Two experiments were performed using the aromatase inhibitor (AI) letrozole (100mg/kg) to promote sex change, from female-to-male, in protogynous dusky grouper. One experiment was performed during the breeding season (spring) and the other at the end of the breeding season (summer). During the spring, AI promoted sex change after 9weeks and the sperm produced was able to fertilize grouper oocytes. During the summer, the sex change was incomplete; intersex individuals were present and sperm was not released by any of the animals. Sex changed gonads had a lamellar architecture; cysts of spermatocytes and spermatozoa in the lumen of the germinal compartment. In the spring, after 4weeks, 11ketotestosterone (11KT) levels were higher in the AI than in control fish, and after 9weeks, coincident with semen release, testosterone levels increased in the AI group, while 11KT returned to the initial levels. Estradiol (E2) levels remained unchanged during the experimental period. Instead of decreasing throughout the period, as in control group, 17 α-OH progesterone levels did not change in the AI-treated fish, resulting in higher values after 9weeks when compared with control fish. fshβ and lhβ gene expression in the AI animals were lower compared with control fish after 9weeks. The use of AI was effective to obtain functional males during the breeding season. The increase in androgens, modulated by gonadotropins, triggered the sex change, enabling the development of male germ cells, whereas a decrease in E2 levels was not required to change sex in dusky grouper. © 2013 Elsevier Inc.

A total of 51,161 records of scrotal circumference measurements at 18 mo of age (SCI 8) and 17,648 records of sperm defects and breeding soundness of Nellore bulls (mean age of 22.5 mo), raised under extensive conditions, were analyzed to estimate coefficients of heritability and genetic correlations of morphological semen traits by Bayesian inference. The observed semen traits were classified as minor (MID). major (MAD), and total sperm defects (TD). The animals were classified according to breeding soundness as satisfactory and unsatisfactory potential breeders. The (co)variance components and breeding values were estimated by Gibbs sampling using the GIBBS2F90 program under an animal model that included contemporary group as fixed effect, age of animal as linear covariate, and direct additive genetic effects as random effects. Heritabilities of 0.40 ± 0.02, 0.16 ± 0.02, 0.04 ± 0.01, 0.15 ± 0.01, and 0.10 ± 0.01 were obtained for SCI8, MID, MAD, TD, and breeding soundness, respectively. The SC18 showed a positive and moderate correlation with breeding soundness (0.56 ± 0.04) and a negative and low correlation with MID (-0.23 ± 0.03), MAD (-0.16 ± 0.02), and TD (-0.24 ± 0.02). In conclusion, scrotal circumference showed the best response to selection among the traits studied and was favorably correlated with breeding soundness and sperm morphology in young Nellore bulls. © 2013 American Society of Animal Science. Climate change is anticipated to have potentially disastrous impacts on the economic viability of the agricultural sector, insomuch as traditional agricultural practices render the agricultural sector climate-dependent. Increased temperatures and increased intensity, timing and occurrence of hydro events are expected to challenge plant and animal viability. Under such circumstances, vector control is expected to become more difficult, which may further prejudice the prosperity of plant, livestock and fisheries growth. The impact is expected to be on the quality of agricultural produce and thereby, indirectly, on human health outcomes. The key threat mechanisms are debilitated plant vitality and increased propagation of pests, as drought periods increase the breeding of vectors through water pooling and soil erosion associated with the increased intensity of hydro events. In addition, climate change is likely to affect crop productivity in specific geographical areas through its impact on growing seasons and crop patterns, to the extent that crop varieties cannot adapt.

A mandioca é cultivada como "mandioca mansa" para consumo in natura e "mandioca para indústria" como fonte de amidos e farinhas. Raças locais foram utilizadas para descoberta de "mutações espontâneas" e desenvolvimento de abordagem evolutiva e de melhoramento para estudos de função gênica. Recursos de Genômica e Proteômica foram obtidos. Análises de expressão gênica por blot de RNA e microarranjos foram desenvolvidos para identificação de expressão diferencial. "Mandioca açucarada" foi identificada, sendo relacionada com falta de expressão do gene da BEI e de uma mutação "nonsence" na sequência do gene GBSSI causando a formação do amido serose. "Mandioca avermelhada" apresentou falta de expressão do gene CasLYB, e a "amarela" uma regulação de repressão do gene CasHYb. Análise Proteômica do complexo carotenóide-proteína, juntamente com a análise de expressão de gene da CAP4, revelaram uma dupla fita de cDNA associada ao elevado acúmulo de carotenóide. Sequenciamento do gene da GBSSI identificou 22 haplótipos e grande diversidade de nucleotídios. Populações segregantes de cruzamentos de fenótipos bioquímicos diferenciados com cultivares elites dos Cerrados foram obtidas.

Distúrbios reprodutivos estão associados a problemas adquiridos (estresse, manejo inadequado e de origem infecciosa), e genéticos (alta consangüinidade existente em alguns rebanhos do Brasil). Embora mostre o grau do potencial de fertilidade do macho, o exame andrológico e, em particular, o teste de funcionalidade, tem sido pouco utilizado, principalmente na seleção dos touros, embora, estes mesmos estudos, mencionem a importância do sêmen para avaliar a eficiência reprodutiva do macho. O objetivo do estudo foi determinar a prevalência de patologias clinicamente notáveis, através de palpação e inspeção e caracterizar o perfil espermático de búfalos criados em sistemas de produção, intensivo e extensivo. O estudo foi realizado em propriedades do Estado do Pará, nos Municípios de Belém, Ipixuna, Mojú, Nova Timbotéua, Paragominas, Santarém Novo e São Caetano de Odivelas, Muaná, Soure, Chaves e Ponta de Pedras. No estado do Amapá, nos Municípios de Cutias, Itaubal, Tartarugalzinho e no Arquipélago do Bailique. Os animais eram criados no sistema intensivo e extensivo, respectivamente. O período do estudo foi de março de 2008 a março de 2009. Foram utilizados 305 touros da raça Murrah, Mediterrânea e mestiços, com idade variando entre 2 a 15 anos, sendo que 160 no Estado do Pará, e 145 no Estado do Amapá. Os dados dos exames clínicos - andrológicos, assim como o espermiograma foram documentados em fichas especiais. A obtenção de sêmen foi realizada pela massagem das ampolas a avaliação física do sêmen contou da cor, aspecto, volume, motilidade, vigor, concentração e a avaliação morfológica e pH. A concentração foi aferida por espectrofotômetro da Central de Biotecnologia Animal (CEBRAN). Para a morfologia espermática foi utilizado o método de Cerovsky. Foram analisadas 200 espermatozóides em microscópio óptico em objetiva de imersão. Para análise estatística, foi empregado o programa estatístico BIOESTAT 5.0, estatísticas descritivas para todas as variáveis estudadas, biometrias testiculares, patologias do sistema reprodutor e características físicas e morfológicas do sêmen, registrando-se as médias, os desvios padrões e a distribuição de freqüências para as características de classes andrológicas. Os dados quantitativos referentes às idades dos touros, biometrias testiculares, avaliações físicas e morfológicas do sêmen foram submetidos à ANOVA, e quando houve efeito significativo pelo teste F, às médias foram comparadas pelo teste de Tukey a 5 % de probabilidade de erro. Dos 305 búfalos, nos animais do Estado do Amapá a media e o desvio padrão da circunferência escrotal foi superior ao do Estado do Pará independente do tipo de manejo. Indiferente de idade e sistemas de criação não foi encontrado nenhuma alteração no pênis. A bolsa escrotal bífida (fenda escrotal) foi a alteração mais prevalente. A maior freqüência de alterações no epidídimo foi epididimite granulomatosa tipo tuberculóide, observada nos animais de criação extensiva no estado do Amapá, com uma tendência aos touros de mais de quatro anos. Entre as alterações do testículo a mais prevalente foi à torção testicular e de causas infecciosa destacamos a ocorrência de casos de orquite interticial crônico ativa, orquite granulomatosa tipo tuberculóide, hipoplasia testicular grau I, hipoplasia testicular grau III associada à orquite tuberculóide e hipoplasia grau II (parcial). Outra observação bem evidente é com relação às percentagens de espermatozóide com defeitos maiores e menores que se apresentam muito altas as recomendadas pelo CBRA. Deste estudo pode-se concluir que os reprodutores criados de forma extensiva apresentam patologias de origem genética e infecciosa muito prevalente.

Soil physical quality is an important factor for the sustainability of agricultural systems. Thus, the aim of this study was to evaluate soil physical properties and soil organic carbon in a Typic Acrudox under an integrated crop-livestock-forest system. The experiment was carried out in Mato Grosso do Sul, Brazil. Treatments consisted of seven systems: integrated crop-livestock-forest, with 357 trees ha-1 and pasture height of 30 cm (CLF357-30); integrated crop-livestock-forest with 357 trees ha-1 and pasture height of 45 cm (CLF357-45); integrated crop-livestock-forest with 227 trees ha-1 and pasture height of 30 cm (CLF227-30); integrated crop-livestock-forest with 227 trees ha-1 and pasture height of 45 cm (CLF227-45); integrated crop-livestock with pasture height of 30 cm (CL30); integrated crop-livestock with pasture height of 45 cm (CL45) and native vegetation (NV). Soil properties were evaluated for the depths of 0-10 and 10-20 cm. All grazing treatments increased bulk density (r b) and penetration resistance (PR), and decreased total porosity (¦t) and macroporosity (¦ma), compared to NV. The values of r b (1.18-1.47 Mg m-3), ¦ma (0.14-0.17 m³ m-3) and PR (0.62-0.81 MPa) at the 0-10 cm depth were not restrictive to plant growth. The change in land use from NV to CL or CLF decreased soil organic carbon (SOC) and the soil organic carbon pool (SOCpool). All grazing treatments had a similar SOCpool at the 0-10 cm depth and were lower than that for NV (17.58 Mg ha-1).

The Gir cattle breed (Bos indicus) is an important genetic resource for milk production throughout the tropics. The small number of Gir animals introduced in Brazil, rapid dissemination of this breed recently, and the intensification of selection practices could contribute to increase of inbreeding level and reduce genetic diversity in this population. The population was analyzed in terms of pedigree completeness level, inbreeding coefficient, coancestry, generation interval, effective population size, effective number of founders and ancestors, among others. Despite the low mean inbreeding (around 2%), minor problems were identified in the population structure of the Brazilian Gir cattle, e.g., trend of narrower bottlenecks in the pedigree in recent years. The effective population sizes based on inbreeding (94) or coancestry (165.9) as well as the effective number of ancestors (76) and founders (143) were relativity high. The major subdivision of the Gir breed was observed between 1993 and 2002 (dairy and dual-purpose herds, wide use of within-herd matings). In this period the level of inbreeding remained at a higher level, there was a small increase in coancestry and the number of equivalent subpopulations was approximately 6. After 2002, there was genetic exchange between subpopulations, reduction in the average inbreeding, pronounced increase in the average coancestry, and the number of equivalent subpopulations was about 2. Furthermore, it was found that the mean generation interval of the population tended to increase in recent years (around 9 years). About 23% of genetic diversity has been lost since the first generation of founders. Based on the effective population size, number of equivalent subpopulations, inbreeding, coancestry, and loss of genetic diversity, the Gir population is still highly structured, but there is ample room for artificial selection. The results regarding the effective number of founders and ancestors in the present population demonstrate the existence of bottlenecks in the pedigree and indicate the need for population structure monitoring.
