A selection of PBDE congeners was analyzed in pooled blubber samples of pilot whale (Globicephala melas), ringed seal (Phoca hispida), minke whale (Balaenoptera acutorostrata), fin whale (Balaenoptera physalus), harbor porpoise (Phocoena phocoena), hooded seal (Cystophora cristata) and Atlantic white-sided dolphin (Lagenorhynchus acutus), covering a time period of more than 20 years (1986-2009). The analytes were extracted and cleaned-up using open column extraction and multi-layer silica gel column chromatography, and the analysis was performed on a GC-MS system operating in the NCI mode. The highest PBDE levels were found in the toothed whale species pilot whale and white-sided dolphin, and the lowest levels in fin whales and ringed seals. One-sided analyses of variance (ANOVA) followed by Tukey comparisons of means were applied to test for differences between years and sampling areas. Due to inter-year sampling variability, only general comparisons of PBDE concentrations between different sampling areas could be made. Differences in PBDE concentrations between three sampling periods, from 1986 to 2007, were evaluated in samples of pilot whales, ringed seals, white-sided dolphins and hooded seals. The highest PBDE levels were found in samples from the late 1990s or beginning of 2000, possibly reflecting the increase in the global production of technical PBDE mixtures in the 1990s. The levels of BDE #153 and #154 increased relative to the total PBDE concentration in some of the species in recent years, which may indicate an increased relative exposure to higher brominated congeners. In order to assess the effect of measures taken in legally binding international agreements, it is important to continuously monitor POPs such as PBDEs in sub-Arctic and Arctic environments.

According to monitoring data gained between 1982-1992, macrobenthos in the Tiksi Bay is characterized by low indices of the total abundance, biomass and taxonomic diversity. 30 macrobenthic species have been recorded in the Tiksi Bay. The bottom biocenoses within the estuarine-arctic water mass consist of widespread eurybiontic boreal-arctic and brackish-water species. The maximal number of species was observed at a depth of 8.5 m. The maximum biomass was recorded on muddy grounds. The studied bottom fauna is characterized by a high population density (from 1160-600 ind/m**2) and low biomass of 15.5-22.4 g/m**2. The predominant benthic animals of the main Lena River channel 4.7 km upstream Stolb Island are Chironomidae, Plecoptera and Oligochaeta. In total, 48 species of macrobenthos were registered here. In spring the average density of macrozoobenthos in the channel is 680, in summer 770, in autumn 720 and in winter 380 ind/m**2, with the average biomass varying between 2.9 g/m**2 in spring, 7.06 in summer, 4.4 in autumn, and 2.6 in winter.

This article examines a new lightweight, slim, high energy efficient, light-transmitting, selfsupporting envelope system, providing for seamless, free-form designs for use in architectural projects. The system exploits vacuum insulation panel technology. The research was based on envelope components already existing on the market and patents and prototypes built by independent laboratories, especially components implemented with silica gel insulation, as this is the most effective transparent thermal insulation there is today.

This article examines a new lightweight, slim, high energy efficient, light-transmitting, self-supporting envelope system, providing for seamless, free-form designs for use in architectural projects. The system exploits vacuum insulation panel technology. The research was based on envelope components already existing on the market and patents and prototypes built by independent laboratories, especially components implemented with silica gel insulation, as this is the most effective transparent thermal insulation there is today. The tests run on these materials revealed that there is not one that has all the features required of the new envelope model, although some do have properties that could be exploited to generate this envelope, namely, the vacuum chamber of vacuum insulation panels, the use of monolithic aerogel as insulation in some prototypes, and reinforced polyester barriers. These three design components have been combined and tested to design a new, variable geometry, energy-saving envelope system that also solves many of the problems that other studies ascribe to the use of vacuum insulation panels.

La fase de establecimiento del regenerado es un proceso crítico para el desarrollo posterior de la masa tanto por las elevadas tasas de mortalidad que habitualmente lleva asociadas, como por proporcionar el material de partida del que van a disponer las fases subsiguientes. Las restricciones a la germinación y establecimiento de la regeneración del pino silvestre varían enormemente entre las distintas regiones de su extensa área de distribución geográfica. La región Mediterránea constituye un hábitat marginal de la especie en el que las condiciones ecológicas son muy diferentes a las del grueso de su área de distribución. Frente a otras limitaciones (frío, luz, encharcamiento…), en el entorno mediterráneo la tasa de mortalidad parece estar asociada a las condiciones micrometeorológicas del período estival - particularmente, a la sequía -, así como a la presencia excesiva de ganado o ungulados silvestres. No obstante, la mayoría de la información disponible sobre el proceso de regeneración de la especie procede del centro y norte de Europa, por lo que no es de aplicación directa en nuestra región, en la que los estudios de este tipo son mucho más escasos. El presente trabajo pretende contribuir a paliar esta relativa escasez a través del estudio del proceso de regeneración natural en el monte “Cabeza de Hierro”, masa irregular por bosquetes de pino silvestre, paradigma de gestión sostenible y uso múltiple. En este entorno, se pretende caracterizar y cuantificar tanto el proceso de germinación y supervivencia de la especie como la influencia de la cobertura vegetal (estratos arbóreo, arbustivo y herbáceo, y capa de restos vegetales) en su desarrollo. Se persigue así mismo analizar el efecto de la compactación del suelo sobre la persistencia de la masa y contrastar y comparar la eficacia de dos tratamientos edáficos de ayuda a la regeneración: escarificado y decapado+acaballonado. Con este fin se han planteado dos diseños experimentales consistentes en sendas redes de muestreo (Red de Muestreo I o RM I y Red de Muestreo II o RM II) integradas, respectivamente, por 192 y 24 parcelas de 1,5x1,5 m ubicadas bajo distintas condiciones de cobertura vegetal. Sobre una parte de estas parcelas (1/4 en la Red de Muestreo I; 1/2 en la Red de Muestreo II) se han aplicado tratamientos de ayuda a la regeneración (RM I: escarificado; RM II: decapado+acaballonado) y, tras llevar a cabo siembras controladas al inicio del período vegetativo, se han practicado controles periódicos de germinación y supervivencia durante uno (RM II) y tres años consecutivos (RM I). Se han realizado así mismo mediciones complementarias de variables micrometeorológicas, espesura, recubrimiento superficial del suelo y compactación. Los resultados obtenidos a partir de las experiencias realizadas en el monte objeto de estudio permiten concluir que, en relación con el proceso de regeneración natural de la especie en este tipo masa y entorno: 1) la regeneración del pino silvestre durante el primer período vegetativo presenta una tasa de éxito muy baja (1,4% de los sembrados), provocada por una elevada mortalidad durante el primer período estival (>92%) subsiguiente a una germinación de en torno al 17% de las semillas viables que llegan al suelo; 2) la mortalidad sigue siendo elevada hasta el tercer período vegetativo, en que comienza a reducirse significativamente hasta alcanzar el 45%; 3) la cobertura vegetal influye significativamente tanto en el proceso de germinación como en el de supervivencia, aunque ambos procesos presentan una baja correlación linear que pone de manifiesto que los lugares idóneos para la germinación no siempre son los más adecuados para la supervivencia; 4) la escarificación del suelo mejora las tasas iniciales de germinación y supervivencia, pero empeora la tasa de supervivencia posterior (años 2 y 3), por lo que su efecto a medio plazo no resulta significativo; 5) el decapado+acaballonado presenta mejores resultados que el escarificado durante el primer verano, aunque sólo resulta efectivo en condiciones intermedias de espesura de masa; 6) la compactación edáfica no resulta limitante para la productividad ni la persistencia de la masa considerada. ABSTRACT Seedling establishment is critical for later stand progress because it involves high mortality rates and the surviving saplings constitute the starting material for all the subsequent stages. Restrictions for Scots pine germination and seedling survival may vary greatly across its geographical range, as it is widely distributed within north latitudes. Mediterranean region is a marginal sector within this species range and its ecological conditions differ greatly from those of the bulk of the area. Mortality rates in Mediterranean environments seem to be related to summer weather (mainly drought) and high livestock stocking rather than to cold, light or flooding. Most available information on scots pine regeneration process comes from north European experiences and is not transferable to Spanish forests, whereas studies on Mediterranean region are much scarcer. The present work aims at broadening Scots pine regeneration knowledge within Mediterranean region by analyzing its establishment process in the “Cabeza de Hierro” forest: a Scots pine uneven-aged forest at blocklevel scale, exemplary managed for multi-services purpose. Germination and surviving processes are to be characterized and quantified as to vegetation cover both in trees, shrubs, grass and litter strata. Soil compaction effects on forest sustainability are also assessed and the efficacy of some site preparation techniques on regeneration success is tested and compared (scarification vs. scalping+mounding). Two sampling networks comprising respectively 198 (SN I) and 24 plots (SN II) of 1.5x1.5m have been established over a wide range of vegetal cover conditions within the forest. Soil preparation techniques have been applied only to some of the sampling points; namely, 1 out of 4 plots have been scarified within Sampling Network I , while 1 out of 2 plots have been object of scalping & mounding within Sampling Network II. After localized sowing prior to growing season, germination and surviving have been periodically sampled for either one (SN II) or three years (SN I). Supplementary measures for micrometeorological variables, stand density, ground vegetal cover and compaction have also been carried out. Results obtained for the studied forest lead to the following insights regarding Scots pine natural regeneration process within this sort of forest and environment: 1) seedling establishment success rate is quite low (0,15% of sowing seeds), due to high mortality during the first summer (>92%), following a prior 17% rate of germination over viable seeds reaching the soil; 2) mortality rate remains high until the third year after emergence and then decreases to the 50% of surviving; 3) although vegetal cover significantly affects both seedling germination and survival, lineal correlation between those two processes is rather low, which may indicate that places fit for emergence are not necessarily suitable for summer surviving; 4) soil scarification improves both germination and survival during the first growing season, but it is associated to higher mortality rates during the next two years; hence it has no significant medium term effect; 5) scalping & mounding treatment is more effective than scarification concerning establishment improving during the first summer; but its effects are only significant under intermediate stand density levels; 6) soil compaction does not restrict either forest productivity or persistence, despite the area’s long history of high livestock stocking rates and mechanized logging.

Este estudo teve por objetivo analisar como o ser-professor se narra e interpretase como sujeito na profissão, tendo em vista compreender de que maneira esse profissional constrói a identidade pessoal/profissional, sendo capaz de afirmar-se ou negar-se como sujeito dentro do sistema-escola? Trata-se de uma pesquisa realizada com educadores de 1º e 2º ano do ciclo I e II do Ensino Fundamental da Rede Pública de São Bernardo do Campo e Mestrandos em Educação. Optou-se por entrevistar seis educadores, de diferentes áreas de ensino, tornando-os autores desta pesquisa. A abordagem metodológica de cunho qualitativo foi utilizadas priorizando a narrativa dos educadores. Para esta metodologia recorreu-se às contribuições de Maria Isabel Cunha; Lüdke; André; Connelly e Clandini que explicitam sobre a importância de trabalhar com essa metodologia na perspectiva de interpretar e compreender a construção da identidade docente, sobretudo, na linha Formação de Educadores. A temática formação docente e construção da identidade foi trazida à luz dos estudos de Pimenta; Brzezinski; Freire; Vianna; Lane; Sung; Nóvoa; Ciampa, entre outros. No âmbito da análise, foram priorizadas algumas categorias ao longo desta temática: razões que levaram à escolha da profissão; percepções que o professor tem de sujeito na profissão; condições de ser e estar na profissão ; sistema e construção da identidade: condições de se sujeitar para se auto-afirmar como sujeito profissional. No trato destas categorias e, ao longo desta pesquisa, foram consideradas as percepções dos professores, como sendo elementos valiosos para identificar da construção de identidade docente. Estas considerações fundam-se no aporte teórico-metodológico que apresentam grandes contribuições referentes à temática. A partir daí, são estabelecidas relações entre Psicologia Social e Sociologia que possibilitam identificar nas narrativas dos educadores uma insatisfação perante o sistema. Alguns professores partem do próprio sentimento ao narrar todo desconforto que as instituições propiciam-lhes; outros enfatizam de modo objetivo seu cotidiano, contornando e explicitando diferenças na condição de ser estar na profissão .

Previously, we showed that the addition of human erythrocyte glycosphingolipids (GSLs) to nonhuman CD4+ or GSL-depleted human CD4+ cells rendered those cells susceptible to HIV-1 envelope glycoprotein-mediated cell fusion. Individual components in the GSL mixture were isolated by fractionation on a silica-gel column and incorporated into the membranes of CD4+ cells. GSL-supplemented target cells were then examined for their ability to fuse with TF228 cells expressing HIV-1LAI envelope glycoprotein. We found that one GSL fraction, fraction 3, exhibited the highest recovery of fusion after incorporation into CD4+ nonhuman and GSL-depleted HeLa-CD4 cells and that fraction 3 contained a single GSL fraction. Fraction 3 was characterized by MS, NMR spectroscopy, enzymatic analysis, and immunostaining with an antiglobotriaosylceramide (Gb3) antibody and was found to be Gal(α1→4)Gal(β1→4)Glc-Cer (Gb3). The addition of fraction 3 or Gb3 to GSL-depleted HeLa-CD4 cells recovered fusion, but the addition of galactosylceramide, glucosylceramide, the monosialoganglioside, GM3, lactosylceramide, globoside, the disialoganglioside, GD3, or α-galactosidase A-digested fraction 3 had no effect. Our findings show that the neutral GSL, Gb3, is required for CD4/CXCR4-dependent HIV-1 fusion.

A requirement for scaffolding complexes containing internalized G protein-coupled receptors and β-arrestins in the activation and subcellular localization of extracellular signal-regulated kinases 1 and 2 (ERK1/2) has recently been proposed. However, the composition of these complexes and the importance of this requirement for function of ERK1/2 appear to differ between receptors. Here we report that substance P (SP) activation of neurokinin-1 receptor (NK1R) stimulates the formation of a scaffolding complex comprising internalized receptor, β-arrestin, src, and ERK1/2 (detected by gel filtration, immunoprecipitation, and immunofluorescence). Inhibition of complex formation, by expression of dominant-negative β-arrestin or a truncated NK1R that fails to interact with β-arrestin, inhibits both SP-stimulated endocytosis of the NK1R and activation of ERK1/2, which is required for the proliferative and antiapoptotic effects of SP. Thus, formation of a β-arrestin-containing complex facilitates the proliferative and antiapoptotic effects of SP, and these effects of SP could be diminished in cells expressing truncated NK1R corresponding to a naturally occurring variant.

Using both confocal immunofluorescence microscopy and biochemical approaches, we have examined the role of β-arrestins in the activation and targeting of extracellular signal-regulated kinase 2 (ERK2) following stimulation of angiotensin II type 1a receptors (AT1aR). In HEK-293 cells expressing hemagglutinin-tagged AT1aR, angiotensin stimulation triggered β-arrestin-2 binding to the receptor and internalization of AT1aR-β-arrestin complexes. Using red fluorescent protein-tagged ERK2 to track the subcellular distribution of ERK2, we found that angiotensin treatment caused the redistribution of activated ERK2 into endosomal vesicles that also contained AT1aR-β-arrestin complexes. This targeting of ERK2 reflects the formation of multiprotein complexes containing AT1aR, β-arrestin-2, and the component kinases of the ERK cascade, cRaf-1, MEK1, and ERK2. Myc-tagged cRaf-1, MEK1, and green fluorescent protein-tagged ERK2 coprecipitated with Flag-tagged β-arrestin-2 from transfected COS-7 cells. Coprecipitation of cRaf-1 with β-arrestin-2 was independent of MEK1 and ERK2, whereas the coprecipitation of MEK1 and ERK2 with β-arrestin-2 was significantly enhanced in the presence of overexpressed cRaf-1, suggesting that binding of cRaf-1 to β-arrestin facilitates the assembly of a cRaf-1, MEK1, ERK2 complex. The phosphorylation of ERK2 in β-arrestin complexes was markedly enhanced by coexpression of cRaf-1, and this effect is blocked by expression of a catalytically inactive dominant inhibitory mutant of MEK1. Stimulation with angiotensin increased the binding of both cRaf-1 and ERK2 to β-arrestin-2, and the association of β-arrestin-2, cRaf-1, and ERK2 with AT1aR. These data suggest that β-arrestins function both as scaffolds to enhance cRaf-1 and MEK-dependent activation of ERK2, and as targeting proteins that direct activated ERK to specific subcellular locations.
