Tutkimuksen tarkoituksena oli selvittää typenoksidien vähennysmahdollisuudet Stora Enson Varkauden tehtaiden sellutehtaalla ja voimalaitoksella. Tutkimuksessa käsiteltiin tehdasalueen suurimpia typenoksidien päästölähteitä: soodakattilaa, meesauunia, kuorikattilaa, öljykattilaa ja muovi-alumiinijakeen kaasutuslaitosta. Tutkimuksessa selvitettiin typenoksidipäästöjen syntymekanismit ja erilaisiin polttotekniikoihin soveltuvat typenoksidien vähennystekniikat. Varkauden tehtaiden typenoksidien vuosipäästö vuonna 2001 oli 836 tonnia. Kansallinen lainsäädäntö, kansainväliset sopimukset sekä paras käytettävissä oleva tekniikka (BAT) huomioiden selvitettiin kuhunkin kohteeseen parhaiten soveltuvat ratkaisut. Tutkimuksen perusteella laadittiin toimenpideohjelma, joka määrittelee suositeltavan toteutusjärjestyksen typenoksidien vähennystoimenpiteille. Toimenpideohjelman tärkeimpinä kriteereinä pidettiin vuonna 2004 tulevan uuden ympäristöluvan arvioituja luparajoja sekä toimenpiteiden kustannustehokkuutta. Toteutusjärjestyksessä ensimmäiseksi valittiin koeajojakson järjestäminen ajon optimoimiseksi kiertopetikattilalla ja toiseksi meesauunin ajon optimointi jatkuvatoimisen NOx-analysaattorin avulla. Seuraaviksi toimenpiteiksi ehdotettiin vertikaali-ilmajärjestelmän käyttöönottoa soodakattilalla sekä SNCR-järjestelmän asennusta kuorikattilalle. Saavutettava NOx-vähennys tulisi olemaan 10 – 45 % ja hinta 30 – 3573 EUR vähennettyä NOx-tonnia kohti. Tutkimuksen osana Ilmatieteen laitoksella teetetyn typenoksidien leviämisselvityksen mukaan Stora Enson tehtaiden NOx-päästöjen vaikutus Varkauden ilmanlaatuun on hyvin pieni. Suurin osa NOx-päästöistä aiheutuu liikenteestä.

The goal of this research was evaluated the effects of potassium and nitrogen fertilization on the plum (Prunus salicina) fresh fruit quality and after cold storage. The experiment was carried out in a five year-old plum orchard 'Reubennel', located at Araucaria County, Parana State, Southern Brazil, in a Haplumbrept Soil. Potassium fertilizer was applied at 55 and 110 kg/ha/year of K2O, as KCl. Nitrogen fertilizer was applied at 40, 80, 120, 160 and 200 kg/ha/year of N, as urea. It was used a split-plot design in a factorial scheme (2x5). One hundred plum fruits were harvested from each plot, in the same day, when 25 to 50% of the peel presented yellow-reddish color. At harvest and after 17, 27 and 37 days of storage at 0 ± 0.5 ºC, the flesh firmness, the total soluble solids, and the titratable acidity were assessed. Fresh fruit quality was affected by N application, with the best results obtained by applying 40 kg/ha/year of N. The N and K rate of 40 and 110 kg/ha/year, respectively, kept superior fruit quality during the storage. 'Reubennel' cold storage can not exceed 27 days. Fresh and stored 'Reubennel' plum fruit qualities depend on the N and K fertilizer rates.

The present study reports arsenic speciation analysis in edible Shiitake (Lentinula edodes) products. The study focused on the extraction, and accurate quantification of inorganic arsenic (iAs), the most toxic form of arsenic, which was selectively separated and determined using anion exchange LC-ICPMS. A wide variety of edible Shiitake products (fresh mushrooms, food supplements, canned and dehydrated) were purchased and analysed. A cultivated Shiitake grown under controlled conditions was also analysed. The extraction method showed satisfactory extraction efficiencies (>90%) and column recoveries (>85%) for all samples. Arsenic speciation revealed that iAs was the major As compound up to 1.38 mg As per kg dm (with a mean percentage of 84% of the total arsenic) and other organoarsenicals were found as minor species. Shiitake products had high proportions of iAs and therefore should not be ignored as potential contributors to dietary iAs exposure in populations with a high intake of Shiitake products.

This study was carried out to evaluate the yield, total N content in leaves and must composition of grapes from the Cabernet Sauvignon variety subjected to the application of urea and organic compost. Cabernet Sauvignon grapevines in Rosário do Sul, RS, Brazil, in 2008, 2009 and 2010 were subjected to annual application of 40 kg N ha-1 in the form of organic compost and urea, and compared to unfertilized grapevines. In the 2008/09, 2009/10 and 2010/11 crop seasons, leaves were collected for analysis of total N content. At maturation of the grapes, the yield and quality attributes of the must were evaluated. The application of N sources, especially organic compost, increased the N content in the whole leaf at full flowering. Application of organic compost and urea has little effect on grape yield and does not affect the total nutrient content in the must, nor the enological attributes.

Tämän työn tarkoituksena oli löytää keinoja erään leijukerroskattilan typenoksidipäästöjen vähentämiseksi. Koska päästöt olivat jo alunperin alhaiset leijukerrostekniikan ja hybridin SNCR/SCR –typenpoistolaitteiston ansiosta, päätettiin päästöjä lähteä vähentämään parantamalla ammoniakkiruiskutuksen säätöä. Alkuperäinen ammoniakkiruiskutuksen säätö oli liian hidas, jotta satunnaisten häiriöiden aiheuttamat typenoksidipiikit olisi pystytty poistamaan. Ammoniakkiruiskutusta parannettiin lisäämällä jokaiseen ammoniakkilinjaan mäntäpumput, joiden avulla ammoniakkia voidaan syöttää sinne, missä sitä eniten tarvitaan. Ammoniakkiruiskutuksen säätöön kehitettiin uusi sumeaan logiikkaan perustuva säätäjä. Myös muita kehittyneitä säätömenetelmiä kuten neuroverkkoa hyödynnettiin säätäjän kehityksessä. Ammoniakkiruiskutuksen säätäjää testattiin menestyksekkäästi Ruotsissa Brista Kraftin Märstassa sijaitsevalla voimalaitoksella

Growth experiments showed that adenine and hypoxanthine can be used as nitrogen sources by several strains of K. pneumoniae under aerobic conditions. The assimilation of all nitrogens from these purines indicates that the catabolic pathway is complete and proceeds past allantoin. Here we identify the genetic system responsible for the oxidation of hypoxanthine to allantoin in K. pneumoniae. The hpx cluster consists of seven genes, for which an organization in four transcriptional units, hpxDE, hpxR, hpxO and hpxPQT, is proposed. The proteins involved in the oxidation of hypoxanthine (HpxDE) or uric acid (HpxO) did not display any similarity to other reported enzymes known to catalyze these reactions, but instead are similar to oxygenases acting on aromatic compounds. Expression of the hpx system is activated by nitrogen limitation and by the presence of specific substrates, with hpxDE and hpxPQT controlled by both signals. Nitrogen control of hpxPQT transcription, which depends on 54, is mediated by the Ntr system. In contrast, neither NtrC nor NAC is involved in the nitrogen control of hpxDE, which is dependent on 70 for transcription. Activation of these operons by the specific substrates is also mediated by different effectors and regulatory proteins. Induction of hpxPQT requires uric acid formation, whereas expression of hpxDE is induced by the presence of hypoxanthine through the regulatory protein HpxR. This LysR-type regulator binds to a TCTGC-N4-GCAAA site in the intergenic hpxD-hpxR region. When bound to this site for hpxDE activation, HpxR negatively controls its own transcription.

In pelagic species inhabiting large oceans, genetic differentiation tends to be mild and populations devoid of structure. However, large cetaceans have provided many examples of structuring. Here we investigate whether the sperm whale, a pelagic species with large population sizes and reputedly highly mobile, shows indication of structuring in the eastern North Atlantic, an ocean basin in which a single population is believed to occur. To do so, we examined stable isotope values in sequential growth layer groups of teeth from individuals sampled in Denmark and NW Spain. In each layer we measured oxygen- isotope ratios (δ18O) in the inorganic component (hydroxyapatite), and nitrogen and carbon isotope ratios (δ15N: δ13C) in the organic component (primarily collagenous). We found significant differences between Denmark and NW Spain in δ15N and δ18O values in the layer deposited at age 3, considered to be the one best representing the baseline of the breeding ground, in δ15N, δ13C and δ18O values in the period up to age 20, and in the ontogenetic variation of δ15N and δ18O values. These differences evidence that diet composition, use of habitat and/or migratory destinations are dissimilar between whales from the two regions and suggest that the North Atlantic population of sperm whales is more structured than traditionally accepted.

This paper reports the method development for the simultaneous determination of methylmercury MeHgþ) and inorganic mercury (iHg) species in seafood samples. The study focused on the extraction and quantification of MeHgþ (the most toxic species) by liquid chromatography coupled to on-line UV irradiation and cold vapour atomic fluorescence spectroscopy (LC-UV-CV-AFS), using HCl 4 mol/L as the extractant agent. Accuracy of the method has been verified by analysing three certified reference materials and different spiked samples. The values found for total Hg and MeHgþ for the CRMs did not differ significantly from certified values at a 95% confidence level, and recoveries between 85% and 97% for MeHgþ, based on spikes, were achieved. The detection limits (LODs) obtained were 0.001 mg Hg/kg for total mercury, 0.0003 mg Hg/kg for MeHgþ and 0.0004 mg Hg/kg for iHg. The quantification limits (LOQs) established were 0.003 mg Hg/kg for total mercury, 0.0010 mg Hg/kg for MeHgþ and 0.0012 mg Hg/kg for iHg. Precision for each mercury species was established, being 12% in terms of RSD in all cases. Finally, the developed method was applied to 24 seafood samples from different origins and total mercury contents. The concentrations for Total Hg, MeHg and iHg ranged from 0.07 to 2.33, 0.003-2.23 and 0.006-0.085 mg Hg/kg, respectively. The established analytical method allows to obtain results for mercury speciation in less than 1 one hour including both, sample pretreatment and measuring step.

BACKGROUND & AIMS: Protein and energy requirements in critically ill children are currently based on insufficient data. Moreover, longitudinal measurements of both total urinary nitrogen (TUN) and resting energy expenditure (REE) are lacking. The aim of this study was to investigate how much protein and energy are needed to equilibrate nitrogen and energy balances in ventilated critically ill children on the basis of daily measurements of TUN, REE and protein and energy intakes. Comparisons were made with the guidelines of the American Society for Parenteral and Enteral Nutrition and the Dietary Reference Intakes. METHODS: Children with an expected duration of mechanical ventilation ≥72 h were prospectively recruited. TUN was measured by chemiluminescence, and REE was measured by indirect calorimetry. Generalised linear models for longitudinal data were used to study the relation between protein intake and nitrogen balance and to calculate the minimum intake of protein needed to achieve nitrogen equilibrium. A similar approach was used for energy. Results were compared to the recommended values. RESULTS: Based on 402 measurements performed in 74 children (median age: 21 months), the mean TUN was high at 0.20 (95% CI: 0.20, 0.22) g/kg/d and the REE was 55 (95% CI: 54, 57) kcal/kg/d. Nitrogen and energy balances were achieved with 1.5 (95% CI: 1.4, 1.6) g/kg/d of protein and 58 (95% CI: 53, 63) kcal/kg/d for the entire group, but there were differences among children of different ages. Children required more protein and less energy than the Dietary Reference Intakes. CONCLUSIONS: In critically ill children, TUN was elevated and REE was reduced during the entire period of mechanical ventilation. Minimum intakes of 1.5 g/kg/d of protein and 58 kcal/kg/d can equilibrate nitrogen and energy balances in children up to 4 years old. Older children require more protein.

The present paper is a review about basic principles of the molecular mechanics that is the most important tool used in molecular modeling area, and their applications to the calculation of the relative stability and chemical reactivity of organometalic and coordination compounds. We show how molecular mechanics can be successfully applied to a wide variety of inorganic systems.

The main aim of this study was to develop the project management framework model which would serve as the new model to follow for upcoming projects at the Lappeenranta cement plant. The other goal was to execute the SNCR (selective non catalytic reduction) project successfully so that the nitrogen oxides emissions are below the stated emission limit when the new emission limit comes into effect beginning in July, 2008. Nitrogen oxides, project management aspects, SNCR and the invested system are explained in the theory part. In the practical part of the study, the SNCR project in the Lappeenranta cement plant was executed and the findings were documented. In order to reach the aim of this study, a framework of project management was made. The framework is based on the executed SNCR project, previous projects in the cement plant and on the available literature relating to the subject matter. The developed project turned out to be successful.

Human activities have resulted in increased nutrient levels in many rivers all over Europe. Sustainable management of river basins demands an assessment of the causes and consequences of human alteration of nutrient flows, together with an evaluation of management options. In the context of an integrated and interdisciplinary environmental assessment (IEA) of nutrient flows, we present and discuss the application of the nutrient emission model MONERIS (MOdelling Nutrient Emissions into River Systems) to the Catalan river basin, La Tordera (north-east Spain), for the period 1996–2002. After a successful calibration and verification process (Nash-Sutcliffe efficiencies E=0.85 for phosphorus and E=0.86 for nitrogen), the application of the model MONERIS proved to be useful in estimating nutrient loads. Crucial for model calibration, in-stream retention was estimated to be about 50 % of nutrient emissions on an annual basis. Through this process, we identified the importance of point sources for phosphorus emissions (about 94% for 1996–2002), and diffuse sources, especially inputs via groundwater, for nitrogen emissions (about 31% for 1996–2002). Despite hurdles related to model structure, observed loads, and input data encountered during the modelling process, MONERIS provided a good representation of the major interannual and spatial patterns in nutrient emissions. An analysis of the model uncertainty and sensitivity to input data indicates that the model MONERIS, even in data-starved Mediterranean catchments, may be profitably used by water managers for evaluating quantitative nutrient emission scenarios for the purpose of managing river basins. As an example of scenario modelling, an analysis of the changes in nutrient emissions through two different future scenarios allowed the identification of a set of relevant measures to reduce nutrient loads.

Hydrological disturbances, light availability and nutrients are the most relevant factors determining the structure of the biological communities in Mediterranean rivers. While some hydrological disturbances are able to induce catastrophic effects, which may cause a complete reset in physical and biological conditions, continued enrichment or changes in light availability are factors leading to the progressive shift in the communities of autotrophs and heterotrophs in the systems. Primary production in Mediterranean streams shows relevant seasonal changes which mainly follows the variations in light availability. In most forested streams, the algal community is shade-adapted. Nutrient enrichment (especially phosphorus) leads to marked increases in primary production, but this increase is not lineal and there is a saturation of algal biomass even in the most enriched systems. The heterotrophs (bacteria, fungi) are related to the pattern of DOC availability (which most depends on the seasonal discharge and leaf fall dynamics) and to the available substrata in the stream. It has been repeatedly observed that shorttime increases of extracellular enzyme activities are related to the accumulation of autochthonous (algal) and/or allochthonous (leaves) organic matter on the streambed during spring and summer, this being more remarkable in dry than in wetter years. Flow reduction favours detritus concentration in pools, and the subsequent increase in the density and biomass of the macroinvertebrate community. In Mediterranean streams collectors are accounting for the highest density and biomass, this being more remarkable in the least permanent systems, in accordance with the effect of floods on the organic matter availability. Nutrients, through the effect on the primary producers, also affect the trophic food web in the streams by favouring the predominance of grazers

The influences of the spray-drying parameters and the type of nanoparticles (nanocapsules or nanospheres) on the characteristics of nanoparticle-coated diclofenac-loaded microparticles were investigated by using a factorial design 3². Gastrointestinal tolerance following oral administration in rats was evaluated. Formulations were selected considering the best yields, the best encapsulation efficiencies and the lowest water contents, presenting surfaces completely coated by nanostructures and a decrease in the surface areas in relation to the uncoated core. In vitro drug release demonstrated the influence of the nanoparticle-coating on the dissolution profiles of diclofenac. Nanocapsule-coated microparticles presented a protective effect on the gastrointestinal mucosa.

The immune system is the responsible for body integrity and prevention of external invasion. On one side, nanoparticles are no triggers that the immune system is prepared to detect, on the other side it is known that foreign bodies, not only bacteria, viruses and parasites, but also inorganic matter, can cause various pathologies such as silicosis, asbestosis or inflammatory reactions. Therefore, nanoparticles entering the body, after interaction with proteins, will be either recognized as self-agents or detected by the immune system, encompassing immunostimulation or immunosuppression responses. The nature of these interactions seems to be dictated not specially by the composition of the material but by modifications of NP coating (composition, surface charge and structure). Herein, we explore the use of gold nanoparticles as substrates to carry multifunctional ligands to manipulate the immune system in a controlled manner, from undetection to immunostimulation. Murine bone marrow macrophages can be activated with artificial nanometric objects consisting of a gold nanoparticle functionalized with peptides. In the presence of some conjugates, macrophage proliferation was stopped and pro-inflammatory cytokines were induced. The biochemical type of response depended on the type of conjugated peptide and was correlated with the degree of ordering in the peptide coating. These findings help to illustrate the basic requirements involved in medical NP conjugate design to either activate the immune system or hide from it, in order to reach their targets before being removed by phagocytes. Additionally, it opens up the possibility to modulate the immune response in order to suppress unwanted responses resulting from autoimmunity, or allergy or to stimulate protective responses against pathogens.
