Naïve FoxP3-expressing regulatory T-cells (Tregs) are essential to control immune responses via continuous replenishment of the activated-Treg pool with thymus-committed suppressor cells. The mechanisms underlying naïve-Treg maintenance throughout life in face of the age-associated thymic involution remain unclear. We found that in adults thymectomized early in infancy the naïve-Treg pool is remarkably well preserved, in contrast to conventional naïve CD4 T-cells. Naïve-Tregs featured high levels of cycling and pro-survival markers, even in healthy individuals, and contrasted with other circulating naïve/memory CD4 T-cell subsets in terms of their strong γc-cytokine-dependent signaling, particularly in response to IL-7. Accordingly, ex-vivo stimulation of naïve-Tregs with IL-7 induced robust cytokine-dependent signaling, Bcl-2 expression, and phosphatidylinositol 3-kinase (PI3K)-dependent proliferation, whilst preserving naïve phenotype and suppressive capacity. Altogether, our data strongly implicate IL-7 in the thymus-independent long-term survival of functional naïve-Tregs, and highlight the potential of targeting the IL-7 pathway to modulate Tregs in different clinical settings.

On several occasions since 2001 Vladimir Putin has raised the concept of ‘Greater Europe’, a partly-integrated common space comprising mainly Russia and the European Union. This concept has never been recast into a detailed political programme. While it has been championed as‘a Europe without dividing lines’, the concept would in practice permanently split Europe into two geopolitical blocs – the Western bloc of the European Union, with Germany in the dominant role, and the Eastern bloc, consisting of the emerging Eurasian Union, with Russia in a hegemonic position. In recent years Russia has undertaken a number of initiatives aimed at implementing some elements of the concept. However, most of these have failed to become reality. In this context, we should expect Russia’s policy to focus on implementing its priority project of Eurasian integration, based on the structures of the Customs Union/the Eurasian Union. The Greater Europe project, on the other hand, will be postponed until the time when, as Moscow believes, a weakened EU will be ready to accept Russian proposals.

Pressing scientific questions concerning the Greenland ice sheet's climatic sensitivity, hydrology, and contributions to current and future sea level rise require hydrological datasets to resolve. While direct observations of ice sheet meltwater losses can be obtained in terrestrial rivers draining the ice sheet and from lake levels, few such datasets exist. We present a new dataset of meltwater river discharge for the vicinity of Kangerlussuaq, Southwest Greenland. The dataset contains measurements of river stage and discharge for three sites along the Akuliarusiarsuup Kuua (Watson) River's northern tributary, with 30 minute temporal resolution between June 2008 and August 2010. Additional data of water temperature, air pressure, and lake water depth and temperature are also provided. Discharge data were measured at sites with near-ideal properties for such data collection. Regardless, high water bedload and turbulent flow introduce considerable uncertainty. These were constrained and quantified using statistical techniques, thereby providing a high quality dataset from this important site. The greatest data uncertainties are associated with streambed elevation change and measurements. Large portions of stream channels deepened according to statistical tests, but poor precision of streambed depth measurements also added uncertainty. Quality checked data are freely available for scientific use as supplementary online material.
