BACKGROUND: Pseudomonas aeruginosa is the major pathogen associated with chronic and ultimately fatal lung infections in patients with cystic fibrosis (CF). To investigate how P. aeruginosa-derived vesicles may contribute to lung disease, we explored their ability to associate with human lung cells. RESULTS: Purified vesicles associated with lung cells and were internalized in a time- and dose-dependent manner. Vesicles from a CF isolate exhibited a 3- to 4-fold greater association with lung cells than vesicles from the lab strain PAO1. Vesicle internalization was temperature-dependent and was inhibited by hypertonic sucrose and cyclodextrins. Surface-bound vesicles rarely colocalized with clathrin. Internalized vesicles colocalized with the endoplasmic reticulum (ER) marker, TRAPalpha, as well as with ER-localized pools of cholera toxin and transferrin. CF isolates of P. aeruginosa abundantly secrete PaAP (PA2939), an aminopeptidase that associates with the surface of vesicles. Vesicles from a PaAP knockout strain exhibited a 40% decrease in cell association. Likewise, vesicles from PAO1 overexpressing PaAP displayed a significant increase in cell association. CONCLUSION: These data reveal that PaAP promotes the association of vesicles with lung cells. Taken together, these results suggest that P. aeruginosa vesicles can interact with and be internalized by lung epithelial cells and contribute to the inflammatory response during infection.

En el presente escrito, se reportan los resultados de un trabajo de investigación a nivel licenciatura, el cual se centró en el estudio de comportamientos gráficos en funciones algebraicas y trigonométricas, específicamente en f(x)=x , f(x)=x^2 ,f(x)=x^3 , f(x)=sen(x) y f(x)=cos(x), así como las transformaciones de cada una, considerando la expresión Y=Cf(ax+c)+D, con la intención de realizar comparaciones gráficas entre las funciones originales y las transformadas, el propósito general fue analizar si la presentación de funciones algebraicas y trigonométricas en diversos contextos (algebraico, visual, numérico y gráfico), permite al estudiante identificar comportamientos análogos y relacionar éstos con transformaciones gráficas. De acuerdo a los resultados obtenidos, concluimos que el estudiante al producir sus propias gráficas, éste logra identificar por si mismo comportamientos análogos entre las gráficas algebraicas y trigonométricas, además, el uso de diferentes registros de representación coadyuva al desarrollo de dichos resultados.

The purpose of the present study was to use attenuated total reflectance-Fourier transform infrared spectroscopy (ATR-FTIR) and target factor analysis (TFA) to investigate the permeation of model drugs and formulation components through Carbosil® membrane and human skin. Diffusion studies of saturated solutions in 50:50 water/ethanol of methyl paraben (MP), ibuprofen (IBU) and caffeine (CF) were performed on Carbosil® membrane. The spectroscopic data were analysed by target factor analysis, and evolution profiles of the signal for each component (i.e. the drug, water, ethanol and membrane) over time were obtained. Results showed that the data were successfully deconvoluted as correlations between factors from the data and reference spectra of the components, were above 0.8 in all cases. Good reproducibility over three runs for the evolution profiles was obtained. From the evolution profiles it was observed that water diffused better through the Carbosil® membrane than ethanol, confirming the hydrophilic properties of the Carbosil® membrane used. IBU diffused slower compared with MP and CF. The evolution profile of CF was very similar to that of water, probably because of the high solubility of CF in water, indicating that both compounds are diffusing concurrently. The second part of the work involved a study of the evolution profiles of the components of a commercial topical gel containing 5% (w/w) of ibuprofen as it permeated through human skin. Although the system was much more complex, data were still successfully deconvoluted and the different components of the formulation identified except for benzyl alcohol which might be attributed to the low concentrations of benzyl alcohol used in topical formulations. The U&I programme Critical Friends (CFs) are developing guidelines on the role of the Critical Friend and the way in which this links with U&I programme model, projects and outputs. The critical friends are also in the process of building a new online community of shared effective practice for current and future critical friends. The CF Benefits Realisation project aims to synthesise existing CF U&I, JISC Curriculum Design and Delivery, JISC Institutional Innovation and related programmes, activities, methodologies and approaches to produce a range of specialist guidelines and other outputs for effective CF practice, within the context of the aims and objectives of the JISC U&I programme. We aim to disseminate these to a wide range of interests within the JISC HE-FE communities, following consultation.
