A produção de H2S (sulfeto de hidrogênio) é um dos principais problemas na indústria do petróleo, sendo esta uma das causas da corrosão de tanques de estocagem e tubulações. Essa produção é possibilitada devido à injeção de água do mar durante o processo de recuperação secundária do petróleo, onde alguns micro-organismos presentes nessa água, tais como as bactérias anaeróbias heterotróficas totais (BANHT) e as bactérias redutoras de sulfato (BRS), que promovem a redução do sulfato a sulfeto. Atualmente, o método de quantificação destes micro-organismos é a técnica do Número Mais Provável (NMP) que estima o resultado em aproximadamente 28 dias. No presente trabalho foi utilizada a metodologia da produção semi-contínua de sulfeto biogênico pelo período de 15 dias, com o intuito de correlacionar com os resultados das quantificações de BANHT e BRS através da técnica convencional do NMP. Foram analisadas amostras de diferentes procedências da indústria do petróleo, apresentando variação na salinidade de 0 a 16 g.L-1. O objetivo deste procedimento foi avaliar as velocidades específicas e instantâneas de produção de H2S, sugerindo, desta forma, quais amostras apresentam maior potencial para a produção biogênica de sulfeto e em quais condições essa produção se dá. Observou-se que em todas as amostras a geração do H2S se dá de forma crescente até a estabilização desta produção, sendo esta obtida quase sempre em seis dias (144h) do crescimento microbiano. A produção do sulfeto biogênico se deu de forma mais intensa nas amostras do fundo de tanque de estocagem de óleo e da água de formação. A quantificação das BANHT e das BRS foram avaliadas pelo método do NMP de acordo com a tabela de Harrigan, a qual subestima a população microbiana, desconsiderando erros provenientes da técnica

Fungal infection was observed in Catla catla and Labeo rohita cultured in two private fish farms. The later stage of the infection resulted in ulcerations followed by haemorrhage on the dorsal surface of the body. Initially, usual treatments of copper sulphate, potassium permanganate and common salt solution were tried, but no improvement was observed. Then repeated intramuscular injections of homeopathic drug Heaper Sulpher and Arnica spray were given with encouraging results. Infection reported in another farm was also successfully controlled using a similar treatment.

Oxygen consumption in Oreochromis mossambicus, Peters (3-60g in weight) was measured under different stress conditions at a constant temperature of 20±1°C. The rate of oxygen consumption was significantly higher (0.170 ml gˉ¹hˉ¹)at a salinity of 30x10ˉ³ compared with that (0.132ml gˉ¹hˉ¹) in freshwater. The oxygen consumption was also found to be affected by changes in pH. Weight specific rate decreased significantly from 0.113 to 0.045 ml gˉ¹hˉ¹ with increasing body weight. A positive correlation was recorded between availability of dissolved oxygen and the rate of oxygen consumption by the fish. While copper sulphate and malachite green inhibited the respiratory metabolism, formaldehyde treatment raised it from 0.088 to 0.118ml gˉ¹hˉ¹.

The present study deals with the water quality and ichthyofauna of Sadatpur lake. Monthly changes in water quality parameters such as temperature, pH, dissolved oxygen (DO), total alkalinity, chlorides, sulphates, phosphates and nitrites have been studied for a period of one year beginning from January 2005 and were investigated to assess the suitability of this lake for fish and fisheries practices. All the physico-chemical parameters determined, revealed that the fluctuations in water temperature, pH, dissolved oxygen, alkalinity and nitrites, were within the desirable limits. On the other hand, chloride and sulphate were lower whereas phosphate was higher than the desirable limits. Altogether 24 fish species belonging to 17 genera and 2 orders were found to be present in the lake. Among fish species family Cyprinidae dominated the lake. The lake subsistence fishery served the need of local fishermen community.

Aspartate aminotransferase (E.C. 2.6.1.1.) from the skeletal muscle of fresh water fish Cirrhina mrigala has been purified 40 fold by ammonium sulphate fractionation, adsorption on alumina Csub(8) gel and chromatography using DEAE-cellulose column and the properties of the purified enzyme studied. The pH optimum of the enzyme is 7.8. The Km value of aspartic acid and 2-oxoglutaric acid are found to be 2.8 x 10sub(-3) M and 1.0 x 10sub(-4) M respectively. The activity of enzyme is inhibited by p-chloromercurybenzoate, hydroxylamine hydrochloride and sodium cyanide. The inhibition by pchloromercurybenzoate is reversed by reduced glutathione, B-mercaptoethanol and cysteine. Dicarboxylic acids such as maleic acid, malic acid and succinic acid inhibit the enzyme activity. The enzyme is not activated by any of the metal ions tested and heavy metal ions such as mercury and silver strongly inhibit the enzyme activity.

Phosphorylase from muscle of tilapia (Tilapia mosambica) was extracted by water and purified by ammonium sulphate precipitation, centrifugation and repeated recrystallisation. Electro-phorogram of the enzyme preparation showed a single band near origin. The enzyme showed optimum pH and temperature at 6.1 and 37°C respectively. Glucose and glucose-6-phosphate were found to be competitive inhibitors of the enzyme. Maltose and starch acted as primers for the phosphorylase reaction like glycogen.

Sediment and water samples were collected from mangrove and estuarine biotopes at fortnightly intervals. The physico-chemical characters of the overlying water were studied. In the mangrove biotope maximum temperature (31.5°C) and in the estuarine biotope maximum salinity (35.6‰) were recorded during the summer season, whereas in post-monsoon period the sulphate content was increased to 516 p.p.m. and the pH was reduced to 7.4. Invariably both in the enriched sediment and water samples four major peaks (at wavelengths 460, 705, 772 and 850 nm) and two minor peaks (at wavelengths 580 and 663 nm) of absorption spectra were noticed. A pure culture of Chromatium sp., isolated from mangroves sediment, showed three peaks of absorption spectra at wavelengths, 500, 580 and 850 nm. The effect of sodium chloride on the growth of Chromatium sp., was also studied and it was observed that maximum growth occurred in the range 1-3% sodium chloride concentration. This isolate was also capable of utilizing various sulphur and carbon compounds. Glycerol and glucose did not show any specific effect whereas pyruvate, malate and acetate increased the growth.

Thirty-three groups of geochemically related natural lake and river waters collected from 368 sites scattered over East and West Africa, were investigated to determine correlations between their various ions and the patterns of change in their ionic composition across the African continent. The following nine significant correlations were found to exist in the entire area or in a major part of it: sodium and potassium; sodium and chloride; sodium and sulphate; potassium and ammonium; potassium and chloride; ammonium and nitrate; calcium and magnesium; magnesium and chloride; sulphate and chloride. When compared with the world average composition. The concentration of potassium in these waters was found to be high, that of calcium and nitrate was low. With respect to the area of East Africa only, trends in the pattern of change involving the ionic composition of the lake and river water and the longitude at which the sites were present, were found to exist for the ratio of sodium: conductivity (which showed an increase in the direction of West to East), magnesium: conductivity, and for the sum of calcium and magnesium: phosphate was generally found to increase in East Africa in the direction of west to East.

In May, 1971, Lake Mahega had pronounced mesothermy (40.W C at one metre). Solar heating of a bloom of baeteria and the blue-green alga, Synechococcus bacillaris Butch., probably caused the high temperature. A total ionic concentration gradient increasing from 192,600 mg 1itre-1 at the surface to 415,200 mg•litre- 1 at three metres stabilized the thermally inverted water. Nearly equal amounts of chloride and sulphate accounted for about 90% of the anionic composition. Sodium was the major cation. Crystals or the triple salt, northupite (Na2 CO3. MgCO3. NaCl) and of thenardite (Na2SO4) were mixed with the surface sediment. We believe it is possible that primary northupite depnsition is occurring. Lake Mahega is also the first mesothermic, sulphato-chloride lake reported for East Africa.

Many unicellular green algae can become yellow or red in various natural habitats due to mass accumulation of a secondary carotenoid, such as lutein, or astaxanthin. The accumulation of secondary carotenoids is generally thought to be a survival strategy of the algae under photo-oxidative stress or other adverse environmental conditions. The physiological role of the carotenoids in stress response is less well understood at the subcellular or molecular level. In this study, a stable astaxanthin overproduction mutant (MT 2877) was isolated by chemical mutagenesis of a wild type (WT) of the green microalga Haematococcus pluvialis Flotow NIES-144. MT 2877 was identical to the WT with respect to morphology, pigment composition, and growth kinetics during the early vegetative stage of the life cycle. However, it had the ability to synthesize and accumulate about twice the astaxanthin content of the WT under high light, or under high light in the presence of excess amounts of ferrous sulphate and sodium acetate. Under stress, the mutant exhibited higher photosynthetic activities than the WT, based on considerably higher chlorophyll fluorescence induction, chlorophyll autofluorescence intensities, and oxygen evolution rates. Cell mortality caused by stress was reduced by half in the mutant culture compared with the WT. Enhanced protection of the mutant against stress is attributed to its accelerated carotenogenesis and accumulation of astaxanthin. Our results suggest that MT 2877, or other astaxanthin overproduction Haematococcus mutants, may offer dual benefits, as compared with the wild type, by increasing cellular astaxanthin content while reducing cell mortality during stress-induced carotenogenesis.

Outer membrane proteins (OMPs) of bacteria are key molecules interacting with the host environment. Flavobacterium columnare, a pathogen-causing columnaris disease of fish worldwide, was studied in order to understand the composition of its OMPs. The sarcosine-insoluble membrane fraction of the OMPs was analysed using sodium dodecyl sulphate polyacrylamide gel electrophoresis (SDS-PAGE) in combination with reverse-phase high-performance liquid chromatography-tandem mass spectrometry (RP-HPLC MS/MS). Thirty-six proteins were identified, including proteins involved in cell wall/membrane biogenesis, specific transport of various nutrients and in essential metabolism. The present study is the first report on the OMPs of F. columnare, and may serve as the basis for understanding the pathogenesis of the bacterium.

A rhabdovirus associated with a lethal hemorrhagic disease in cultured turbot Scophthalm us maximus Linnaeus was isolated. The virus induced typical cytopathogenic effects (CPE) in 9 of 15 fish cell lines examined and was then propagated and isolated from infected carp leucocyte cells (CLC). Electron microscopy observations revealed that the negatively stained virions had a typical bullet-shaped morphology with one rounded end and one flat base end. The bullet-shaped morphology was more obvious and clear in ultrathin sections of infected cells. Experimental infections also indicated that the S. maximus rhabdovirus (SMRV) was not only a viral pathogen for cultured turbot, but also had the ability to infect other fish species, such as freshwater grass carp. A partial nucleotide sequence of the SMRV polymerase gene was determined by RT-PCR using 2 pairs of degenerate primers designed according to the conserved sequences of rhabdovirus polymerase genes. Homology analysis, amino acid sequence alignment, and phylogenetic relationship analysis of the partial SMRV polymerase sequence indicated that SMRV was genetically distinct from other rhabdoviruses. Sodium dodecyl sulphate polyacrylamide gel electrophoresis (SDS-PAGE) analysis of the purified SMRV revealed 5 major structural proteins, and their molecular masses were estimated to be about 250, 58, 47, 42, and 28 kDa. Significant serological reactivity differences were also observed between SMRV and its nearest neighbor, spring viremia of carp virus (SVCV). The data suggest that SMRV is likely a novel fish rhabdovirus, although it is closely related to rhabdoviruses in the genus Vesiculovirus.

The chondroitin AC lyase gene, cslA, was cloned for the first time from the fish bacterial pathogen F. columnare G(4). From the first transcription initiation site, the cslA extends 2620 nucleotides to the end of the 3' region. The open reading frame of cslA transcript has 2286 nucleotides encoding 762 amino acids with a 16 residues long signal peptide at the N-terminus. The gene, cslA was then successfully expressed in Escherichia coli and recombinant chondroitin AC lyase, rChonAC was purified, with its lytic activity analyzed. Zymography analysis copolymerized with chondroitin sulphate revealed the lytic activity of rChonAC and also the crude native ChonAC isolated from periplamic space of cultured F. columnare G(4). The low level of lytic activity observed in crude native ChonAC may be due possibly to the low level of expression of this gene in the cultured condition. The expression and the role of this virulence factor is of interest for further research on the pathogenesis of F. columnare.

PURPOSE: Low inspiratory force in patients with lung disease is associated with poor deagglomeration and high throat deposition when using dry powder inhalers (DPIs). The potential of two reverse flow cyclone prototypes as spacers for commercial carrier-based DPIs was investigated. METHODS: Cyclohaler®, Accuhaler® and Easyhaler® were tested with and without the spacers between 30 and 60 Lmin−1. Deposition of particles in the next generation impactor and within the devices was determined by high performance liquid chromatography. RESULTS: Reduced induction port deposition of the emitted particles from the cyclones was observed due to the high retention of the drug within the spacers (e.g. salbutamol sulphate (SS): 67.89 ± 6.51% at 30 Lmin−1 in Cheng 1). Fine particle fractions of aerosol as emitted from the cyclones were substantially higher than the DPIs alone. Moreover, the aerodynamic diameters of particles emitted from the cyclones were halved compared to the DPIs alone (e.g. SS from the Cyclohaler® at 4 kPa: 1.08 ± 0.05 μm vs. 3.00 ± 0.12 μm, with and without Cheng 2, respectively) and unaltered with increased flow rates. CONCLUSION: This work has shown the potential of employing a cyclone spacer for commercial carrier-based DPIs to improve inhaled drug delivery.
