Durante a transformação maligna dos melanócitos, padrões de metilação do DNA encontram-se alterados, afetando a expressão gênica. As citosinas metiladas (5mC) podem ser hidroxiladas pelas dioxigenases dependentes de 2-oxoglutarato e Fe(II), Tet1, 2 e 3, resultando na formação de 5-hidroximetilcitosina (5hmC) (Globisch et al., 2010; Ko et al., 2010). A 5hmC pode agir tanto como um produto intermediário na desmetilação ativa do DNA quanto na desmetilação passiva ao longo de sucessivas replicações, o que pode contribuir com a dinâmica da metilação do DNA (Tahiliani et al., 2009). Além disso, demonstrou-se que Tet1 e Tet2 são expressas em células tronco embrionárias (Ito et al., 2010) e que 5hmC desempenha importante papel na manutenção e diferenciação destas células (Koh et al., 2011). No entanto, o papel da 5hmC e das proteínas Tet tanto na fisiologia normal quanto em doenças como o câncer ainda não está claro. Utilizando um modelo linear de progressão do melanoma, observamos aumento na expressão gênica de Tet1 nas linhagens celulares correspondendo a melanócitos pré-malignos (4C) e células de melanoma não metastático (4C11-), comparado a melanócitos não tumorigênicos (melan-a) e células de melanoma metastático (4C11+). Em relação à expressão proteica, há um aumento da expressão de Tet1 nas linhagens 4C e 4C11+, o que pode ser relacionado às fases de transição epitélio-mesênquima (EMT) e mesênquima-epitélio (MET), respectivamente. Além disso, constatamos aumento na expressão gênica e proteica de Tet2 na linhagem metastática 4C11+, o que pode indicar diferenças funcionais entre as proteínas Tet1 e Tet2. Também verificamos aumento significativo do conteúdo de 5hmC na linhagem 4C11+, o que se correlaciona com maior expressão gênica e proteica de Tet2. Por fim, linhagens humanas de melanoma metastático menos agressivas apresentaram aumento significativo na expressão dos genes TET1 ...

Photodynamic therapy is a method of healing skin lesions , in addition to radiotherapy and brachytherapy are commonly used in the treatment of skin cancers , but it is still little known in Brazil , consisting of the application to the skin of a drug in the form of cream or lotion ( photosensitizer ) . It is absorbed by tumor cells and subsequently activated by light device , that wavelength of light in the order of 630 nanometers , promoting the selective destruction of cancer cells while preserving healthy cells around the lesion . It can be considered highly effective for treatment of skin cancer types of non-melanoma ( basal cell carcinoma , 85-100 % success in curing [ 3 ] , and Bowen's disease), actinic keratoses , which are precancerous lesions and still no studies that prove also action in the treatment of acne and aging caused by the sun . And the advantage of this treatment is that , when indicated , replace the surgical procedure , selectively treating the diseased cells . Thus, it offers better cosmetic results, reducing the risk of scarring [ 4 ]

Destruction of bone and periodontal ligament as a result of periodontal disease can lead to anatomical defects in the furcation area. Treatment of these lesions is a major challenge to the clinician. Periodontal instruments have limited access to this area and plaque and calculus removal from root surfaces are extremely difficult. For proper treatment planning a number of factors must be taken into consideration to achieve immediate and long term success. Surgical therapy associated with bone grafts may be a viable option in the treatment of class II furcation defects, aiming to restore lost tissues. The aim of this paper is to report a clinical case where a simplified surgical approach with the use of autogenous graft was used to treat a class II furcation defect Twelve months after the surgery, an increase in clinical attachment level and pocket depth reduction resulted in a complete closure of the furcation lesion.

The aim of this study was to evaluate in vitro the sealing ability of the root canal using different techniques: cold lateral compaction, modified Hybrid Tagger, Thermafil and System B. Material and methods. Were used 116 human single-roots, included in resin blocks, sectioned longitudinally, attached to a device with screws and divided into four groups (n=29). After instrumentation using a bur, depression was made in each third of the root canal in each of the halves. Teeth were filled upon the different techniques. For evaluation of the quality, pictures were taken with a 1.5X magnification, photos and radiographs were utilized. After statistical analysis, were performed both macroscopic and radiographic evaluation, by Chi-square test (χ2). Results. The Thermafil technique showed better results in terms of sealing capacity, failures and homogeneity followed by modified Hybrid Tagger, the System B and cold lateral compaction. Conclusions. Thermafil was the technique that shown to be macroscopic, radiographic and homogeneously best over the other techniques evaluated in regard to the filling of the depressions made (AU)

Objectives: The objective of this study was to evaluate the ability of endodontists and general practioners to perform the differential radiographic diagnosis between internal and external root resorptions, using digitized periapical radiographs. Methods: Thirty periapical radiographic images of external and internal resorption were selected. The evaluators were comprised of 15 endodontists and 29 general practitioners who answered a questionnaire with the following question: “What type of resorption in the following radiographics do you diagnose?” (1) Inflammatory external root resorption. (2) Internal root resorption. Statistical analysis was performed based on the Mann-Whitney test, with significance level of 5%. Results: There was no statistically significant difference in the percentage of correct answers from specialists in endodontics and general practitioners. Conclusions: Both groups demonstrated satisfactory skill in the radiographic interpretation of internal and external resorption and there was a higher percentage of correct diagnosis of internal resorption than the external.

Periodontal treatment of teeth with furcation lesions often have unpredictable results due to several factors that may hinder therapy or influence the progress of periodontal disease. Of these, the length of the root trunk may contribute to the early injury of the furcations or even cause complications for the application of the treatment; the opening of the roots can guide the type of treatment to be instituted and cervical enamel projections may hinder the instrumentation of the region or even act as a retention plate niche. Thus, the purpose of this study was measured using a digital caliper, root logs database 400 molar both the 1st and the 2nd and both upper and lower as well as analyze using appropriate index type projection cervical enamel and through appropriate classification the opening of the roots in order to guide the diagnosis and treatment plan. We conclude that there is variation in measures of root trunk between the faces of the teeth examined and that greater cervical enamel projection (ECP) correlates with a decrease in the length of the root trunk in the sample evaluated as the first molars have higher occurrence of roots with a degree of opening larger than the second molars.
