Com o objetivo de contribuir com conhecimentos sobre a reprodução em laboratório de ostras nativas do gênero Crassostrea e estudar as possÃveis interações entre as espécies cultivadas no Brasil, o presente trabalho avaliou: 1) um método de anestesia e amostragem de tecido gonádico sem sacrifÃcio de animais para a análise do estado de desenvolvimento sexual; 2) a hibridação entre a espécie de ostra do PacÃfico, Crassostrea gigas, e as espécies nativas, C. rhizophorae e C. gasar; e 3) o uso de marcadores de DNA mitocondrial e nuclear para a identificação de hÃbridos. Como resultados, o uso de Cloreto de Magnésio (50 g.L-1), aplicado à água do mar, promoveu o relaxamento muscular e a abertura das valvas nas três espécies de ostras estudadas, permitindo biópsias de tecido gonádico com seringas e agulhas e a determinação do sexo dos animais. Os procedimentos de anestesia e amostragem de tecido não causaram mortalidade nestes indivÃduos que apresentaram 100% de sobrevivência após 10 dias. Após o uso do anestésico, também não foram observadas alterações na atividade reprodutiva e na geração de larvas-D de C. gigas. A partir dos cruzamentos recÃprocos entre C. gigas, C. rhizophorae e C. gasar, houve sucesso assimétrico na fecundação de oócitos de C. rhizophorae (R) com espermatozoides de C. gigas (G), oócitos de C. gasar (B) com espermatozoides de C. gigas (G) e oócitos de C. rhizophorae (R) com espermatozoides de C. gasar (B). A compatibilidade unidirecional de gametas entre as três espécies resultou na formação de larvas hÃbridas que apresentaram crescimento similar à espécie materna até sete dias de idade. Após este perÃodo, as larvas pararam de crescer e morreram. As análises de marcadores moleculares confirmaram que as progênies RG eram hÃbridos verdadeiros e continham o DNA de ambas as espécies parentais em seu genoma. A inviabilidade no desenvolvimento de larvas hÃbridas interespecÃficas em laboratório sugere que a incompatibilidade genômica é suficiente para evitar o risco de hibridação natural entre C. gigas e as espécies nativas C. rhizophorae e C. gasar.

A Histologia, o estudo de tecidos, é uma das áreas fundamentais da Biologia que permitiu enormes avanços cientÃficos. Sendo uma tarefa exigente, meticulosa e demorada, será importante aproveitar a existência de ferramentas e algoritmos computacionais no seu auxÃlio, tornando o processo mais rápido e possibilitando a descoberta de informação que poderá não estar visÃvel à partida. Esta dissertação tem como principal objectivo averiguar se um animal foi ou não sujeito à ingestão de um xenobiótico. Com esse objectivo em vista, utilizaram-se técnicas de processamento e segmentação de imagem aplicadas a imagens de tecido renal de ratos saudáveis e ratos que ingeriram o xenobiótico. Destas imagens extraÃram-se inúmeras caracterÃsticas do corpúsculo renal que após serem analisadas através de vários algoritmos de classificação mostraram ser possÃvel saber se o animal ingeriu ou não o xenobiótico, com um reduzido grau de incerteza. ABSTRACT: Histology, the study of tissues, is one of the key areas of Biology that has allowed huge advances in Science. Being a demanding, meticulous and time consuming task, it is important to use the existence of computational tools and algorithms in its aid, making the process faster and enabling the discovery of information that may not be initially visible. The main goal of this thesis is to ascertain if an animal was subjected or not to the ingestion of a xenobiotic. With this in mind, were used image processing and segmentation techniques applied on images of kidney tissue from healthy rats and rats that ingested the xenobiotic. From these images were extracted several features of renal glomeruli that after being analyzed by various classification algorithms had shown to be possible to know, with an acceptable degree of certainty, if the animal ingested or not the xenobiotic.

O Diabetes mellitus é uma das doenças mais freqüentes nas sociedades modernas. Diabéticos apresentam condições periodontais alteradas quando comparados aos não diabéticos. O presente estudo teve como objetivo compreender a relação entre doença periodontal e diabetes mellitus, identificando os efeitos sobre a saúde do indivÃduo com a finalidade de organizar um protocolo para atendimento desse paciente na saúde bucal. Trata-se de uma revisão da literatura, com abordagem descritiva, realizada em base de dados. Considerou-se periódicos, livros, dissertações e teses na lÃngua portuguesa e que estivessem disponÃveis em sua Ãntegra. Utilizou-se como descritores: saúde bucal, periodontia, diabetes mellitus. Para análise do material bibliográfico utilizado nesta pesquisa, consideraram-se os critérios de inclusão o tipo de estudo, ano e texto completo e as referências quanto ao atendimento dos objetivos propostos pelo estudo. A Doença Periodontal se constitui numa afecção bucal crônica, caracterizada por inflamação e/ou destruição de tecido de suporte. A doença periodontal tem sido considerada a sexta complicação do diabetes. A estreita relação entre a doença periodontal e diabetes tem sido motivo de preocupação entre os cirurgiões-dentistas. Considerando a Unidade Básica de Saúde como porta de entrada do paciente com diabetes para o sistema de saúde, um protocolo de atendimento ao paciente diabético na saúde bucal permite aos dentistas instrumentos norteadores para ações em saúde eficazes e integrais, junto à equipe saúde da famÃlia.

OBJECTIVE: To describe the role of magnetic resonance imaging (MRI) in the evaluation of patients with chronic and recurrent aseptic meningitis.METHOD: A retrospective study of five patients with aseptic meningoencefalitis diagnosed by clinical and CSF findings. CT scans showed without no relevant findings. RESULTS: MRI showed small multifocal lesions hyperintense on T2 weighted images and FLAIR, with mild or no gadolinium enhancement, mainly in periventricular and subcortical regions. Meningoencephalitis preceded the diagnosis of the underlying disease in four patients (Behçet´s disease or systemic lupus erythematosus). After the introduction of adequate treatment for the rheumatic disease, they did not present further symptoms of aseptic meningoencephalitis. CONCLUSION: Aseptic meningoencephalitis can be an early presentation of an autoimmune disease. It is important to emphasize the role of MRI in the diagnosis and follow-up of these patients.

Oligodendrocytes and Schwann cells are engaged in myelin production, maintenance and repairing respectively in the central nervous system (CNS) and the peripheral nervous system (PNS). Whereas oligodendrocytes act only within the CNS, Schwann cells are able to invade the CNS in order to make new myelin sheaths around demyelinated axons. Both cells have some limitations in their activities, i.e. oligodendrocytes are post-mitotic cells and Schwann cells only get into the CNS in the absence of astrocytes. Ethidium bromide (EB) is a gliotoxic chemical that when injected locally within the CNS, induce demyelination. In the EB model of demyelination, glial cells are destroyed early after intoxication and Schwann cells are free to approach the naked central axons. In normal Wistar rats, regeneration of lost myelin sheaths can be achieved as early as thirteen days after intoxication; in Wistar rats immunosuppressed with cyclophosphamide the process is delayed and in rats administered cyclosporine it may be accelerated. Aiming the enlightening of those complex processes, all events concerning the myelinating cells in an experimental model are herein presented and discussed.

The aim of this work was to describe the morphology and ontogeny of P. riedelii fruits to aid in taxonomic, ecological and phylogenetic studies in Apocynaceae. Fruits were fixed in FAA, embedded in plastic resin, sectioned at 10 ìm and stained with toluidine blue, for structural analysis. The fruit of P. riedelii is a follicarium, with two follicular fruitlets. The epicarp is one-cell-layered, with trichomes and thick cuticle. The mesocarp, originating from fundamental ovary tissue, is parenchymatous with laticifers, non-lignified fibers and vascular bundles. The endocarp sensu lato is two-celllayered of crossed sclereids, originating from the inner ovary epidermis and from a single layer of parenchyma cells of fundamental ovary tissue. Follicle dehiscence is lateral and the dehiscence process involves anatomical characteristics such as a dehiscence zone with thin-walled cells, non-lignified fibers in the mesocarp and crossed sclereids in the endocarp.

Aflatoxins are hepatotoxic metabolites produced by Aspergillus flavus and A. parasiticus on a number of agricultural commodities. This research was carried out to evaluate the ability of thermolysed and active Saccharomyces cerevisiae to attenuate liver damage caused by aflatoxin. Diets were prepared containing 0 aflatoxin; 400 mug kg-1 aflatoxin; 400 mug kg-1 aflatoxin plus 1% of dehydrated active yeast, and 400 mug kg-1 aflatoxin plus 1% of thermolysed yeast. A bioassay with Wistar rats was conducted for 28 days, and body organs were weighted and analyses of the liver tissue of the animals were performed. The relative weight of heart, kidneys and liver from animals submitted to the different treatments did not show any difference, and liver tissue of animals feeding on the aflatoxin-free diet was adopted as a toxicity-free pattern. Hepatic tissue of animals feeding on diets containing 400 mug kg-1 aflatoxin or the diet supplemented with 1% thermolysed yeast showed clear signs of toxicity and damage. Hepatic tissue of animals feeding on the diet containing 1% of dehydrated active yeast showed less toxicity signs and damage than those receiving the diet containing 400 mug kg-1 aflatoxin. Active, dehydrated yeast had the ability to reduce toxic effects caused by aflatoxin, but thermolysed yeast was not able to alleviate the effects of aflatoxin toxicity.

The copolymer poly (L-co-D,L lactic acid), PLDLA, has gained prominence in the field of temporary prostheses due to the fact that their time of degradation is quite compatible with the requirement in the case of osseous fracture. In this work the in vivo degradation of devices from copolymer, as a system of plates and screws, used in fixation of the tibia of rabbits was studied. The devices were implanted in 15 adult rabbits, albinos, New Zealand race, and they were used as control devices of alloys of titanium (Ti-6Al-4V/ V grade). The use of copolymers, synthesized in the laboratory, was tested in the repair of fracture in rabbits'tibias, being assessed in the following times: 2 weeks, 2 months and 3 months. Morphological analysis of tissue surrounding the plate and screw system, for 2 weeks of implantation, showed the presence of osteoblasts, indicating a pre bone formation. After 2 months there was new bone formation in the region in contact with the polymer. This bone growth occurred simultaneously with the process of PLDLA degradation, invading the region where there was polymer and after 3 months there was an intense degradation of the copolymer and hence greater tissue invasion compared to 2 months which characterized bone formation in a region where the polymer degraded. The in vivo degradation study of the devices for PLDLA by means of histological evaluations during the period of consolidation of the fracture showed the efficiency of plate and screw system, and it was possible to check formation of bone tissue at the implantation site, without the presence of inflammatory reaction

The present work evaluated the effect of low doses of X-irradiation on the repairing process of sutured and nonsutured skin wounds in rats. For that, rats underwent a surgical proceedure, in which a 20 x 5-millimeter rectangular wound approximately 2-millimeter-deep was made in the dorsal region of each animal, and were divided in four groups: nonirradiated nonsutured; irradiated nonsutured ; nonirradiated sutured and irradiated sutured. The animals under irradiation were protected, during exposure, with a 2-millimeter-thick lead apron in such a way that only the incision was irradiated. Each animal was submitted to 18 seconds of exposure, undergoing a total of 7.4 rads. The evaluation of the effects of X-rays on the repairing process was carried out through microscopic observation by means of hematoxylin-eosin staining for morphological evaluation, and silver impregnation under polarized light for the observation of collagen synthesis. The results have shown that X-irradiation has caused delay in the repairing process, but it did not stop its development. The irradiated nonsutured group was considered to show the greater delay when compared with the other groups.

INTRODUCTION: Like in humans, lower amounts of glycogen are present in tissues of diabetic rats. However, training or drugs that lower glycemia can improve the metabolic control. Metformin increased glycogen while decreased glycemia in normal rats stressed by exercise. OBJECTIVE: In this work we investigated if regular exercise and metformin effects improve the metabolism of diabetic rats. METHODS: Alloxan diabetic Wistar rats treated with metformin (DTM) or not (DT) were trained. Training consisted of 20 sessions of 30 min, 5 days a week. Sedentary diabetic rats served as control (SD and SDM). Metformin (5.6 µg/g) was given in the drinking water. After 48 h resting, glucose (mg/dl) and insulin (ng/mL) was measured in plasma and glycogen (mg/100 mg of wet tissue) in liver, soleus and gastrocnemius. RESULTS: Glycemia decreased in DM group from 435±15 to 230±20, in DT group to 143±8.1 and in DTM group to 138±19 mg/dl. DM group had proportional increase in the hepatic glycogen from 1.69±0.22 to 3.53±0.24, and the training increased to 3.36 ± 0.16 mg/100 mg. Metformin induced the same proportional increase in the muscles (soleus from 0.21±0.008 to 0.42±0.03 and gastrocnemius from 0.33±0.02 to 0.46±0.03), while the training promoted increase on gastrocnemius to 0,53 ± 0,03, only. A high interaction was observed in liver (glycogen increased to 6.48±0.34). CONCLUSION: Very small oral doses of metformin and/or, partially restored glycemia in diabetic rats and decreased glycogen in tissues. Its association with an exercise program was beneficial, helping lower glycemia further and increase glycogen stores on liver of diabetic rats.

PURPOSES: 1) To verify the impact of the creation of the Single Technical Record (STR) at the University of Campinas (Unicamp) Hospital das ClÃnicas, on the preservation period of corneas which were used in elective penetrating keratoplasties, and 2) to compare the primary failure incidence in cornea penetrating keratoplasties regarding the periods before and after the creation of STR. METHODS: A retrospective study was conducted at the Unicamp Hospital, which evaluated 15 consecutive cornea penetrating keratoplasties between January 1st and April 30th, 2000 and 24 consecutive penetrating keratoplasties between May 1st and September 20th of the same year (corneas under the control of the STR), totaling 39 keratoplasties. RESULTS: The mean time between cornea preparation and transplantation was 3.8 days (±1.78) in the period before STR creation, and 6.0 days (±2.97) after STR creation, representing a 36.7% increase in the preservation time. There was a statistically significant difference (p=0.02) between the two groups. No corneal primary failure was observed among the 39 transplanted patients in both groups. CONCLUSION: Based on the results of this study, it can be concluded that this new concept of the State Transplantation System has caused a statistically significant increase in the conservation period of corneas, which may reduce the period of a clear transplant due to an increased loss of endothelial cells, as well as increase the primary failure incidence or result in a high number of corneas that cannot be used due to having exceeded the preservation time recommended by the literature.
