Schistosomiasis is still an endemic disease in many regions, with 250 million people infected with Schistosoma and about 500,000 deaths per year. Praziquantel (PZQ) is the drug of choice for schistosomiasis treatment, however it is classified as Class II in the Biopharmaceutics Classification System, as its low solubility hinders its performance in biological systems. The use of cyclodextrins is a useful tool to increase the solubility and bioavailability of drugs. The aim of this work was to prepare an inclusion compound of PZQ and methyl-beta-cyclodextrin (MeCD), perform its physico-chemical characterization, and explore its in vitro cytotoxicity. SEM showed a change of the morphological characteristics of PZQ:MeCD crystals, and IR data supported this finding, with changes after interaction with MeCD including effects on the C-H of the aromatic ring, observed at 758 cm(-1). Differential scanning calorimetry measurements revealed that complexation occurred in a 1:1 molar ratio, as evidenced by the lack of a PZQ transition temperature after inclusion into the MeCD cavity. In solution, the PZQ UV spectrum profile in the presence of MeCD was comparable to the PZQ spectrum in a hydrophobic solvent. Phase solubility diagrams showed that there was a 5.5-fold increase in PZQ solubility, and were indicative of a type A(L) isotherm, that was used to determine an association constant (K(a)) of 140.8 M(-1). No cytotoxicity of the PZQ:MeCD inclusion compound was observed in tests using 3T3 cells. The results suggest that the association of PZQ with MeCD could be a good alternative for the treatment of schistosomiasis.

Robot-Assisted Rehabilitation (RAR) is relevant for treating patients affected by nervous system injuries (e.g., stroke and spinal cord injury) -- The accurate estimation of the joint angles of the patient limbs in RAR is critical to assess the patient improvement -- The economical prevalent method to estimate the patient posture in Exoskeleton-based RAR is to approximate the limb joint angles with the ones of the Exoskeleton -- This approximation is rough since their kinematic structures differ -- Motion capture systems (MOCAPs) can improve the estimations, at the expenses of a considerable overload of the therapy setup -- Alternatively, the Extended Inverse Kinematics Posture Estimation (EIKPE) computational method models the limb and Exoskeleton as differing parallel kinematic chains -- EIKPE has been tested with single DOFmovements of the wrist and elbow joints -- This paper presents the assessment of EIKPEwith elbow-shoulder compoundmovements (i.e., object prehension) -- Ground-truth for estimation assessment is obtained from an optical MOCAP (not intended for the treatment stage) -- The assessment shows EIKPE rendering a good numerical approximation of the actual posture during the compoundmovement execution, especially for the shoulder joint angles -- This work opens the horizon for clinical studies with patient groups, Exoskeleton models, and movements types --

Stable isotope analyses were applied to explore the relative dietary nitrogen contributions from fish meal and pea meal (Pisum sativum) to muscle tissue of Pacific white shrimp postlarvae (141 ± 31 mg) fed low protein diets having different proportions of both ingredients as the sole dietary protein sources. A negative control diet was formulated to contain 100% pea meal and six more isoproteic diets to have decreasing levels of pea meal-derived nitrogen: 95%, 85%, 70%, 55%, 40% and 0% of the initial level. Growth rates were negatively correlated to dietary pea protein inclusion due to progressive essential amino acid deficiencies (sulphur amino acids, threonine, lysine, histidine). The nitrogen turnover rate significantly increased in muscle tissue of shrimps fed diets having high levels of pea meal; however, contrary to observations from a previous study using soy protein, the relative contributions of dietary nitrogen from pea meal to shrimp muscle tissue were equal or higher than expected contributions established by the dietary formulations. Results highlight the effectiveness of stable isotope analysis in assessing the nutritional contributions of alternative ingredients for aquaculture feeds and the potential suitability of pea as a source of protein (provided the diets are nutritionally balanced)

Detection canines represent the fastest and most versatile means of illicit material detection. This research endeavor in its most simplistic form is the improvement of detection canines through training, training aids, and calibration. This study focuses on developing a universal calibration compound for which all detection canines, regardless of detection substance, can be tested daily to ensure that they are working with acceptable parameters. Surrogate continuation aids (SCAs) were developed for peroxide based explosives along with the validation of the SCAs already developed within the International Forensic Research Institute (IFRI) prototype surrogate explosives kit. Storage parameters of the SCAs were evaluated to give recommendations to the detection canine community on the best possible training aid storage solution that minimizes the likelihood of contamination. Two commonly used and accepted detection canine imprinting methods were also evaluated for the speed in which the canine is trained and their reliability. As a result of the completion of this study, SCAs have been developed for explosive detection canine use covering: peroxide based explosives, TNT based explosives, nitroglycerin based explosives, tagged explosives, plasticized explosives, and smokeless powders. Through the use of these surrogate continuation aids a more uniform and reliable system of training can be implemented in the field than is currently used today. By examining the storage parameters of the SCAs, an ideal storage system has been developed using three levels of containment for the reduction of possible contamination. The developed calibration compound will ease the growing concerns over the legality and reliability of detection canine use by detailing the daily working parameters of the canine, allowing for Daubert rules of evidence admissibility to be applied. Through canine field testing, it has been shown that the IFRI SCAs outperform other commercially available training aids on the market. Additionally, of the imprinting methods tested, no difference was found in the speed in which the canines are trained or their reliability to detect illicit materials. Therefore, if the recommendations discovered in this study are followed, the detection canine community will greatly benefit through the use of scientifically validated training techniques and training aids.

In this paper, dedicated to Prof. Lou Kauffman, we determine the Thurston’s geometry possesed by any Seifert fibered conemanifold structure in a Seifert manifold with orbit space (Formula presented.) and no more than three exceptional fibers, whose singular set, composed by fibers, has at most three components which can include exceptional or general fibers (the total number of exceptional and singular fibers is less than or equal to three). We also give the method to obtain the holonomy of that structure. We apply these results to three families of Seifert manifolds, namely, spherical, Nil manifolds and manifolds obtained by Dehn surgery on a torus knot (Formula presented.). As a consequence we generalize to all torus knots the results obtained in [Geometric conemanifolds structures on (Formula presented.), the result of (Formula presented.) surgery in the left-handed trefoil knot (Formula presented.), J. Knot Theory Ramifications 24(12) (2015), Article ID: 1550057, 38pp., doi: 10.1142/S0218216515500571] for the case of the left handle trefoil knot. We associate a plot to each torus knot for the different geometries, in the spirit of Thurston.

Il compound a matrice poliolefinica studiato è un materiale espandibile e reticolabile, usato per la produzione di manufatti espansi a celle chiuse mediante stampaggio ad iniezione. In questo progetto di tesi il compound viene studiato dal punto di vista reologico, per cercare di prevedere il grado di espansione del manufatto stampato. La correlazione tra l’analisi reometrica e il coefficiente di espansione del manufatto finale potrebbe essere sfruttata per un controllo di qualità del materiale, permettendo così di prevedere il comportamento e le dimensioni finali del manufatto prima dello stampaggio. Lo scopo dello studio è quello di prevedere le dimensioni del manufatto finale partendo da un campione derivante dai granuli del compound a matrice poliolefinica. La prima parte dello studio si concentra sull’analisi del materiale in condizioni ideali, cioè campioni che contengono solo agente reticolante e campioni che contengono esclusivamente agente espandente, valutando l’influenza di questi additivi singolarmente. La seconda parte si concentra sull’analisi del materiale in condizioni reali, co-presenza di reticolante ed espandente, e sulla ricerca di una correlazione tra le analisi reometriche e il coefficiente di espansione. L’ultima parte dello studio si focalizza sull’applicazione di un metodo di previsione lineare delle dimensioni di manufatti stampati, partendo da mescole incognite. Le tecniche di analisi impiegate spaziano da quelle termogravimetriche (TGA), calorimetriche (DSC) a quelle reometriche sui provini derivanti dai granuli, mentre sui campioni stampati sono state eseguite prove dimensionali, prove di densità, prove di durezza (SHORE A e Ascker C) e prove di abrasione.

Il seguente lavoro si propone come analisi degli operatori convoluzionali che caratterizzano le graph neural networks. ln particolare, la trattazione si divide in due parti, una teorica e una sperimentale. Nella parte teorica vengono innanzitutto introdotte le nozioni preliminari di mesh e convoluzione su mesh. In seguito vengono riportati i concetti base del geometric deep learning, quali le definizioni degli operatori convoluzionali e di pooling e unpooling. Un'attenzione particolare è stata data all'architettura Graph U-Net. La parte sperimentare riguarda l'applicazione delle reti neurali e l'analisi degli operatori convoluzionali applicati al denoising di superfici perturbate a causa di misurazioni imperfette effettuate da scanner 3D.

Questa tesi propone una panoramica sul funzionamento interno delle architetture alla base del deep learning e in particolare del geometric deep learning. Iniziando a discutere dalla storia degli algoritmi di intelligenza artificiale, vengono introdotti i principali costituenti di questi. In seguito vengono approfonditi alcuni elementi della teoria dei grafi, in particolare il concetto di laplaciano discreto e il suo ruolo nello studio del fenomeno di diffusione sui grafi. Infine vengono presentati alcuni algoritmi utilizzati nell'ambito del geometric deep learning su grafi per la classificazione di nodi. I concetti discussi vengono poi applicati nella realizzazione di un'architettura in grado di classficiare i nodi del dataset Zachary Karate Club.

La crescente disponibilità di scanner 3D ha reso più semplice l’acquisizione di modelli 3D dall’ambiente. A causa delle inevitabili imperfezioni ed errori che possono avvenire durante la fase di scansione, i modelli acquisiti possono risultare a volte inutilizzabili ed affetti da rumore. Le tecniche di denoising hanno come obiettivo quello di rimuovere dalla superficie della mesh 3D scannerizzata i disturbi provocati dal rumore, ristabilendo le caratteristiche originali della superficie senza introdurre false informazioni. Per risolvere questo problema, un approccio innovativo è quello di utilizzare il Geometric Deep Learning per addestrare una Rete Neurale in maniera da renderla in grado di eseguire efficacemente il denoising di mesh. L’obiettivo di questa tesi è descrivere il Geometric Deep Learning nell’ambito del problema sotto esame.

La seguente tesi propone un’introduzione al geometric deep learning. Nella prima parte vengono presentati i concetti principali di teoria dei grafi ed introdotta una dinamica di diffusione su grafo, in analogia con l’equazione del calore. A seguire, iniziando dal linear classifier verranno introdotte le architetture che hanno portato all’ideazione delle graph convolutional networks. In conclusione, si analizzano esempi di alcuni algoritmi utilizzati nel geometric deep learning e si mostra una loro implementazione sul Cora dataset, un insieme di dati con struttura a grafo.

Snakebite is a neglected disease and serious health problem in Brazil, with most bites being caused by snakes of the genus Bothrops. Although serum therapy is the primary treatment for systemic envenomation, it is generally ineffective in neutralizing the local effects of these venoms. In this work, we examined the ability of 7,8,3'-trihydroxy-4'-methoxyisoflavone (TM), an isoflavone from Dipteryx alata, to neutralize the neurotoxicity (in mouse phrenic nerve-diaphragm preparations) and myotoxicity (assessed by light microscopy) of Bothrops jararacussu snake venom in vitro. The toxicity of TM was assessed using the Salmonella microsome assay (Ames test). Incubation with TM alone (200 μg/mL) did not alter the muscle twitch tension whereas incubation with venom (40 μg/mL) caused irreversible paralysis. Preincubation of TM (200 μg/mL) with venom attenuated the venom-induced neuromuscular blockade by 84% ± 5% (mean ± SEM; n = 4). The neuromuscular blockade caused by bothropstoxin-I (BthTX-I), the major myotoxic PLA2 of this venom, was also attenuated by TM. Histological analysis of diaphragm muscle incubated with TM showed that most fibers were preserved (only 9.2% ± 1.7% were damaged; n = 4) compared to venom alone (50.3% ± 5.4% of fibers damaged; n = 3), and preincubation of TM with venom significantly attenuated the venom-induced damage (only 17% ± 3.4% of fibers damaged; n = 3; p < 0.05 compared to venom alone). TM showed no mutagenicity in the Ames test using Salmonella strains TA98 and TA97a with (+S9) and without (-S9) metabolic activation. These findings indicate that TM is a potentially useful compound for antagonizing the neuromuscular effects (neurotoxicity and myotoxicity) of B. jararacussu venom.

The reproductive capacity between Triatoma lenti and Triatoma sherlocki was observed in order to verify the fertility and viability of the offspring. Cytogenetic, morphological and morphometric approaches were used to analyze the differences that were inherited. Experimental crosses were performed in both directions. The fertility rate of the eggs in crosses involving T. sherlocki females was 65% and 90% in F1 and F2 offspring, respectively. In reciprocal crosses, it was 7% and 25% in F1 and F2 offspring, respectively. The cytogenetic analyses of the male meiotic process of the hybrids were performed using lacto-acetic orcein, C-banding and Feulgen techniques. The male F1 offspring presented normal chromosome behavior, a finding that was similar to those reported in parental species. However, cytogenetic analysis of F2 offspring showed errors in chromosome pairing. This post-zygotic isolation, which prevents hybrids in nature, may represent the collapse of the hybrid. This phenomenon is due to a genetic dysregulation that occurs in the chromosomes of F1. The results were similar in the hybrids from both crosses. Morphological features, such as color and size of connexive and the presence of red-orange rings on the femora, were similar to T. sherlocki, while wins size was similar to T. lenti in F1 offspring. The eggshells showed characteristics that were similar to species of origin, whereas the median process of the pygophore resulted in intermediate characteristics in the F1 and a segregating pattern in F2 offspring. Geometric morphometric techniques used on the wings showed that both F1 and F2 offspring were similar to T. lenti. These studies on the reproductive capacity between T. lenti and T. sherlocki confirm that both species are evolutionarily closed; hence, they are included in the brasiliensis subcomplex. The extremely reduced fertility observed in the F2 hybrids confirmed the specific status of the species that were analyzed.

To evaluate patients with transverse fractures of the shaft of the humerus treated with indirect reduction and internal fixation with plate and screws through minimally invasive technique. Inclusion criteria were adult patients with transverse diaphyseal fractures of the humerus closed, isolated or not occurring within 15 days of the initial trauma. Exclusion criteria were patients with compound fractures. In two patients, proximal screw loosening occurred, however, the fractures consolidated in the same mean time as the rest of the series. Consolidation with up to 5 degrees of varus occurred in five cases and extension deficit was observed in the patient with olecranon fracture treated with tension band, which was not considered as a complication. There was no recurrence of infection or iatrogenic radial nerve injury. It can be concluded that minimally invasive osteosynthesis with bridge plate can be considered a safe and effective option for the treatment of transverse fractures of the humeral shaft. Hybrid bioisoster derivatives from N-acylhydrazones and furoxan groups were designed with the objective of obtaining at least a dual mechanism of action: cruzain inhibition and nitric oxide (NO) releasing activity. Fifteen designed compounds were synthesized varying the substitution in N-acylhydrazone and in furoxan group as well. They had its anti-Trypanosoma cruzi activity in amastigotes forms, NO releasing potential and inhibitory cruzain activity evaluated. The two most active compounds (6, 14) both in the parasite amastigotes and in the enzyme contain the nitro group in para position of the aromatic ring. The permeability screening in Caco-2 cell and cytotoxicity assay in human cells were performed for those most active compounds and both showed to be less cytotoxic than the reference drug, benznidazole. Compound 6 was the most promising, since besides activity it showed good permeability and selectivity index, higher than the reference drug. Thereby the compound 6 was considered as a possible candidate for additional studies.
