Silver Creek is a warm water stream resource located in one of the most intensely cropped portions of Clayton County. The stream has been included on Iowa’s 303(d) list of impaired waters since 2002. Aquatic life, which should be present in Silver Creek, isn’t there. According to the Draft Total Maximum Daily Load (TMDL) for Silver Creek, the primary nonpoint pollution sources are soil erosion from agricultural land uses and direct deposition of ammonia by livestock with access to the stream. The Clayton Soil & Water Conservation District has begun efforts to remove Silver Creek from the impaired waters list. The District has promoted stream corridor and sinkhole protection, and the installation of buffer practices along Silver Creek and its tributaries. Conservation practices have been targeted to crop fields to reduce sediment delivery to the stream. A series of news articles, newsletters, and field days have been utilized to increase public understanding of water quality issues. Landowner interest has outweighed available cost share resources. Additional financial support will allow the project to build upon its early successes, to further address the identified impairments, and to respond to a long list of landowners that are interested in conservation work on their farms.

BACKGROUND: 2013 AHA/ACC guidelines on the treatment of cholesterol advised to tailor high-intensity statin after ACS, while previous ATP-III recommended titration of statin to reach low-density lipoprotein cholesterol (LDL-C) targets. We simulated the impact of this change of paradigm on the achievement of recommended targets. METHODS: Among a prospective cohort study of consecutive patients hospitalized for ACS from 2009 to 2012 at four Swiss university hospitals, we analyzed 1602 patients who survived one year after recruitment. Targets based on the previous guidelines approach was defined as (1) achievement of LDL-C target < 1.8 mmol/l, (2) reduction of LDL-C ≥ 50% or (3) intensification of statin in patients who did not reach LDL-C targets. Targets based on the 2013 AHA/ACC guidelines approach was defined as the maximization of statin therapy at high-intensity in patients aged ≤75 years and moderate- or high-intensity statin in patients >75 years. RESULTS: 1578 (99%) patients were prescribed statin at discharge, with 1120 (70%) at high-intensity. 1507 patients (94%) reported taking statin at one year, with 909 (57%) at high-intensity. Among 482 patients discharged with sub-maximal statin, intensification of statin was only observed in 109 patients (23%). 773 (47%) patients reached the previous LDL-C targets, while 1014 (63%) reached the 2013 AHA/ACC guidelines targetsone year after ACS (p value < 0.001). CONCLUSION: The application of the new 2013 AHA/ACC guidelines criteria would substantially increase the proportion of patients achieving recommended lipid targets one year after ACS. Streptococcus tigurinus is responsible for systemic infections in humans including infective endocarditis. We investigated whether the invasive trait of S. tigurinus in humans correlated with an increased ability to induce IE in rats. Rats with catheter-induced aortic vegetations were inoculated with 10⁴ CFU/ml of either of four S. tigurinus strains AZ_3a(T), AZ_4a, AZ_8 and AZ_14, isolated from patients with infective endocarditis or with the well known IE pathogen Streptococcus gordonii (Challis). Aortic infection was assessed after 24 h. S. tigurinus AZ_3a(T), AZ_4a and AZ_14 produced endocarditis in ≥80% of rats whereas S. gordonii produced endocarditis in only 33% of animals (P<0.05). S. tigurinus AZ_8 caused vegetation infection in 56% of the animals. The capacity of S. tigurinus to induce aortic infection was not related to their ability to bind extracellular matrix proteins (fibrinogen, fibronectin or collagen) or to trigger platelet aggregation. However, all S. tigurinus isolates showed an enhanced resistance to phagocytosis by macrophages and two of them had an increased ability to enter endothelial cells, key attributes of invasive streptococcal species.

The Muchakinock Creek Watershed Project began in February of 2005 to treat upland soil erosion in the creek that has lead to a 303(d) impairment. The Mahaska SWCD is currently administering this cost-share program to promote terraces, basins, and grade stabilization structures. The District is seeking funding from WIRB to treat specific abandoned mine lands in the Muchakinock Creek Watershed. These areas contribute sediment to the creek at levels second only to agricultural lands as well as acid mine drainage from open pits mines that have been left to decay across the county. The WIRB funding would be used to compliment Federal Abandoned Mine Land (AML) funding in the reclamation of these areas.

O objetivo deste trabalho foi determinar a influência do reprodutor nas características de crescimento de 303 cordeiros para abate, dos grupos genéticos ½ sangue Somalis Brasileira x Sem Raça Definida (SRD) e ½ sangue Santa Inês x SRD. Os animais foram pesados a intervalos de 14 dias, até o desmame na época seca, e até 140 dias de idade na época chuvosa. O grupo genético e o ano de nascimento não influenciaram (P>0,05) nenhuma das características avaliadas. O mês de nascimento influenciou (P<0,05) os pesos ao nascer, aos 56 dias de idade e ao desmame, e o ganho em peso até o desmame. Fevereiro foi o melhor mês para nascimento de cordeiros. Animais de nascimentos simples pesaram mais e ganharam mais peso (P<0,05) que os de nascimentos múltiplos, em todas as idades e intervalos de ganhos estudados, exceto no período compreendido entre o desmame e a idade de 140 dias. Os machos pesaram mais e ganharam mais peso que as fêmeas, tanto no período de cria como no pós-desmame. Os dois grupos genéticos avaliados são semelhantes quanto ao peso corporal e ao ganho em peso, tanto no período pré-desmame como no pós-desmame.

Resultados experimentais seguros exigem um plano experimental apropriado, obtido através da utilização de adequado tamanho e forma de parcelas, número de repetições e de amostras. O objetivo deste trabalho foi avaliar a influência de diferentes bases genéticas de milho, híbrido simples (DAS 9560), duplo (AG 6018), triplo (AG 303) e variedade cultivada (Pampa), sobre a estimativa do tamanho de parcela experimental e do número de repetições. O delineamento experimental foi de blocos ao acaso com quatro repetições e amostragem nas parcelas. As parcelas experimentais foram constituídas por oito fileiras com 12 metros de comprimento, formando 96 unidades básicas. O tamanho de parcela e o número de repetições foram estimados por meio dos métodos de Lessman & Atkins e de Hatheway. O tamanho ótimo de parcela variou entre 4,5 m² (híbrido simples) e 7,2 m² (híbrido duplo). A heterogeneidade do solo e a variabilidade genética influenciam no tamanho ótimo de parcela e número de repetições necessárias.

Pluripotency in human embryonic stem cells (hESCs) and induced pluripotent stem cells (iPSCs) is regulated by three transcription factors-OCT3/4, SOX2, and NANOG. To fully exploit the therapeutic potential of these cells it is essential to have a good mechanistic understanding of the maintenance of self-renewal and pluripotency. In this study, we demonstrate a powerful systems biology approach in which we first expand literature-based network encompassing the core regulators of pluripotency by assessing the behavior of genes targeted by perturbation experiments. We focused our attention on highly regulated genes encoding cell surface and secreted proteins as these can be more easily manipulated by the use of inhibitors or recombinant proteins. Qualitative modeling based on combining boolean networks and in silico perturbation experiments were employed to identify novel pluripotency-regulating genes. We validated Interleukin-11 (IL-11) and demonstrate that this cytokine is a novel pluripotency-associated factor capable of supporting self-renewal in the absence of exogenously added bFGF in culture. To date, the various protocols for hESCs maintenance require supplementation with bFGF to activate the Activin/Nodal branch of the TGFβ signaling pathway. Additional evidence supporting our findings is that IL-11 belongs to the same protein family as LIF, which is known to be necessary for maintaining pluripotency in mouse but not in human ESCs. These cytokines operate through the same gp130 receptor which interacts with Janus kinases. Our finding might explain why mESCs are in a more naïve cell state compared to hESCs and how to convert primed hESCs back to the naïve state. Taken together, our integrative modeling approach has identified novel genes as putative candidates to be incorporated into the expansion of the current gene regulatory network responsible for inducing and maintaining pluripotency.
