The psychrotrophic Antarctic alga, Chlorella vulgaris NJ-7, grows under an extreme environment of low temperature and high salinity. In an effort to better understand the correlation between fatty acid metabolism and acclimation to Antarctic environment, we analyzed its fatty acid compositions. An extremely high amount of Delta(12) unsaturated fatty acids was identified which prompted us to speculate about the involvement of Delta(12) fatty acid desaturase in the process of acclimation. A full-length cDNA sequence, designated CvFAD2, was isolated from C. vulgaris NJ-7 via reverse transcription polymerase chain reaction (RT-PCR) and RACE methods. Sequence alignment and phylogenetic analysis showed that the gene was homologous to known microsomal Delta(12)-FADs with the conserved histidine motifs. Heterologous expression in yeast was used to confirm the regioselectivity and the function of CvFAD2. Linoleic acid (18:2), normally not present in wild-type yeast cells, was detected in transformants of CvFAD2. The induction of CvFAD2 at an mRNA level under cold stress and high salinity is detected by real-time PCR. The results showed that both temperature and salinity motivated the upregulation of CvFAD2 expression. The accumulation of CvFAD2 increased 2.2-fold at 15A degrees C and 3.9-fold at 4A degrees C compared to the alga at 25A degrees C. Meanwhile a 1.7- and 8.5-fold increase at 3 and 6% NaCl was detected. These data suggest that CvFAD2 is the enzyme responsible for the Delta(12) fatty acids desaturation involved in the adaption to cold and high salinity for Antarctic C. vugaris NJ-7.

In the course of a screening program, we have isolated the new natural product, 5,7-dihydroxy-5,6,7,8-tetrahydroazocin-2(IH)-one (1), from the staurosporine producing marine-derived Streptomyces sp. strain QD518. Here we report the isolation and structure elucidation of 1 and the artifacts 3 and 4 resulting from I by acid catalyzed intra- and inter-molecular reactions.

Eolian flux in the Chinese Loess Plateau was reconstructed by measuring the dry bulk density and CaCO3 content of the late Cenozoic loess-paleosol-red clay sequences in the Lingtai profile. Comparison of eolian flux variation between the Lingtai profile and the ODP sites 885/886 in the North Pacific shows a significant wet-dry variability in addition to a gradual drying trend in the dust source regions in interior Asia. Especially, the increase of eolian fluxes from both continental and pelagic eolian sediments indicates a sharp drying of the dust source regions between 3.6 and 2.6 MaBP, which might be attributed to the tectonic uplift of the Tibetan Plateau, which cut down the moisture input to the interior Asia. The average value and variability of eolian flux are higher after 2.6 MaBP than before, which may be related to the Quaternary climatic fluctuations on the glacial-interglacial timescale after the commencement of major Northern Hemisphere Glaciations. The eolian fluxes of the Lingtai profile and Core V21-146 in northwest Pacific show a synchronous variation on the 10(4)-10(5) a timescale, indicating that the flux variations from both continental and marine records are closely correlated to the Quaternary climatic fluctuation forced by the ice volume changes on a global scale.

本课题组自1999年以来,将培养条件优化、生物活性跟踪及化学跟踪技术应用到胶州湾海洋放线菌的次级代谢产物研究中,发现了一批具有生物活性的新结构化合物。 本研究从青岛胶州湾分离出各类海洋微生物423株,包括海洋放线菌287株,海洋细菌116株,海洋真菌20株,对胶州湾海洋微生物的资源和分布规律有了基本的认识。对其中海洋放线菌的次级代谢产物进行了活性和化学筛选,获得了它们对八种病原微生物的抑制活性数据。发现胶州湾海洋放线菌对至少一种受试微生物具有中等以上拮抗能力的比例为50%。 从三株海洋放线菌的发酵粗提物中鉴定出7个新结构化合物和31个已知化合物。具体是:①从菌株M518中鉴定出21个化合物,包括3个新结构天然化合物:N-Foramidostaurosporine,5,7-Dihydroxy-5,6,7,8-tetrahydro-1H-azocin-2-one,Selina-4(14),7(11)-diene-8,9-diol和1个新结构衍生物Trimer 3。其中化合物5,7-Dihydroxy-5,6,7,8-tetrahydro-1H-azocin-2-one为新骨架类型的天然化合物。② 从菌株M491 中鉴定出10个化合物,包括3个新结构化合物10α-14-Dihydroxyamorph-4-en-3-one,10α,11-Dihydroxyamorph-4-ene,5α,10α,11-Triydroxyamorphan-3-one; 并首次从微生物中分离得到已知化合物10α-Hydroxyamorph-4-en-3-one;③从菌株M311中鉴定出7个已知化合物。 发现天然化合物5,7-Dihydroxy-5,6,7,8-tetrahydro-1H-azocin-2-one很容易发生重排反应,分离出了两个重排反应产物,并得出该化合物可能的重排反应过程及三聚体形成的方式。 生物活性实验结果表明新型星形孢菌素N-Formamidostaurosporine具有很强的抗肿瘤活性:对于37株人类肿瘤细胞的平均IC50,IC70和IC90分别为0.016µg/ml,0.171µg/ml和2.352µg/ml。对37株肿瘤细胞株的抗肿瘤选择性为27%。还首次发现Staurosporine类化合物具有抑制微藻生长的活性。 对产生新化合物的菌株M518 和M491 进行了形态、生理生化及分子鉴定,提交两株菌的16S rDNA 序列到GenBank(DQ184649 和DQ184648)。结合形态、生理生化特征及系统 发育树分析表明:菌株M518 属于链霉菌属,并可能属于该属中的粉红孢类群;菌株M491属于链霉菌属,并可能属于该属中的青色类群。

A new lead(II) phosphonate, Pb[(PO3)(2)C(OH)CH3]center dot H2O (1) was hydrothermally synthesized and characterized by IR, elemental analysis, UV, TGA, SEM, and single crystal X-ray diffraction analysis. X-ray crystallographic study showed that complex 1 has a two-dimensional double layered hybrid structure containing interconnected 4- and 12-membered rings and shows an unusual (5,5)-connected (4(7) . 6(3)) (4(8) .6(2)) topology. 针对我国青藏高原草地大面积退化及由此引发的一系列生态环境问题,从土壤生态功能恢复和区域可持续发展的角度出发,将原生高寒嵩草草甸封育系统作为对照,研究了土地退化对土壤碳氮含最的影响,检验了不同人工重建措施(3个人工种植处理:混播、松耙单播、翻耕单播和1个自然恢复处理)对土壤碳含量的相对影响程度。研究结果如下:原生植被封育处理每平方米土壤平均碳、氮含量分别为7.47kg和0.647kg,而重度退化地碳、氮含量分别为3.67和0.448kg•m^-2.可以推算,由于土地退化而造成的土壤(0-20cm层)碳氮丢失量分别为3.80kg•m^-2和0.199kg•m^-2,即高寒草甸土地退化导致0~20cm土壤层中50.87%的有机碳和30.75%的氮流失,可以看出高寒草甸土壤退化后流失的碳比氮多;混播处理、松耙单播处理、翻耕单播处理和自然恢复处理土壤单位面积有机碳含量分别是原牛植被土壤有机碳的70.5%,69.0%,49.0%和80%,单位面积氮含量分别是原生植被土壤全氮的86.9%,88.7%,71.1%和91.7%。但是,与重度退化地相比,除翻耕单播处理外,其它恢复重建措施均能部分恢复系统的碳氮含量,因此,将重度退化地进行自然恢复或松耙混播重建多年生植被可以作为系统固定碳(碳汇)的一个途径。

调查草毡寒冻雏形土生物量及土壤有机质,利用涡度相关技术观测该区域作用层与大气CO2通量.结果表明:地下90%生物量集中于0~10 cm的表土层,年总净初级生产量约935.0 g/m2;土壤有机质含量在6.401~7.060%之间;净CO2通量呈明显的日变化和季节变化规律;5月中旬到9月底为CO2的净吸收(780 g CO2/m2),其中以7月最高,净吸收量明显高于非生长季的,10月到翌年5月初CO2的净排放量(383 g CO2/m2);全年固定碳高达397 g/m2.

络石藤为夹竹桃科络石属植物络石藤Trachelospernum jisminoides(Lindl.)Lem.的干燥带叶藤茎,有祛风通络、凉血消肿之功,主治风湿热痹、筋脉拘挛、腰膝酸痛等症。主要成分为黄酮、木质素类化合物。王杰松等的研究表明,芹菜素类化合物具有骨关节保护作用。因此,本研究采用反相高效液相色谱法测定其中的主要黄酮——芹菜素7-O-β-新橙皮糖苷(AN),为络石藤质量标准的制定提供依据。

目的通过观察藏药7号水提液对大鼠离体胸动脉条收缩作用的影响,研究藏药7号的降压机制.方法以生理盐水作为对照组,观察藏药7号水提液(6 g/L)和维拉帕米(Ver 0.013 g/L)对高K+液引起的主动脉条收缩的时效影响,观察对KCl、NE及CaCl2引起的大鼠主动脉条收缩的量效曲线的影响,以及对NE引起的依赖于细胞内钙及细胞外钙收缩的影响.结果藏药7号水提液抑制高K+液引起的主动脉收缩(P<0.001);而且可以使KCl、NE及CaCl2引起的大鼠主动脉条收缩的量效曲线非平行右移,最大效应降低,呈非竞争性拮抗作用(P<0.05);与维拉帕米相似,并且对NE引起的依赖于细胞内钙及细胞外钙的收缩均有抑制作用(P<0.05).结论提示藏药7号的降压机制与钙离子通道拮抗剂一致,而且其作用效果比Ver平稳,其最大作用与Ver相近.

To investigate the changes in the chemical weathering intensity of dust source region in Asia continent, ~(87)Sr/~(86)Sr ratios of acid-washed residues from the loess deposits in China and Tajikistan were analyzed, respectively. The results and conclusions are listed as below. 1. The oscillation of ~(87)Sr/~(86)Sr ratios of acid-insoluble residues in the Chinese Loess Plateau was mainly attributed to the chemical weathering intensity of the source region and the grain size. Counteracted the effect of particle size, the calibrated 87Sr/S6Sr ratio can be used as a proxy for the chemical weathering intensity of the source region. 2. The Sr/ Sr ratios of red clay-loess sequence from the Loess Plateau indicate that the chemical weathering intensity of the dust source region between 7.0 and 2.6 Ma is stronger than that in the Quaternary period. This also suggests a general decline in chemical weathering intensity of the source region from 2.6 Ma to the present. Such pattern is more remarkable since 1.0 Ma BP. 3. The ~(87)Sr/~(86)Sr ratios of the Tajik loess during 0.8-1.8 Ma is much more higher than those from 0.8 Ma to the present. This implies that the chemical weathering intensity of the source region in Central Asia is much stronger during 0.8-1.8 Ma than the period since 0.8 Ma. 4. The record of Sr isotope ratios from both sections shows an accelerating course of aridity of the Asian dust source region over the Quaternary period.
