Urban problems have several features that make them inherently dynamic. Large transaction costs all but guarantee that homeowners will do their best to consider how a neighborhood might change before buying a house. Similarly, stores face large sunk costs when opening, and want to be sure that their investment will pay off in the long run. In line with those concerns, different areas of Economics have made recent advances in modeling those questions within a dynamic framework. This dissertation contributes to those efforts.

Chapter 2 discusses how to model an agent’s location decision when the agent must learn about an exogenous amenity that may be changing over time. The model is applied to estimating the marginal willingness to pay to avoid crime, in which agents are learning about the crime rate in a neighborhood, and the crime rate can change in predictable (Markovian) ways.

Chapters 3 and 4 concentrate on location decision problems when there are externalities between decision makers. Chapter 3 focuses on the decision of business owners to open a store, when its demand is a function of other nearby stores, either through competition, or through spillovers on foot traffic. It uses a dynamic model in continuous time to model agents’ decisions. A particular challenge is isolating the contribution of spillovers from the contribution of other unobserved neighborhood attributes that could also lead to agglomeration. A key contribution of this chapter is showing how we can use information on storefront ownership to help separately identify spillovers.

Finally, chapter 4 focuses on a class of models in which families prefer to live

close to similar neighbors. This chapter provides the first simulation of such a model in which agents are forward looking, and shows that this leads to more segregation than it would have been observed with myopic agents, which is the standard in this literature. The chapter also discusses several extensions of the model that can be used to investigate relevant questions such as the arrival of a large contingent high skilled tech workers in San Francisco, the immigration of hispanic families to several southern American cities, large changes in local amenities, such as the construction of magnet schools or metro stations, and the flight of wealthy residents from cities in the Rust belt, such as Detroit.

This dissertation consists of three separate essays on job search and labor market dynamics. In the first essay, “The Impact of Labor Market Conditions on Job Creation: Evidence from Firm Level Data”, I study how much changes in labor market conditions reduce employment fluctuations over the business cycle. Changes in labor market conditions make hiring more expensive during expansions and cheaper during recessions, creating counter-cyclical incentives for job creation. I estimate firm level elasticities of labor demand with respect to changes in labor market conditions, considering two margins: changes in labor market tightness and changes in wages. Using employer-employee matched data from Brazil, I find that all firms are more sensitive to changes in wages rather than labor market tightness, and there is substantial heterogeneity in labor demand elasticity across regions. Based on these results, I demonstrate that changes in labor market conditions reduce the variance of employment growth over the business cycle by 20% in a median region, and this effect is equally driven by changes along each margin. Moreover, I show that the magnitude of the effect of labor market conditions on employment growth can be significantly affected by economic policy. In particular, I document that the rapid growth of the national minimum wages in Brazil in 1997-2010 amplified the impact of the change in labor market conditions during local expansions and diminished this impact during local recessions.

In the second essay, “A Framework for Estimating Persistence of Local Labor

Demand Shocks”, I propose a decomposition which allows me to study the persistence of local labor demand shocks. Persistence of labor demand shocks varies across industries, and the incidence of shocks in a region depends on the regional industrial composition. As a result, less diverse regions are more likely to experience deeper shocks, but not necessarily more long lasting shocks. Building on this idea, I propose a decomposition of local labor demand shocks into idiosyncratic location shocks and nationwide industry shocks and estimate the variance and the persistence of these shocks using the Quarterly Census of Employment and Wages (QCEW) in 1990-2013.

In the third essay, “Conditional Choice Probability Estimation of Continuous- Time Job Search Models”, co-authored with Peter Arcidiacono and Arnaud Maurel, we propose a novel, computationally feasible method of estimating non-stationary job search models. Non-stationary job search models arise in many applications, where policy change can be anticipated by the workers. The most prominent example of such policy is the expiration of unemployment benefits. However, estimating these models still poses a considerable computational challenge, because of the need to solve a differential equation numerically at each step of the optimization routine. We overcome this challenge by adopting conditional choice probability methods, widely used in dynamic discrete choice literature, to job search models and show how the hazard rate out of unemployment and the distribution of the accepted wages, which can be estimated in many datasets, can be used to infer the value of unemployment. We demonstrate how to apply our method by analyzing the effect of the unemployment benefit expiration on duration of unemployment using the data from the Survey of Income and Program Participation (SIPP) in 1996-2007.

Monitoring seawater CO2 for a full year with seasonal observations of community metabolism in Ishigaki Island, Japan, revealed seasonal variation and anomalous values owing to the bleaching event in 1998. The daily average pCO2 showed a seasonal pattern on an annual scale, 280 to 320 ?atm in winter and 360 to 400 ?atm in summer, which was determined primarily by the seasonal change in seawater temperature. By contrast, the range in the diel variation in pCO2, 400 to 500 ?atm in summer 200 to 300 ?atm in winter, was attributed to the seasonal variation in community metabolism: Gross primary production (P g ) and respiration (R) were high in summer and low in winter. During the 1998 bleaching event, although P g and R increased, community excess organic production (E) decreased by three quarters compared with the same month in 1999, when the coral community showed high recovery. This change in metabolism led to large diel range and increased average value of pCO2 levels in the seawater on the reef flat. The decrease in the range and increase in the average value of pCO2 were observed by monitoring the Palau barrier reef flat, where overall mortality of corals occurred after the bleaching. All the metabolic parameters, P g , R, E and calcification (G) were reduced by half after the bleaching, which increased the average pCO2 value by 10 ?atm and decreased its diel range from 200-400 ?atm to 100-200 ?atm. Bleaching and resultant mortality of coral reefs led to degradation of their metabolic performance, and thus resulted in the loss of their active interaction with the carbon cycle.

Community metabolism was investigated using a Lagrangian flow respirometry technique on 2 reef flats at Moorea (French Polynesia) during austral winter and Yonge Reef (Great Barrier Reef) during austral summer. The data were used to estimate related air-sea CO2 disequilibrium. A sine function did not satisfactorily model the diel light curves and overestimated the metabolic parameters. The ranges of community gross primary production and respiration (Pg and R; 9 to 15 g C m-2 d-1) were within the range previously reported for reef flats, and community net calcification (G; 19 to 25 g CaCO3 m-2 d-1) was higher than the 'standard' range. The molar ratio of organic to inorganic carbon uptake was 6:1 for both sites. The reef flat at Moorea displayed a higher rate of organic production and a lower rate of calcification compared to previous measurements carried out during austral summer. The approximate uncertainty of the daily metabolic parameters was estimated using a procedure based on a Monte Carlo simulation. The standard errors of Pg,R and Pg/R expressed as a percentage of the mean are lower than 3% but are comparatively larger for E, the excess production (6 to 78%). The daily air-sea CO2 flux (FCO2) was positive throughout the field experiments, indicating that the reef flats at Moorea and Yonge Reef released CO2 to the atmosphere at the time of measurement. FCO2 decreased as a function of increasing daily irradiance.

Maerl community respiration, photosynthesis and calcification were measured seasonally in the Bay of Brest (France). The dynamics of oxygen, carbon and carbonate fluxes at the water-sediment interface were assessed using benthic chambers. Community respiration (CR) fluctuated in accordance with the seasonal changes in water temperature, from 1.5 mmol C m**-2 h**-1 in winter to 8.7 mmol C m**-2 h**-1 in summer. Mean gross community production (GCP) varied significantly among seasons, according to incident irradiance and temperature, from 3.4 mmol C m**-2 h**-1 in winter to 12.7 mmol C m-2 h-1 in summer. Mean annual Pmax for the P-E curve was estimated to 13.3 mmol C m-2 h-1. Carbonate precipitation only occurred during light incubations and varied seasonally from 0.7 mmol CaCO3 m-2 h-1 in winter to 4.2 mmol CaCO3 m-2 h-1 in summer. Mean annual Pmax was 3.2 mmol CaCO3 m-2 h-1. Annual CR was estimated to 407.4 g C m**-2 yr**-1, and GCP, to 240.9 g C m**-2 yr**-1. Maerl communities are, therefore, heterotrophic systems (GCP:CR = 0.6), and are a source of CO2 for surrounding environments. In addition, CO2 released by calcification averaged 39.2 g C m**-2 yr**-1. Maerl community annual carbonate production was estimated to 486.7 g CaCO3 m**-2 yr**-1; they are therefore one of the most important carbonate producers in shallow coastal waters.

Carbon dioxide and oxygen fluxes were measured in 0.2 m2 enclosures placed at the water sediment interface in the SW lagoon of New Caledonia. Experiments, performed at several stations in a wide range of environments, were carried out both in darkness to estimate respiration and at ambient light, to assess the effects of primary production. The community respiratory quotient (CRQ = CO2 production rate/02 consumption rate) and the community photosynthetic quotient (CPQ= gross O2 production rate/gross CO2 consumption rate) were calculated by functional regressions. The CRQ value, calculated from 61 incubations, was 1.14 (S.E. 0.05) and the CPQ value, obtained from 18 incubations, was 1.03 (S.E. 0.08). The linearity of the relationship between the O2 and the CO2 fluxes suggests that these values are representative for the whole lagoon

The pressures on honeybee (Apis mellifera) populations, resulting from threats by modern pesticides, parasites, predators and diseases, have raised awareness of the economic importance and critical role this insect plays in agricultural societies across the globe. However, the association of humans with A. mellifera predates post-industrial-revolution agriculture, as evidenced by the widespread presence of ancient Egyptian bee iconography dating to the Old Kingdom (approximately 2400 bc)1. There are also indications of Stone Age people harvesting bee products; for example, honey hunting is interpreted from rock art2 in a prehistoric Holocene context and a beeswax find in a pre-agriculturalist site3. However, when and where the regular association of A. mellifera with agriculturalists emerged is unknown4. One of the major products of A. mellifera is beeswax, which is composed of a complex suite of lipids including n-alkanes, n-alkanoic acids and fatty acyl wax esters. The composition is highly constant as it is determined genetically through the insect’s biochemistry. Thus, the chemical ‘fingerprint’ of beeswax provides a reliable basis for detecting this commodity in organic residues preserved at archaeological sites, which we now use to trace the exploitation by humans of A. mellifera temporally and spatially. Here we present secure identifications of beeswax in lipid residues preserved in pottery vessels of Neolithic Old World farmers. The geographical range of bee product exploitation is traced in Neolithic Europe, the Near East and North Africa, providing the palaeoecological range of honeybees during prehistory. Temporally, we demonstrate that bee products were exploited continuously, and probably extensively in some regions, at least from the seventh millennium cal bc, likely fulfilling a variety of technological and cultural functions. The close association of A. mellifera with Neolithic farming communities dates to the early onset of agriculture and may provide evidence for the beginnings of a domestication process.

BACKGROUND: In the previously reported ALSYMPCA trial in patients with castration-resistant prostate cancer and symptomatic bone metastases, overall survival was significantly longer in patients treated with radium-223 dichloride (radium-223) than in patients treated with placebo. In this study, we investigated safety and overall survival in radium-223 treated patients in an early access programme done after the ALSYMPCA study and before regulatory approval of radium-223.

METHODS: We did an international, prospective, interventional, open-label, single-arm, phase 3b study. Enrolled patients were aged 18 years or older with histologically or cytologically confirmed progressive bone-predominant metastatic castration-resistant prostate cancer with two or more skeletal metastases on imaging (with no restriction as to whether they were symptomatic or asymptomatic; without visceral disease but lymph node metastases were allowed). Patients received intravenous injections of radium-223, 50 kBq/kg (current recommendation 55 kBq/kg after implementation of National Institute of Standards and Technology update on April 18, 2016) every 4 weeks for up to six injections. Other concomitant anticancer therapies were allowed. Primary endpoints were safety and overall survival. The safety and efficacy analyses were done on all patients who received at least one dose of the study drug. The study has been completed, and we report the final analysis here. This study is registered with ClinicalTrials.gov, number NCT01618370, and the European Union Clinical Trials Register, EudraCT number 2012-000075-16.

FINDINGS: Between July 22, 2012, and Dec 19, 2013, 839 patients were enrolled from 113 sites in 14 countries. 696 patients received one or more doses of radium-223; 403 (58%) of these patients had all six planned injections. Any-grade treatment-emergent adverse events occurred in 523 (75%) of 696 patients; any-grade treatment-emergent adverse events deemed to be related to treatment were reported in 281 (40%) patients. The most common grade 3 or worse treatment-related treatment-emergent adverse events were anaemia in 32 (5%) patients, thrombocytopenia in 15 (2%) patients, neutropenia in ten (1%) patients, and leucopenia in nine (1%) patients. Any grade of serious adverse events were reported in 243 (35%) patients. Median follow-up was 7·5 months (IQR 5-11) and 210 deaths were reported; median overall survival was 16 months (95% CI 13-not available [NA]). In an exploratory analysis of overall survival with predefined factors, median overall survival was longer for: patients with baseline alkaline phosphatase concentration less than the upper limit of normal (ULN; median NA, 95% CI 16 months-NA) than for patients with an alkaline phosphatase concentration equal to or greater than the ULN (median 12 months, 11-15); patients with baseline haemoglobin levels 10 g/dL or greater (median 17 months, 14-NA) than for patients with haemoglobin levels less than 10 g/dL (median 10 months, 8-14); patients with a baseline Eastern Cooperative Oncology Group performance status (ECOG PS) of 0 (median NA, 17 months-NA) than for patients with an ECOG PS of 1 (median 13 months, 11-NA) or an ECOG PS of 2 or more (median 7 months, 5-11); and for patients with no reported baseline pain (median NA, 16 months-NA) than for those with mild pain (median 14 months, 13-NA) or moderate-severe pain (median 11 months, 9-13). Median overall survival was also longer in patients who received radium-223 plus abiraterone, enzalutamide, or both (median NA, 95% CI 16 months-NA) than in those who did not receive these agents (median 13 months, 12-16), and in patients who received radium-223 plus denosumab (median NA, 15 months-NA) than in patients who received radium-223 without denosumab (median 13 months, 12-NA).

INTERPRETATION: Our findings show that radium-223 can be safely combined with abiraterone or enzalutamide, which are now both part of the standard of care for patients with metastatic castration-resistant prostate cancer. Furthermore, our findings extend to patients who were asymptomatic at baseline, unlike those enrolled in the pivotal ALSYMPCA study. The findings of prolonged survival in patients treated with concomitant abiraterone, enzalutamide, or denosumab require confirmation in prospective randomised trials.

O bioetanol constitui uma alternativa renovável aos combustíveis fósseis. Contudo, o bioetanol de primeira geração, produzido a partir de matérias-primas alimentares, desencadeou sérios problemas económicos e sociais, pelo que é fundamental encontrar estratégias que permitam a viabilidade comercial do bioetanol de segunda geração, produzido a partir de matérias-primas lenho-celulósicas. O licor de cozimento ao sulfito ácido de árvores folhosas (HSSL) é um subproduto da indústria papeleira que, devido ao seu elevado conteúdo em açúcares, pode ser utilizado como substrato para a produção de bioetanol de segunda geração. No entanto, a maior fração dos açúcares do HSSL é composta por pentoses. Por isso, a fermentação do HSSL é realizada pela levedura Scheffersomyces stipitis, pois esta é capaz de fermentar tanto as hexoses como as pentoses. Todavia, a S. stipitis só produz etanol sob condições microaerófilas, pelo que o maior desafio da produção de bioetanol por S. stipitis reside no estabelecimento das condições ótimas de arejamento. Este trabalho teve assim por objetivo estabelecer uma estratégia de arejamento que permita a eficiente produção de bioetanol a partir de HSSL por S. stipitis C4, a qual é uma estirpe adaptada a este substrato. Deste modo, foram realizados ensaios em Erlenmeyer, de modo a caracterizar o crescimento da S. stipitis C4, e ensaios em biorreator, com vista a estudar a produção de etanol por S. stipitis C4 em duas estratégias de arejamento diferentes. Na primeira estratégia foi usado apenas um único estágio de arejamento, com controlo da tensão de oxigénio dissolvido, DOT (%), e na segunda estratégia foram usados dois estágios de arejamento, com controlo da DOT no primeiro estágio e com restrição de oxigénio no segundo estágio. Nos ensaios em Erlenmeyer com HSSL o crescimento da S. stipitis C4 foi completamente inibido. Por sua vez, nos ensaios em biorreator com um único estágio de arejamento o controlo da DOT não permitiu a produção de etanol. No entanto, nos ensaios com dois estágios de arejamento em meio sintético foi possível produzir etanol de forma eficiente. Nesta estratégia, a utilização de um maior valor de DOT no primeiro estágio de arejamento permitiu aumentar a taxa específica de crescimento máxima e o rendimento em biomassa do primeiro estágio. Para além disso, a utilização de um maior valor de DOT no primeiro estágio também permitiu aumentar a produtividade em etanol durante o segundo estágio de arejamento. Por sua vez, no segundo estágio de arejamento verificou-se que a restrição de oxigénio evitou a reassimilação de etanol pela S. stipitis C4. Deste modo, os melhores resultados para a produção de etanol foram obtidos com controlo da DOT a 50% durante o primeiro estágio e com 0 mLAr.min-1 e 250 rpm durante o segundo estágio de arejamento. A aplicação desta estratégia de arejamento a 60% HSSL/40% meio sintético permitiu obter, no primeiro estágio de arejamento, uma taxa específica de crescimento máxima de 0,17 h-1, o que demonstra que a elevada disponibilidade de oxigénio durante o primeiro estágio aumenta a tolerância da S. stipitis C4 aos inibidores. Para além disso, a taxa volumétrica de produção de etanol e o rendimento em etanol de toda a fermentação foi de respetivamente de 0,03 g.L-1.h-1 e 0,38 g.g-1. Assim, a elevada eficiência de conversão dos açúcares em etanol (74,4%) demostra que a fermentação com dois estágios de arejamento constitui uma estratégia promissora para a produção de bioetanol de segunda geração a partir de HSSL.

A Refinaria de Matosinhos é um dos complexos industriais da Galp Energia. A sua estação de tratamento de águas residuais industriais (ETARI) – designada internamente por Unidade 7000 – é composta por quatro tratamentos: o pré-tratamento, o tratamento físico-químico, o tratamento biológico e o pós-tratamento. Dada a interligação existente, é fundamental a otimização de cada um dos tratamentos. Este trabalho teve como objetivos a identificação dos problemas e/ou possibilidades de melhoria do pré-tratamento, tratamento físico-químico e pós-tratamento e principalmente a otimização do tratamento biológico da ETARI. No pré-tratamento verificou-se que a separação de óleos e lamas não era eficaz uma vez que se formam emulsões destas duas fases. Como solução, sugeriu-se a adição de agentes desemulsionantes, que se revelou economicamente inviável. Assim, sugeriu-se como alternativa o recurso a técnicas de tratamento da emulsão gerada, tais como a extração com solvente, centrifugação, ultrassons e micro-ondas. No tratamento físico-químico constatou-se que o controlo da unidade de saturação de ar na água era feito com base na análise visual dos operadores, o que pode conduzir a condições de operação afastadas das ótimas para este tratamento. Assim, sugeriu-se a realização de um estudo de otimização desta unidade com vista à determinação da razão ar/sólidos ótima para este efluente. Para além disto, constatou-se, ainda, que os consumos de coagulante aumentaram cerca de -- % no último ano, pelo que foi sugerido o estudo da viabilidade do processo de eletrocoagulação como substituto do sistema de coagulação existente. No pós-tratamento identificou-se o processo de lavagem dos filtros como sendo a etapa com possibilidade de ser otimizada. Através de um estudo preliminar concluiu-se que a lavagem contínua de um filtro por cada turno melhorava o desempenho dos mesmos. Constatou-se, ainda, que a introdução de ar comprimido na água de lavagem promove uma maior remoção de detritos do leito de areia, no entanto esta prática parece influenciar negativamente o desempenho dos filtros. No caso do tratamento biológico, identificaram-se problemas ao nível do tempo de retenção hidráulico do tratamento biológico II, que apresentou elevada variabilidade. Apesar de identificado concluiu-se que este problema era de difícil solução. Verificou-se, também, que o oxigénio dissolvido não era monitorizado, pelo que se sugeriu a instalação de uma sonda de oxigénio dissolvido numa zona de baixa turbulência do tanque de arejamento. Concluiu-se que o oxigénio era distribuído de forma homogénea por todo o tanque de arejamento e tentou-se identificar quais os fatores que influenciariam este parâmetro, no entanto, dada a elevada variabilidade do efluente e das condições de tratamento, tal não foi possível. Constatou-se, também, que o doseamento de fosfato para o tratamento biológico II era pouco eficiente já Otimização dos sistemas biológicos e melhorias nos tratamentos da ETARI da Refinaria de Matosinhos que em -- % dos dias se verificaram níveis baixos de fosfato no licor misto (< - mg/L). Foi, por isso, proposta a alteração do atual sistema de doseamento por gravidade para um sistema de bomba doseadora. Para além disso verificou-se que os consumos deste nutriente aumentaram significativamente no último ano (cerca de --%), situação que se constatou estar relacionada com um aumento da população microbiana para este período. Foi possível relacionar-se o aparecimento frequente de lamas à superfície dos decantadores secundários com incrementos repentinos de condutividade, pelo que se sugeriu o armazenamento do efluente nas bacias de tempestade, nestas situações. Verificou-se que a remoção de azoto era praticamente ineficaz uma vez que a conversão de azoto amoniacal em nitratos foi muito baixa. Assim, sugeriu-se o recurso à técnica de bio-augmentação ou a transformação do sistema de lamas ativadas num sistema bietápico. Por fim, constatou-se que a temperatura do efluente à entrada da ETARI apresenta valores bastante elevados para o tratamento biológico (aproximadamente de --º C) pelo que se sugeriu a instalação de uma sonda de temperatura no tanque de arejamento de modo a controlar de forma mais eficaz a temperatura do licor misto. Ainda no que diz respeito ao tratamento biológico, foi possível desenvolver-se um conjunto de ferramentas que visaram o funcionamento otimizado deste tratamento. Nesse sentido, foram apresentadas várias sugestões de melhoria: a utilização do índice volumétrico de lamas como indicador da qualidade das lamas em alternativa à percentagem de lamas; foi desenvolvido um conjunto de fluxogramas para a orientação dos operadores de exterior na resolução de problemas; foi criada uma “janela de operação” que pretende ser um guia de apoio à operação; foi ainda proposta a monitorização frequente da idade das lamas e da razão alimento/microrganismo.
