The goal of this work was to establish the origins of the chemical engineering course at the National School of Chemistry of the University of Brazil (at present, the School of Chemistry of the Federal University of Rio de Janeiro). There was a previous Industrial Chemistry course at the Higher School of Agriculture and Veterinary, established in 1920, following the French model. This course was the basis of the creation of the National School of Chemistry in 1933. During the 1940s, teachers and students claimed for a new course in true connection with the needs of the country and to reach full valorization of their job. The structure of the new Chemical Engineering course was approved in 1946 and the first class begun in 1952. This course was of great importance during the Brazilian industrial development during the 1950s and 1960s.

The development of new tools for chemoinformatics, allied to the use of different algorithms and computer programmes for structure elucidation of organic compounds, is growing fast worldwide. Massive efforts in research and development are currently being pursued both by academia and the so-called chemistry software development companies. The demystification of this environment provoked by the availability of software packages and a vast array of publications exert a positive impact on chemistry. In this work, an overview concerning the more classical approaches as well as new strategies on computer-based tools for structure elucidation of organic compounds is presented. Historical background is also taken into account since these techniques began to develop around four decades ago. Attention will be paid to companies which develop, distribute or commercialize software as well as web-based and open access tools which are currently available to chemists.

A sample of hydroxyapatite was synthesized and its crystalline structure was analyzed by X-ray diffraction by means of the Rietveld method. Two functions were used to fit the peak profiles, modified Voigt (TCHZ) and Pearson VII. The occupational factors and lattice parameters obtained by both models show that the sample does not contain relevant cationic substitutions. The interatomic distances from Ca1 to oxygens O1, O2 and O3 were adequate for a pure hydroxyapatite without defect at site Ca1. Besides, the use of multiple lines in planes (300) and (002) associated with the model Pearson VII resulted in good agreement with the TCHZ model with respect to the size-strain effectswith an ellipsoidal shape of crystallites. In conclusion, the procedures adopted in the synthesis of hydroxyapatite produced a pure and crystalline material. The experimental results of transmission electron microscopy confirmed the predicted shape of crystals.

This work presents three operationally simple laboratory protocols for monocrystal growth of small-molecule organic compounds, which have been applied with success in the last ten years for the formation of single crystals for X-ray structural studies. In addition, five structure hints were formulated as general guidelines for selecting a small-molecule organic compound as a candidate for monocrystal growth: molecular weight >200 D, melting point >100 ºC, two or more aromatic rings in the structure, at least two sites for intermolecular hydrogen bond formation, and a halogen or other heavy atom in the structure.

In this study, novel Chitosan/PVA based films were chemically crosslinked by glutaraldehyde, under pH=(4,00 ±0,05), in order to achieve structures tailored for wound tissue engineering applications. Both precursors and developed films were characterized by FTIR, SEM and XRD in order to determine the presence of chemicals groups and nanostructural order, respectively. The results have shown that the glutaraldehyde crosslinking have altered the crystallinity of pure chitosan and the increase on the C=N bands and simultaneous decrease on NH2 bands suggested that Chitosan/GA crosslinking has preference to occur in carbon-2 of the saccharide ring by the Schiff's base reaction. Also, FTIR spectroscopy clearly showed that crosslinking has also taken place with blends of PVA and chitosan. The mechanical properties presented high degree dependence with on the increase of the content of chitosan and glutaraldehyde. The results have indicated that, by controlling the ratio [PVA]/[chitosan] in the blends and the extent of chemical crosslinking, it was possible to tailor the hybrid network produced aiming to obtain properties of interest for the specific application.

Several reasons motivated the development of new generations of antibiotics, such as their high ability to develop resistance to virtually all kinds of anti-infective agents and the crescent market demand for new drugs to treat special demanding patients. After penicillin discovery, several antibiotics were developed from fungal metabolites, since antibacterial secondary metabolites consists on a fungal endogenous protective mechanism against natural competitors. The aim of this review is to present the structural diversity of antibacterial and antifungal metabolites produced by fungi, mentioning sources of fungal isolates, cultivation process and details on the scope of their antibiotic activity.

This work deals with an evaluation of an experimental application about polarimetry for pharmacy and food engineering courses. Foods obtained from the undergraduate students were used for demonstrating multidisciplinary concepts and these concepts were associated to the teaching of polarimetry. According to the results, the benefits of the contextualization are beyond the class and the undergraduating students became interested in control of quality of foods. From these results, it can be concluded that the experimental emphasis given is valid and creates motivation and interest for learning physico-chemistry, in comparison with the traditional methodology applied to teach polarimetry.

The NMR (RMN¹H, 13C, COSY, HMQC, HMBC, NOE-DIFF, NOESY) and mass spectra data analysis of sixteen flavonoids, including nine natural, 7-O-methylkanferol (ramnocitrin), 3,7-di-O-methylkanferol (kumatakenin), 3-O-methylquercetin, 3,7,3',4'-tetra-O-methylquercetin (retusin), 3,7,8,4'-tetra-O-methylgossipetin, 3,7,8,3',4'-penta-O-methylgossipetin, 7-O-methylapigenin (genkwanin), 3,7,8-tri-O-methylherbacetin, 7,4'- di-O-methylquercetin (ombuine), isolated from Solanum paludosum and S. jabrense, and seven prepared methyl and acetyl derivatives, are discussed according the substitution on the rings A, B and C.

The reaction of 4-(phenyl)thiosemicarbazide with isatin yielded a new ligand, isatin-3-(N4-benzylthiosemicarbazone). Isatin-3-(N4-benzylthiosemicarbazone) deprotonated in ethanol/KOH reacts with an ethanolic solution of Hg(NO3)2 to give a mercury complex. The compounds were characterized by IR and X-ray single crystal structure determination. The X-ray studies revealed that the complex possesses a tetrahedral geometry with two deprotonated thiosemicarbazone ligands coordenated. The ligand and its mercury complex crystallize in the monoclinic (P2(1)/c) and triclinic (P-1) crystal system, respectively.

The knowledge of the structure characteristic of the Organic Matter is important for the understanding of the natural process. In this context aquatic humic substances (principal fraction) were isolated from water sample collected from the two distinct rivers, using procedure recommended for International Humic Substances Society and characterized by elemental analysis, electron paramagnetic resonance and nuclear magnetic resonance (13C NMR). The results were interpreted using principal component analysis (PCA) and the statistical analyses showed different in the structural characteristics of the aquatic humic substances studied.

Three samples of hydrotalcite-like materials (HTC) were synthesized and their structural characteristics were compared with two HTCs obtained commercially. Thermal analyses, FT-IR, PXRD and textural analyses were used to investigate the structural differences between commercial and synthetic samples. Particularly, the memory effect was observed at temperature higher than 600 ºC. The Rietveld refinements were obtained with expressive accuracy and the statistical parameters of goodness of fit are quite satisfactory. In conclusion, the procedures adopted in synthesis of HTC produced crystalline materials with high surface area materials.

In this work, a novel device based on polyacrylamide (PAAm) hydrogels and KL - DeOH - H2O lyotropic liquid crystal (LLC), with potential for application as Polymer Dispersed Liquid Crystals (PDLC), was proposed and its properties characterized. The confinement of LLC promoted significant changes in spectroscopic, morphological, optical, hydrophilic, structural and mechanical properties due to the interaction between the LLC-PAAm matrix and entropic parameter changes. The mechanical and structural properties can be controlled by adjusting AAm, temperature and presence of LLC, which can be useful for technological applications of these systems in optical devices.

O estudo do efeito da complexidade estrutural da fonte de nitrogênio no transporte de amônio em Saccharomyces cerevisiae foi realizado cultivando-se o microrganismo em um meio mínimo contendo glicose e fontes de nitrogênio, variando de um simples sal de amônio (sulfato de amônio) a aminoácidos livres (casaminoácidos) e peptídeos (peptona). O transporte de amônio foi avaliado acompanhando-se a entrada do análogo metilamônio, utilizando duas metodologias diferentes: transporte de metilamônio radioativo e efluxo de potássio acoplado ao transporte de metilamônio em células crescidas em diferentes condições de cultivo. A cinética de transporte de amônio é detectada nos meios contendo peptona e amônio e não no meio suplementado com casaminoácidos, e o transporte medido em diferentes fases de crescimento sugere que o processo é mais estável em células crescidas em peptona. Os resultados descritos neste trabalho indicam que a complexidade estrutural interfere com a expressão do transportador do íon amônio e que a complementação do meio de cultura com uma fonte de nitrogênio na forma de peptídeos é a mais eficiente não só para a expressão do transportador de amônio, mas também de conferir maior estabilidade ao processo.

O cultivo das helicônias vem sendo afetado pela murcha, causada por Fusarium oxysporum f.sp. cubense. Este trabalho objetivou identificar fontes de resistência, verificar a detecção da resistência através de método alternativo e avaliar a lignificação como mecanismo de defesa do hospedeiro ao patógeno. As espécies utilizadas na identificação de resistência foram Heliconia bihai, H. psittacorum cvs. Golden Torch e Golden Torch Adrian, H. rostrata, H. stricta cvs. Capri e Fire Bird, H. psittacorum cvs. Sassy e Alan Carle, H. caribea, H. latispatha, H. wagneriana e H. chartacea cv. Sexy Pink. A avaliação dos sintomas foi realizada aos 40 dias após a inoculação baseada em escala de notas variando de 1 a 6. As espécies consideradas resistentes foram H. bihai, H. psittacorum cvs. Golden Torch e Golden Torch Adrian, H. rostrata, H. stricta cv. Capri, H. psittacorum cv. Sassy e H. caribea. O método alternativo de detecção de resistência consistiu na utilização de filtrado fúngico obtido a partir do cultivo em meio Czapek, utilizando várias concentrações do mesmo depositando em folhas destacadas das cultivares resistentes e suscetíveis H. psittacorum cvs. Golden Torch e Alan Carle, respectivamente. A avaliação foi feita após 48 horas de incubação, onde a concentração em 50% do filtrado foi a mais eficiente na distinção da resistência. O mecanismo estrutural foi observado em secções histológicas nas raízes das espécies utilizadas no estudo de resistência, inoculadas com o método de injeção e não inoculadas, que permitiram verificar a ausência de relação entre a resistência e a lignificação.
