Predatory Bdellovibrio bacteriovorus bacteria are remarkable in that they attach to, penetrate and digest other Gram-negative bacteria, living and replicating within them until all resources are exhausted, when they escape the prey ghost to invade fresh prey. Remarkable remodeling of both predator and prey cell occurs during this process to allow the Bdellovibrio to exploit the intracellular niche they have worked so hard to enter, keeping the prey "bdelloplast" intact until the end of predatory growth. If one views motile non-predatory bacteria in a light microscope, one is immediately struck by how rare it is for bacteria to collide. This highlights how the cell surface of Bdellovibrio must be specialized and adapted to allow productive collisions and further to allow entry into the prey periplasm and subsequent secretion of hydrolytic enzymes to digest it. Bdellovibrio can, however, also be made to grow artificially without prey; thus, they have a large genome containing both predatory genes and genes for saprophytic heterotrophic growth. Thus, the membrane and outer surface layers are a patchwork of proteins encompassing not only those that have a sole purpose in heterotrophic growth but also many more that are specialized or employed to attach to, enter, remodel, kill and ultimately digest prey cells. There is much that is as yet not understood, but molecular genetic and post-genomic approaches to microbial physiology have enhanced the pioneering biochemical work of four decades ago in characterizing some of the key events and surface protein requirements for prey attack.

We investigate the generation mechanisms of MHD Poynting flux in the magnetized solar photosphere. Using radiative MHD modeling of the solar photosphere with initial magnetic configurations that differ in their field strength and geometry, we show the presence of two different mechanisms for MHD Poynting flux generation in simulations of solar photospheric magnetoconvection. The weaker mechanism is connected to vertical transport of weak horizontal magnetic fields in the convectively stable layers of the upper photosphere, while the stronger is the production of Poynting flux in strongly magnetized intergranular lanes experiencing horizontal vortex motions. These mechanisms may be responsible for the energy transport from the solar convection zone to the higher layers of the solar atmosphere.

The Faroe-Shetland channel is situated in the main path of the inflow of warm North Atlantic surface water to the Nordic seas and further provides an escape route for the cold Norwegian Sea Deep Water. AMS 14C dates of planktonic foraminifera covering Marine Isotope Stage 3 from two cores in the Faroe-Shetland channel will be used to trace past variability of the Atlantic Meridional Overturning Circulation (AMOC). The reservoir age R shows considerable variability ranging between 50 to 2750 14C years. In particular high R values are observed during Heinrich event 4 (H4) with values around 1550 14C years and during the Laschamp magnetic excursion with R values as high as 2700 14C years. The period between Greenland interstadial 8 (GI8) and GI5 show highly variable R values with interstadial R values around 500 – 650 14C years, i.e. slightly higher than ‘normal’, whereas stadials show either significantly higher or lower R values. From GI5 towards the Last Glacial Maximum R values are generally around 1000 14C years or higher. Using magnetic susceptibility, IRD and δ13C and δ18O values measured on the planktic foraminifera species Neogloboquadrina pachyderma, we compare the observed R variability with reconstructed changes in the Atlantic Meridional Overturning Circulation (AMOC). Furthermore a climate model of intermediate complexity (GENIE) including 14C is used as conceptual tool for identifying oceanographic configuration explaining the observed R variability.

Superluminous supernovae (SLSNe) of Type Ic have a tendency to occur in faint host galaxies which are likely to have low mass and low metallicity. PTF12dam is one of the closest and best-studied superluminous explosions that has a broad and slowly fading light curve similar to SN 2007bi. Here we present new photometry and spectroscopy for PTF12dam from 200-500 d (rest frame) after peak and a detailed analysis of the host galaxy (SDSS J142446.21+461348.6 at z = 0.107). Using deep templates and image subtraction we show that the light curve can be fit with a magnetar model if escape of high-energy gamma rays is taken into account. The full bolometric light curve from -53 to +399 d (with respect to peak) cannot be fit satisfactorily with the pair-instability models. An alternative model of interaction with a dense circumstellar material (CSM) produces a good fit to the data although this requires a very large mass (˜13 M⊙) of hydrogen-free CSM. The host galaxy is a compact dwarf (physical size ˜1.9 kpc) and with Mg = -19.33 ± 0.10, it is the brightest nearby SLSN Ic host discovered so far. The host is a low-mass system (2.8 × 108 M⊙) with a star formation rate (5.0 M⊙ yr-1), which implies a very high specific star formation rate (17.9 Gyr-1). The remarkably strong nebular emission provide detections of the [O III] λ4363 and [O II] λλ7320, 7330auroral lines and an accurate oxygen abundance of 12 + log (O/H) = 8.05 ± 0.09. We show here that they are at the extreme end of the metallicity distribution of dwarf galaxies and propose that low metallicity is a requirement to produce these rare and peculiar SNe.

Allogeneic blood or bone marrow transplantation is a successful treatment for leukaemia and severe aplastic anaemia (SAA). Graft rejection following transplantation for leukaemia is a rare event but leukaemic relapse may occur at varying rates, depending upon the stage of leukaemia at which the transplant was undertaken and the type of leukaemia. Relapse is generally assumed to occur in residual host cells, which are refractory to, or escape from the myeloablative conditioning therapy. Rare cases have been described, however, in which the leukaemia recurs in cells of donor origin. Lack of a successful outcome of blood or bone marrow transplantation for severe aplastic anaemia (SAA), however, is due to late graft rejection or graft-versus-host disease. Leukaemia in cells of donor origin has rarely been reported in patients following allogeneic bone marrow transplantation for SAA. This report describes leukaemic transformation in donor cells following a second allogeneic BMT for severe aplastic anaemia. PCR of short tandem repeats in bone marrow aspirates and in colonies derived from BFUE and CFU-GM indicated the donor origin of leukaemia. Donor leukaemia is a rare event following transplantation for severe aplastic anaemia but may represent the persistence or perturbation of a stromal defect in these patients inducing leukaemic change in donor haemopoietic stem cells.

The proto-oncogene c-Myc paradoxically activates both proliferation and apoptosis. In the pathogenic state, c-Myc-induced apoptosis is bypassed via a critical, yet poorly understood escape mechanism that promotes cellular transformation and tumorigenesis. The accumulation of unfolded proteins in the ER initiates a cellular stress program termed the unfolded protein response (UPR) to support cell survival. Analysis of spontaneous mouse and human lymphomas demonstrated significantly higher levels of UPR activation compared with normal tissues. Using multiple genetic models, we demonstrated that c-Myc and N-Myc activated the PERK/eIF2α/ATF4 arm of the UPR, leading to increased cell survival via the induction of cytoprotective autophagy. Inhibition of PERK significantly reduced Myc-induced autophagy, colony formation, and tumor formation. Moreover, pharmacologic or genetic inhibition of autophagy resulted in increased Myc-dependent apoptosis. Mechanistically, we demonstrated an important link between Myc-dependent increases in protein synthesis and UPR activation. Specifically, by employing a mouse minute (L24+/-) mutant, which resulted in wild-type levels of protein synthesis and attenuation of Myc-induced lymphomagenesis, we showed that Myc-induced UPR activation was reversed. Our findings establish a role for UPR as an enhancer of c-Myc-induced transformation and suggest that UPR inhibition may be particularly effective against malignancies characterized by c-Myc overexpression.

PURPOSE: To review key clinical issues underlying the assessment of in vivo efficacy when using antiangiogenic therapies for cancer treatment.
METHODS: Literature relevant to use of antiangiogenic therapies in cancer was reviewed, with particular emphasis on the assessment of in vivo efficacy of these agents, as well as additional angiogenic factors that could play a role in escape from angiogenesis inhibition.
RESULTS: In order to grow and metastasize, tumors need to continually acquire new blood supplies; therefore, therapeutic inhibition of angiogenesis has become a component of anticancer treatment for many tumor types. Bevacizumab, a humanized monoclonal antibody directed at vascular endothelial growth factor A (VEGF-A), has shown activity in combination with chemotherapy in metastatic colorectal cancer. Nevertheless, the use of antiangiogenic therapies remains suboptimal; specifically, optimal dose, duration of therapy, and combination of agents remain unknown. Also, at present, it is not possible to determine which patients are most likely to respond to a given form of antiangiogenic therapy. There has been increased recognition of alternative pathways possibly associated with disease progression in patients undergoing antiangiogenic therapy targeted at VEGF-A. Multiligand-targeted antiangiogenic therapies, such as ziv-aflibercept (formerly known as aflibercept, VEGF Trap), are currently undergoing clinical evaluation. Ziv-aflibercept forms monomeric complexes with VEGF-A, VEGF-B, and PlGF, which have a long half-life, allowing optimization of ziv-aflibercept doses and angiogenic blockage.
CONCLUSIONS: Although antiangiogenic therapies have increased treatment options for cancer patients, their use is limited by a lack of established and standardized methodology to evaluate their efficacy in vivo. Circulating endothelial cells, hypertension, and several molecular and imaging-based markers have potential for use as biomarkers in these patients and may better define appropriate patient populations.

Set in the borderlands between Letterkenny and Derry-Londonderry, a landscape scarred by geological fold, river and cartographer’s pen, the Ulster crime novelist Brian McGilloway chronicles the hopes and fears of a contemporary society unable to escape a complicated history, redolent and entwined with the voices of its ‘ghosts of its past.’ Through his choice of chief protagonist, An Garda Síochána officer Benedict Devlin, McGilloway turns detective to critically investigate the both the seemingly straightforward and the unseen dwelling in the rural Ulster landscape. Following in the footsteps of Nordic and Tartan Noir in making commentary on current societ,y McGilloway recognises the importance of the past in trying to reach an understanding of the present. His critique however goes beyond criminal behaviour motivated primarily by politics or religion, allowing a deeper and more meaningful diagnosis of the ‘state of the nation’. Place, name and event become especially important in contextualising the liminality of McGilloway’s real rural border settings. In doing so, McGilloway continues in the rich tradition of Ulster poet such as Heaney, MacNiece, Muldoon and Hewitt in trying to rationalise the man-made amidst the elemental in the land of both the ‘Planter & The Gael.’ History, language, tradition and the sacral are all instruments of investigation in helping McGilloway present a revealing pathology and atlas of our times to his readers. Turning literary investigator, the author contends that there is much to learn from this physiography, not just for the borderlands region, but for the wider countryside and society beyond. This work investigated the differences in the reactivity of Sarda (primiparous n= 18, multiparous n= 17) and Dorset (multiparous n= 8) breeds of sheep and their singleton lambs to two challenging test situations involving a mother-lamb partial separation test and an isolation test. Non-parametric analysis used single behavioural variables and fear scores to evaluate the effect of parity, sex of lambs, and the association between mother-lamb behaviour. Amongst ewes, Dorset were characterised by a more calm temperament while Sarda (especially primiparous ewes) were more active in their response to challenge (i.e. more attempts to escape). As with their dams, lambs reflected to a certain extent this divergence and overall during isolation lamb fear score was on average significantly higher than dams. Correlations between measures of behavioural reactivity across tests were carried out to search for predictive measures of fear. A very strong correlation emerged linking vocalisation to locomotor activity. Vocalisation could be a good candidate as predictor factor of an active reaction of sheep to a fearful situation.

Background: Oncogenic mutations in BRAF occur in 8% of patients with advanced colorectal cancer (CRC) and have been shown to correlate with poor prognosis. In contrast to BRAF mutant (MT) melanoma, where the BRAF inhibitor Vemurafenib (PLX4032) has shown significant increases in response rates and overall survival, only minor responses to Vemurafenib treatment have been reported in BRAFMT CRC. Clear understanding of the vulnerabilities of BRAFMT CRC is important, and identification of druggable targets uniquely required by BRAFMT CRC tumours has the potential to fill a gap in the therapeutic armamentarium of advanced CRC. The aim of this study was to identify novel resistance mechanisms to MEK inhibition in BRAFMT CRC. Methods: Paired BRAFMT/WT RKO and VACO432 CRC cells and non-isogenic BRAFMT LIM2405, WiDR, HT-29 and COLO205 CRC cells were used. Changes in protein expression/activity were assessed by Western Blotting. Interactions between MEK1/2 and JAK1/2 or c-MET inhibition were assessed using the MTT cell viability assays and Flow Cytometry. Apoptosis was measured using Western Blotting for PARP, cleaved caspase 3, 8 and 9, and caspase 3/7 and 8 activity assays. Results: Treatment with MEK1/2 inhibitors AZD6244, trametinib, UO126 and PD98059 resulted in acute increases in STAT3 activity in the BRAFMT RKO and VACO432 cells but not in their BRAFWT clones and this was associated with increases in JAK2 activity. Inhibition of JAK/STAT3 activation using gene specific siRNA or small molecule inhibitors TG101348 or AZD1480, abrogated this survival response and resulted in synergy and significant increases in cell death when combined with MEK1/2 inhibitors AZD6244 or trametinib in BRAFMT CRC cells. The RTK c-MET is activated upstream of STAT3 following MEK1/2 inhibition. Inhibition of c-MET and MEK1/2, using pharmacological inhibitors (crizotinib and AZD6244), results in synergy and increased cell death in BRAFMT CRC cells. Conclusions: We have identified JAK/STAT3 activation as an important escape mechanism for BRAFMT CRC following MEK1/2 inhibition in vitro. Combinations of JAK/MEKi or MET/MEKi can be a potential novel treatment strategy for poor prognostic BRAFMT advanced CRC patients.

Growing demands for marine fish products is leading to increased pressure on already depleted wild populations and a rise in aquaculture production. Consequently, more captive-bred fish are released into the wild through accidental escape or deliberate releases. The increased mixing of captive-bred and wild fish may affect the ecological and/or genetic integrity of wild fish populations. Unambiguous identification tools for captive-bred fish will be highly valuable to manage risks (fisheries management) and tracing of escapees and seafood products (wildlife forensics). Using single nucleotide polymorphism (SNP) data from captive-bred and wild populations of Atlantic cod Gadus morhua L. and sole Solea solea L., we explored the efficiency of population and parentage assignment techniques for the identification and tracing of captive-bred fish. Simulated and empirical data were used to correct for stochastic genetic effects. Overall, parentage assignment performed well when a large effective population size characterized the broodstock and escapees originated from early generations of captive breeding. Consequently, parentage assignments are particularly useful from a fisheries management perspective to monitor the effects of deliberate releases of captive-bred fish on wild populations. Population assignment proved to be more efficient after several generations of captive breeding, which makes it a useful method in forensic applications for well-established aquaculture species. We suggest the implementation of a case-by-case strategy when choosing the best method.

Inhibition of JAK/STAT3 activation using gene specific siRNA or small molecule inhibitors TG101348 or AZD1480, abrogated this survival response and resulted in significant increases in cell death when combined with MEK1/2 inhibitors AZD6244 or GSK1120212 in BRAFMT CRC cells. In addition, combination of MEK1/2 and JAK/STAT3 inhibition resulted in strong synergy with CI values between 0.3 and 0.7 in BRAFMT CRC cells.
Conclusions: We have identified JAK/STAT3 activation as an important escape mechanism for BRAFMT CRC following MEK1/2 inhibition. These data provide a strong rationale for further investigation of combination of MEK1/2 and JAK/STAT3 inhibition in BRAFMT in vivo models.

Introduction: Fewer than 50% of adults and 40% of youth meet US CDC guidelines for physical activity (PA) with the built environment (BE) a culprit for limited PA. A challenge in evaluating policy and BE change is the forethought to capture a priori PA behaviors and the ability to eliminate bias in post-change environments. The present objective was to analyze existing public data feeds to quantify effectiveness of BE interventions. The Archive of Many Outdoor Scenes (AMOS) has collected 135 million images of outdoor environments from 12,000 webcams since 2006. Many of these environments have experienced BE change. Methods: One example of BE change is the addition of protected bike lanes and a bike share program in Washington, DC.Weselected an AMOS webcam that captured this change. AMOS captures a photograph from eachwebcamevery half hour.AMOScaptured the 120 webcam photographs between 0700 and 1900 during the first work week of June 2009 and the 120 photographs from the same week in 2010. We used the Amazon Mechanical Turk (MTurk) website to crowd-source the image annotation. MTurk workers were paid US$0.01 to mark each pedestrian, cyclist and vehicle in a photograph. Each image was coded 5 unique times (n=1200). The data, counts of transportation mode, was downloaded to SPSS for analysis. Results: The number of cyclists per scene increased four-fold between 2009 and 2010 (F=36.72, p=0.002). El proceso de transición del familiar, a cuidador de la persona con depresión Método: diseño de la investigación es de naturaleza cualitativa y inductiva, usando la Teoría Fundamentada. Investigación tuvo lugar en 2 polos de las consultas externas del Departamento de Psiquiatría y Salud Mental, Hospital de Évora, Portugal, en 2 ciudades diferentes, durante Febrero y Julio, 2009. La selección de participantes se hizo mediante la muestra no probabilística intencional de acuerdo con los criterios: adulto y/o anciano con un diagnóstico clínico de reacción depresiva breve o prolongada (CIE-9); vivir con familiares; capacidad cognitiva para permitir la recogida de datos; participación voluntaria del paciente y su familia. Se realizaron entrevistas narrativas semiestructuradas a 8 familias, correspondientes a 20 participantes. Resultados: la codificación axial nos permitió identificar 2 categorías: 1ª- narrativa de la enfermedad (inicio, causas, manifestaciones y la caracterización de la depresión). El inicio es identificado por la familia y el paciente, mientras que las causas se identifican sólo por el paciente. Para el familiar la depresión se manifiesta por comportamientos agresivos, aislamiento, desinversión y pasividad; para el paciente, se manifiesta por señales somáticas, pérdida de la voluntad, tristeza, miedo y aislamiento. Para el familiar es una cosa mala porque destruye las relaciones familiares y manipuladora porque es una manera de conseguir lo que el paciente quiere; para el paciente es una cosa de la cabeza, que no se ve, es inconstante y cambia a la persona. 2ª- estrategias de cuidado (escape, indiferencia, conflicto, chantaje).Como manera de cuidar, el familiar está menos tiempo en el hogar para evitar conflictos, aumenta el consumo de alcohol como vía de escape, ignora la conducta del paciente para no molestarlo, como una forma de estimular exige que el paciente haga actividades, y niega actividades sociales si el paciente no lo acompaña, para que él se aísle menos. A veces responsabiliza al paciente por la inestabilidad de la familia, intentando así ayudarle a ser consciente de su comportamiento. No participa en las consultas médicas para que el paciente se sienta más cómodo y rara vez participa en las tareas del hogar, también para que el paciente haga alguna actividad. Discusión: de alguna manera los familiares cambian, para atender a las necesidades del paciente y el papel de cuidador se construye cada día en la interacción entre las personas que cohabitan, de modo que el cuidado tendrá características particulares. Todo lo que el familiar hace, tiene la intención de ayudar al paciente a mejorar y superar su depresión a través de un modo diferente de cuidado. Algunas veces hay un sentimiento de culpa, como consecuencia no de haber cometido una mala acción, sino por haber tenido un comportamiento más duro con el paciente y no saber se eso es lo correcto. Así, es muy importante la intervención de enfermería para que los cuidadores reciban orientaciones y apoyo y la transición se haga armoniosamente.

As zonas costeiras, estuarinas e lagunares são consideradas áreas muito produtivas e dotadas de grande biodiversidade sendo, por isso, consideradas de elevado valor ecológico e económico. No entanto, nas últimas décadas tem vindo a verificar-se um aumento da contaminação destes ecossistemas como resultado de diversas actividades antrópicas. As abordagens actualmente disponíveis para avaliação do impacto da poluição em ecossistemas estuarinos e lagunares apresentam diversos tipos de lacunas, pelo que é importante desenvolver metodologias mais eficazes com organismos autóctones. Neste contexto, o objectivo central desta dissertação consistiu em desenvolver e validar métodos ecologicamente relevantes para avaliação da contaminação estuarina e dos seus efeitos, utilizando o góbio-comum (Pomatoschistus microps), quer como organismo-teste quer como espécie sentinela, devido à importante função que desempenha nas cadeias tróficas de diversos estuários da costa Portuguesa. A Ria de Aveiro foi seleccionada como área de estudo principalmente pelo facto de possuir zonas com diferentes tipos de contaminação predominante e de haver conhecimento científico de base abundante e de elevada qualidade sobre este ecosistema. Na primeira fase do estudo, foram investigados os efeitos agudos de dois hidrocarbonetos aromáticos policíclicos (HAPs) (benzo[a]pireno e antraceno), de um fuel-óleo e de dois metais (cobre e mercúrio) em P. microps, utilizando ensaios laboratoriais baseados em biomarcadores e em parâmetros comportamentais, os quais foram avaliados utilizando um dispositivo expressamente desenvolvido para o efeito, designado por speed performance device (SPEDE). Como biomarcadores foram utilizados parâmetros envolvidos em funções fisiológicas determinantes para a sobrevivência e desempenho dos animais (neurotransmissão, obtenção de energia, destoxificação e defesas anti-oxidantes), nomeadamente a actividade das enzimas acetilcolinesterase, lactato desidrogenase, CYP1A1, glutationa S-transferases, glutationa reductase, glutationa peroxidase, superóxido dismutase, catalase, tendo ainda sido determinados os níveis de peroxidação lipídica como indicador de danos oxidativos. De forma global, os resultados indicaram que os agentes e a mistura testados têm a capacidade de interferir com a função neurológica, de alterar as vias utilizadas para obtenção de energia celular, induzir as defesas antioxidantes e, no caso do cobre e do mercúrio, de causarem peroxidação lipídica. Foram ainda obtidas relações concentração-resposta a nível dos parâmetros comportamentais testados, nomeadamente a capacidade de nadar contra a corrente e a distância percorrida a nadar contra o fluxo de água, sugerindo que os agentes testados podem, por exemplo, diminuir a capacidade de fuga aos predadores, as probabilidades de captura de presas e o sucesso reprodutivo. Na segunda fase, tendo sido já adaptadas técnicas para determinação de vários biomarcadores em P. microps e estudada a sua resposta a dois grupos de poluentes particularmente relevantes em ecossistemas estuarinos e lagunares (metais e HAPs), foi efectuado um estudo de monitorização utilizando P. microps como bioindicador e que incluiu diversos parâmetros ecológicos e ecotoxicológicos, nomedamente: 20 parâmetros indicativos da qualidade da água e do sedimento, concentração de 9 metais em sedimentos e no corpo de P. microps, 8 biomarcadores e 2 índices de condição na espécie seleccionada. A amostragem foi efectuada em quatro locais da Ria de Aveiro, um considerado como referência (Barra) e três com diferentes tipos predominantes de contaminação (Vagueira, Porto de Aveiro e Cais do Bico), sazonalmente, durante um ano. Os resultados obtidos permitiram uma caracterização ecotoxicológica dos locais, incluindo informação sobre a qualidade da água, concentrações de contaminantes ambientais prioritários nos sedimentos e nos tecidos de P. microps, capacidade desta espécie para bioacumular metais, efeitos exercidos pelas complexas misturas de poluentes presentes em cada uma das zonas de amostragem nesta espécie e possíveis consequências para a população. A análise multivariada permitiu analisar de forma integrada todos os resultados, proporcionando informação que não poderia ser obtida analisando os dados de forma compartimentalizada. Em conclusão, os resultados obtidos no âmbito desta dissertação indicam que P. microps possui características adequadas para ser utilizado como organismoteste em ensaios laboratoriais (e.g. abundância, fácil manutenção, permite a determinação de diferentes tipos de critérios de efeito utilizando um número relativamente reduzido de animais, entre outras) e como organismo sentinela em estudos de monitorização da poluição e da qualidade ambiental, estando portanto de acordo com estudos de menor dimensão previamente efectuados. O trabalho desenvolvido permitiu ainda adaptar a P. microps diversas técnicas bioquímicas vulgarmente utilizadas como biomarcadores em Ecotoxicologia e validá-las quer no laboratório quer em cenários reais; desenvolver um novo bioensaio, utilizando um dispositivo de teste especialmente concebido para peixes epibentónicos baseado na performance natatória de uma espécie autóctone e em biomarcadores; relacionar os efeitos a nível bioquímico com parâmetros comportamentais que ao serem afectados podem reduzir de forma drástica e diversificada (e.g. aumento da mortalidade, diminuição do sucesso reprodutivo, redução do crescimento) a contribuição individual para a população. Finalmente, foi validada uma abordagem multidisciplinar, combinando metodologias ecológicas, ecotoxicológicas e químicas que, quando considerada de forma integrada utilizando análises de estatística multivariada, fornece informação científica da maior relevância susceptível de ser utilizada como suporte a medidas de conservação e gestão em estuários e sistemas lagunares.
