A novel approach for lossless as well as lossy compression of monochrome images using Boolean minimization is proposed. The image is split into bit planes. Each bit plane is divided into windows or blocks of variable size. Each block is transformed into a Boolean switching function in cubical form, treating the pixel values as output of the function. Compression is performed by minimizing these switching functions using ESPRESSO, a cube based two level function minimizer. The minimized cubes are encoded using a code set which satisfies the prefix property. Our technique of lossless compression involves linear prediction as a preprocessing step and has compression ratio comparable to that of JPEG lossless compression technique. Our lossy compression technique involves reducing the number of bit planes as a preprocessing step which incurs minimal loss in the information of the image. The bit planes that remain after preprocessing are compressed using our lossless compression technique based on Boolean minimization. Qualitatively one cannot visually distinguish between the original image and the lossy image and the value of mean square error is kept low. For mean square error value close to that of JPEG lossy compression technique, our method gives better compression ratio. The compression scheme is relatively slower while the decompression time is comparable to that of JPEG.

Guanylyl cyclase C (GC-C) is a membrane-associated form of guanylyl cyclase and serves as the receptor for the heat-stable enterotoxin (ST) peptide and endogenous ligands guanylin, uroguanylin, and lymphoguanylin. The major site of expression of GC-C is the intestinal epithelial cell, although GC-C is also expressed in extraintestinal tissue such as the kidney, airway epithelium, perinatal liver, stomach, brain, and adrenal glands. Binding of ligands to GC-C leads to accumulation of intracellular cGMP, the activation of protein kinases G and A, and phosphorylation of the cystic fibrosis transmembrane conductance regulator (CFTR), a chloride channel that regulates salt and water secretion. We examined the expression of GC-C and its ligands in various tissues of the reproductive tract of the rat. Using reverse transcriptase and the polymerase chain reaction, we demonstrated the presence of GC-C, uroguanylin, and guanylin mRNA in both male and female reproductive organs. Western blot analysis using a monoclonal antibody to GC-C revealed the presence of differentially glycosylated forms of GC-C in the caput and cauda epididymis. Exogenous addition of uroguanylin to minced epididymal tissue resulted in cGMP accumulation, suggesting an autocrine or endocrine activation of GC-C in this tissue. Immunohistochemical analyses demonstrated expression of GC-C in the tubular epithelial cells of both the caput epididymis and cauda epididymis. Our results suggest that the GC-C signaling pathway could converge on CFTR in the epididymis and perhaps control fluid and ion balance for optimal sperm maturation and storage in this tissue.

Using in situ x-ray diffraction and Raman scattering techniques, we have investigated the behaviour of single-walled carbon nanotubes bundles under non-hydrostatic pressures. It is seen that the diffraction line corresponding to the two-dimensional triangular lattice in the bundles is not reversible for pressures beyond 5 GPa, in sharp contrast to earlier results under hydrostatic pressure conditions. Most interestingly, radial breathing and tangential Raman modes of the pressure-cycled samples from 21 and 30 GPa match very well with those of the starting sample. Raman and x-ray results put together clearly suggest that the ordering of tubes in the bundles is only marginally regained with a very short coherence length on decompression.

Summary form only given. A scheme for code compression that has a fast decompression algorithm, which can be implemented using simple hardware, is proposed. The effectiveness of the scheme on the TMS320C62x architecture that includes the overheads of a line address table (LAT) is evaluated and obtained compression rates ranging from 70% to 80%. Two schemes for decompression are proposed. The basic idea underlying the scheme is a simple clustering algorithm that partially maps a block of instructions into a set of clusters. The clustering algorithm is a greedy algorithm based on the frequency of occurrence of various instructions.

BACKGROUND Familial diarrhea disorders are, in most cases, severe and caused by recessive mutations. We describe the cause of a novel dominant disease in 32 members of a Norwegian family. The affected members have chronic diarrhea that is of early onset, is relatively mild, and is associated with increased susceptibility to inflammatory bowel disease, small-bowel obstruction, and esophagitis. METHODS We used linkage analysis, based on arrays with single-nucleotide polymorphisms, to identify a candidate region on chromosome 12 and then sequenced GUCY2C, encoding guanylate cyclase C (GC-C), an intestinal receptor for bacterial heat-stable enterotoxins. We performed exome sequencing of the entire candidate region from three affected family members, to exclude the possibility that mutations in genes other than GUCY2C could cause or contribute to susceptibility to the disease. We carried out functional studies of mutant GC-C using HEK293T cells. RESULTS We identified a heterozygous missense mutation (c.2519G -> T) in GUCY2C in all affected family members and observed no other rare variants in the exons of genes in the candidate region. Exposure of the mutant receptor to its ligands resulted in markedly increased production of cyclic guanosine monophosphate (cGMP). This may cause hyperactivation of the cystic fibrosis transmembrane regulator (CFTR), leading to increased chloride and water secretion from the enterocytes, and may thus explain the chronic diarrhea in the affected family members. CONCLUSIONS Increased GC-C signaling disturbs normal bowel function and appears to have a proinflammatory effect, either through increased chloride secretion or additional effects of elevated cellular cGMP. Further investigation of the relevance of genetic variants affecting the GC-C-CFTR pathway to conditions such as Crohn's disease is warranted. The Southern Marginal Zone of the Limpopo Complex is composed of granite-greenstone cratonic rocks reworked by a Neoarchean high-grade tectono-metamorphic event. Petrographic and mineral chemical characterization of an Al-Mg granulite from this zone is presented here. The granulite has a gneissic fabric with distinct Al-rich and Si-rich layers, with the former preserving the unusual lamellar (random and regular subparallel) intergrowths of corundum and symplectic intergrowth of spinel with orthopyroxene. The Al-rich layer preserves mineral assemblages such as rutile with orthopyroxene + sillimanite +/- A quartz, Al-rich orthopyroxene (similar to 11 wt%), spinel + quartz, and corundum in possible equilibrium with quartz, while the Si-rich layer preserves antiperthites and orthopyroxene + sillimanite +/- A quartz, all considered diagnostic of ultrahigh-temperature metamorphism. Application of Al-in-opx thermometry, ternary feldspar thermometry and construction of suitable pressure-temperature phase diagrams, compositional and model proportion isopleth results indicate P-T conditions as high as similar to 1,050-1,100 A degrees C, and similar to 10-12 kbars for the Al-Mg granulite. Our report of ultrahigh-temperature conditions is significant considering that the very high temperature was reached during decompression of an otherwise high-pressure granulite complex (clockwise P-T path), whereas most other ultrahigh-temperature granulites are linked to magma underplating at the base of the crust (counterclockwise P-T path).

We report detailed evidence for a new paleo-suture zone (the Kumta suture) on the western margin of southern India. The c. 15-km-wide, westward dipping suture zone contains garnet-biotite, fuchsite-haematite, chlorite-quartz, quartz-phengite schists, biotite augen gneiss, marble and amphibolite. The isochemical phase diagram estimations and the high-Si phengite composition of quartz-phengite schist suggest a near-peak condition of c. 18 kbar at c. 550 degrees C, followed by near-isothermal decompression. The detrital SHRIMP U-Pb zircon ages from quartz-phengite schist give four age populations ranging from 3280 to 2993 Ma. Phengite from quartz-phengite schist and biotite from garnet-biotite schist have K-Ar metamorphic ages of ca. 1326 and ca. 1385 Ma respectively. Electron microprobe-CHIME ages of in situ zircons in quartz-phengite schist (ca. 3750 Ma and ca. 1697 Ma) are consistent with the above results. The Bondla ultramafic-gabbro complex in the west of the Kumta suture compositionally represents an arc with K-Ar biotite ages from gabbro in the range 1644-1536 Ma. On the eastern side of the suture are weakly deformed and unmetamorphosed shallow westward-dipping sedimentary rocks of the Sirsi shelf, which has the following upward stratigraphy: pebbly quartzite/sandstone, turbidite, magnetite iron formation, and limestone; farther east the lower lying quartzite has an unconformable contact with ca. 2571 Ma quartzo-feldspathic gneisses of the Dharwar block with a ca. 1733 Ma biotite cooling age. To the west of the suture is a c. 60-km-wide Karwar block mainly consisting of tonalite-trondhjemite-granodiorite (TTG) and amphibolite. The TTGs have U-Pb zircon magmatic ages of ca. 3200 Ma with a rare inherited core age of ca. 3601 Ma. The K-Ar biotite cooling age from the TTGs (1746 Ma and 1796 Ma) and amphibolite (ca. 1697 Ma) represents late-stage uplift. Integration of geological, structural and geochronological data from western India and eastern Madagascar suggest diachronous ocean closure during the amalgamation of Rodinia; in the north at around ca. 1380 Ma, and a progression toward the south until ca. 750 Ma. Satellite imagery based regional structural lineaments suggests that the Betsimisaraka suture continues into western India as the Kumta suture and possibly farther south toward a suture in the Coorg area, representing in total a c. 1000 km long Rodinian suture. The variation of electrical resistivity in the system of glasses Ge17Te83-xTlx, with (1 <= x <= 13), has been studied as a function of high pressure for pressures up to 10 GPa. It is found that the normalized electrical resistivity decreases continuously with the increase in pressure and shows a sudden drop at a particular pressure (transition pressure), indicating the presence of a transition from semiconductor to near-metallic at these pressures which are in the range 3.0-5.0 GPa. This transition pressure is seen to decrease with the increase in the percentage content of thallium due to increasing metallicity of the thallium. The transition is reversible under application of pressure and X-ray diffraction of samples recovered after pressurization show that they remain amorphous after undergoing a pressurization decompression cycle.

Many bacteria secrete a highly hydrated framework of extracellular polymer matrix on suitable substrates and embed within the matrix to form a biofilm. Bacterial biofilms are observed on many medical devices, endocarditis, periodontitis and lung infections in cystic fibrosis patients. Bacteria in biofilm are protected from antibiotics and >1,000 times of the minimum inhibitory concentration may be required to treat biofilm infections. Here, we demonstrated that shock waves could be used to remove Salmonella, Pseudomonas and Staphylococcus biofilms in urinary catheters. The studies were extended to a Pseudomonas chronic pneumonia lung infection and Staphylococcus skin suture infection model in mice. The biofilm infections in mice, treated with shock waves became susceptible to antibiotics, unlike untreated biofilms. Mice exposed to shock waves responded to ciprofloxacin treatment, while ciprofloxacin alone was ineffective in treating the infection. These results demonstrate for the first time that, shock waves, combined with antibiotic treatment can be used to treat biofilm infection on medical devices as well as in situ infections.

The occurrence of high-pressure mafic-ultramafic bodies within major shear zones is one of the indicators of paleo-subduction. In mafic granulites of the Andriamena complex (north-eastern Madagascar) we document unusual textures including garnet-clinopyroxene-quartz coronas that formed after the breakdown of orthopyroxene-plagioclase-ilmenite. Textural evidence and isochemical phase diagram calculations in the Na2O-CaO-K2O-FeO-MgO-Al2O3-SiO2-H2O-TiO2 system indicate a pressure-temperature (P-T) evolution from an isothermal (780 degrees C) pressure up to c. 24 kbar to decompression and cooling. Such a P-T trajectory is typically attained in a subduction zone setting where a gabbroic/ultramafic complex is subducted and later exhumed to the present crustal level during oceanic closure and final continental collision. The present results suggest that the presence of such deeply subducted rocks of the Andriamena complex is related to formation of the Betsimisaraka suture. LA-ICPMS U-Pb zircon dating of pelitic gneisses from the Betsimisaraka suture yields low Th/U ratios and protolith ages ranging from 2535 to 2625 Ma. A granitic gneiss from the Alaotra complex yields a zircon crystallization age of ca. 818 Ma and Th/U ratios vary from 1.08 to 2.09. K-Ar dating of muscovite and biotite from biotite-kyanite-sillimanite gneiss and garnet-biotite gneiss yields age of 486 +/- 9 Ma and 459 +/- 9 Ma respectively. We have estimated regional crustal thicknesses in NE Madagascar using a flexural inversion technique, which indicates the presence of an anomalously thick crust (c. 43 km) beneath the Antananarivo block. This result is consistent with the present concept that subduction beneath the Antananarivo block resulted in a more competent and thicker crust. The textural data, thermodynamic model, and geophysical evidence together provide a new insight to the subduction history, crustal thickening and evolution of the high-pressure Andriamena complex and its link to the terminal formation of the Betsimisaraka suture in north-eastern Madagascar. Resumen: El 4 de junio de 2010 se solicita al Comité de Bioética del Hospital Privado de Córdoba, Argentina, una consulta para que se analicen los aspectos éticos de un trasplante pulmonar lobar bilateral, con donante vivo relacionado para dos hermanas gemelas de 17 años con diagnóstico de fibrosis quística (Acta 172). Este caso nos permite revisar dos ejes esenciales de la constitución de la Bioética como son las funciones y actuación de los comités de Bioética en las instituciones hospitalarias, y la licitud de los trasplantes de órganos cuando hay donantes vivos. Se justificará la actuación del Comité de Bioética en este caso, analizando los puntos a tener en cuenta y los recaudos y recomendaciones que habrían de considerarse frente a esta situación planteada.

A fibrose cística (FC) é a doença autossômica recessiva mais comum na população branca que leva à redução na expectativa de vida. A doença pulmonar é a maior causa de morbidade e mortalidade. A relevância do presente estudo se dá diante de alguns fatores: aumento drástico da sobrevida média nos últimos 60 anos na FC, a fisiopatologia pulmonar não é bem compreendida, ausência de estudos reportados na literatura, até o momento, utilizando a técnica de oscilações forçadas (TOF) exclusivamente em adultos com FC. Assim sendo os objetivos deste estudo são: analisar as alterações da mecânica respiratória em adultos com FC através da espirometria, pletismografia e TOF; correlacionar os resultados da TOF aos espirométricos e pletismográficos e avaliar a sensibilidade e especificidade da TOF nestes indivíduos. É um estudo de corte transversal descritivo, no qual foram analisados dois grupos de indivíduos: controle (n=23) e FC (n=27). Os resultados foram expressos através média desvio-padrão. As técnicas funcionais respiratórias foram realizadas na seguinte sequência: TOF, espirometria, pletismografia. Na pletismografia foram avaliados os parâmetros: CPT (capacidade pulmonar total), CRF (capacidade residual funcional) e VR (volume residual), CRF/CPT e VR/CPT, resistência (Rva) e condutância específica das vias aéreas (SGva). Na espirometria: volume expiratório forçado no primeiro segundo (VEF1), capacidade vital forçada (CVF), fluxo expiratório entre 25% e 75% (FEF25%-75%) da CVF (FEF25%-75%) e razões VEF1/CVF (%) e FEF/CVF (%). Na TOF: propriedades resistivas do sistema respiratório- R0 (resistência no intercepto), Rm (resistência média) e S (inclinação da reta de resistência) e propriedades reativas: Cdin,sr (complacência dinâmica do sistema respiratório), Xm (reatância média), frequência de ressonância (fr); e o módulo da impedância em 4 Hz (׀Zrs4Hz׀). Na espirometria o distúrbio ventilatório obstrutivo (DVO) com CVF reduzida foi predominante, com marcante redução do FEF25%-75% no grupo FC (p<0,0001) em relação ao controle. Na pletismografia: destacou-se a elevação de VR, na presença de CPT normal e elevação da Rva e redução da SGva no grupo FC. Alterações da TOF ocorridas no grupo FC em relação ao controle: aumento de R0 e Rm (p<0,0001) e fr (p<0,0002), relacionados à obstrução das vias aéreas; redução de S (p<0,0006), Xm (p<0,0001) associadas à não-homogeneidade do sistema respiratório e Cdin,sr (p<0,0001), relacionada à redução da complacência pulmonar; aumento do módulo da impedância em 4 Hz (׀Zrs4Hz׀) (representando a carga mecânica total do sistema respiratório) resultante da interação das demais alterações da TOF citadas. Os parâmetros da TOF apresentaram correlações muito boas com a espirometria e moderadas com a pletismografia. Rm foi o único parâmetro que não se relacionou com nenhuma destas técnicas. A sensibilidade e especificidade da TOF em adultos com FC apresentaram valores elevados, sobretudo nos parâmetros reativos, em especial, Xm (85,2% e 73,9% respectivamente e área sob a curva de 0,86).

All major geochemical cycles on the Earth’s surface are mediated by microorganisms. Our understanding of how these microbes have interacted with their environments (and vice versa) throughout Earth's history, and how they will respond to changes in the future, is primarily based on studying their activity in different environments today. The overarching questions that motivate the research presented in the two parts of this thesis -- how do microorganisms shape their environment (and vice versa)? and how can we best study microbial activity in situ? -- have arisen from the ultimate goal of being able to predict microbial activity in response to changes within their environments both past and future.

Part one focuses on work related to microbial processes in iron-rich Lake Matano and, more broadly, microbial interactions with the biogeochemical cycling of iron. Primarily, we find that the chelation of ferrous iron by organic ligands can affect the role of iron in anoxic environmental systems, enabling photomixotrophic growth of anoxygenic microorganisms with ferrous iron, as well as catalyzing the oxidation of ferrous iron by denitrification intermediates. These results imply that the ability to grow photomixotrophically on ferrous iron might be more widespread than previously assumed, and that the co-occurrence of chemical and biological processes involved in the coupled biogeochemical cycling of iron and nitrogen likely dominate organic-rich environmental systems.

Part two switches focus to in situ measurements of growth activity and comprises work related to microbial processes in the Cystic Fibrosis lung, and more broadly, the physiology of slow growth. We introduce stable isotope labeling of microbial membrane fatty acids and whole cells with heavy water as a new technique to measure microbial activity in a wide range of environments, demonstrate its application in continuous culture in the laboratory at the population and single cell level, and apply the tool to measure the in situ activity of the opportunistic pathogen Staphylococcus aureus within the environment of expectorated mucus from cystic fibrosis patients. We find that the average in situ growth rates of S. aureus fall into a range of generation times between ~12 hours and ~4 days, with substantial heterogeneity at the single-cell level. These data illustrate the use of heavy water as a universal environmental tracer for microbial activity, and highlight the crucial importance of studying the physiology of slow growth in representative laboratory systems in order to understand the role of these microorganisms in their native environments.

Nesta dissertação foram estudas rochas máficas dos complexos alcalinos de Morro de São João, Rio Bonito, Tanguá, Gericinó-Mendanha, Morro Redondo, Itatiaia e Passa Quatro. Essas rochas ocorrem na forma de diques e/ou sills. As amostras coletadas foram classificadas como lamprófiros, fonolitos, gabros e diabásios alcalinos. A análise geoquímica permitiu identificar um trend fortemente insaturado e um trend moderadamente alcalino para os complexos estudados. O primeiro é caracterizado por foiditos e fonolitos como membros parentais e mais evoluídos, respectivamente, enquanto o segundo tem basaltos alcalinos como membros parentais e traquitos como os mais evoluídos. Todas as amostras plotam no campo da série alcalina, sendo majoritariamente miaskíticas, sódicas ou potássicas. Adicionalmente, o estudo geoquímico indicou que os complexos alcalinos representam câmaras magmáticas distintas, onde diferentes processos evolutivos tiveram lugar. As modelagens apontaram dois processos de diferenciação distintos nos complexos estudados. Os complexos alcalinos de Morro de São João, Morro Redondo, Gericinó-Mendanha e Itatiaia estariam relacionados a processos de cristalização fracionada. Por outro lado, o Complexo Alcalino de Passa Quatro teria sido diferenciado por processos de cristalização fracionada com esvaziamento e posterior reabastecimento da câmara magmática (RTF). De um modo geral, esses modelos indicaram a presença de mais do que uma série magmática nos complexos estudados e a não cogeneticidade entre as séries agpaíticas e miaskíticas. A discriminação de fontes foi feita com base na análise dos elementos terras raras das amostras parentais de cada um dos complexos (gabro em Morro de São João e lamprófiro nos demais). No entanto, este procedimento não foi aplicado para o Complexo Alcalino de Morro Redondo, uma vez que todas as suas amostras apresentaram valores de MgO muito abaixo do típico para líquidos parentais. O líquido parental do Complexo Alcalino do Gericinó-Mendanha apresentou razões de La/Yb e La/Nb, maior e menor que a unidade, respectivamente, típicas de derivação a partir fontes férteis. Os líquidos parentais dos outros complexos alcalinos tiveram suas razões La/Yb e La/Nb maiores que a unidade, típicas de derivação a partir de fontes enriquecidas. Os modelos desenvolvidos revelaram que os líquidos parentais de cada um dos complexos estudados estariam relacionados a fontes lherzolíticas com granada residual. Além disso, a fusão parcial destas fontes teria ocorrido num intervalo de 1 a 7% dentro da zona da granada. Finalmente, as modelagens petrogenéticas elaboradas permitiram a proposição de um cenário geodinâmico, envolvendo a descompressão adiabática do manto litosférico e sublitosférico anomalamente aquecidos. As características geoquímicas dos líquidos parentais parecem ter sido controladas essencialmente pela mistura desses dois tipos de fontes.
