Les modifications post-transcriptionnelles de l’ARN messager (ARNm), comme l’épissage alternatif, jouent un rôle important dans la régulation du développement embryonnaire, de la fonction cellulaire et de l’immunité. De nouvelles évidences révèlent que l’épissage alternatif serait également impliqué dans la régulation de la maturation et de l’activation des cellules du système hématopoïétique. Le facteur hnRNP L a été identifié comme étant le principal régulateur de l’épissage alternatif du gène codant pour le récepteur CD45 in vitro. Le récepteur CD45 est une tyrosine phosphatase exprimée par toutes les cellules du système hématopoïétique qui contrôle le développement et l’activation des lymphocytes T. Dans un premier temps, nous avons étudié la fonction du facteur hnRNP L dans le développement des lymphocytes T et dans l’épissage de l’ARNm de CD45 in vivo en utilisant des souris dont le gène de hnRNP L a été supprimé spécifiquement dans les cellules T. La délétion de hnRNP L dans les thymocytes résulte en une expression aberrante des différents isoformes de CD45 avec une prédominance de l'isoforme CD45RA qui est généralement absent dans le thymus. Une conséquence de la délétion de hnRNP L est une diminution de la cellularité du thymus causée par un blocage partiel du développement des cellules pré-T au stade DN4. Cette réduction du nombre de cellules dans le thymus n’est pas liée à une hausse de la mort cellulaire. Les thymocytes déficients pour hnRNP L démontrent plutôt une prolifération augmentée comparée aux thymocytes sauvages due à une hyper-activation des kinases Lck, Erk1/2 et Akt. De plus, la délétion de hnRNP L dans le thymus cause une perte des cellules T en périphérie. Les résultats des expériences in vitro suggèrent que cette perte est principalement due à un défaut de migration des thymocytes déficients pour hnRNP L du thymus vers la périphérie en réponse aux chimiokines. L’épissage alternatif de CD45 ne peut expliquer ce phénotype mais l’identification de cibles par RNA-Seq a révélé un rôle de hnRNP L dans la régulation de l’épissage alternatif de facteurs impliqués dans la polymérisation de l’actine. Dans un second temps, nous avons étudié le rôle de hnRNP L dans l’hématopoïèse en utilisant des souris dont la délétion de hnRNP L était spécifique aux cellules hématopoïétiques dans les foies fœtaux et la moelle osseuse. L’ablation de hnRNP L réduit le nombre de cellules progénitrices incluant les cellules progénitrices lymphocytaires (CLPs), myéloïdes (CMPs, GMPs) et mégakaryocytes-érythrocytaires (MEPs) et une perte des cellules hématopoïétiques matures. À l’opposé des cellules progénitrices multipotentes (MPPs) qui sont affectées en absence de hnRNP L, la population de cellules souches hématopoïétiques (HSCs) n’est pas réduite et prolifère plus que les cellules contrôles. Cependant, les HSCs n’exprimant pas hnRNP L sont positives pour l'Annexin V et expriment CD95 ce qui suggère une mort cellulaire prononcée. Comme pour les thymocytes, une analyse par RNA-Seq des foies fœtaux a révélé différents gènes cibles de hnRNP L appartenant aux catégories reliées à la mort cellulaire, la réponse aux dommages à l’ADN et à l’adhésion cellulaire qui peuvent tous expliquer le phénotype des cellules n’exprimant pas le gène hnRNP L. Ces résultats suggèrent que hnRNP L et l’épissage alternatif sont essentiels pour maintenir le potentiel de différenciation des cellules souches hématopoïétiques et leur intégrité fonctionnelle. HnRNP L est aussi crucial pour le développement des cellules T par la régulation de l’épissage de CD45 ainsi que pour leur migration.

Le glaucome est la première cause de cécité irréversible à travers le monde. À présent il n’existe aucun remède au glaucome, et les thérapies adoptées sont souvent inadéquates. La perte de vision causée par le glaucome est due à la mort sélective des cellules rétiniennes ganglionnaires, les neurones qui envoient de l’information visuelle de la rétine au cerveau. Le mécanisme principal menant au dommage des cellules rétiniennes ganglionnaires lors du glaucome n’est pas bien compris, mais quelques responsables putatifs ont été proposés tels que l’excitotoxicité, le manque de neurotrophines, la compression mécanique, l’ischémie, les astrocytes réactifs et le stress oxidatif, parmis d’autres. Indépendamment de la cause, il est bien établi que la perte des cellules rétiniennes ganglionnaires lors du glaucome est causée par la mort cellulaire programmée apoptotique. Cependant, les mécanismes moléculaires précis qui déclenchent l’apoptose dans les cellules rétiniennes ganglionnaires adultes sont mal définis. Pour aborder ce point, j’ai avancé l’hypothèse centrale que l’identification de voies de signalisations moléculaires impliquées dans la mort apoptotique des cellules rétiniennes ganglionnaires offrirait des avenues thérapeutiques pour ralentir ou même prévenir la mort de celles-ci lors de neuropathies oculaires telles que le glaucome. Dans la première partie de ma thèse, j’ai caractérisé le rôle de la famille de protéines stimulatrices d’apoptose de p53 (ASPP), protéines régulatrices de la famille p53, dans la mort apoptotique des cellules rétiniennes ganglionnaires. p53 est un facteur de transcription nucléaire impliqué dans des fonctions cellulaires variant de la transcription à l’apoptose. Les membres de la famille ASPP, soit ASPP1, ASPP2 et iASPP, sont des protéines de liaison de p53 qui régulent l’apoptose. Pourtant, le rôle de la famille des ASPP dans la mort des cellules rétiniennes ganglionnaires est inconnu. ASPP1 et ASPP2 étant pro-apoptotiques, l’hypothèse de cette première étude est que la baisse ciblée de ASPP1 et ASPP2 promouvrait la survie des cellules rétiniennes ganglionnaires après une blessure du nerf optique. Nous avons utilisé un modèle expérimental bien caractérisé de mort apoptotique neuronale induite par axotomie du nerf optique chez le rat de type Sprague Dawley. Les résultats de cette étude (Wilson et al. Journal of Neuroscience, 2013) ont démontré que p53 est impliqué dans la mort apoptotique des cellules rétiniennes ganglionnaires, et qu’une baisse ciblée de ASPP1 et ASPP2 par acide ribonucléique d’interference promeut la survie des cellules rétiniennes ganglionnaires. Dans la deuxième partie de ma thèse, j’ai caractérisé le rôle d’iASPP, le membre anti-apoptotique de la famille des ASPP, dans la mort apoptotique des cellules rétiniennes ganglionnaires. L’hypothèse de cette seconde étude est que la surexpression d’iASPP promouvrait la survie des cellules rétiniennes ganglionnaires après axotomie. Mes résultats (Wilson et al. PLoS ONE, 2014) démontrent que le knockdown ciblé de iASPP exacerbe la mort apoptotique des cellules rétiniennes ganglionnaires, et que la surexpression de iASPP par virus adéno-associé promeut la survie des cellules rétiniennes ganglionnaires. En conclusion, les résultats présentés dans cette thèse contribuent à une meilleure compréhension des mécanismes régulateurs sous-jacents la perte de cellules rétiniennes ganglionnaires par apoptose et pourraient fournir des pistes pour la conception de nouvelles stratégies neuroprotectrices pour le traitement de maladies neurodégénératives telles que le glaucome.

The thesis deals with the preparation of chemical, optical, thermal and electrical characterization of five compounds, namely metal free naphthalocyanine, vanadyl napthalocyanine, zinc naphlocyanine, europium dinaphthalocyanine, and europium diphthalocyanine in the pristine and iodine-doped forms. Two important technological properties of these compounds have been investigated. The electrical properties are important in applications sensors and semiconductor lasers. Opto-thermal properties assume significance for optical imaging and data recording. The electrical properties were investigated by dc and ac techniques. This work has revealed some novel information on the conduction mechanism in five macrocyclic compounds and their iodine-doped forms. Also useful data on the thermal diffusivity of the target compounds have been obtained by optical techniques.

The research work has been in the area of compounding and characterization of rubbers for use in under water electro acoustic transducers. The study also covers specific material system such as encapsulation materials, baffle material, seal material, etc. Life prediction techniques of under water rubbers in general have been established with reference to more than one functional property. Ranges of passive materials, besides the active sensing material go into the construction of underwater electro acoustic transducers. Reliability of the transducer is critically dependent on these passive materials. Rubbers are a major class of passive materials. The present work concentrates on these materials. Conventional rubbers are inadequate to meet many of the stringent function specific requirements. There exists a large gap of information in the rubber technology of underwater rubbers, particularly relating to underwater electro acoustic transducers. This study is towards filling up the gaps of information in this crucial area. Water intake into rubber is considered as the single most important issue for the long-term performance of rubbers, especially Neoprene. In this study, the cause and effects of a range of parameters affecting the water absorption by diffusion and permeation have been investigated.

In this communication, we discuss the details of fabricating an off-line fibre optic sensor (FOS) based on evanescent wave absorption for detecting trace amounts of Fe3+ in water. Two types of FOS are developed; one type uses the unclad portion of a multimode silica fibre as the sensing region whereas the other employs the microbent portion of a multimode plastic fibre as the sensing region. Sensing is performed by measuring the absorption of the evanescent wave in a reagent medium surrounding the sensing region. To evaluate the relative merits of the two types of FOS in Fe3+ sensing, a comparative study of the sensors is made, which reveals the superiority of the latter in many respects, such as smaller sensing length, use of a double detection scheme (for detecting both core and cladding modes) and higher sensitivity of cladding mode detection at an intermediate range of concentration along with the added advantage that plastic fibres are inexpensive. A detection limit of 1 ppb is observed in both types of fibre and the range of detection can be as large as 1 ppb–50 ppm. All the measurements are carried out using a LabVIEW set-up.

This optical fibre based sensor works on the principle of the evanescent wave penetration of waveguide modes into the uncladded portion of a multimode fibre. The utility of this optical fibre sensor is demonstrated in the case of PLD of silver thin films obtained by a Q-switched Nd:YAG laser which is used to irradiate a silver target at the required conditions for the preparation of thin films. This paper describes the performance and characteristics of the sensor and shows how the device can be used as an effective tool for the monitoring of the deposition rate of silver thin films. The fibre optic sensor is very simple, inexpensive and highly sensitive compared with existing techniques for thin film deposition rate measurements

A fibre optic technique for detecting trace amounts of nitrite compounds in water is described. The off-line fibre optic sensor outlined here is based on evanescent field absorption in a test solution formed by the reaction of nitrite compounds in water with suitable chemical reagents. A short unclad portion of a plastic clad silica fibre acts as the sensing region. The experimental results clearly establish the usefulness of the present technique for detecting very low concentrations of the order of 1 ppb (parts per billion) of nitrite compounds with a large dynamic range of 1–1000 ppb. Such a high sensitivity enables the present device to be used for measuring the nitrite content in drinking water.

The design and development of a fibre optic evanescent wave refractometer for the detection of trace amounts of paraffin oil and palm oil in coconut oil is presented. This sensor is based on a side-polished plastic optical fibre. At the sensing region, the cladding and a small portion of the core are removed and the fibre nicely polished. The sensing region is fabricated in such a manner that it sits perfectly within a bent mould. This bending of the sensing region enhances its sensitivity. The oil mixture of different mix ratios is introduced into the sensing region and we observed a sharp decrease in the output intensity. The observed variation in the intensity is found to be linear and the detection limit is 2% (by volume) paraffin oil/palm oil in coconut oil. The resolution of this refractometric sensor is of the order of 10−3. Since coconut oil is consumed in large volumes as edible oil in south India, this fibre optic sensor finds great relevance for the detection of adulterants such as paraffin oil or palm oil which are readily miscible in coconut oil. The advantage of this type of sensor is that it is inexpensive and easy to set up. Another attraction of the side-polished fibre is that only a very small amount of analyte is needed and its response time is only 7 s.
