When the summer of 1935 rolled around, Gaines was contemplating his next step. He was graduating in August, with honors no less, and he was pondering a career. After graduation, he filed papers with placement office at Lincoln but he could not find an immediate position. Although he minored in education, Gaines was not primarily searching for a teaching position.

A história do Senhor Bom Jesus da Cana Verde: conflitos e celebrações em torno de uma imagem religiosa (Siqueira Campos PR, 1933)

Essa dissertação abordou os conflitos que ocorreram no espaço geográfico denominado Norte Pioneiro . A imagem do Bom Jesus, de propriedade da família Pinto, foi expropriada pelo vigário da paróquia do Distrito de Salto do Itararé, padre Alfredo Simon, que reuniu cerca de vinte homens aramados para capturar esse santo. Nesse conflito religioso que ocorreu no dia 26/4/1933 duas pessoas foram mortas: o comerciante do Arraial dos Pintos, João Moreira, e o herdeiro do Bom Jesus, José Pinto de Oliveira. Esse último veio a falecer meses depois do conflito. Ao redor dessa imagem foi sendo criada uma história oficial e vigiada pelos donos do poder simbólico, mantenedora da ordem e da tradição. No entanto, a história do Bom Jesus foi compreendida numa concepção mais ampla, pois na esfera religiosa ocorria um fenômeno denominado de romanização. A Igreja Católica seguia o Código de Direito Canônico de 1917, não reconhecendo o Código de Direito Civil do Estado Nacional Brasileiro. Na esfera política, o governo paranaense colocou em prática o sistema de terras devolutas. No setor dos transportes, a estrada de ferro RVPRSC (Rede Viária Paraná Santa Catarina) já se encontrava na região desde 1919. E nesse ínterim, as novas relações sócio-culturais e econômicas foram introduzidas no campo, isto é, o capitalismo agrário.(AU)

CP12 provides a new mode of light regulation of Calvin cycle activity in higher plants

CP12 is a small nuclear encoded chloroplast protein of higher plants, which was recently shown to interact with NAD(P)H–glyceraldehyde-3-phosphate dehydrogenase (GAPDH; EC 1.2.1.13), one of the key enzymes of the reductive pentosephosphate cycle (Calvin cycle). Screening of a pea cDNA library in the yeast two-hybrid system for proteins that interact with CP12, led to the identification of a second member of the Calvin cycle, phosphoribulokinase (PRK; EC 2.7.1.19), as a further specific binding partner for CP12. The exchange of cysteines for serines in CP12 demonstrate that interaction with PRK occurs at the N-terminal peptide loop of CP12. Size exclusion chromatography and immunoprecipitation assays reveal the existence of a stable 600-kDa PRK/CP12/GAPDH complex in the stroma of higher plant chloroplasts. Its stoichiometry is proposed to be of two N-terminally dimerized CP12 molecules, each carrying one PRK dimer on its N terminus and one A2B2 complex of GAPDH subunits on the C-terminal peptide loop. Incubation of the complex with NADP or NADPH, in contrast to NAD or NADH, causes its dissociation. Assays with the stromal 600-kDa fractions in the presence of the four different nicotinamide-adenine dinucleotides indicate that PRK activity depends on complex dissociation and might be further regulated by the accessible ratio of NADP/NADPH. From these results, we conclude that light regulation of the Calvin cycle in higher plants is not only via reductive activation of different proteins by the well-established ferredoxin/thioredoxin system, but in addition, by reversible dissociation of the PRK/CP12/GAPDH complex, mediated by NADP(H).

Acclimation of Arabidopsis Leaves Developing at Low Temperatures. Increasing Cytoplasmic Volume Accompanies Increased Activities of Enzymes in the Calvin Cycle and in the Sucrose-Biosynthesis Pathway1

Photosynthetic and metabolic acclimation to low growth temperatures were studied in Arabidopsis (Heynh.). Plants were grown at 23°C and then shifted to 5°C. We compared the leaves shifted to 5°C for 10 d and the new leaves developed at 5°C with the control leaves on plants that had been left at 23°C. Leaf development at 5°C resulted in the recovery of photosynthesis to rates comparable with those achieved by control leaves at 23°C. There was a shift in the partitioning of carbon from starch and toward sucrose (Suc) in leaves that developed at 5°C. The recovery of photosynthetic capacity and the redirection of carbon to Suc in these leaves were associated with coordinated increases in the activity of several Calvin-cycle enzymes, even larger increases in the activity of key enzymes for Suc biosynthesis, and an increase in the phosphate available for metabolism. Development of leaves at 5°C also led to an increase in cytoplasmic volume and a decrease in vacuolar volume, which may provide an important mechanism for increasing the enzymes and metabolites in cold-acclimated leaves. Understanding the mechanisms underlying such structural changes during leaf development in the cold could result in novel approaches to increasing plant yield.

A Functional Calvin Cycle Is Not Indispensable for the Light Activation of C4 Phosphoenolpyruvate Carboxylase Kinase and Its Target Enzyme in the Maize Mutant bundle sheath defective2-mutable11

We used a pale-green maize (Zea mays L.) mutant that fails to accumulate ribulose-1,5-bisphosphate carboxylase/oxygenase (Rubisco) to test the working hypothesis that the regulatory phosphorylation of C4 phosphoenolpyruvate carboxylase (PEPC) by its Ca2+-insensitive protein-serine/threonine kinase (PEPC kinase) in the C4 mesophyll cytosol depends on cross-talk with a functional Calvin cycle in the bundle sheath. Wild-type (W22) and bundle sheath defective2-mutable1 (bsd2-m1) seeds were grown in a controlled environment chamber at 100 to 130 μmol m−2 s−1 photosynthetic photon flux density, and leaf tissue was harvested 11 d after sowing, following exposure to various light intensities. Immunoblot analysis showed no major difference in the amount of polypeptide present for several mesophyll- and bundle-sheath-specific photosynthetic enzymes apart from Rubisco, which was either completely absent or very much reduced in the mutant. Similarly, leaf net CO2-exchange analysis and in vitro radiometric Rubisco assays showed that no appreciable carbon fixation was occurring in the mutant. In contrast, the sensitivity of PEPC to malate inhibition in bsd2-m1 leaves decreased significantly with an increase in light intensity, and there was a concomitant increase in PEPC kinase activity, similar to that seen in wild-type leaf tissue. Thus, although bsd2-m1 mutant plants lack an operative Calvin cycle, light activation of PEPC kinase and its target enzyme are not grossly perturbed.

Aportes de la Asociación Rural del Uruguay a la viticultura en Uruguay (1872-1900)

En este artículo se analizan desde una perspectiva agronómica los contenidos de los discursos de los viticultores y técnicos en Uruguay para el periodo 1872-1900, presentes en la Revista de la Asociación Rural del Uruguay. Los contenidos se organizan según dimensiones del manejo de la viña en la etapa de campo. Se expresa la preponderancia de los primeros actores en la constitución de la temprana viticultura nacional -productores pertenecientes a una elite empresarial de inmigrantes- al adaptar sus conocimientos previos desde procesos de experimentación. Se indaga en el papel que cumplió la Asociación Rural del Uruguay en esta fase

La divulgación radiofónica de la alimentación y la higiene infantil en la España de la Segunda República (1933-1935)

El trabajo analiza la incorporación de medios tecnológicos, en particular la radio, en el proceso de divulgación del conocimiento sobre la salud infantil durante la Segunda República española. En el marco de la campaña sanitaria contra la mortalidad infantil que puso en marcha la Segunda República en la década del 30, la Asociación Española de Médicos Puericultores Titulados, en colaboración con Unión Radio, organizó dos ciclos de conferencias radiofónicas que pretendían explicar, a través de un discurso divulgativo de tono descriptivo dirigido a las clases populares, las nociones fundamentales de la higiene y la alimentación infantil, e impulsar una cultura de la prevención, guiada y orientada por los dictados de los expertos. Se trataba de una acción de propaganda y divulgación que tenía su continuidad en el medio familiar y comunitario a través de la actuación de las enfermeras visitadoras puericultoras, contemplando a las madres como población diana.
