P>Vegetable oils can be extracted using ethanol as solvent. The main goal of this work was to evaluate the ethanol performance on the extraction process of rice bran oil. The influence of process variables, solvent hydration and temperature was evaluated using the response surface methodology, aiming to maximise the soluble substances and gamma-oryzanol transfer and minimise the free fatty acids extraction and the liquid content in the underflow solid. It can be noted that oil solubility in ethanol was highly affected by the water content. The free fatty acids extraction is improved by increasing the moisture content in the solvent. Regarding the gamma-oryzanol, it can be observed that its extraction is affected by temperature when low level of water is added to ethanol. On the other hand, the influence of temperature is minimised with high levels of water in the ethanol.

P>In this study, physical characteristics of goat milk powder produced with the addition of soy lecithin at levels of 0 (control), 0.4, 0.8 and 1.0 g lecithin/100 g of total solids in concentrated milk before the spray drying process were investigated. Goat milk was pasteurised, concentrated at 40% of total solids, spray dried and packed in plastic bags under vaccum conditions. Lecithin addition decreased the wetting time of milk powders, although no influence was observed on dispersibility, water sorption, water activity and particle size distribution of the powders. Powders with higher levels of lecithin showed significantly lower brightness, with a greater intensity of yellow colour. It was concluded that lecithin addition before spray drying process at the minimal proportion in concentrated milk of 0.4 g lecithin/100 g of total solids in concentrated milk is useful for achieving more rapid wetting time of goat milk powder.

Background and objectives: There have been few studies investigating acute kidney injury (AKI) in patients infected with the 2009 pandemic influenza A (H1N1) virus. Therefore, the objective of this study was to identify the factors associated with AKI in H1N1-infected patients. Design, setting, participants, & measurements: This was a study of 47 consecutive critically ill adult patients with reverse transcriptase-PCR-confirmed H1N1 infection in Brazil. Outcome measures were AKI (as defined by the Risk, Injury, Failure, Loss, and End-stage renal failure [RIFLE] criteria) and in-hospital death. Results: AKI was identified in 25 (53%) of the 47 H1N1-infected patients. AKI was associated with vasopressor use, mechanical ventilation, high Acute Physiology and Chronic Health Evaluation II (APACHE II) scores, and severe acidosis as well as with higher levels of C-reactive protein and lactic dehydrogenase upon intensive care unit (ICU) admission. A nephrology consultation was requested for 16 patients (64%), and 8 (50%) required dialysis. At ICU admission, 7 (15%) of the 25 AKI patients had not yet progressed to AKI. However, by 72 hours after ICU admission, no difference in RIFLE score was found between AKI survivors and nonsurvivors. Of the 47 patients, 9 (19%) died, all with AKI. Mortality was associated with mechanical ventilation, vasopressor use, dialysis, high APACHE II score, high bilirubin levels, and a low RIFLE score at ICU admission. Conclusions: Among critically ill H1N1-infected patients, the incidence of AKI is high. In such patients, AKI is mainly attributable to shock. Alzheimer`s Disease (AD) is the most common type of dementia among the elderly, with devastating consequences for the patient, their relatives, and caregivers. More than 300 genetic polymorphisms have been involved with AD, demonstrating that this condition is polygenic and with a complex pattern of inheritance. This paper aims to report and compare the results of AD genetics studies in case-control and familial analysis performed in Brazil since our first publication, 10 years ago. They include the following genes/markers: Apolipoprotein E (APOE), 5-hidroxytryptamine transporter length polymorphic region (5-HTTLPR), brain-derived neurotrophin factor (BDNF), monoamine oxidase A (MAO-A), and two simple-sequence tandem repeat polymorphisms (DXS1047 and D10S1423). Previously unpublished data of the interleukin-1 alpha (IL-1 alpha) and interleukin-1 beta (IL-1 beta) genes are reported here briefly. Results from others Brazilian studies with AD patients are also reported at this short review. Four local families studied with various markers at the chromosome 21, 19, 14, and 1 are briefly reported for the first time. The importance of studying DNA samples from Brazil is highlighted because of the uniqueness of its population, which presents both intense ethnical miscegenation, mainly at the east coast, but also clusters with high inbreeding rates in rural areas at the countryside. We discuss the current stage of extending these studies using high-throughput methods of large-scale genotyping, such as single nucleotide polymorphism microarrays, associated with bioinformatics tools that allow the analysis of such extensive number of genetics variables, with different levels of penetrance. There is still a long way between the huge amount of data gathered so far and the actual application toward the full understanding of AD, but the final goal is to develop precise tools for diagnosis and prognosis, creating new strategies for better treatments based on genetic profile.

Rats with unilateral lesion of the substantia nigra pars compacta (SNpc) have been used as a model of Parkinson`s disease. Depending on the lesion protocol and on the drug challenge, these rats rotate in opposite directions. The aim of the present study was to propose a model to explain how critical factors determine the direction of these turns. Unilateral lesion of the SNpc was induced with 6-hydroxydopamine (6-OHDA) or 1-methyl-4-phenyl-1,2,3,6-tetrahydropyridine (MPTP). Separate analysis showed that neither the type of neurotoxin nor the site of lesion along the nigrostriatal. pathway was able to predict the direction of the turns these rats made after they were challenged with apomorphine. However, the combination of these two factors determined the magnitude of the lesion estimated by tyrosine-hydroxylase immunohistochemistry and HPLC-ED measurement of striatal dopamine. Very small lesions did Dot cause turns, medium-size lesions caused ipsiversive turns, and large lesions caused contraversive turns. Large-size SNpc lesions resulted in an increased binding of [H-3] raclopride to D2 receptors, while medium-size lesions reduced the binding of [H-3]SCH-23390 D1 receptors in the ipsilateral striatum. These results are coherent with the model proposing that after challenged with a dopamine receptor agonist, unilaterally SNpc-lesioned rats rotate toward the side with the weaker activation of dopamine receptors. This activation is weaker on the lesioned side in animals with small SNpc lesions due to the loss of dopamine, but stronger in animals with large lesions due to dopamine receptor supersensitivity. The literature reports research efforts allowing the editing of interactive TV multimedia documents by end-users. In this article we propose complementary contributions relative to end-user generated interactive video, video tagging, and collaboration. In earlier work we proposed the watch-and-comment (WaC) paradigm as the seamless capture of an individual`s comments so that corresponding annotated interactive videos be automatically generated. As a proof of concept, we implemented a prototype application, the WACTOOL, that supports the capture of digital ink and voice comments over individual frames and segments of the video, producing a declarative document that specifies both: different media stream structure and synchronization. In this article, we extend the WaC paradigm in two ways. First, user-video interactions are associated with edit commands and digital ink operations. Second, focusing on collaboration and distribution issues, we employ annotations as simple containers for context information by using them as tags in order to organize, store and distribute information in a P2P-based multimedia capture platform. We highlight the design principles of the watch-and-comment paradigm, and demonstrate related results including the current version of the WACTOOL and its architecture. We also illustrate how an interactive video produced by the WACTOOL can be rendered in an interactive video environment, the Ginga-NCL player, and include results from a preliminary evaluation.

The synthesis of FDU-1 silica with large cage-like mesopores prepared with a new triblock copolymer Vorasurf 504 (R) (Eo)(38)(BO)(46)(EO)(38) was developed. The hydrothermal treatment temperature, the dissolution of the copolymer in ethanol, the HCl concentration, the solution stirring time and the hydrothermal treatment time in a microwave oven were evaluated with factorial design procedures. The dissolution in ethanol is important to produce a material with better porous morphology. Increases in the hydrothermal temperature (100 degrees C) and HCl concentration (2 M) improved structural, textural and chemical properties of the cubic ordered mesoporous silica. Also, longer times induced better physical and chemical property characteristics. (C) 2010 Elsevier Inc. All rights reserved.

Topological interactions will be generated in theories with compact extra dimensions where fermionic chiral zero modes have different localizations. This is the case in many warped extra dimension models where the right-handed top quark is typically localized away from the left-handed one. Using deconstruction techniques, we study the topological interactions in these models. These interactions appear as trilinear and quadrilinear gauge boson couplings in low energy effective theories with three or more sites, as well as in the continuum limit. We derive the form of these interactions for various cases, including examples of Abelian, non-Abelian and product gauge groups of phenomenological interest. The topological interactions provide a window into the more fundamental aspects of these theories and could result in unique signatures at the Large Hadron Collider, some of which we explore.

The Amazonian craton in the Sao Felix do Xingu city, southeast region of the Para state, north of Brazil, hosts exceptionally well-preserved Paleoproterozoic bimodal magmatic units grouped in the Sobreiro and Santa Rosa formations. These formations are correlated to the Uatuma magmatic event, which is largely distributed in the Amazonian craton occupying more than 1,500,000 km(2). Geological mapping and petrographical observations reveal distinct spectra of volcanic facies in both formations. The basal calc-alkaline Sobreiro Formation is composed mainly of andesitic and dacitic lava flows and associated volcaniclastic facies of autoclastic origin, with subordinate pyroclastic flow deposits. This formation shows inferred eruption style that is similar to those in Flood Basalt Provinces, with rare scutulum-type lava shields. The upper A-type Santa Rosa Formation was generated by multicyclic explosive and effusive episodes predominantly associated with large fissures and is materialized by voluminous ignimbrites with subordinated ash-fall tuff, crystal tuff, lapilli-tuff, co-ignimbritic breccias, rhyolitic dikes and domes, and associated granitic porphyries and equigranular granitic intrusions. Ignimbrite and rhyolite dikes reveal conspicuous vertical flow pattern pointing to a fissure-controlled eruption, similar to Sierra Madre Occidental ignimbrite province. The proposed evolutionary model for the Sao Felix do Xingu units differs from those of other occurrences related to the Uatuma magmatic event in the Amazonian craton, characterized by predominance of A-type volcanism and contemporaneous granites. Oxidized bases are common types of DNA modifications. Their accumulation in the genome is linked to aging and degenerative diseases. These modifications are commonly repaired by the base excision repair (BER) pathway. Oxoguanine DNA glycosylase (OGG1) initiates BER of oxidized purine bases. A small number of protein interactions have been identified for OGG1, while very few appear to have functional consequences. We report here that OGG1 interacts with the recombination protein RAD52 in vitro and in vivo. This interaction has reciprocal functional consequences as OGG1 inhibits RAD52 catalytic activities and RAD52 stimulates OGG1 incision activity, likely increasing its turnover rate. RAD52 colocalizes with OGG1 after oxidative stress to cultured cells, but not after the direct induction of double-strand breaks by ionizing radiation. Human cells depleted of RAD52 via small interfering RNA knockdown, and mouse cells lacking the protein via gene knockout showed increased sensitivity to oxidative stress. Moreover, cells depleted of RAD52 show higher accumulation of oxidized bases in their genome than cells with normal levels of RAD52. Our results indicate that RAD52 cooperates with OGG1 to repair oxidative DNA damage and enhances the cellular resistance to oxidative stress. Our observations suggest a coordinated action between these proteins that may be relevant when oxidative lesions positioned close to strand breaks impose a hindrance to RAD52 catalytic activities.

Cdc25 phosphatases involved in cell cycle checkpoints are now active targets for the development of anti-cancer therapies. Rational drug design would certainly benefit from detailed structural information for Cdc25s. However, only apo- or sulfate-bound crystal structures of the Cdc25 catalytic domain have been described so far. Together with previously available crystalographic data, results from molecular dynamics simulations, bioinformatic analysis, and computer-generated conformational ensembles shown here indicate that the last 30-40 residues in the C-terminus of Cdc25B are partially unfolded or disordered in solution. The effect of C-terminal flexibility upon binding of two potent small molecule inhibitors to Cdc25B is then analyzed by using three structural models with variable levels of flexibility, including an equilibrium distributed ensemble of Cdc25B backbone conformations. The three Cdc25B structural models are used in combination with flexible docking, clustering, and calculation of binding free energies by the linear interaction energy approximation to construct and validate Cdc25B-inhibitor complexes. Two binding sites are identified on top and beside the Cdc25B active site. The diversity of interaction modes found increases with receptor flexibility. Backbone flexibility allows the formation of transient cavities or compact hydrophobic units on the surface of the stable, folded protein core that are unexposed or unavailable for ligand binding in rigid and densely packed crystal structures. The present results may help to speculate on the mechanisms of small molecule complexation to partially unfolded or locally disordered proteins.

Several conditions have been used in the coupling reaction of stepwise SPPS at elevated temperature (SPPS-ET), but we have elected the following as our first choice: 2.5-fold molar excess of 0.04-0.08 M Boc or Fmoc-amino acid derivative, equimolar amount of DIC/HOBt (1:1)or TBTU/DIPEA(1:3), 25% DMSO/toluene, 60 degrees C, conventional heating. In this study, aimed to further examine enantiomerization under such condition and study the applicability of our protocols to microwave-SPPS, peptides containing L-Ser, L-His, L-Cys and/or L-Met were manually synthesized traditionally, at 60 degrees C using conventional heating and at 60 degrees C using microwave heating. Detailed assessment of all crude peptides (in their intact and/or fully hydrolyzed forms) revealed that, except for the microwave-assisted coupling of L-Cys, all other reactions occurred with low levels of amino acid enantiomerization (<2%). Therefore, herein we (i) provide new evidences that our protocols for SPPS at 60 degrees C using conventional heating are suitable for routine use, (ii) demonstrate their appropriateness for microwave-assisted SPPS by Boc and Fmoc chemistries, (iii) disclose advantages and limitations of the three synthetic approaches employed. Thus, this study complements our past research on SPPS-ET and suggests alternative conditions for microwave-assisted SPPS. Copyright (C) 2009 European Peptide Society and John Wiley & Sons, Ltd.

The aim of this thesis is to examine the early vocabulary development of a sample of Swedish children in relation to parental input and early communicative skills. Three studies are situated in an overall description of early language development in children. The data analyzed in the thesis was collected within a larger project at Stockholm University (SPRINT- “Effects of enhanced parental input on young children’s vocabulary development and subsequent literacy development” [VR 2008-5094]). Data analysis was based on parental report via SECDI, the Swedish version of the MacArthur-Bates Communicative Development Inventories, and audio recordings. One study examined parental verbal interaction characteristics in three groups of children with varying vocabulary size at 18 months. The stability of vocabulary development at 18 and 24 months was investigated in a larger study, with focus on children’s vocabulary composition and grammatical abilities. The third study examined interrelations among early gestures, receptive and productive vocabulary, and grammar measured with M3L, i.e. three longest utterances, from 12 to 30 months. Overall results of the thesis highlight the importance of early language development. Variability in different characteristics in parental input is associated with variability in child vocabulary size. Children with large early vocabularies exhibit the most stability in vocabulary composition and the earliest grammatical development. Children’s vocabulary composition may reflect individual stylistic variation. Use of early gestures is associated differentially with receptive and productive vocabulary. Results of the thesis have implications for parents, child- and healthcare personnel, as well as researchers and educational practitioners. The results underscore the importance of high quality in adult-child interaction, with rich input fine-tuned to children’s developmental levels and age, together with high awareness of early language development.

The importance of understanding the process by which a result was generated in an experiment is fundamental to science. Without such information, other scientists cannot replicate, validate, or duplicate an experiment. We define provenance as the process that led to a result. With large scale in-silico experiments, it becomes increasingly difficult for scientists to record process documentation that can be used to retrieve the provenance of a result. Provenance Recording for Services (PReServ) is a software package that allows developers to integrate process documentation recording into their applications. PReServ has been used by several applications and its performance has been benchmarked.

Introdução: Esta tese de doutorado é mais uma contribuição do Grupo de Qualidade de Vida (Centro Brasileiro) do Programa de Pós-Graduação em Ciências Médicas: Psiquiatria, da UFRGS. Esse grupo têm trabalhado em projetos transculturais de elaboração de instrumentos de qualidade de vida (QV) sob a coordenação da Organização Mundial de Saúde. Entre os instrumentos genéricos de QV já desenvolvidos estão o WHOQOL-100 e o WHOQOL-Bref e entre os instrumentos específicos, para populações especiais, estão o WHOQOL-HIV, o WHOQOL-SRPB e, mais recentemente, o WHOQOL-OLD. O desenvolvimento de uma escala de QV para idosos é especialmente importante tendo em vista especificidades deste grupo etário, bem como o aumento da proporção de idosos na população mundial. Objetivos: O objetivo maior deste estudo é desenvolver uma escala de QV para idosos (WHOQOL-OLD). Entretanto, como trata-se de um processo com longa duração de tempo, foram gerados objetivos específicos a partir da revisão da literatura e coleta de dados, que deram origem a 5 artigos, cada um com sua proposta, a saber: artigo 1. Apresentar a metodologia utilizada e os resultados dos grupos focais para avaliação de QV do idoso, artigo 2. Identificar variáveis relevantes na QV de pessoas idosas, artigo 3. Investigar a relação da percepção de QV do idoso com a percepção de QV do idoso na opinião de seu cuidador, artigo 4. Investigar variáveis associadas com percepção subjetiva de saúde em idosos internados e artigo 5. Pesquisar um possível viés nas respostas de idosos no Inventário de Depressão de Beck (BDI). Métodos: O primeiro estudo teve um desenho qualitativo enquanto os demais foram quantitativos. As amostras variaram para cada estudo. Em todos os estudos idosos(as) acima de 60 anos foram entrevistados. Para os estudos 1 e 2, a amostra contou com profissionais da área da saúde (1) e cuidadores (1 e 2). E, para o estudo 5, adultos acima de 18 anos também foram pesquisados. A coleta dos dados foi realizada em hospitais, lares e grupos comunitários, residências e recrutamento utilizando a técnica de "snow-ball" (bola de neve) em que cada sujeito indicava um outro sujeito. Todos os entrevistados preencheram o Termo de Consentimento Informado e, a partir daí, foram convidados a responder acerca de informações sociodemográficas, QV percebida (WHOQOL-100) e sintomatologia depressiva (BDI), com pequena variação para os cuidadores. O último estudo contou apenas com os dados sociodemográficos e com as respostas ao BDI. Resultados: De forma abreviada, os resultados dos 5 artigos confirmam as especificidades do idoso e portanto a necessidade de desenvolvimento de instrumentos específicos para esta população. O artigo 1 teve como resultado a sugestão de novos itens para idosos, a partir das respostas espontâneas e análise dos domínios e facetas do WHOQOL-100. O artigo 2, por sua vez, mostrou associações da percepção de QV geral com níveis de depressão, percepção subjetiva de saúde e sexo. No artigo 3, foi possível verificar uma tendência, em todos os domínios e na medida QV geral, de o cuidador responder pior percepção de QV do idoso do que o próprio idoso cuidado, apesar de algumas concordâncias (domínios físico, nível de independência, meio ambiente e espiritualidade/religião). Também observou-se que a intensidade de depressão do idoso exerceu forte influência tanto na sua própria percepção de QV quanto na percepção do cuidador sobre o idoso. O artigo 4 mostrou uma prevalência alta e não esperada de idosos internados que se percebiam como saudáveis. Foi possível observar, ainda, uma associação significativa entre percepção saudável e menor intensidade de sintomas depressivos, bem como melhor percepção de QV no domínio nível de independência. E por último, o artigo 5 discute o viés da subescala somático e de desempenho nas respostas do idoso ao BDI. Conclusões: Idosos constituem um grupo particular e, como tal, apresentam especificidades relevantes. A avaliação dos idosos em relação às suas percepções de QV está associada a sexo, idade, estado civil, classe social, percepção de saúde e mais fortemente associada a níveis de sintomas depressivos. Explorando o cuidador como avaliador da QV do idoso observou-se uma tendência de o cuidador perceber a QV do idoso pior do que a própria percepção do idoso, apesar de fortes correlações para todos os domínios e na medida QV geral na percepção do par idoso-cuidador. Já na avaliação de percepção de saúde em idosos foi verificada a influência da intensidade dos sintomas depressivos bem como da dimensão independência: quanto menor a intensidade de depressão e quanto maior o nível de independência, maior associação com percepção de saúde entre idosos. Além desses, características próprias da população idosa podem interferir nos resultados do BDI fazendo com que seus achados sejam maximizados por questões somáticas e de desempenho sugerindo pontos de corte especiais para os idosos. Novos estudos são sugeridos a fim de atender a demanda específica do idoso.
