The study discusses an approach that allows simultaneous determination of boronic acid and its anhydride without the need for tedious physical separation of the mixture. The assignment of the proton spectra of monomer, dimer and trimer was achieved by combining utility of 1D and 2D experimental techniques including 2D DOSY. The differential intensities of NMR peaks and supplementary resonances were detected in low polar solvents, such as, chloroform, toluene and in a non-polar solvent benzene. A fascinating phenomenon is observed at lower temperature where there is a formation of aryl boronic acid with the disappearance of boraxine formation. (C) 2015 Elsevier B.V. All rights reserved.

Breast cancer is one of the leading cause of cancer related deaths in women and early detection is crucial for reducing mortality rates. In this paper, we present a novel and fully automated approach based on tissue transition analysis for lesion detection in breast ultrasound images. Every candidate pixel is classified as belonging to the lesion boundary, lesion interior or normal tissue based on its descriptor value. The tissue transitions are modeled using a Markov chain to estimate the likelihood of a candidate lesion region. Experimental evaluation on a clinical dataset of 135 images show that the proposed approach can achieve high sensitivity (95 %) with modest (3) false positives per image. The approach achieves very similar results (94 % for 3 false positives) on a completely different clinical dataset of 159 images without retraining, highlighting the robustness of the approach.

Metastasis is clinically the most challenging and lethal aspect of breast cancer. While animal-based xenograft models are expensive and time-consuming, conventional two-dimensional (2D) cell culture systems fail to mimic in vivo signaling. In this study we have developed a three-dimensional (3D) scaffold system that better mimics the topography and mechanical properties of the breast tumor, thus recreating the tumor microenvironment in vitro to study breast cancer metastasis. Porous poly(e-caprolactone) (PCL) scaffolds of modulus 7.0 +/- 0.5 kPa, comparable to that of breast tumor tissue were fabricated, on which MDA-MB-231 cells proliferated forming tumoroids. A comparative gene expression analysis revealed that cells growing in the scaffolds expressed increased levels of genes implicated in the three major events of metastasis, viz., initiation, progression, and the site-specific colonization compared to cells grown in conventional 2D tissue culture polystyrene (TCPS) dishes. The cells cultured in scaffolds showed increased invasiveness and sphere efficiency in vitro and increased lung metastasis in vivo. A global gene expression analysis revealed a significant increase in the expression of genes involved in cell cell and cell matrix interactions and tissue remodeling, cancer inflammation, and the PI3K/Akt, Wnt, NF-kappaB, and HIFI signaling pathways all of which are implicated in metastasis. Thus, culturing breast cancer cells in 3D scaffolds that mimic the in vivo tumor-like microenvironment enhances their metastatic potential. This system could serve as a comprehensive in vitro model to investigate the manifold mechanisms of breast cancer metastasis.

Macro-steatosis in deceased donor livers is increasingly prevalent and is associated with poor or non-function of the liver upon reperfusion. Current assessment of the extent of steatosis depends upon the macroscopic assessment of the liver by the surgeon and histological examination, if available. In this paper we demonstrate electrical and optical spectroscopy techniques which quantitatively characterize fatty infiltration in liver tissue. Optical spectroscopy showed a correlation coefficient of 0.85 in humans when referenced to clinical hematoxylin and eosin (H&E) sections in 20 human samples. With further development, an optical probe may provide a comprehensive measure of steatosis across the liver at the time of procurement.

The acoustic response of conventional mechanical oscillators, such as a piezoelectric crystal, is predominantly harmonic at modest amplitudes. However, here, we observe from the electrical response that significant motional anharmonicity is introduced in the presence of attached analyte. Experiments were conducted with streptavidin-coated polystyrene microbeads of various sizes attached to a quartz crystal resonator via specific and nonspecific molecular tethers in liquid. Quantitative analysis reveals that the deviation of odd Fourier harmonics of the response caused by introduction of microbeads as a function of oscillation amplitude presents a unique signature of the molecular tether. Hence, the described anharmonic detection technique (ADT) based on this function allows screening of biomolecules and provides an additional level of selectivity in receptor-based detection that is often associated with nonspecific interactions. We also propose methods to extract mechanical force-extension characteristics of the molecular tether and activation energy using this technique.

Microbe-Associated Molecular Patterns and virulence effectors are recognized by plants as a first step to mount a defence response against potential pathogens. This recognition involves a large family of extracellular membrane receptors and other immune proteins located in different sub-cellular compartments. We have used phage-display technology to express and select for Arabidopsis proteins able to bind bacterial pathogens. To rapidly identify microbe-bound phage, we developed a monitoring method based on microarrays. This combined strategy allowed for a genome-wide screening of plant proteins involved in pathogen perception. Two phage libraries for high-throughput selection were constructed from cDNA of plants infected with Pseudomonas aeruginosa PA14, or from combined samples of the virulent isolate DC3000 of Pseudomonas syringae pv. tomato and its avirulent variant avrRpt2. These three pathosystems represent different degrees in the specificity of plant-microbe interactions. Libraries cover up to 26107 different plant transcripts that can be displayed as functional proteins on the surface of T7 bacteriophage. A number of these were selected in a bio-panning assay for binding to Pseudomonas cells. Among the selected clones we isolated the ethylene response factor ATERF-1, which was able to bind the three bacterial strains in competition assays. ATERF-1 was rapidly exported from the nucleus upon infiltration of either alive or heat-killed Pseudomonas. Moreover, aterf-1 mutants exhibited enhanced susceptibility to infection. These findings suggest that ATERF-1 contains a microbe-recognition domain with a role in plant defence. To identify other putative pathogen-binding proteins on a genome-wide scale, the copy number of selected-vs.-total clones was compared by hybridizing phage cDNAs with Arabidopsis microarrays. Microarray analysis revealed a set of 472 candidates with significant fold change. Within this set defence-related genes, including well-known targets of bacterial effectors, are over-represented. Other genes non-previously related to defence can be associated through this study with general or strain-specific recognition of Pseudomonas.

Wide field-of-view (FOV) microscopy is of high importance to biological research and clinical diagnosis where a high-throughput screening of samples is needed. This thesis presents the development of several novel wide FOV imaging technologies and demonstrates their capabilities in longitudinal imaging of living organisms, on the scale of viral plaques to live cells and tissues.

The ePetri Dish is a wide FOV on-chip bright-field microscope. Here we applied an ePetri platform for plaque analysis of murine norovirus 1 (MNV-1). The ePetri offers the ability to dynamically track plaques at the individual cell death event level over a wide FOV of 6 mm × 4 mm at 30 min intervals. A density-based clustering algorithm is used to analyze the spatial-temporal distribution of cell death events to identify plaques at their earliest stages. We also demonstrate the capabilities of the ePetri in viral titer count and dynamically monitoring plaque formation, growth, and the influence of antiviral drugs.

We developed another wide FOV imaging technique, the Talbot microscope, for the fluorescence imaging of live cells. The Talbot microscope takes advantage of the Talbot effect and can generate a focal spot array to scan the fluorescence samples directly on-chip. It has a resolution of 1.2 μm and a FOV of ~13 mm2. We further upgraded the Talbot microscope for the long-term time-lapse fluorescence imaging of live cell cultures, and analyzed the cells’ dynamic response to an anticancer drug.

We present two wide FOV endoscopes for tissue imaging, named the AnCam and the PanCam. The AnCam is based on the contact image sensor (CIS) technology, and can scan the whole anal canal within 10 seconds with a resolution of 89 μm, a maximum FOV of 100 mm × 120 mm, and a depth-of-field (DOF) of 0.65 mm. We also demonstrate the performance of the AnCam in whole anal canal imaging in both animal models and real patients. In addition to this, the PanCam is based on a smartphone platform integrated with a panoramic annular lens (PAL), and can capture a FOV of 18 mm × 120 mm in a single shot with a resolution of 100─140 μm. In this work we demonstrate the PanCam’s performance in imaging a stained tissue sample.

As lesões impalpáveis da mama que muitas das vezes são assintomáticas, podem corresponder à um estágio de progressão de câncer difícil de ser detectado, durante os exames de rotina de palpação da mulher. O único método possível para a descoberta dessas lesões é através dos exames de imagem da mama, de modo geral, através da mamografia, que geralmente ocorre após os 45 anos. Devido a esses fatores, lesões impalpáveis, são frequentemente, descobertas apenas quando o estágio de desenvolvimento da doença já está avançado e as intervenções terapêuticas são menos reparadoras. Com a finalidade de iniciar a caracterização de tumores impalpáveis iniciais, objetivamos analisar o perfil genético (mutação) e epigenético (metilação de região promotora) de regiões do DNA relacionadas ao gene supressor tumoral TP53, provenientes de biópsias de mulheres residentes do Estado do Rio de Janeiro. Neste trabalho, foram investigadas 34 amostras de tecido de tumor de mama, por sequenciamento de DNA, nos exons de 5 a 8 do gene TP53. Nesta região, não foi encontrada nenhuma mutação. Este resultado pode estar relacionado ao tipo inicial de lesão, de acordo com os dados radiológicos das lesões de categorias 3 e 4 da escala BIRADS. Para verificar o estado de metilação da região promotora do gene TP53, analisamos 30 pares de amostras (sangue e tumor) de pacientes com suspeita de câncer de mama, pela técnica MSP-PCR. Nenhuma amostra tumoral apresentou alteração no estado de metilação na região promotora do gene TP53, quando comparada à amostra normal. Um motivo possível para a disparidade de resultados em relação à outros trabalhos pode ter sido a utilização da técnica. A caracterização das lesões impalpáveis apenas foi iniciada neste trabalho, no qual pudemos constatar que a mutação em TP53 pode ser um evento mais tardio. Portanto, a lesão mamária, em suas diferentes formas, continuará a ser o assunto investigado por nosso grupo, ampliando o número de amostras e alcançando melhor conexão da conduta e dos métodos clínicos já existentes, com as novas possibilidades de diagnóstico via marcadores moleculares em tumores e fluidos biológicos

O câncer de colo de útero é o terceiro tipo de neoplasia mais comum em mulheres no estado do Rio de Janeiro, perdendo apenas para câncer de mama e pulmão. Não há informação disponível sobre a qualidade dos exames utilizados para prevenção do câncer do colo do útero nos municípios fluminenses. O trabalho teve como objetivo avaliar a distribuição geográfica das unidades laboratoriais e a cobertura de exames e a performance da rede de laboratórios de citopatologia do estado do Rio de Janeiro, segundo regiões mesopolitanas e municípios, no que diz respeito aos exames citopatológicos realizados no âmbito do SUS, considerando a qualidade, a normalização dos procedimentos e a gestão do laboratório e propor melhorias. A pesquisa usa um desenho de estudo do tipo transversal incluindo dados relativos ao funcionamento dos laboratórios que fazem exame papanicolaou no âmbito do SUS no estado do Rio de Janeiro. Trata-se de um estudo com dados secundários, já coletados para atender a ação de avaliação dos laboratórios de citopatologia no âmbito do SUS, nesse trabalho também é usada a abordagem ecológica para estimar a cobertura dos exames em relação à população-alvo do programa de rastreamento de câncer de colo de útero. A rede de laboratórios de citopatologia do estado do Rio de Janeiro não teve desempenho satisfatório. As regiões que tiveram laboratórios com melhor desempenho foram Baia da Ilha Grande e Baixada Litorânea e aquelas com laboratórios de pior desempenho foram Médio Paraíba e Noroeste. Os critérios avaliados com melhor desempenho foram da dimensão qualidade e o mais fraco desempenho foi observado para os critérios da dimensão normalização. A dimensão de gestão de laboratórios teve desempenho regular. Um relevante achado desse estudo foi a insuficiente qualidade da leitura de lâminas, inclusive procedimentos de releitura, que pode ser explicado pela suposta falta de capacitação dos profissionais em todo o processo exigido desde a identificação, fixação, e formas de encaminhamento do material até a chegada aos laboratórios. Com relação à cobertura, alguns municípios se aproximam do parâmetro (0,30) porém, embora este dado revele a capacidade da rede estadual do rio de Janeiro de ofertar exames, é preciso que ele seja analisado em conjunto com as situações de citologia anterior e tempo da citologia anterior para verificação da periodicidade da oferta e o melhor dimensionamento do alcance da população alvo assim como a abrangência da rede laboratorial de cada município. Conclusão: Os dados mostram repetição desnecessária de exames citopatológicos, o que implica custos injustificados e uma situação ainda mais deficitária de alcance das ações do que tem revelado o indicador razão. Ocorrendo principalmente em regiões com predominância de laboratórios privados. A expansão da cobertura com base na periodicidade recomendada do exame é relevante no quadro estadual encontrado e deve vir associada a iniciativas que garantam a qualidade no processo de coleta e análise do material, bem como a adequada capacitação dos profissionais para adoção de condutas recomendadas para as lesões identificadas.

In estrogen receptor-negative breast cancer patients, metastatic relapse usually occurs in the lung and is responsible for the fatal outcome of the disease. Thus, a better understanding of the biology of metastasis is needed. In particular, biomarkers to identify patients that are at risk of lung metastasis could open the avenue for new therapeutic opportunities. Here we characterize the biological activity of RARRES3, a new metastasis suppressor gene whose reduced expression in the primary breast tumors identifies a subgroup of patients more likely to develop lung metastasis. We show that RARRES3 downregulation engages metastasis-initiating capabilities by facilitating adhesion of the tumor cells to the lung parenchyma. In addition, impaired tumor cell differentiation due to the loss of RARRES3 phospholipase A1/A2 activity also contributes to lung metastasis. Our results establish RARRES3 downregulation as a potential biomarker to identify patients at high risk of lung metastasis who might benefit from a differentiation treatment in the adjuvant programme.
