Wave energy conversion has an essential difference from other renewable energies since the dependence between the devices design and the energy resource is stronger. Dimensioning is therefore considered a key stage when a design project of Wave Energy Converters (WEC) is undertaken. Location, WEC concept, Power Take-Off (PTO) type, control strategy and hydrodynamic resonance considerations are some of the critical aspects to take into account to achieve a good performance. The paper proposes an automatic dimensioning methodology to be accomplished at the initial design project stages and the following elements are described to carry out the study: an optimization design algorithm, its objective functions and restrictions, a PTO model, as well as a procedure to evaluate the WEC energy production. After that, a parametric analysis is included considering different combinations of the key parameters previously introduced. A variety of study cases are analysed from the point of view of energy production for different design-parameters and all of them are compared with a reference case. Finally, a discussion is presented based on the results obtained, and some recommendations to face the WEC design stage are given.

We present a multichannel tomographic technique to detect fluorescent objects embedded in thick (6.4 cm) tissue-like turbid media using early-arriving photons. The experiments use picosecond laser pulses and a streak camera with single photon counting capability to provide short time resolution and high signal-to-noise ratio. The tomographic algorithm is based on the Laplace transform of an analytical diffusion approximation of the photon migration process and provides excellent agreement between the actual positions of the fluorescent objects and the experimental estimates. Submillimeter localization accuracy and 4- to 5-mm resolution are demonstrated. Moreover, objects can be accurately localized when fluorescence background is present. The results show the feasibility of using early-arriving photons to image fluorescent objects embedded in a turbid medium and its potential in clinical applications such as breast tumor detection.

An Fcα receptor probe of human origin was used to identify novel members of the Ig gene superfamily in mice. Paired Ig-like receptors, named PIR-A and PIR-B, are predicted from sequence analysis of the cDNAs isolated from a mouse splenic library. Both type I transmembrane proteins possess similar ectodomains with six Ig-like loops, but have different transmembrane and cytoplasmic regions. The predicted PIR-A protein has a short cytoplasmic tail and a charged Arg residue in the transmembrane region that, by analogy with the FcαR relative, suggests the potential for association with an additional transmembrane protein to form a signal transducing unit. In contrast, the PIR-B protein has an uncharged transmembrane region and a long cytoplasmic tail containing four potential immunoreceptor tyrosine-based inhibitory motifs. These features are shared by the related killer inhibitory receptors. PIR-A proteins appear to be highly variable, in that predicted peptide sequences differ for seven randomly selected PIR-A clones, whereas PIR-B cDNA clones are invariant. Southern blot analysis with PIR-B and PIR-A-specific probes suggests only one PIR-B gene and multiple PIR-A genes. The PIR-A and PIR-B genes are expressed in B lymphocytes and myeloid lineage cells, wherein both are expressed simultaneously. The characteristics of the highly-conserved PIR-A and PIR-B genes and their coordinate cellular expression suggest a potential regulatory role in humoral, inflammatory, and allergic responses.

Our global impact is finally receiving the scientific attention it deserves. The outcome will largely determine the future course of evolution. Human-modified ecosystems are shaped by our activities and their side effects. They share a common set of traits including simplified food webs, landscape homogenization, and high nutrient and energy inputs. Ecosystem simplification is the ecological hallmark of humanity and the reason for our evolutionary success. However, the side effects of our profligacy and poor resource practices are now so pervasive as to threaten our future no less than that of biological diversity itself. This article looks at human impact on ecosystems and the consequences for evolution. It concludes that future evolution will be shaped by our awareness of the global threats, our willingness to take action, and our ability to do so. Our ability is presently hampered by several factors, including the poor state of ecosystem and planetary knowledge, ignorance of human impact, lack of guidelines for sustainability, and a paucity of good policies, practices, and incentives for adopting those guidelines in daily life. Conservation philosophy, science, and practice must be framed against the reality of human-dominated ecosystems, rather than the separation of humanity and nature underlying the modern conservation movement. The steps scientists can take to imbed science in conservation and conservation in the societal process affecting the future of ecosystems and human well-being are discussed.

Secretory granules store neuropeptides and hormones and exhibit regulated exocytosis upon appropriate cellular stimulation. They are generated in the trans-Golgi network as immature secretory granules, short-lived vesicular intermediates, which undergo a complex and poorly understood maturation process. Due to their short half-life and low abundance, real-time studies of immature secretory granules have not been previously possible. We describe here a pulse/chase-like system based on the expression of a human chromogranin B-GFP fusion protein in neuroendocrine PC12 cells, which permits direct visualization of the budding of immature secretory granules and their dynamics during maturation. Live cell imaging revealed that newly formed immature secretory granules are transported in a direct and microtubule-dependent manner within a few seconds to the cell periphery. Our data suggest that the cooperative action of microtubules and actin filaments restricts immature secretory granules to the F-actin-rich cell cortex, where they move randomly and mature completely within a few hours. During this maturation period, secretory granules segregate into pools of different motility. In a late phase of maturation, 60% of secretory granules were found to be immobile and about half of these underwent F-actin-dependent tethering.

Parathyroid hormone-related peptide (PTHrP) was initially identified as a product of malignant tumors that mediates paraneoplastic hypercalcemia. It is now known that the parathyroid hormone (PTH) and PTHrP genes are evolutionarily related and that the products of these two genes share a common receptor, the PTH/PTHrP receptor. PTHrP and the PTH/PTHrP receptor are widely expressed in both adult and fetal tissues, and recent gene-targeting and disruption experiments have implicated PTHrP as a developmental regulatory molecule. Apparent PTHrP functions include the regulation of endochondral bone development, of hair follicle formation, and of branching morphogenesis in the breast. Herein, we report that overexpression of PTHrP in chondrocytes using the mouse type II collagen promoter induces a novel form of chondrodysplasia characterized by short-limbed dwarfism and a delay in endochondral ossification. This features a delay in chondrocyte differentiation and in bone collar formation and is sufficiently marked that the mice are born with a cartilaginous endochondral skeleton. In addition to the delay, chondrocytes in the transgenic mice initially become hypertrophic at the periphery of the developing long bones rather than in the middle, leading to a seeming reversal in the pattern of chondrocyte differentiation and ossification. By 7 weeks, the delays in chondrocyte differentiation and ossification have largely corrected, leaving foreshortened and misshapen but histologically near-normal bones. These findings confirm a role for PTHrP as an inhibitor of the program of chondrocyte differentiation. PTHrP may function in this regard to maintain the stepwise differentiation of chondrocytes that initiates endochondral ossification in the midsection of endochondral bones early in development and that also permits linear growth at the growth plate later in development.

The collective behavior of interconnected spiking nerve cells is investigated. It is shown that a variety of model systems exhibit the same short-time behavior and rapidly converge to (approximately) periodic firing patterns with locally synchronized action potentials. The dynamics of one model can be described by a downhill motion on an abstract energy landscape. Since an energy landscape makes it possible to understand and program computation done by an attractor network, the results will extend our understanding of collective computation from models based on a firing-rate description to biologically more realistic systems with integrate-and-fire neurons.

Partindo das considerações de que os termos sujeito e subjetividade permeiam o discurso psicanalítico contemporâneo e de que são, direta e indiretamente, atribuídos a Freud a despeito de ele próprio nunca tê-los conceituado, esta pesquisa tem como objetivo caracterizar um perfil de sujeito a partir do discurso freudiano. O trabalho orientou-se pela metodologia da Análise Institucional do Discurso, uma analítica do domínio subjetivo que toma o discurso em seu caráter de ato e acontecimento. Primeiramente se realizou um estudo de As técnicas de si, de Michel Foucault, de modo a permitir o circunstanciamento da psicanálise como uma técnica que produz um si, um sujeito este circunstanciamento permitiu, então, tomar sujeito e subjetividade na qualidade de produções histórica, geográfica e analiticamente contextualizadas, não como formas de imanência ou transcendência. A partir desse pressuposto, elaborou-se uma análise institucional do discurso de O mal-estar na civilização que privilegiou não apenas seu conteúdo, mas principalmente seu modo de produção, colocando em relevo o contexto presente no texto, as interlocuções que se criam, os lugares atribuídos e ocupados, as expectativas assim mobilizadas, as estratégias discursivas utilizadas, os jogos de poder e verdade exercidos, bem como os efeitos de reconhecimento e desconhecimento então facultados. Esta análise mostrou que Freud exerce uma perspectiva de interioridade, pois o mal-estar que acomete a civilização é compreendido em analogia à concepção psicanalítica de desenvolvimento individual, explicando, em suma, a cultura pelo prisma do indivíduo; evidenciou que as teorizações sobre a vida instintiva são a principal sustentação do discurso sobre o mal-estar da civilização; apontou como as estratégias discursivas utilizadas por Freud promovem a subjetivação, por parte do leitor, daquilo que seu discurso produz como verdade; e que o conceito de indivíduo é ocasião de exercício daquela perspectiva de interioridade e de atualização dos pressupostos teóricos. Com base nisso, pôde-se caracterizar um sujeito universal; psicologizado; determinado sobretudo pelos movimentos da sexualidade e da agressividade; cuja tônica recai sobre o dito mundo interno; dotado de origens e propósitos concernentes à vida instintiva; e cujo perfil é delimitado pela tarefa de administração dos instintos, isto é, cujo perfil se dá entre os imperativos superegóicos de renúncia e a margem de liberdade de que dispõe para satisfazer as exigências do princípio de prazer. Observou-se também, na esteira do pontuado por Guirado (2010), que em geral Freud naturaliza os termos do discurso teórico, fazendo de sua universalização a condição e o limite para se pensar o domínio subjetivo e a singularidade; diferentemente de Foucault, que compreende esse domínio em referência às relações de poder e saber, de forma contextualizada. Do ponto de vista da análise institucional do discurso freudiano, concluiu-se, finalmente, que o si ao qual a técnica psicanalítica dá lastro é efeito da perspectiva exercida por Freud, que promove o reconhecimento da interioridade instintiva como crivo da civilização, de um modo de vida e de si mesmo

In this project we review the effects of reputation within the context of game theory. This is done through a study of two key papers. First, we examine a paper from Fudenberg and Levine: Reputation and Equilibrium Selection in Games with a Patient Player (1989). We add to this a review Gossner’s Simple Bounds on the Value of a Reputation (2011). We look specifically at scenarios in which a long-run player faces a series of short-run opponents, and how the former may develop a reputation. In turn, we show how reputation leads directly to both lower and upper bounds on the long-run player’s payoffs.

The European Union (EU) was the frontrunner for the establishment of the world’s first multinational emissions trading scheme (ETS). Committed to combating climate change, the EU sought to overcome the multilateral paralysis within the International Civil Aviation Organization (ICAO) to mitigate aviation emissions. Unsuccessful in pushing for a global market-based measure (MBM) within the organisation, the EU was ready for take-off to include the sector in the EU emissions trading system (EU ETS). The geographical scope, however, including all flights from and to Europe in their entire trajectory, caused frictions with the international community about sovereignty issues. Ultimately, Climate Commissioner Connie Hedegaard announced a ‘stop-the-clock’ for international flights, a temporary derogation until the 2013 ICAO Assembly in order to find a global agreement. The ’stop-the-clock’ initiative provides ample opportunity to analyse EU leadership in curbing aviation emissions based on an analytical framework specifying different types of leadership. Its shows the global challenge to the EU’s claim of structural leadership on various levels in and beyond ICAO. The paper aims to analyse to what extent the EU is a global leader in mitigating aviation emissions and to identify the kind of EU leadership according to a threefold analytical framework. In addition, it will factor in the 'stop-the-clock' initiative and to what extent it altered the perception of EU leadership in the field. The paper comes to the conclusion that EU leadership in mitigating aviation emissions is not stalling. On the contrary, the EU, by pursuing the extension of the EU ETS, has put aviation emissions on everybody’s radar – and thus showed idea-based leadership. Proving the scheme’s feasibility further underlined EU leadership, in the form of directional leadership. The 'stop-the-clock' decision, however, already indicated what was later on confirmed in the 38th ICAO Assembly: Unilateral structural leadership of the EU in the field of aviation emissions is not credible at the moment.

This paper analyses the extent of European Union (EU) actorness and effectiveness at the fifteenth United Nations Framework Convention on Climate Change (UNFCCC) Conference of the Parties (COP) meeting in Copenhagen in December 2009. For over a decade the European Union has been characterised as a leader in international climate policy-making and as an important actor in international climate change negotiations. The COP15 meeting in Copenhagen has overall brought about disappointing outcomes, especially from the perspective of the European Union. This casts doubts on EU leadership and begs the question of what has happened to EU actorness and effectiveness in this field. In terms of actorness we take Jupille and Caporaso (1998) as a point of departure and then specify a more parsimonious actorness framework that consists of cohesion and autonomy. Effectiveness (i.e. goal attainment) is seen as conceptually separate from actorness. Effectiveness is conceptualised as the result of actorness conditioned by the ‘opportunity structure’, i.e. the external context (of other actors, events and ideas) that enables or constrains EU actions. We hold that the EU’s actorness has been only moderate, especially given somewhat limited preference cohesion. In terms of the opportunity structure in Copenhagen we argue that the high degree of politicisation constrained the EU’s ability to negotiate and thus to attain its goals. Another external factor that had a substantial adverse impact on the EU’s effectiveness at the Copenhagen negotiations was the strong involvement of other actors with rather different positions, namely the United States (US) and the BASIC countries (Brazil, South Africa, India, and China).

Underwater video transects have become a common tool for quantitative analysis of the seafloor. However a major difficulty remains in the accurate determination of the area surveyed as underwater navigation can be unreliable and image scaling does not always compensate for distortions due to perspective and topography. Depending on the camera set-up and available instruments, different methods of surface measurement are applied, which make it difficult to compare data obtained by different vehicles. 3-D modelling of the seafloor based on 2-D video data and a reference scale can be used to compute subtransect dimensions. Focussing on the length of the subtransect, the data obtained from 3-D models created with the software PhotoModeler Scanner are compared with those determined from underwater acoustic positioning (ultra short baseline, USBL) and bottom tracking (Doppler velocity log, DVL). 3-D model building and scaling was successfully conducted on all three tested set-ups and the distortion of the reference scales due to substrate roughness was identified as the main source of imprecision. Acoustic positioning was generally inaccurate and bottom tracking unreliable on rough terrain. Subtransect lengths assessed with PhotoModeler were on average 20% longer than those derived from acoustic positioning due to the higher spatial resolution and the inclusion of slope. On a high relief wall bottom tracking and 3-D modelling yielded similar results. At present, 3-D modelling is the most powerful, albeit the most time-consuming, method for accurate determination of video subtransect dimensions.

Material and data were collected at 41 sites in the subpolar North Atlantic Ocean between Scotland and Newfoundland, during the RRS CharlesDarwin CD159 cruise in July 2004 (McCave, 2005). Sites were selected to reflect the major inputs of water that becomes the North Atlantic Deep Water (NADW); the Iceland-Scotland Overflow Water (ISOW), the Denmark Strait Overflow Water (DSOW) and the Labrador Sea Water (LSW). Areas cored were the south Iceland Rise, SE Greenland slope/rise and Eirik Drift, and the Labrador margin. A total of 29 box cores, 19 piston cores, 6 kasten cores, 9 short gravity cores and 20 CTD casts as well as 28 surface water samples were collected during the cruise. Here we present sediment core-top sample ages. The cores were sampled at 1 or 0.5 cm intervals and we used the top 1 or 2 cm, depending on availability of foraminifera in the samples. Sediment samples were disaggregated on an end-over-end wheel, wet sieved at >63 um, and dry sieved to 63-150 and >150 um. Accelerator Mass Spectrometer (AMS) radiocarbon dating was done for each core top based on between 900-1600 monospecific planktonic foraminifera (Globigerina bulloides or Neogloboquadrina pachyderma (sinistral)). All dates were of modern or late Holocene age except site RAPID-08-5B (9806 ± 38 uncorrected 14C years BP) and site RAPID-14-10B (11543 ± 40 uncorrected 14C years BP). The >150 um fraction was split until approximately 300 foraminifera remained and counted for number of lithic grains, benthic foraminifera, planktonic foraminifera and foraminifera fragments. In all but the shallowest sample (Greenland rise, 761m water depth) benthic foraminifera constituted less than 2% of the total >150 um fraction of the sample.

A detailed Pliocene oxygen isotope record from the Ontong Java Plateau, based on measurements of the surface-dwelling planktonic foraminifer Globigerinoides sacculifer, was produced for the period from 5 to 2 Ma. The record documents major long- and short-term climate changes. The results show periods of enhanced ice volume at 4.6 to 4.3 Ma and after 2.85 Ma, a long-term warming trend from 4.1 to 3.7 Ma, and a distinct cooling trend that was initiated at 3.5 Ma and progressed through the initiation of large-scale Northern Hemisphere glaciation after 2.85 Ma (according to the time scale of Shackleton and others proposed in 1990). Periods of high average ice volumes also show the highest d18O amplitudes. The pattern of climate cyclicity changed markedly at about 2.85 Ma. Earlier times were marked by high-frequency variability at the precessional frequencies or even higher frequencies, pointing to low-latitude processes as a main controlling factor driving planktonic d18O variability in this period. The high-frequency variability is not coherent with insolation and points to strong nonlinearity in the way the climate system responded to orbital forcing before the onset of large scale Northern Hemisphere glaciation. After 3 Ma, stronger 41-k.y. cyclicity appears in the record. The shift in pattern is clearest around 2.85 Ma (according to the time scale proposed by Shackleton and others in 1990), 100-200 k.y. before the most dramatic spread of Northern Hemisphere ice sheets. This indicates that high-latitude processes from this point on began to take over and influence most strongly the d18O record, which now reflects ice-volume fluctuations related to the climatic effects of obliquity forcing on the seasonality of high-latitude areas, most probably in the Northern Hemisphere. The general Pliocene trend is that high-latitude climate sensitivity and instability was increasing, and the causal factors producing the intensified glacial cyclicity during the Pliocene must be factors that enhance cooling and climate sensitivity in the subarctic areas.
