Introduction: A standardized three-dimensional ultrasonographic (3DUS) protocol is described that allows fetal face reconstruction. Ability to identify cleft lip with 3DUS using this protocol was assessed by operators with minimal 3DUS experience. Material and Methods: 260 stored volumes of fetal face were analyzed using a standardized protocol by operators with different levels of competence in 3DUS. The outcomes studied were: (1) the performance of post-processing 3D face volumes for the detection of facial clefts; (2) the ability of a resident with minimal 3DUS experience to reconstruct the acquired facial volumes, and (3) the time needed to reconstruct each plane to allow proper diagnosis of a cleft. Results: The three orthogonal planes of the fetal face (axial, sagittal and coronal) were adequately reconstructed with similar performance when acquired by a maternal-fetal medicine specialist or by residents with minimal experience (72 vs. 76%, p = 0.629). The learning curve for manipulation of 3DUS volumes of the fetal face corresponds to 30 cases and is independent of the operator's level of experience. Discussion: The learning curve for the standardized protocol we describe is short, even for inexperienced sonographers. This technique might decrease the length of anatomy ultrasounds and improve the ability to visualize fetal face anomalies.

Com o objetivo de estudar o efeito de três espaçamentos entre fileiras (30, 40 e 50 cm) e três densidades de semeadura (100, 150 e 200 sementes viáveis/m²) sobre o desenvolvimento da planta, os componentes da produção e a produtividade do arroz irrigado por aspersão até a tensão de reposição de água de -0,070 MPa, foi instalado um experimento em condições de campo, em um Latossolo Vermelho-Escuro, epieutrófico, textura argilosa, em Selvíria, MS. A cultivar avaliada foi a IAC 201. Esta cultivar apresenta suscetibilidade ao acamamento, no sistema de irrigação por aspersão, até uma tensão de reposição de água, no solo, de -0,070 Mpa. O número de colmos e de panículas é incrementado com a redução do espaçamento. A densidade de 100 sementes/m² é a mais indicada para a cultivar IAC 201 irrigada por aspersão, por proporcionar menor gasto de sementes. O espaçamento de 30 cm entre fileiras de plantas proporciona maior produtividade de grãos da cultivar IAC 201 no sistema de irrigação por aspersão.

Fetal MRI reconstruction aims at finding a high-resolution image given a small set of low-resolution images. It is usually modeled as an inverse problem where the regularization term plays a central role in the reconstruction quality. Literature has considered several regularization terms s.a. Dirichlet/Laplacian energy, Total Variation (TV)- based energies and more recently non-local means. Although TV energies are quite attractive because of their ability in edge preservation, standard explicit steepest gradient techniques have been applied to optimize fetal-based TV energies. The main contribution of this work lies in the introduction of a well-posed TV algorithm from the point of view of convex optimization. Specifically, our proposed TV optimization algorithm or fetal reconstruction is optimal w.r.t. the asymptotic and iterative convergence speeds O(1/n2) and O(1/√ε), while existing techniques are in O(1/n2) and O(1/√ε). We apply our algorithm to (1) clinical newborn data, considered as ground truth, and (2) clinical fetal acquisitions. Our algorithm compares favorably with the literature in terms of speed and accuracy.

We describe herein some immunological properties of human fetal bone cells recently tested for bone tissue-engineering applications. Adult mesenchymal stem cells (MSCs) and osteoblasts were included in the study for comparison. Surface markers involved in bone metabolism and immune recognition were analyzed using flow cytometry before and after differentiation or treatment with cytokines. Immunomodulatory properties were studied on activated peripheral blood mononuclear cells (PBMCs). The immuno-profile of fetal bone cells was further investigated at the gene expression level. Fetal bone cells and adult MSCs were positive for Stro-1, alkaline phosphatase, CD10, CD44, CD54, and beta2-microglobulin, but human leukocyte antigen (HLA)-I and CD80 were less present than on adult osteoblasts. All cells were negative for HLA-II. Treatment with recombinant human interferon gamma increased the presence of HLA-I in adult cells much more than in fetal cells. In the presence of activated PBMCs, fetal cells had antiproliferative effects, although with patterns not always comparable with those of adult MSCs and osteoblasts. Because of the immunological profile, and with their more-differentiated phenotype than of stem cells, fetal bone cells present an interesting potential for allogeneic cell source in tissue-engineering applications.

Trisomy 13 was detected in 10% of mouse embryos obtained from pregnant females which were doubly heterozygous for Robertsonian chromosomes involving chromosome 13. The developing dorsal root ganglia and spinal cords were examined in trisomy 13 and littermate control mice between days 12 and 18 of gestation (E12-18). The overall size of the dorsal root ganglia and number of ganglion cells within a given ganglion were not altered, but the number of neurons immunoreactive for calbindin and calretinin was reduced. The trisomic spinal cord was reduced in size with neurons lying in a tightly compact distribution in the gray matter. In trisomic fetuses, the extent of the neuropil of the spinal cord was reduced, and may represent a diminished field of interneuronal connectivity, due to reduced arborization of dendritic processes of the neurons present, particularly of calbindin-immunostained neurons. Furthermore, the subpopulation of calretinin-immunoreactive neurons and axons was also reduced in developing trisomic gray and white matter, respectively. Thus, overexpression of genes on mouse chromosome 13 exerts a deleterious effect on the development of neuropil, affecting both dendritic and axonal arborization in the trisomy 13 mouse. The defect of calbindin or calretinin expression by subsets of dorsal root ganglion or spinal cord neurons may result from deficient cell-to-cell interactions with targets which are hypoplastic.

Weakening of cardiac function in patients with heart failure results from a loss of cardiomyocytes in the damaged heart. Cell replacement therapies as a way to induce myocardial regeneration in humans could represent attractive alternatives to classical drug-based approaches. However, a suitable source of precursor cells, which could produce a functional myocardium after transplantation, remains to be identified. In the present study, we isolated cardiovascular precursor cells from ventricles of human fetal hearts at 12 weeks of gestation. These cells expressed Nkx2.5 but not late cardiac markers such as α-actinin and troponin I. In addition, proliferating cells expressed the mesenchymal stem cell markers CD73, CD90, and CD105. Evidence for functional cardiogenic differentiation in vitro was demonstrated by the upregulation of cardiac gene expression as well as the appearance of cells with organized sarcomeric structures. Importantly, differentiated cells presented spontaneous and triggered calcium signals. Differentiation into smooth muscle cells was also detected. In contrast, precursor cells did not produce endothelial cells. The engraftment and differentiation capacity of green fluorescent protein (GFP)-labeled cardiac precursor cells were then tested in vivo after transfer into the heart of immunodeficient severe combined immunodeficient mice. Engrafted human cells were readily detected in the mouse myocardium. These cells retained their cardiac commitment and differentiated into α-actinin-positive cardiomyocytes. Expression of connexin-43 at the interface between GFP-labeled and endogenous cardiomyocytes indicated that precursor-derived cells connected to the mouse myocardium. Together, these results suggest that human ventricular nonmyocyte cells isolated from fetal hearts represent a suitable source of precursors for cell replacement therapies.

Prenatal ultrasound can often reliably distinguish fetal anatomic anomalies, particularly in the hands of an experienced ultrasonographer. Given the large number of existing syndromes and the significant overlap in prenatal findings, antenatal differentiation for syndrome diagnosis is difficult. We constructed a hierarchic tree of 1140 sonographic markers and submarkers, organized per organ system. Subsequently, a database of prenatally diagnosable syndromes was built. An internet-based search engine was then designed to search the syndrome database based on a single or multiple sonographic markers. Future developments will include a database with magnetic resonance imaging findings as well as further refinements in the search engine to allow prioritization based on incidence of syndromes and markers.

O objetivo deste trabalho foi avaliar os efeitos da compactação do solo sobre suas propriedades morfoestruturais e sobre o desenvolvimento radicular da soja, sob sistema plantio direto, em um experimento conduzido num Latossolo Vermelho distroférrico, em Londrina, PR. O delineamento utilizado foi o de blocos ao acaso, com parcelas subdivididas e quatro repetições. Foram constituídos dois tratamentos de manejo nas parcelas: um em solo escarificado à profundidade de 20-25 cm, com densidade do solo de 1,17 Mg m-3; e outro em solo compactado, a 12 cm de profundidade, e densidade de 1,38 Mg m-3. Nas subparcelas, foram semeadas duas cultivares de soja, Embrapa-4 e BR-16. A presença de camadas mais compactas no perfil do solo provocou o desenvolvimento radicular em zonas de menor resistência, em poros biológicos e nas fissuras do solo. A produção de grãos não apresentou diferença entre os tratamentos, graças à adequada disponibilidade hídrica durante o período avaliado. A cultivar Embrapa-4 apresentou maior crescimento radicular em estruturas compactas, comparada à cultivar BR-16.

We report a case of abdominal eventration associated with cystic fibrosis, diagnosed by mid-trimester ultrasonography. The defect concerned the abdominal muscles and their aponevrotic sheath, but respected the skin. There was no associated malformation. The outcome was favorable after surgery, and the infant is well at the age of 6 months.

The aim of this study was to culture human fetal bone cells (dedicated cell banks of fetal bone derived from 14 week gestation femurs) within both hyaluronic acid gel and collagen foam, to compare the biocompatibility of both matrices as potential delivery systems for bone engineering and particularly for oral application. Fetal bone cell banks were prepared from one organ donation and cells were cultured for up to 4 weeks within hyaluronic acid (Mesolis(®)) and collagen foams (TissueFleece(®)). Cell survival and differentiation were assessed by cell proliferation assays and histology of frozen sections stained with Giemsa, von Kossa and ALP at 1, 2 and 4 weeks of culture. Within both materials, fetal bone cells could proliferate in three-dimensional structure at ∼70% capacity compared to monolayer culture. In addition, these cells were positive for ALP and von Kossa staining, indicating cellular differentiation and matrix production. Collagen foam provides a better structure for fetal bone cell delivery if cavity filling is necessary and hydrogels would permit an injectable technique for difficult to treat areas. In all, there was high biocompatibility, cellular differentiation and matrix deposition seen in both matrices by fetal bone cells, allowing for easy cell delivery for bone stimulation in vivo. Erythrina falcata Benth. tem sido usada na recuperação de ecossistemas degradados, como planta ornamental e em sistemas agroflorestais. Entretanto, a produção de mudas por sementes é difícil. O objetivo deste trabalho foi avaliar a propagação vegetativa de E. falcata utilizando estacas caulinares provenientes de árvores adultas (estacas herbáceas, semilenhosas e de rebrota) e mudas, coletadas em quatro épocas do ano e o efeito do ácido indolbutírico (AIB) no enraizamento de estacas dessa espécie. Depois de preparadas, as estacas foram imersas em ácido indolbutírico (AIB, 0, 1,5 e 3 g L-1). O material se desenvolveu em tubetes de plástico (55 mL), em casa de vegetação climatizada, a uma temperatura de 25 a 30ºC e umidade relativa do ar acima de 80%, em substrato de vermiculita granulometria média. Maior porcentagem de enraizamento (73%), comprimento das quatro maiores raízes (46 mm) e número de raízes (6,2) foram obtidos em estacas de mudas coletadas no verão. Não houve enraizamento em estacas semilenhosas, provenientes de árvores adultas. A imersão das estacas em AIB não exerceu qualquer influência no enraizamento. As estacas de mudas coletadas no verão são as de melhor propagação vegetativa em virtude de melhor porcentagem de enraizamento e sobrevivência.

For tissue engineering, several cell types and tissues have been proposed as starting material. Allogenic skin products available for therapeutic usage are mostly developed with cell culture and with foreskin tissue of young individuals. Fetal skin cells offer a valuable solution for effective and safe tissue engineering for wounds due to their rapid growth and simple cell culture. By selecting families of genes that have been reported to be implicated in wound repair and particularly for scarless fetal wound healing including transforming growth factor-beta (TGF-beta) superfamily, extracellular matrix, and nerve/angiogenesis growth factors, we have analyzed differences in their expression between fetal skin and foreskin cells, and the same passages. Of the five TGF-beta superfamily genes analyzed by real-time reverse transcription-polymerase chain reaction, three were found to be significantly different with sixfold up-regulated for TGF-beta2, and 3.8-fold for BMP-6 in fetal cells, whereas GDF-10 was 11.8-fold down-regulated. For nerve growth factors, midkine was 36-fold down-regulated in fetal cells, and pleiotrophin was 4.76-fold up-regulated. We propose that fetal cells present technical and therapeutic advantages compared to foreskin cells for effective cell-based therapy for wound management, and overall differences in gene expression could contribute to the degree of efficiency seen in clinical use with these cells.

A fotoinibição pode ser intensificada por fatores de estresse como alta temperatura. O objetivo deste trabalho foi determinar os efeitos da luminosidade e temperatura na fotossíntese e na recuperação da fotoinibição sob baixa irradiância em mogno (Swietenia macrophylla King) e acariquara (Minquartia guianensis Aubl.). As plantas cresceram sob baixa irradiância: 0,20 (acariquara) e 1,43 mol m-2 dia-1 (mogno). A fotossíntese líquida e as características da fluorescência foram avaliadas em duas irradiâncias, 1.000 e 1.700 mmol m-2 s-1 , e duas temperaturas foliares, 32 e 38ºC. A 32ºC, a fotossíntese máxima (Amáx) foi maior em mogno do que em acariquara. Em mogno, a exposição a 38ºC causou redução gradativa na fotossíntese, sendo a taxa fotossintética após 50 min similar à encontrada em acariquara. A 32ºC, o aumento na irradiância, de 1.000 para 1.700 mmol m-2 s-1 , intensificou a fotoinibição, mas Amáx não foi alterada. Em 1.700 mmol m-2 s-1, o aumento da temperatura, de 32 para 38ºC, não afetou a fotoinibição. Após 72 horas em baixa irradiância, a recuperação da fotoinibição foi de 80% em acariquara e 89% em mogno. Não houve efeito da temperatura da folha na fotoinibição.

O trabalho teve como objetivo avaliar a distribuição de formas de cobre, em função do pH e da adição de cama-de-frango, em amostras da camada superficial (0 a 10 cm) de um Neossolo Litólico distrófico típico e de um Cambissolo Húmico alumínico típico, da região da Serra Gaúcha, cultivados por mais de 15 anos com parreirais que receberam aplicações sucessivas de produtos fitossanitários à base de cobre. Para avaliar os efeitos da variação do pH e do teor de matéria orgânica do solo nas formas de cobre, cinco doses de HNO3 (50, 100, 200, 300 e 400 mmol kg-1) e quatro doses de cama-de-frango em base seca (10, 20, 40 e 80 t ha-1) foram adicionadas aos solos. As frações observadas foram: trocável; ligada à matéria orgânica; ligada a óxidos de Fe, de Al e de Mn; e fração residual. A maior proporção do cobre dos solos esteve ligada à fração orgânica (entre 70 e 80%). Com a diminuição do pH (de 6,8 para 3,8), os teores de cobre trocável aumentaram, enquanto os teores de cobre ligados às frações matéria orgânica, óxidos e residual diminuíram. A adição de cama-de-frango não afetou significativamente a distribuição das formas de cobre nos solos, com exceção dos teores trocáveis.

The chemical functionalization of cell-surface proteins of human primary fetal bone cells with hydrophilic bioorthogonal intermediates was investigated. Toward this goal, chemical pathways were developed for click reaction-mediated coupling of alkyne derivatives with cellular azido-expressing proteins. The incorporation via a tetraethylene glycol linker of a dipeptide and a reporter biotin allowed the proof of concept for the introduction of cell-specific peptide ligands and to follow the reaction in living cells. Tuning the conditions of the click reaction resulted in chemical functionalization of living human fetal osteoblasts with excellent cell survival.
