc basement gneisses. Within these metamorphic rocks are a variety of sub-economic Fe skarns. The mineral assemblages and fluid inclusions from these rocks have been used to derive age, pressure, temperature and fluid composition constraints for two Variscan events. Metamorphic hornblendes within the assemblages from the basement amphibolites and iron sk:lms have been dated using Ar-40/Ar-39, and indicate that these metamorphic events have a minimum age of approximately 334 Ma. Garnet-hornblende-plagioclase thermobarometry and stable isotope data obtained from the basement amphibolites are consistent with metamorphic temperatures in the range 515 to 580 degrees C, and pressures ranging from 5 to 8 kbar. Garnet-hornblende-magnetite thermobarometry and fluid inclusion studies indicate that the iron skarns formed at slightly lower temperatures, ranging from 400 to 500 degrees C in the presence of saline fluids at formational pressures similar to those experienced by the basement amphibolites. Late Paleozoic minimum uplift rates and geothermal gradients calculated using these data and the presence of Ladinien ichnofossils are on the order of 0.32 mm/year and 20 degrees C/km respectively. These uplift rates and geothermal gradients differ from those obtained from the neighbouring Aiguilles Rouges massif and indicate that these two massifs experienced different metamorphic conditions during the Carboniferous and Permian periods. During the early to late Carboniferous period the relative depths of the two massifs were reversed with the Aiguilles Rouges being initially unroofed at a much greater rate than the Mont Blanc, but experiencing relatively slower uplift rates near the termination of the Variscan orogeny.

Com o objetivo de estabelecer parâmetros para avaliação de tolerância à seca, 15 genótipos de feijoeiro (Phaseolus vulgaris L.) foram monitorados durante a indução de estresse hídrico na pré-floração, por meio do potencial hídrico nas folhas (psif) e de outras variáveis. Os genótipos A 222, A 285, BAT 477, BAT 561 e IPA 7 apresentaram valores de psif superiores ao dos outros genótipos, durante o estresse, sugerindo um maior controle da abertura estomática. Os genótipos com tolerância à antracnose mostraram possuir este mecanismo de controle, enquanto as cultivares Carioca e Ouro Negro recuperaram-se mais rapidamente, após a reidratação. O potencial de fixação biológica do nitrogênio, avaliado pelo número e peso seco de nódulos, foi significativamente pouco afetado pelo estresse. Quanto aos parâmetros morfológicos, aqueles que sofreram maior efeito do déficit hídrico foram a área foliar e o peso seco da parte aérea. Nos componentes de rendimento, o número de vagens por planta foi afetado somente nos genótipos Xodó e CNF 0145, enquanto os genótipos Carioca, A 285, CNF 0145, BAT 477 e cv. 121 e a linhagem LM 30036 sofreram diminuição no peso de grãos por planta, devido à deficiência hídrica. Dos genótipos estudados, somente A 222 e IPA 7 mantiveram o psif mais alto do que os outros e não sofreram redução significativa na área foliar e no peso de grãos, apesar de o primeiro genótipo apresentar baixa produção por planta, em condições de estresse.

The effect of heterogeneous environments upon the dynamics of invasion and the eradication or control of invasive species is poorly understood, although it is a major challenge for biodiversity conservation. Here, we first investigate how the probability and time for invasion are affected by spatial heterogeneity. Then, we study the effect of control program strategies (e.g. species specificity, spatial scale of action, detection and eradication efficiency) on the success and time of eradication. We find that heterogeneity increases both the invasion probability and the time to invasion. Heterogeneity also reduces the probability of eradication but does not change the time taken for successful eradication. We confirm that early detection of invasive species reduces the time until eradication, but we also demonstrate that this is true only if the local control action is sufficiently efficient. The criterion of removal efficiency is even more important for an eradication program than simply ensuring control effort when the invasive species is not abundant.

Background/Purpose: Patient-reported outcomes (PROs) in patients with rheumatoid arthritis (RA) are critical in evaluating RA treatment effects on function and health-related quality of life (HR-QoL). Significant improvement in PROs has been reported in RA studies of biologic agents, including etanercept (ETN), but most studies have been conducted in patients with established disease. In addition to assessing treatment effects in early RA, there is interest in therapeutic strategies that allow dose reduction or withdrawal of biologic therapy (biologic-free) after induction of response. The PRIZE trial is an ongoing, 3-period study to evaluate the efficacy of combined ETN and methotrexate (MTX) therapy in patients with early, moderate-to-severe RA and to assess whether efficacy (remission) can be maintained with ETN dose reduction or biologic-free (Period 2) or drug-free (Period 3). Herein we report PROs associated with ETN 50 mg QW plus MTX (ETN50/MTX) therapy administered for 52 wks in Period 1 (induction) of the PRIZE trial. Methods: In Period 1, MTX- and biologic-naı‥ve patients with early, active RA (symptom onset 12 mo from enrollment; DAS28 _3.2) received open-label ETN50/MTX for 52 wks. The starting dose of MTX was 10 mg QW; at the discretion of the investigator, titration was permitted up to a maximum of 25 mg QW to achieve remission. Corticosteroid boosts were administered to patients not achieving low disease state at wks 13 and 26, unless contraindicated or not tolerated. PROs were assessed using the Health Assessment Questionnaire (HAQ) total score; Patient Acceptable Symptom State (PASS); EuroQol-5 Dimensions (EQ-5D) total index; Short Form Health Survey (SF-36); Functional Assessment of Chronic Illness Therapy (FACIT)-Fatigue; Work Instability Scale for Rheumatoid Arthritis (RAWIS); and Work Productivity and Activity Impairment Questionnaire: Rheumatoid Arthritis (WPAI:RA). Results: A total of 306 patients received treatment in Period 1 (mITT population); 222 (73%) patients completed the period. The majority of patients were female (70%), with a mean age of 50 y, mean DAS28 of 6.0 (median, 6.0), and duration of disease symptoms from onset of 6.5 months (median, 6.3 mo). Significant and clinically meaningful improvements in PROs, including in HAQ, EQ-5D, SF-36, and FACIT-Fatigue, were demonstrated with ETN50/MTX therapy from baseline to the final on therapy visit (Table; P_0.0001). Similar improvements were observed in all dimensions of RA-WIS and WPAI:RA (Table; P_0.0001). Conclusion: Combination therapy with ETN50/MTX for 52 wks in patients with _12 mo of symptomatic, active RA resulted in significant, clinically important improvements in measures of physical function, including normal HAQ (66.6% of patients), HR-QoL, fatigue, and work productivity. These outcomes are consistent with those reported in prior studies in patients with more established disease.

Regulation of renal Na(+) transport is essential for controlling blood pressure, as well as Na(+) and K(+) homeostasis. Aldosterone stimulates Na(+) reabsorption by the Na(+)-Cl(-) cotransporter (NCC) in the distal convoluted tubule (DCT) and by the epithelial Na(+) channel (ENaC) in the late DCT, connecting tubule, and collecting duct. Aldosterone increases ENaC expression by inhibiting the channel's ubiquitylation and degradation; aldosterone promotes serum-glucocorticoid-regulated kinase SGK1-mediated phosphorylation of the ubiquitin-protein ligase Nedd4-2 on serine 328, which prevents the Nedd4-2/ENaC interaction. It is important to note that aldosterone increases NCC protein expression by an unknown post-translational mechanism. Here, we present evidence that Nedd4-2 coimmunoprecipitated with NCC and stimulated NCC ubiquitylation at the surface of transfected HEK293 cells. In Xenopus laevis oocytes, coexpression of NCC with wild-type Nedd4-2, but not its catalytically inactive mutant, strongly decreased NCC activity and surface expression. SGK1 prevented this inhibition in a kinase-dependent manner. Furthermore, deficiency of Nedd4-2 in the renal tubules of mice and in cultured mDCT(15) cells upregulated NCC. In contrast to ENaC, Nedd4-2-mediated inhibition of NCC did not require the PY-like motif of NCC. Moreover, the mutation of Nedd4-2 at either serine 328 or 222 did not affect SGK1 action, and mutation at both sites enhanced Nedd4-2 activity and abolished SGK1-dependent inhibition. Taken together, these results suggest that aldosterone modulates NCC protein expression via a pathway involving SGK1 and Nedd4-2 and provides an explanation for the well-known aldosterone-induced increase in NCC protein expression.

Foram estimadas repetibilidades (r) e herdabilidades (h²) de características reprodutivas de três populações de galinhas para corte do Setor de Aves da ESALQ/USP Piracicaba, SP. Cento e quinze galinhas foram inseminadas segundo um esquema hierárquico, com sêmen de 23 galos amostrados das populações I, II e III no experimento E1 e de outros 23 galos no experimento E2. Foram utilizadas quatro incubações, ou blocos, em cada experimento. Estimativas de r e h² foram obtidas dentro de experimento, pela análise de variância de médias de galinha em cada incubação. As estimativas de r da produção de ovos (POST) foram significativas (P<0,01) e iguais a 0,19±0,05 e 0,32±0,04 nos experimentos E1 e E2, respectivamente. Quanto a eclodibilidade (ECLOD), as h² estimadas de componentes de variância de galos foram 0,14±0,14 no E1 (P>0,05) e 0,26±0,17 no E2 (P<0,05). No tocante a nascimento de pintos (NASC), as h² estimadas (P<0,05) foram 0,18±0,15 (E1) e 0,26±0,17 (E2). No entanto, h² estimadas com base em componentes de fêmea foram significativas (P<0,01) e maiores: no tocante a ECLOD, as estimativas foram 0,77±0,09 e 0,48±0,20 no experimento E1 e E2, respectivamente, e 0,58±0,21 e 0,48±0,20 quanto a nascimento (NASC). Os resultados confirmam que ECLOD e NASC apresentam variabilidade genética aditiva baixa.

Régner, Escribe, and Dupeyrat (2007) recently demonstrated that not only performance-approach and performance-avoidance goals (respectively, the desire to outperform others and not to be outperformed by others) but also mastery goals (the desire to acquire knowledge) were related to social comparison orientation (SCO, the tendency to search for social comparison information). In the present article, the possibility of a link between mastery goals and social comparison that depends on the level of performance-approach goals-a possibility supported by a multiple-goal perspective-was tested by examining the interaction effect between mastery and performance-approach goals. This is an important endeavor, as educational settings are rarely free from performance-approach goals, even when mastery goals are promoted. In Study 1, we tested self-set achievement goals (mastery, performance-approach, and performance-avoidance goals) as predictors of SCO; the interaction between mastery goals and performance-approach goals indicated that the higher the performance-approach goal endorsement, the stronger the link between mastery goals and SCO. In Study 2, we manipulated goal conditions; mastery goals predicted interest in social comparison in the performance-approach goal condition only. The freeze-thaw resistance of concretes was studied. Nine concrete mixes, made with five cements and cement-Class C fly ash combinations, were exposed to freeze-thaw cycling following 110 to 222 days of moist curing. Prior to the freeze-thaw cycling, the specimens were examined by a low-vacuum scanning electron microscope (SEM) for their microstructure. The influence of a wet/dry treatment was also studied. Infilling of ettringite in entrained air voids was observed in the concretes tested. The extent of the infilling depends on the period of moist curing as well as the wet/dry treatment. The concretes with 15% Class C fly ash replacement show more infilling in their air voids. It was found that the influence of the infilling on the freeze-thaw durability relates to the air spacing factor. The greater the spacing factor, the more expansion under the freeze-thaw cycling. The infilling seems to decrease effective air content and to increase effective spacing factor. The infilling also implies that the filled air voids are water-accessible. These might lead to concrete more vulnerable to the freeze-thaw attack. By combining the above results with field observations, one may conclude that the freeze-thaw damage is a factor related to premature deterioration of portland cement concrete pavements in Iowa.

Objetivou-se avaliar a influência de diferentes densidades e arranjos de plantio sobre os componentes de produção e a relação ráquis/cacho de bananeiras 'Nanicão', estabelecidas em Piracicaba, SP. Foram estudadas quatro densidades: 1.333, 1.666, 2.222 e 3.333 plantas ha-1, e dois arranjos de plantio: retângulo e triângulo (quincôncio). Independentemente do arranjo utilizado, o aumento da densidade elevou a produção de frutos de 37,24 para 75,83 t ha-1 e reduziu a massa do cacho de 30,30 para 24,79 kg, por causa da redução na massa do fruto. A porcentagem da massa da ráquis em relação à massa do cacho permaneceu em torno de 8%, independentemente da densidade e arranjo. Na maior densidade (3.333 plantas ha-1) podem ser exportadas do bananal até 6,80 t ha-1 de ráquis,material que pode ser aproveitado como matéria-prima para a fabricação de papel.

O objetivo do presente trabalho foi verificar o efeito de diferentes densidades de plantio e sistemas de espaçamento sobre a produção da bananeira 'Nanicão', avaliando-se os primeiros quatro ciclos. Quatro densidades (3.333, 2.222, 1.666 e 1.333 plantas ha-1) e dois sistemas de espaçamento (retângulo e triângulo) foram testados para as condições de Piracicaba,SP. O aumento da densidade de 1.333 para 3.333 plantas ha-1 diminuiu a massa do cacho em 15% a 20%, em decorrência do menor número de frutos por cacho, massa e tamanho do fruto. A produção foi sempre maior com o aumento da densidade, porém não ocorreu o mesmo quanto à produtividade. Até o terceiro ciclo, a produtividade das plantas no tratamento de maior densidade superou a das demais. No quarto ciclo, não houve diferença de produtividade entre as plantas, nas densidades testadas, em razão do aumento na duração do ciclo de produção do plantio mais denso. Comparado com o retângulo, o sistema de espaçamento em triângulo promoveu maior massa do cacho no primeiro ciclo e produtividade levemente superior ao longo dos quatro ciclos.

Context: Cross-cultural clinical competence (CCC) requires a mixtureof "knowledge" (K), "attitude" (A) and "skills" (S), in order to develop theability to give quality care to patients of different cultures. Theseattributes allow, while providing medical care, consideration of thepatient's medical, social, cultural and language needs. The LausanneUniversity Medical Policlinic (PMU) provides approximately 30000consultations per year to migrant patients and over the past five yearshas implemented a training course on CCC that focuses on trialogue,stereotypes and administrative procedures for the healthcare ofmigrants.Method: A quantitative survey of 18 new residents, was carried outusing a validated questionnaire, the "Multicultural AssessmentQuestionnaire" (the MAQ, 16 questions on K, A and S) to evaluate theimpact of CCC training. The questionnaire was distributed before theCCC course (J-1), three days after (J+3) and three months later (J+90).A descriptive univariate analysis of the difference in MAQ scoresbetween the times J-1 - J+3 and J-1 - J+90 was made. Three FocusGroups were conducted, at three months, to explore residents' thoughtsabout the course.Results: A significant increase was observed in global performancedeclared by residents. Following the intervention, the score of the MAQincreased from 31.4 points to 38.0 points at three days (p = 0.004) andto 37.7 points at three months (p = 0.003). This increase was mostnoticeable in the field of acquiring K: total score J-1: 118, J+3: 189,J+90: 190 (difference J-1 - J+3 and J-1 - J+90: p <0.005). There was nosignificant difference in acquiring A (J-1: 222, J+3: 228, J+90: 229), andS increases in a significant way at first (J-1: 222, J+3: 265, J-1 - J+3:p = 0.035), then comes back to the start value (J+90: 217). The residentswere interested by the course which they felt provides useful informationfor clinical practice. They had a great number of expectations in varyingfields (medical anthropology, cultural differences, epidemiology, etc.),hoping a "ready-made" solution for the approach of migrant patients.Conclusions: A unique training of CCC at the post-graduate level,upgraded K, and to a lesser extent A and S, for these 18 residents. Theywere interest and they had many expectations. Subsequent coursesshould consolidate these acquisitions. Future study should demonstratethe impact on patients' clinical outcome.
