[EN]The number of eggs obtained in natural spawning (25.60 million) is higher than those obtained by injections (12.9 million) and implants (10.5 million). The number of eggs per female was 12.80 million, in natural spawning, and 4.30 and 3.51 million, in induced with injections and implants, respectively. In number of eggs per spawn, significant differences was observed, between natural spawns (1.11 million), and induced spawning’s (0.44 and 0.21 millions, injected and implants respectively). Significant differences was also found, in number of eggs per Kg female/spawn, between natural spawning’s (56,700 eggs), injection induced (37,200 eggs) and implants (25,200 eggs).

La temperatura influenza molti dei processi fisiologici degli organismi marini e, considerato che la riproduzione dei coralli sembrerebbe essere sensibile agli stress, è necessario comprendere come questa possa reagire ai cambiamenti climatici globali per riuscire a prevedere le future risposte delle popolazioni. Leptopsammia pruvoti (Scleractinia, Dendrophylliidae) è un corallo solitario non zooxantellato presente in Mediterraneo e lungo le coste Atlantiche dal Portogallo alla Gran Bretagna meridionale, dalla superficie fino a 70 metri di profondità.. È un organismo gonocorico con fecondazione interna. In questo lavoro di tesi sono stati analizzati gli aspetti della gametogenesi di L. pruvoti a diverse latitudini per ottenere risultati preliminari riguardanti le possibili correlazioni tra attività riproduttiva e parametri ambientali (temperatura e irradianza). Tale studio si colloca all’interno del progetto europeo sul riscaldamento globale e biologia dei coralli FP7–IDEAS-ERC “Corals and Global Warming: The Mediterranean versus the Red Sea” (CoralWarm). I risultati presentati in questo lavoro sono relativi a cinque popolazioni di L. pruvoti (Genova, Calafuria, Palinuro, Scilla e Pantelleria) disposte lungo un gradiente latitudinale di temperatura e irradianza nel versante occidentale della penisola italiana. I campioni sono stati raccolti mediante campionamenti mensili effettuati tramite immersioni subacquee. Su ogni campione sono state effettuate misurazioni biometriche e analisi cito-istometriche. Nelle popolazioni analizzate i parametri riproduttivi (fecondità, abbondanza, indice gonadico, dimensione) sono stati messi in relazione con la temperatura di fondo (DT, °C) e l’irradianza solare (W/m2) peculiari di ogni sito. L. pruvoti ha mostrato una sessualità gonocorica in tutti i siti considerati. In tutte le popolazioni, la presenza di due stock di ovociti e la distribuzione degli stadi di maturazione degli spermiari durante le fasi di attività riproduttiva, nell’arco dell’anno, hanno permesso di definire un periodo di reclutamento e uno di maturità gonadica, suggerendo che tali eventi siano influenzati dai cambiamenti stagionali della temperatura dell’acqua e del fotoperiodo. Nel periodo di reclutamento, la fecondità presentava una debole correlazione positiva con i parametri ambientali considerati. Al momento della maturità gonadica gli ovociti erano di dimensioni maggiori e meno numerosi rispetto al periodo precedente, nelle popolazioni con una maggiore temperatura e irradianza. Una possibile interpretazione potrebbe essere l’esistenza di un processo di fusione degli ovociti nel periodo di maturità gonadica che sembrerebbe più marcato nelle popolazioni più calde e maggiormente irradiate. La spermatogenesi, diversamente, non ha delineato finora un pattern chiaramente interpretabile. Con i dati attualmente in nostro possesso, non possiamo tuttavia escludere che le differenze riscontrate nelle dimensioni di ovociti e spermiari lungo il gradiente latitudinale considerato siano dovute a un lieve sfasamento nel raggiungimento della maturità gonadica, pur mantenendo la medesima stagionalità nel ciclo riproduttivo delle diverse popolazioni. Ulteriori studi saranno necessari per poter meglio comprendere i complessi meccanismi che regolano e controllano i processi riproduttivi al variare dei parametri ambientali. In questo modo sarà possibile considerare i risulti ottenuti nel contesto dei cambiamenti climatici globali.

In this study, conditions of deposition and stratigraphical architecture of Neogene (Tortonian, 11-6,7Ma) sediments of southern central Crete were analysed. In order to improve resolution of paleoclimatic data, new methods were applied to quantify environmental parameters and to increase the chronostratigraphic resolution in shallow water sediments. A relationship between paleoenvironmental change observed on Crete and global processes was established and a depositional model was developed. Based on a detailed analysis of the distribution of non geniculate coralline red algae, index values for water temperature and water depth were established and tested with the distribution patterns of benthic foraminifera and symbiont-bearing corals. Calcite shelled bivalves were sampled from the Algarve coast (southern Portugal) and central Crete and then 87Sr/86Sr was measured. A high resolution chronostratigraphy was developed based on the correlation between fluctuations in Sr ratios in the measured sections and in a late Miocene global seawater Sr isotope reference curve. Applying this method, a time frame was established to compare paleoenvironmental data from southern central Crete with global information on climate change reflected in oxygen isotope data. The comparison between paleotemperature data based on red algae and global oxygen isotope data showed that the employed index values reflect global change in temperature. Data indicate a warm interval during earliest Tortonian, a second short warm interval between 10 and 9,5Ma, a longer climatic optimum between 9 and 8Ma and an interval of increasing temperatures in the latest Tortonian. The distribution of coral reefs and carpets shows that during the warm intervals, the depositional environment became tropical while temperate climates prevailed during the cold interval. Since relative tectonic movements after initial half-graben formation in the early Tortonian were low in southern central Crete, sedimentary successions strongly respond to global sea-level fluctuation. A characteristic sedimentary succession formed during a 3rd order sea-level cycle: It comprises mixed siliciclastic-limestone deposited during sea-level fall and lowstand, homogenous red algal deposits formed during sea-level rise and coral carpets formed during late rise and highstand. Individual beds in the succession reflect glacioeustatic fluctuations that are most prominent in the mixed siliciclastic-limestone interval. These results confirm the fact that sedimentary successions deposited at the critical threshold between temperate and tropical environments develop characteristic changes in depositional systems and biotic associations that can be used to assemble paleoclimatic datasets.

In questa tesi sono illustrate alcune sperimentazioni finalizzate alla standardizzazione del ciclo produttivo della sogliola comune (Solea solea) in cattività. E’ stato creato un parco di riproduttori selvatici ed è stata standardizzata la riproduzione ad un livello compatibile con la realtà produttiva del settore. Indagini genetiche di assegnazione parentale hanno evidenziato come alcuni esemplari siano stati predominanti negli accoppiamenti e nel conseguente contributo alla generazione della prole. Ciò ha determinato una diminuzione della variabilità genetica dei discendenti. La composizione quali-quantitativa degli acidi grassi delle uova è stata correlata con la sopravvivenza larvale nel corso di un’intera stagione riproduttiva. Tale composizione non ha subito importanti variazioni su scala temporale e sembra essere stata influenzata dall’alimentazione somministrata ai riproduttori nel periodo precedente alla riproduzione. Le analisi di interazione tra momento riproduttivo e qualità delle uova hanno confermato che è stato possibile ottenere uova di buona qualità in termini di sopravvivenza larvale nel corso di tutta la stagione riproduttiva. Larve di sogliola sono state svezzate precocemente 13 giorni dopo la schiusa riducendo l’impiego di cibo vivo a favore di micro diete commerciali. Tale svezzamento ha ridotto le performance di accrescimento, ma non la sopravvivenza e lo sviluppo della metamorfosi quando comparati ad un trattamento standard. La riduzione del cibo vivo ha ottimizzato i costi di produzione e migliorato l’igiene in vasca. L’ontogenesi di precursori di enzimi digestivi è stata determinata tramite PCR quantitativa. I risultati di espressione di tripsinogeno, chimotripsinogeno e amilasi hanno mostrato come tali enzimi rivestano un ruolo chiave nei processi digestivi delle prime fasi larvali. Esemplari giovanili hanno ottenuto un significativo maggiore indice di accrescimento e migliore indice di conversione quando alimentati con diete sperimentali contenenti un elevato tenore proteico. Un aumento dell’incidenza di vacuoli lipidici a livello epatico è stato osservato all’aumentare del tenore proteico della dieta.

The motivating problem concerns the estimation of the growth curve of solitary corals that follow the nonlinear Von Bertalanffy Growth Function (VBGF). The most common parameterization of the VBGF for corals is based on two parameters: the ultimate length L∞ and the growth rate k. One aim was to find a more reliable method for estimating these parameters, which can capture the influence of environmental covariates. The main issue with current methods is that they force the linearization of VBGF and neglect intra-individual variability. The idea was to use the hierarchical nonlinear model which has the appealing features of taking into account the influence of collection sites, possible intra-site measurement correlation and variance heterogeneity, and that can handle the influence of environmental factors and all the reliable information that might influence coral growth. This method was used on two databases of different solitary corals i.e. Balanophyllia europaea and Leptopsammia pruvoti, collected in six different sites in different environmental conditions, which introduced a decisive improvement in the results. Nevertheless, the theory of the energy balance in growth ascertains the linear correlation of the two parameters and the independence of the ultimate length L∞ from the influence of environmental covariates, so a further aim of the thesis was to propose a new parameterization based on the ultimate length and parameter c which explicitly describes the part of growth ascribable to site-specific conditions such as environmental factors. We explored the possibility of estimating these parameters characterizing the VBGF new parameterization via the nonlinear hierarchical model. Again there was a general improvement with respect to traditional methods. The results of the two parameterizations were similar, although a very slight improvement was observed in the new one. This is, nevertheless, more suitable from a theoretical point of view when considering environmental covariates.

My PhD project was focused on Atlantic bluefin tuna, Thunnus thynnus, a fishery resource overexploited in the last decades. For a better management of stocks, it was necessary to improve scientific knowledge of this species and to develop novel tools to avoid collapse of this important commercial resource. To do this, we used new high throughput sequencing technologies, as Next Generation Sequencing (NGS), and markers linked to expressed genes, as SNPs (Single Nucleotide Polymorphisms). In this work we applied a combined approach: transcriptomic resources were used to build cDNA libreries from mRNA isolated by muscle, and genomic resources allowed to create a reference backbone for this species lacking of reference genome. All cDNA reads, obtained from mRNA, were mapped against this genome and, employing several bioinformatics tools and different restricted parameters, we achieved a set of contigs to detect SNPs. Once a final panel of 384 SNPs was developed, following the selection criteria, it was genotyped in 960 individuals of Atlantic bluefin tuna, including all size/age classes, from larvae to adults, collected from the entire range of the species. The analysis of obtained data was aimed to evaluate the genetic diversity and the population structure of Thunnus thynnus. We detect a low but significant signal of genetic differentiation among spawning samples, that can suggest the presence of three genetically separate reproduction areas. The adult samples resulted instead genetically undifferentiated between them and from the spawning populations, indicating a presence of panmictic population of adult bluefin tuna in the Mediterranean Sea, without different meta populations.

The present-day climate in the Mediterranean region is characterized by mild, wet winters and hot, dry summers. There is contradictory evidence as to whether the present-day conditions (“Mediterranean climate”) already existed in the Late Miocene. This thesis presents seasonally-resolved isotope and element proxy data obtained from Late Miocene reef corals from Crete (Southern Aegean, Eastern Mediterranean) in order to illustrate climate conditions in the Mediterranean region during this time. There was a transition from greenhouse to icehouse conditions without a Greenland ice sheet during the Late Miocene. Since the Greenland ice sheet is predicted to melt fully within the next millennia, Late Miocene climate mechanisms can be considered as useful analogues in evaluating models of Northern Hemispheric climate conditions in the future. So far, high resolution chemical proxy data on Late Miocene environments are limited. In order to enlarge the proxy database for this time span, coral genus Tarbellastraea was evaluated as a new proxy archive, and proved reliable based on consistent oxygen isotope records of Tarbellastraea and the established paleoenvironmental archive of coral genus Porites. In combination with lithostratigraphic data, global 87Sr/86Sr seawater chronostratigraphy was used to constrain the numerical age of the coral sites, assuming the Mediterranean Sea to be equilibrated with global open ocean water. 87Sr/86Sr ratios of Tarbellastraea and Porites from eight stratigraphically different sampling sites were measured by thermal ionization mass spectrometry. The ratios range from 0.708900 to 0.708958 corresponding to ages of 10 to 7 Ma (Tortonian to Early Messinian). Spectral analyses of multi-decadal time-series yield interannual δ18O variability with periods of ~2 and ~5 years, similar to that of modern records, indicating that pressure field systems comparable to those controlling the seasonality of present-day Mediterranean climate existed, at least intermittently, already during the Late Miocene. In addition to sea surface temperature (SST), δ18O composition of coral aragonite is controlled by other parameters such as local seawater composition which as a result of precipitation and evaporation, influences sea surface salinity (SSS). The Sr/Ca ratio is considered to be independent of salinity, and was used, therefore, as an additional proxy to estimate seasonality in SST. Major and trace element concentrations in coral aragonite determined by laser ablation inductively coupled plasma mass spectrometry yield significant variations along a transect perpendicular to coral growth increments, and record varying environmental conditions. The comparison between the average SST seasonality of 7°C and 9°C, derived from average annual δ18O (1.1‰) and Sr/Ca (0.579 mmol/mol) amplitudes, respectively, indicates that the δ18O-derived SST seasonality is biased by seawater composition, reducing the δ18O amplitude by 0.3‰. This value is equivalent to a seasonal SSS variation of 1‰, as observed under present-day Aegean Sea conditions. Concentration patterns of non-lattice bound major and trace elements, related to trapped particles within the coral skeleton, reflect seasonal input of suspended load into the reef environment. δ18O, Sr/Ca and non-lattice bound element proxy records, as well as geochemical compositions of the trapped particles, provide evidence for intense precipitation in the Eastern Mediterranean during winters. Winter rain caused freshwater discharge and transport of weathering products from the hinterland into the reef environment. There is a trend in coral δ18O data to more positive mean δ18O values (–2.7‰ to –1.7‰) coupled with decreased seasonal δ18O amplitudes (1.1‰ to 0.7‰) from 10 to 7 Ma. This relationship is most easily explained in terms of more positive summer δ18O. Since coral diversity and annual growth rates indicate more or less constant average SST for the Mediterranean from the Tortonian to the Early Messinian, more positive mean and summer δ18O indicate increasing aridity during the Late Miocene, and more pronounced during summers. The analytical results implicate that winter rainfall and summer drought, the main characteristics of the present-day Mediterranean climate, were already present in the Mediterranean region during the Late Miocene. Some models have argued that the Mediterranean climate did not exist in this region prior to the Pliocene. However, the data presented here show that conditions comparable to those of the present-day existed either intermittently or permanently since at least about 10 Ma.

Paracentrotus lividus è un echinoderma bentonico marino appartenente alla classe Echinoidea, comune in tutto il Mar Mediterraneo e lungo le coste atlantiche dalla Scozia al Marocco. Questa specie è estremamente importante dal punto di vista ecologico in quanto specie chiave nella strutturazione della comunità bentonica macroalgale. Inoltre in tempi recenti il mercato delle sue gonadi ha subito una forte espansione rendendolo una specie di interesse economico in numerosi paesi europei. Il crescente prelievo sta gradualmente portando al depauperamento della risorsa, per questo motivo il riccio di mare è oggetto di numerosi studi e programmi a fini conservazionistici. Una maggiore conoscenza del ciclo riproduttivo della specie potrebbe risultare utile per una migliore gestione della risorsa. Pertanto, questo studio si propone di indagare sulla biologia di P. lividus in due aree distinte della Sardegna: Su Pallosu, nella Penisola del Sinis Sardegna centro-occidentale, e nell’Area Marina Protetta di Tavolara – Punta Coda Cavallo, Sardegna nordorientale, al fine di far luce sulle dinamiche del ciclo riproduttivo. Le informazioni bibliografiche a riguardo evidenziano l’influenza dei fattori ambientali quali fotoperiodo, idrodinamismo, temperatura e tipologia di pascolo sull’andamento del ciclo biologico e il momento di spawning. Diversi autori hanno osservato in Mar Mediterraneo sia un unico ciclo riproduttivo che due eventi riproduttivi annuali a conferma dell’ampia variabilità dell’andamento gonadico. Nel presente lavoro al fine di mettere in relazione le caratteristiche dell’habitat con il ciclo riproduttivo delle popolazioni in oggetto è stato analizzato l’andamento dell’Indice Gonadosomatico (IGS) su individui di diverse taglie/età, come rapporto fra il peso delle gonadi e il peso dell’individuo; inoltre a conferma dello stadio di maturazione è stata effettuata l’analisi istologica delle gonadi su campioni fissati e inclusi in paraffina, sezionati a 7μm e colorati con ematossilina-eosina prima di essere osservati al microscopio ottico. Mediante analisi d’immagine di foto subacquee utilizzando il programma Seascape è stata caratterizzata la componente macroalgale bentonica e la componente abiotica degli habitat (sabbia e roccia non vegetata) con particolare attenzione al calcolo della percentuale di ricoprimento delle specie vegetali. Inoltre i siti sono stati caratterizzati rispetto all’esposizione dei venti dominanti con elaborazione dei dati del periodo di campionamento per direzione, frequenza e intensità.

L’anguilla europea, è una specie eurialina catadroma con un complesso ciclo biologico: l’area di riproduzione, unica, si trova molto distante da quella di distribuzione. La specie necessita di una gestione dello stock a fini conservazionistici. Il problema è europeo: lo stock è unico, distribuito in Europa e nell’Africa settentrionale, si riproduce in Atlantico ed è panmittico. C’è preoccupazione per il declino del reclutamento e delle catture di adulti. Lo scopo del progetto è di individuare possibili unità di stock nella penisola italiana. La ricerca è basata sullo studio degli otoliti mediante analisi morfometrica e microchimica. I contorni degli otoliti sono sottoposti ad analisi ellittica di Fourier per individuare eventuali gruppi. Gli otoliti sono stati levigati per effettuare: letture d’età, indagini microstrutturali al SEM delle fasi larvali, analisi microchimiche LA-ICP-MS del nucleo, studiarne l’origine e valutare l’ambiente di sviluppo. Le indagini morfometriche mostrano evidenti pattern ontogenetici, ma non legati ocorrelati alla località, sesso o anno di nascita. Le indagini microstrutturali hanno evidenziano l’alto contenuto organico nucleare, un pattern comune di crescita ed eventi chiave delle fasi larvali, con una media di 212 anelli giornalieri. La microchimica rivela che le larve si sviluppano in acque salate fino alla metamorfosi, poi migrano verso acque meno salate. Le analisi su campioni nati nello stesso anno, evidenziano due gruppi: individui di rimonta naturale e individui di ripopolamento. I profili nucleo bordo evidenziano la permanenza a salinità intermedie degli adulti. L’attività di ricerca si è dimostrata proficua dal punto di vista tecnico con la messa a punto di protocolli innovativi e con forti ricadute sulla riduzione dei tempi e costi d’analisi. Il debole segnale di possibili unità di stock andrà verificato in futuro mediante analisi più dettagliate discriminando meglio la storia di ogni singolo individuo.

The objective of this theses is to contribute to the wide discussion about the biological control level on the biomineralization operated by calcifying organisms. In particular the intra-crystalline organic matrix associated with different coral species was studied and its role in the process was investigated. The main goals obtained from the research on corals included: (i) the discovery of the species specific role of the intra-crystalline organic matrix molecules in the precipitation of calcium carbonate; (ii) the definition of the role of magnesium ions in the control of the macromolecules assembly/aggregation and in the consequent calcium carbonate polymorphic selectivity; (iii) the discovery that in corals the biomineralization process is not affected by the sea water acidity, as consequence corals are able to construct their skeletons independently from the environmental conditions as far they survive. At the same time, investigations on different kind of vaterite, biogenic and synthetic, were also carried out and confirm the importance of the organism control on the biomineralization process and in particular on the co-existence of different crystalline structures of vaterite for enabling optimization of specific functions, through the employment of OM and acidic macromolecules.

As a large and long-lived species with high economic value, restricted spawning areas and short spawning periods, the Atlantic bluefin tuna (BFT; Thunnus thynnus) is particularly susceptible to over-exploitation. Although BFT have been targeted by fisheries in the Mediterranean Sea for thousands of years, it has only been in these last decades that the exploitation rate has reached far beyond sustainable levels. An understanding of the population structure, spatial dynamics, exploitation rates and the environmental variables that affect BFT is crucial for the conservation of the species. The aims of this PhD project were 1) to assess the accuracy of larval identification methods, 2) determine the genetic structure of modern BFT populations, 3) assess the self-recruitment rate in the Gulf of Mexico and Mediterranean spawning areas, 4) estimate the immigration rate of BFT to feeding aggregations from the various spawning areas, and 5) develop tools capable of investigating the temporal stability of population structuring in the Mediterranean Sea. Several weaknesses in modern morphology-based taxonomy including demographic decline of expert taxonomists, flawed identification keys, reluctance of the taxonomic community to embrace advances in digital communications and a general scarcity of modern user-friendly materials are reviewed. Barcoding of scombrid larvae revealed important differences in the accuracy of the taxonomic identifications carried out by different ichthyoplanktologists following morphology-based methods. Using a Genotyping-by-Sequencing a panel of 95 SNPs was developed and used to characterize the population structuring of BFT and composition of adult feeding aggregations. Using novel molecular techniques, DNA was extracted from bluefin tuna vertebrae excavated from late iron age, ancient roman settlements Byzantine-era Constantinople and a 20th century collection. A second panel of 96 SNPs was developed to genotype historical and modern samples in order to elucidate changes in population structuring and allele frequencies of loci associated with selective traits.

Die neogene Lagos-Portimão Formation (Unter- bis Mittelmiozän) bildet einen Teil der Steilküste der Algarve (S-Portugal) und besteht aus einer zyklischen Wechsellagerung von Karbonaten und Sand-steinen. Die vorliegende Arbeit bietet ein Modell zur sedimentologischen, faziellen und stratigraphischen Entwicklung dieser Einheit an. Basierend auf Profilen entlang der gesamten lateralen Erstreckung der Einheit wurden verschiedene Gelände- und Labormethoden angewandt, um ein Modell entwickeln zu können. Messungen des Sr87/86-Isotopenverhältnisses sollten Klarheit bezüglich der stratigraphischen Position bringen. Die laterale Korrelation der Profile erfolgte über lithologische und fazielle Ansprachen. Unterstützend wurden einzelne Profile mit einem tragbaren Gammaray-Spektrometer gemessen. Es wurden vier Leithorizonte etabliert, die sich durch fazielle Merkmale und spezielle Fossilführung defi-nieren lassen. Die Mikrofazies wurde qualitativ und quantitativ analysiert. Als statistisches Verfahren wurde unter anderem eine hierarchische Clusteranalyse durchgeführt, über welche fünf Biofaziestypen des warm-temperierten Klimabereichs unterschieden werden. Die Fossilführung wird von Mollusken, Bryozoen und Rotalgen dominiert. Ausnahmen bilden stratigraphisch isolierte Vorkommen von kolo-nialen Korallen, die jedoch keine Riffkörper aufbauen. Die Ergebnisse aller zuvor erwähnten Untersuchungen deuten auf Ablagerungen eines nicht-tropischen Hochenergie-Schelfs hin. Sedimentäre Zyklen sind oftmals unvollständig, es treten Hartgründe und Auf-arbeitungs- bzw- Kondesationshorizonte auf. Die geochemische Altersdatierung weist Alterssprünge und -inversionen auf. Ein Vergleich mit dem SW-australischen Schelf und dem von James et al. (1994) eingeführten Modell des shaved shelf bietet sich aufgrund der Ähnlichkeit der Sedimentgesteine und des ozeanographischen Settings an. Weiterhin werden zeitgleiche bzw. faziell ähnliche Becken vergleichend diskutiert. Das Sedimentationsgeschehen der Lagos-Portimão Formation wird maßgeblich durch eine halokinetisch bedingte unregelmäßige Subsidenz und Hebung beeinflußt. Der Salzdom von Albufeira war während der Sedimentation der Einheit mehrfach in Bewegung. Rutschungspakete, Entlastungsspalten und Sanddikes zeugen davon. Die sequenzstratigraphische Interpretation bietet einen neuen Ansatz, in dem sie von Hochstand-Sandsteinen und Tiefstand-Karbonaten ausgeht.

Global climate change is impacting coral reefs worldwide, with approximately 19% of reefs being permanently degraded, 15% showing symptoms of imminent collapse, and 20% at risk of becoming critically affected in the next few decades. This alarming level of reef degradation is mainly due to an increase in frequency and intensity of natural and anthropogenic disturbances. Recent evidence has called into question whether corals have the capacity to acclimatize or adapt to climate changes and some groups of corals showed inherent physiological tolerance to environmental stressors. The aim of the present study was to evaluate mRNA expression patterns underlying differences in thermal tolerance in specimen of the common reef-building coral Pocillopora verrucosa collected at different locations in Bangka Island waters (North Sulawesi, Indonesia). Part of the experimental work was carried out at the CoralEye Reef Research Outpost (Bangka Island). This includes sampling of corals at selected sites and at different depths (3 and 12 m) as well as their experimental exposure to an increased water temperature under controlled conditions for 3 and 7 days. Levels of mRNAs encoding ATP synthase (ATPs) NADH dehydrogenase (NDH) and a 70kDa Heat Shock Protein (HSP70) were evaluated by quantitative real time PCR. Transcriptional profiles evaluated under field conditions suggested an adaptation to peculiar local environmental conditions in corals collected at different sites and at the low depth. Nevertheless, high–depth collected corals showed a less pronounced site-to-site separation suggesting more homogenous environmental conditions. Exposure to an elevated temperature under controlled conditions pointed out that corals adapted to the high depth are more sensitive to the effects of thermal stress, so that reacted to thermal challenge by significantly over-expressing the selected gene products. Being continuously exposed to fluctuating environmental conditions, low-depth adapted corals are more resilient to the stress stimulus, and indeed showed unaffected or down-regulated mRNA expression profiles. Overall these results highlight that transcriptional profiles of selected genes involved in cellular stress response are modulated by natural seasonal temperature changes in P. verrucosa. Moreover, specimens living in more variable habitats (low-depth) exhibit higher basal HSP70 mRNA levels, possibly enhancing physiological tolerance to environmental stressors.

Indo-Pacific region encompasses about 75% of world's coral reefs, but hard coral cover in this region experienced a 32% region-wide decline since 1970s. This great change is primarily ascribable to natural and anthropogenic pressures, including climate change and human activities effects. Coral reef conservation requires management strategies oriented to maintain their diversity and the capacity to provide ecosystem goods and services. Coral reef resilience, i.e. the capacity to recover after disturbances, is critical to their long-term persistence. The aims of the present study were to design and to test field experiments intended to measure changes in recruitment processes, as a fundamental aspect of the coral reef resilience. Recruitment experiments, using artificial panels suspended in the water column, were carried out in two Indo-Pacific locations affected by different disturbances: a new mine in Bangka Island (Indonesia), and the increased sedimentation due to coastal dynamics in Vavvaru Island (Maldives). One (or more) putatively disturbed site(s) was selected to be tested against 3 randomly selected control sites. Panels’ arrangement simulates 2 proximities to living corals, i.e. the sources of propagules: few centimetres and 2 meters over. Panels were deployed simultaneously at each site and left submerged for about five months. Recruits were identified to the lowest possible taxonomic level and recruited assemblages were analysed in terms of percent cover. In general it was not possible to detect significant differences between the benthic assemblages recruited in disturbed and control sites. The high variability observed in recruits assemblages structure among control sites may be so large to mask the possible disturbance effects. Only few taxa showed possible effects of the disturb they undergo. The field tests have highlighted strengths and weaknesses of the proposed approach and, based on these results, some possible improvements were suggested.

Whitefish, genus Coregonus, show exceptional levels of phenotypic diversity with sympatric morphs occurring in numerous postglacial lakes in the northern hemisphere. Here, we studied the effects of human-induced eutrophication on sympatric whitefish morphs in the Swiss lake, Lake Thun. In particular, we addressed the questions whether eutrophication (i) induced hybridization between two ecologically divergent summer-spawning morphs through a loss of environmental heterogeneity, and (ii) induced rapid adaptive morphological changes through changes in the food web structure. Genetic analysis based on 11 microsatellite loci of 282 spawners revealed that the pelagic and the benthic morph represent highly distinct gene pools occurring at different relative proportions on all seven known spawning sites. Gill raker counts, a highly heritable trait, showed nearly discrete distributions for the two morphs. Multilocus genotypes characteristic of the pelagic morph had more gill rakers than genotypes characteristic of benthic morph. Using Bayesian methods, we found indications of recent but limited introgressive hybridization. Comparisons with historical gill raker data yielded median evolutionary rates of 0.24 haldanes and median selection intensities of 0.27 for this trait in both morphs for 1948-2004 suggesting rapid evolution through directional selection at this trait. However, phenotypic plasticity as an alternative explanation for this phenotypic change cannot be discarded. We hypothesize that both the temporal shifts in mean gill raker counts and the recent hybridization reflect responses to changes in the trophic state of the lake induced by pollution in the 1960s, which created novel selection pressures with respect to feeding niches and spawning site preferences.
