The commensal microbiota impacts specific immune cell populations and their functions at peripheral sites, such as gut mucosal tissues. However, it remains unknown whether gut microbiota control immunity through regulation of hematopoiesis at primary immune sites. We reveal that germ-free mice display reduced proportions and differentiation potential of specific myeloid cell progenitors of both yolk sac and bone marrow origin. Homeostatic innate immune defects may lead to impaired early responses to pathogens. Indeed, following systemic infection with Listeria monocytogenes, germ-free and oral antibiotic-treated mice display increased pathogen burden and acute death. Recolonization of germ-free mice with a complex microbiota restores defects in myelopoiesis and resistance to Listeria. These findings reveal that gut bacteria direct innate immune cell development via promoting hematopoiesis, contributing to our appreciation of the deep evolutionary connection between mammals and their microbiota.

Tanzania Fisheries Research Institute being currently the only fisheries research institute in Tanzania has sole responsibility of marking the presence and distribution of fish species in the country. This work is expected to be completed by the year 2005. In order that this work can be accomplished, both published works and field observations (including geographical location using GPS) have to be compiled. Since the institute has centers in all the Great Lakes and one along the Indian Ocean coast, it is expected that field observations and identifications will be obtained and incorporated into the document to be produced by 2005. Whereas some species may rely on periodic drought conditions for part of their life histories, or have life strategies suited to exploiting the habitat or changed environmental conditions that are created by drought, for other organisms it is a time of stress. Periodic drought conditions therefore generate a series of waves of colonization and extinctions. Studies on lowland wet grassland, in winterbournes and in the toiche zone of both ponds and rivers, also demonstrate that different organisms are competitively favoured with changing hydrological conditions, and that this process prevents any one species from overwhelming its competitors. Competitive impacts may be inter- and intraspecific. It is therefore apparent that the death of organisms such as adult fish during severe drought conditions, though traumatic for human onlookers and commercial interests, may be merely a regular occurrence to which the ecosystem is adapted. The variability of climatic conditions thereby provides a direct influence on the maintenance of biological diversity, and it is this very biodiversity that provides the ecosystem with the resilience to respond to environmental changes in both the short and the longer term.

The Naaf River estuary is one of the large estuaries in the Bangladesh coastal region not to have been affected by extensive human disturbance. This research provides information about the fisheries diversity status by Estuarine Set Bag Net (ESBN) sampling relation to physicochemical variables in both spatio-temporal scales. About 25 km of the lower estuary was divided into six zones for sample collection by considering the accessibility and availability of the ESBN operation, fish landing centers and location of the fishing villages. In total 48 samples have been analyzed which were taken throughout March to October 2006. To quantify the species diversity, all fisheries data were analyzed by using EstimateS and EcoSim software which accounts the different diversity indices viz., species richness, Shannon–Wiener diversity Index, Dominance and Evenness index. The research results demonstrate that the Naaf River estuary is a habitat of 161 (species richness, Sobs=161, Choa 1=162±2.34, ACE=161.73) different species which belong to 98 fin fishes, 23 shrimps and prawns,13 crabs, 11 molluscs, 3 echinoderms, 4 other crustaceans;while 9 remain unidentified. Results on the aquatic environment,mainly salinity and turbidity were found to have a major influence on their occurrence and distribution. All the findings indicated that the Naaf River estuary is a highly productive system and provides a favourable environment for large variety of estuarine species assemblages.

Understanding the roles of microorganisms in environmental settings by linking phylogenetic identity to metabolic function is a key challenge in delineating their broad-scale impact and functional diversity throughout the biosphere. This work addresses and extends such questions in the context of marine methane seeps, which represent globally relevant conduits for an important greenhouse gas. Through the application and development of a range of culture-independent tools, novel habitats for methanotrophic microbial communities were identified, established settings were characterized in new ways, and potential past conditions amenable to methane-based metabolism were proposed. Biomass abundance and metabolic activity measures – both catabolic and anabolic – demonstrated that authigenic carbonates associated with seep environments retain methanotrophic activity, not only within high-flow seep settings but also in adjacent locations exhibiting no visual evidence of chemosynthetic communities. Across this newly extended habitat, microbial diversity surveys revealed archaeal assemblages that were shaped primarily by seepage activity level and bacterial assemblages influenced more substantially by physical substrate type. In order to reliably measure methane consumption rates in these and other methanotrophic settings, a novel method was developed that traces deuterium atoms from the methane substrate into aqueous medium and uses empirically established scaling factors linked to radiotracer rate techniques to arrive at absolute methane consumption values. Stable isotope probing metaproteomic investigations exposed an array of functional diversity both within and beyond methane oxidation- and sulfate reduction-linked metabolisms, identifying components of each proposed enzyme in both pathways. A core set of commonly occurring unannotated protein products was identified as promising targets for future biochemical investigation. Physicochemical and energetic principles governing anaerobic methane oxidation were incorporated into a reaction transport model that was applied to putative settings on ancient Mars. Many conditions enabled exergonic model reactions, marking the metabolism and its attendant biomarkers as potentially promising targets for future astrobiological investigations. This set of inter-related investigations targeting methane metabolism extends the known and potential habitat of methanotrophic microbial communities and provides a more detailed understanding of their activity and functional diversity.

This short interim progress report builds on previous progress reports which have described the quantification of the process both within and between lakes of different degrees of eutrophication. These data indicated that slight changes in methodology, particularly when investigating sediment deposits, could grossly affect the measured activity. The aim of the present research was an attempt to rationalize these differences. If this could be achieved it would enable meaningful interpretation of published data obtained using different methods and therefore enlarge the available database. In addition some observations have been made on the production of nitrite by Grasmere profundal sediment slurries sampled during the circulation period.

Obtaining a reliable estimate of the bacterial population is one of the main problems facing the bacterial ecologist. The author discusses the various methods available and concludes that the observed variability in bacterial populations depends on the sampling interval used.

This review discusses the processes involved in the decomposition of organic carbon derived initially from structural components of algae and other primary producers. It describes how groups of bacteria interact in time and space in a eutrophic lake. The relative importance of anaerobic and aerobic processes are discussed. The bulk of decomposition occurs within the sediment. The role of bacteria in the nitrogen cycle and the iron cycle, and in sulphate reduction and methanogenesis as the terminal metabolism of organic carbon are described.

It is widely recognised that conventional culture techniques may underestimate true viable bacterial numbers by several orders of magnitude. The basis of this discrepancy is that a culture in or on media of high nutrient concentration is highly selective (either through ”nutrient shock” or failure to provide vital co-factors) and decreases apparent diversity; thus it is unrepresentative of the natural community. In addition, the non-culturable but viable state (NCBV) is a strategy adopted by some bacteria as a response to environmental stress. The basis for the non-culturable state is that cells placed in conditions present in the environment cannot be recultured but can be shown to maintain their viability. Consequently, these cells would not be detected by standard water quality techniques that are based on culture. In the case of pathogens, it may explain outbreaks of disease in populations that have not come into contact with the pathogen. However, the NCBV state is difficult to attribute, due to the failure to distinguish between NCBV and non-viable cells. This article will describe experiences with the fish pathogen Aeromonas salmonicida subsp. salmonicida and the application of molecular techniques for its detection and physiological analysis.

Protozoa feed on and regulate the abundance of most types of aquatic microorganisms, and they are an integral part of all aquatic microbial food webs. Being so small, aerobic protozoa thrive at low oxygen tensions, where they feed (largely unaffected by metazoan grazing) on the abundance of other microorganisms. In anaerobic environments, they are the only phagotrophic organisms, and they live in unique symbiotic consortia with methanogens, sulphate reducers and non-sulphur purple bacteria. The number of extant species of protozoa may be quite modest (the global number of ciliate species is estimated at 3000), and most of them probably have cosmopolitan distributions. This will undoubtedly make it easier to carry out further tasks, e.g. understanding the role of protozoan species diversity in the natural environment.

O manto polar antártico retêm informação paleoclimatologica por entres suas camadas de neve e gelo. O gelo antártico tem revelado a base de dados paleoclimática de maior resolução para os últimos 800 mil anos. Os padrões de transporte atmosférico refletem a composição e a fonte do particulado encontrado na neve e no gelo do continente Antártico. Estando relacionado a processos climáticos, as características desse transporte alteram em quantidade e qualidade as espécies químicas que se depositam sobre o manto de gelo. Dessa forma, o estudo dos depósitos de particulado ao longo das camadas de neve/gelo na Antártica pode sugerir mudanças nos padrões de transporte atmosférico. Atualmente a comunidade científica discute as diferenças de padrões climáticos entre o leste e o oeste antártico. Enquanto de forma geral observa-se instabilidade no setor oeste, o clima da antártica oriental demonstra relativa estabilidade climática. Neste estudo, analisamos dois testemunhos de gelo recente de duas regiões com características climáticas diferentes do continente Antártico. No Platô Detroit situado na Península Antártica (6410′S/0600′O), analisamos a variabilidade de Black Carbon (BC) ao longo de 20 metros de neve. O BC encontrado na Península Antártica apresentou baixas concentrações comparáveis as encontradas no gelo do Artico período pré-industrial. Nossos resultados sugerem que sua variabiliade corresponde à sazonalidade dos períodos de queimada nos continentes do Hemisfério Sul. No interior do continente Antártico, analisamos o particulado em geral por um processo de microanálise ao longo de um testemunho de 40 metros extraído em Mont Johns (79o55′S/09423′O). Encontramos uma tendência negativa na deposição de poeira mineral (AlSi) entre 1967 e 2007. Nossos resultados sugerem que esta tendência seja resultado de um crescente isolamento atmosférico da região central do continente antártico pelo aumento da intensidade dos ventos ao redor da Antártica. Este aumento na intensidade dos ventos reflete por sua vez o resfriamento da alta atmosfera no centro antártico causado pela depleção da camada de ozônio na região. Adicionalmente, amostras de diferentes microambientes de Patriot Hills (8018′S/08121′O) foram coletadas de maneira asséptica para análise microbiológica. As amostras foram cultivadas em meio R2 e paralelamente o DNA total extraído foi sequenciado pela técnica de pirosequenciamento. Os resultados preliminares desta analise mostram grande riqueza de espécies dos mais variados grupos. Os resultados deste trabalho caracterizam três diferentes parâmetros relacionados a deposição atmosférica em duas áreas pouco exploradas e de grande interesse científico do continente antártico.

Lipoproteins are responsible for cholesterol traffic in humans. Low density lipoprotein (LDL) delivers cholesterol from liver to peripheral tissues. A misleading delivery can lead to the formation of atherosclerotic plaques. LDL has a single protein, apoB-100, that binds to a specific receptor. It is known that the failure associated with a deficient protein-receptor binding leads to plaque formation. ApoB-100 is a large single lipid-associated polypeptide difficulting the study of its structure. IR spectroscopy is a technique suitable to follow the different conformational changes produced in apoB-100 because it is not affected by the size of the protein or the turbidity of the sample. We have analyzed LDL spectra of different individuals and shown that, even if there are not big structural changes, a different pattern in the intensity of the band located around 1617 cm 21 related with strands embedded in the lipid monolayer, can be associated with a different conformational rearrangement that could affect to a protein interacting region with the receptor.
