A simple method of rubella antigen production by treatment with sodium desoxycholate for use in enzyme immunoassay (IMT-ELISA) is presented. When this assay was compared with a commercial test (Enzygnost-Rubella, Behring), in the study of 108 sera and 118 filter paper blood samples, 96.9% (219/226) overall agreement and correlation coefficient of 0.90 between absorbances were observed. Seven samples showed discordant results, negative by the commercial kit and positive by our test. Four of those 7 samples were available, being 3 positive by HI.

ABSTRACT – Background: According to the Report on Carcinogens, formaldehyde ranks 25th in the overall U.S. chemical production, with more than 5 million tons produced each year. Given its economic importance and widespread use, many people are exposed to formaldehyde environmentally and/or occupationally. Presently, the International Agency for Research on Cancer classifies formaldehyde as carcinogenic to humans (Group 1), based on sufficient evidence in humans and in experimental animals. Manyfold in vitro studies clearly indicated that formaldehyde can induce genotoxic effects in proliferating cultured mammalian cells. Furthermore, some in vivo studies have found changes in epithelial cells and in peripheral blood lymphocytes related to formaldehyde exposure. Methods: A study was carried out in Portugal, using 80 workers occupationally exposed to formaldehyde vapours: 30 workers from formaldehyde and formaldehyde-based resins production factory and 50 from 10 pathology and anatomy laboratories. A control group of 85 non-exposed subjects was considered. Exposure assessment was performed by applying simultaneously two techniques of air monitoring: NIOSH Method 2541 and Photo Ionization Detection equipment with simultaneously video recording. Evaluation of genotoxic effects was performed by application of micronucleus test in exfoliated epithelial cells from buccal mucosa and peripheral blood lymphocytes. Results: Time-weighted average concentrations not exceeded the reference value (0.75 ppm) in the two occupational settings studied. Ceiling concentrations, on the other hand, were higher than reference value (0.3 ppm) in both. The frequency of micronucleus in peripheral blood lymphocytes and in epithelial cells was significantly higher in both exposed groups than in the control group (p < 0.001). Moreover, the frequency of micronucleus in peripheral blood lymphocytes was significantly higher in the laboratories group than in the factory workers (p < 0.05). A moderate positive correlation was found between duration of occupational exposure to formaldehyde (years of exposure) and micronucleus frequency in peripheral blood lymphocytes (r = 0.401; p < 0.001) and in epithelial cells (r = 0.209; p < 0.01). Conclusions: The population studied is exposed to high peak concentrations of formaldehyde with a long-term exposure. These two aspects, cumulatively, can be the cause of the observed genotoxic endpoint effects. The association of these cytogenetic effects with formaldehyde exposure gives important information to risk assessment process and may also be used to assess health risks for exposed worker

Primary cultures of human keratinocytes were challenged with increasing doses from 10 ng/mL to 2 mg/mL of Loxosceles gaucho venom, responsible for dermonecrotic lesion in humans. TNF-a was investigated by bioassay and ELISA in the supernatant of the cultures challenged with 100 ng/mL, 500 ng/mL, 1 and 2 mg/mL of venom. TNF-a was detected by bioassay in the supernatant of cultures challenged with 100 ng/mL, after 6 h. The cytokine was detected by ELISA in the supernatant of the cells challenged with doses of l mg/mL, after 6 and 12 h. The results point out the capacity of this venom to activate the keratinocytes in primary cultures to produce TNF-a. The production of cytokines could contribute to the local inflammatory process in patients bitten by Loxosceles sp.

We describe the production of the potential monoclonal antibodies (MoAbs) using BALB/c mice immunized with vesicular fluid (VF)-Tcra (T. crassiceps) antigen. Immune sera presented anti-VF-Tcra (<20kD) IgG and IgM antibodies with cross-reactivity with T. solium (Tso) antigen (8-12, 14, and 18 kD). After cell fusion, we selected 33 anti-Tcra and anti-Tso reactive IgM-clones and 53 anti-Tcra specific IgG-clones, 5 of them also recognizing Tso antigens. Two clones identified the 8-14 and 18kD peptides of VF-Tcra.

The objective of the article is to evaluate the impact of the friction force mouse-pad in the contraction level of the forearm muscles M. extensor carpi ulnaris, M. extensor digitorum and M. extensor carpi radialis longus. A standard protocol of mouse movements was performed involving horizontal, vertical and diagonal mouse displacements drag-and-drop type. The operators were instructed to execute the protocol with their normal working speed. The movements protocol were performed by each subject (n=17) with three selected pairs mouse-pad, classified as low, medium and high friction force pairs. The mean time to execute the protocol with each mouse was ~138s. Mean values of ~13%MVE and ~17%MVE were found in the M. extensor carpi ulnaris and in the M. extensor digitorum respectively when performing the movements’ protocol. A 8.1% increase in %MVE was observed in the M. extensor digitorum and a 9.4% increase in %MVE was observed in the M. extensor carpi ulnaris when the high friction force pair was operated, relatively to the low friction force pair (p<0.05). The main preliminary conclusions of this study is that operating a high friction force mouse-pad may increase the risk to symptoms or disorders in the wrist due to an increase in the forearm muscles contraction levels, particularly during work with drawing application

Contemporaneamente o Homem depara-se com um dos grandes desafios que é o de efetivar a transição para um futuro sustentável. Assim, o setor da energia tem um papel fundamental neste processo de transição, com principal enfoque no setor dos automóveis, sendo este um setor que contribui com elevadas quantidades de gases de efeito estufa libertados para a atmosfera. Também a escassez dos recursos petrolíferos constitui um ponto fundamental no tema apresentado. Com a necessidade de combater esses problemas é que se tem vindo a tentar desenvolver combustíveis renováveis e neutros quanto às emissões. A primeira geração de biocombustíveis obtidos através de culturas agrícolas terrestres preenche em parte esses requisitos, porém, não atinge os valores da procura e ainda competem com a produção de alimentos. Daí o interesse na aposta de uma segunda geração de biocombustíveis produzidos de fontes que não pertencem à cadeia alimentar e são residuais mas, que mesmo assim não permitem satisfazer as necessidades de matériaprima. A terceira geração de biocombustíveis vem justamente responder a estas questões pois assenta em matérias-primas que não competem pela utilização do solo agrícola nem são usadas para fins alimentares, tendo produtividades areais substancialmente superiores às que as culturas convencionais ou biomassas residuais conseguem assegurar. A matéria prima de terceira geração são portanto as microalgas, cujas produtividades em biomassa são extremamente elevadas, para além de produtividades muito superiores em lípidos, hidratos de carbono e/ou outros produtos de valor elevado. No entanto, este tipo de produção de biocombustível ainda enfrenta alguns problemas técnicos que o tornam num processo dispendioso para competir economicamente com outros tipos de produção de biodiesel. Na linha do que foi dito anteriormente, este trabalho apresenta um estudo de viabilidade económica e energética do biodiesel produzido através da Chlorella vulgaris, apresentando as técnicas e resultados de cultivo da Chlorella vulgaris e posteriormente de produção do biodiesel através dos lípidos obtidos através da mesma. Para melhorar a colheita das microalgas, que é uma das fases mais dispendiosas, testou-se o aumento de pH e a adição de um floculante (Pax XL-10), sendo que o primeiro não permitiu obter resultados satisfatórios, enquanto o segundo permitiu obter resultados de rendimento na ordem dos 90%. Mesmo com a melhoria da etapa da colheita, o preço mínimo do biodiesel produzido a partir do óleo de Chlorella vulgaris, com as condições ótimas de cultivo e produtividades máximas encontradas na literatura, foi de 8,76 €/L, pois, na análise económica, o Pax XL-10 revelou-se extremamente caro para utilizar na floculação de microalgas para obtenção de um produto de baixo valor, como é o biodiesel. A não utilização da floculação reduz o preço do biodiesel para 7,85 €/L. O que se pode concluir deste trabalho é que face às técnicas utilizadas, a produção de biodiesel Chlorella vulgaris apenas, não é economicamente viável, pelo que para viabilizar a sustentabilidade do processo seria ainda necessário desenvolver mais esforços no sentido de otimizar a produção de biodiesel, eventualmente associando-a à produção de um outro biocombustível produzido a partir da biomassa extraída residual e/ou da recuperação de outros produtos de maior valor.

This work proposes a novel approach for a suitable orientation of antibodies (Ab) on an immunosensing platform, applied here to the determination of 8-hydroxy-2′-deoxyguanosine (8OHdG), a biomarker of oxidative stress that has been associated to chronic diseases, such as cancer. The anti-8OHdG was bound to an amine modified gold support through its Fc region after activation of its carboxylic functions. Non-oriented approaches of Ab binding to the platform were tested in parallel, in order to show that the presented methodology favored Ab/Ag affinity and immunodetection of the antigen. The immunosensor design was evaluated by quartz-crystal microbalance with dissipation, atomic force microscopy, electrochemical impedance spectroscopy (EIS) and square-wave voltammetry. EIS was also a suitable technique to follow the analytical behavior of the device against 8OHdG. The affinity binding between 8OHdG and the antibody immobilized in the gold modified platform increased the charge transfer resistance across the electrochemical set-up. The observed behavior was linear from 0.02 to 7.0 ng/mL of 8OHdG concentrations. The interference from glucose, urea and creatinine was found negligible. An attempt of application to synthetic samples was also successfully conducted. Overall, the presented approach enabled the production of suitably oriented Abs over a gold platform by means of a much simpler process than other oriented-Ab binding approaches described in the literature, as far as we know, and was successful in terms of analytical features and sample application.
