Este relatório está inserido no âmbito da disciplina Prática de Ensino Supervisionada (PES), que permitirá a obtenção do grau de Mestre em Ensino de Educação Física nos Ensinos Básico e Secundário. O relatório de estágio aborda o trabalho desenvolvido realizado durante o ano lectivo 2009/201O, como professor estagiário, na Escola EB 2,3 Conde de Vilalva, na Escola Secundária Gabriel Pereira e na Escola Básica 1-Jardim de Infância do Bacelo. No final do estágio foi possível afirmar que o que aprendemos ao longo deste ano lectivo nos acompanhará para o resto da vida profissional e social, uma vez que este estágio curricular veio complementar a formação adquirida durante a Licenciatura, auxiliando na incrementação das competências técnico científicas, sociais e pessoais, indispensáveis a esta actividade profissional. ABSTRACT: This report is inserted in the supervised teaching subject, as known as Prática de Ensino Supervisionada (PES), which purpose is to obtain Master's degree in Physical Education in primary and secondary education. The stage report deals with all the work developed during 2009/201O school year, as a student teacher, in Escola EB 2,3 Conde de Vilalva, Escola Secundária Gabriel Pereira and in Escola Básica 1-Jardim de Infância do Bacelo. At the end of the stage, it was possible to state that what we have learned past this year will be useful and will be with us for the rest of our professional and social life due the formation acquired during graduation, improving technical, scientific, social and personal skills that are vital and indispensable to this professional activity.

A Prática de Ensino Supervisionada desenvolveu-se no Agrupamento de Escolas nº 4 de Évora- EB 2,3 Conde Vilalva e na Escola Secundária André de Gouveia, com as turmas do 8º ano e 12° ano respectivamente. Para elucidar o trabalho realizado ao longo do ano desenvolvi o presente relatório onde abordei a preparação cientifica, pedagógica e didáctica que permitiu um conhecimento dos alunos e do currículo; a Planificação e condução das aulas e respectiva avaliação que faz referencia ao planeamento por etapas e às vantagens do mesmo e ainda à forma como foram leccionadas as aulas; uma análise da prática de ensino; a participação na escola e as actividades desenvolvidas nas mesmas; e o desenvolvimento profissional com referências ás competências que o professor deve assumir dentro da comunidade escolar. Com os parâmetros acima referidos demonstra-se a grande importância que o estágio teve na aquisição das competências essenciais inerentes à docência. ABSTRACT: Supervised Teaching Practice was developed in School Cluster nº 4 of Évora Elementary School Conde Vilalva and in Secondary School André de Gouveia, with classes of 8th and 12th grade, respectively. To elucidate the work I done over this school year, I developed this report which discussed the scientific, educational and teaching preparation, allowing an understanding of students and the curriculum; the planning and conduct of lessons and their assessment that refers to the planning stages, and to advantage of it and, on the other hand, the way that were taught lessons; an analysis of teaching practice; participation in school and activities carried therein; and professional development with reference to the competences that the teacher should take within the school community. With the above parameters is shown the importance of this stage in the acquisition of essential skills related to teaching.

Este trabalho tem por base o relato de um ano lectivo 2009/201O de Prática de Ensino Supervisionada (PES) para a especialidade do grau de Mestre em Ensino de Educação Física nos Ensinos Básico e Secundário, leccionado na Universidade de Évora e contribui para a avaliação do mesmo. O estágio foi realizado no agrupamento de Escolas n° 4 de Évora - EB 2,3 Conde de Vilalva; EB1 Jardim de Infância e Escola Secundária Gabriel Pereira, abrangendo os três ciclos de aprendizagem dando assim cumprimento à exigência curricular. Relata de forma sucinta a preparação científica, pedagógica e didáctica desde o conhecimento do currículo, do conteúdo e dos alunos. Desenvolve de forma fundamentada a planificação, condução de aulas e avaliação das aprendizagens. Faz uma análise das actividades desenvolvidas como professor de uma disciplina e como participante activo na escola e na comunidade escolar. Tenta fundamentar as estratégias, metodologias e decisões tomadas. Por último, realiza uma análise da prática de ensino. O que foi constatado nesta pequena experiência foi uma prática mais consciente e um respeito maior, por nós mesmo, enquanto profissional da educação, e pelos alunos, enquanto sujeitos activos no processo de ensino/aprendizagem. ABSTRACT: This work is based on the report of an academic year 2009/2010 Supervised Teaching Practice (PES) for the specialty Master's degree in Teaching Physical Education in Primary and Secondary Schools, taught at the University of Évora and contribute to the evaluation the same. The group stage was held at school n8 4 EB 2,3 Conde Vilalva e Kindergarten and Secondary school Gabriel Pereira, covering the three cycles of learning thus fulfilling the curricular requirement. Reports briefly the scientific training and didactic knowledge from the curriculum the content and pupils. Develop and informed opinion to planning, conducting classes and learning assessment. Makes and analyses of the activities as a teacher of a discipline and as an active participant in school and school community. Try the strategies, methodologies and decisions. Finally makes an analysis of teaching practice. What was found on this little experiment was a more conscious practice and a greater respect for ourselves as professional education, and students as active participants in the teaching /learning.

A presente dissertação é uma sinopse do trabalho desenvolvido no âmbito do estágio de Prática de Ensino Supervisionada em Educação Física. O estágio decorreu em duas escolas dos Ensinos Básico e Secundário. A dissertação encontra-se dividida por partes: preparação e planificação de aulas, conhecimento das turmas, condução das aulas, avaliação e processo reflexivo das várias etapas. Foram desenvolvidas actividades para as comunidades escolares de ambas as escolas e foi feita uma reflexão final do estágio que teve como objectivo analisar todos os procedimentos que foram desenvolvidos ao longo do ano lectivo. A passagem pelos quatro ciclos de ensino foi bastante produtiva para o enriquecimento do processo enquanto professor. As aulas seguiram o planeamento anual que estava delineado e foi esta ferramenta que possibilitou a existência de uma grande autonomia dos alunos, um maior controlo na prática pedagógica e a presença sistemática do jogo, trazendo uma motivação e empenhamento suplementar nas aulas e nas actividades desenvolvidas. ABSTRACT: The following dissertation is a synopsis of what was done at the scope of Supervised Practices for Physical Education beginning teachers. The traineeship took place in two schools from Primary to Secondary levels. The dissertation is divided into several parts: preparation and lessons planning, students' profile, lessons, evaluation and reflexive process on the different stages. lt was developed activities for the communities of the booth schools and was made a final reflection of the traineeship stage that have as a propose analyze all the procedures that was developed in all the academic year. The passage for the four cycles of school was very productive for the enrichment as a professor. The classes were follow as was expected and was that possibilities the existence of a great autonomy of the students, a bigger contrail in the pedagogy practice and systematic presence of the game that bring a motivation and supplementary interest in the classes and activities developed.

O presente relatório surge como um complemento à minha Prática de Ensino Supervisionada, realizada no ano lectivo de 2009/201O, visando essencialmente a reflexão e documentação do trabalho por mim desenvolvido ao longo deste ano. A minha Prática de Ensino Supervisionada debruçou-se sobre a disciplina de Educação Física, e decorreu na Escola EB 2,3 Conde de Vilalva, onde leccionei a disciplina a uma turma de 8° ano, e na Escola Secundária André de Gouveia, tendo leccionado a disciplina a um 12° ano. Durante este mesmo ano lectivo, leccionei ainda entre 8 intervenções a uma turma do 1° Ciclo de Ensino Básico, 1° ano; e 9 intervenções a uma turma do 2° Ciclo de Ensino Básico, o 5° ano. ABSTRACT: This document comes as a complement to my Supervised Teaching Practice, held in the 2009/201O academic year, aiming primarily to reflection and documentation of the work developed throughout this year. My Supervised Teaching Practice was focused on the discipline of Physical Education, and held at EB 2,3 Conde de Vilalva where I taught a class in 8th grade, and in high school André de Gouveia, where I taught the discipline to a class in 12th grade. During that same academic year, I taught between 8 and 1O interventions with a class of 1st cycle of education, 1st grade, and with a class of 2nd cycle of education, a 5th grade.

The present work is part of a research project that involves the study of new copper based complexes to be employed as photosensitizer in carbon dioxide photoreduction reaction. My research project is focused on the synthesis and characterization of 1,2,3 triazoles with a quinoline or pyridine in the lateral chain, which have been successively utilized to synthesize heteroleptic Cu(I) complexes. Redox potential and photophysic properties have been studied.

A recent integral-field spectroscopic (IFS) survey, the MASSIVE survey (Ma et al. 2014), observed the 116 most massive (MK < −25.3 mag, stellar mass M∗ > 10^11.6 M⊙) early-type galaxies (ETGs) within 108 Mpc, out to radii as large as 40 kpc, that correspond to ∼ 2 − 3 effective radii (Re). One of the major findings of the MASSIVE survey is that the galaxy sample is split nearly equally among three groups showing three different velocity dispersion profiles σ(R) outer of a radius ∼ 5 kpc (falling, flat and rising with radius). The purpose of this thesis is to model the kinematic profiles of six ETGs included in the MASSIVE survey and representative of the three observed σ(R) shapes, with the aim of investigating their dynamical structure. Models for the chosen galaxies are built using the numerical code JASMINE (Posacki, Pellegrini, and Ciotti 2013). The code produces models of axisymmetric galaxies, based on the solution of the Jeans equations for a multicomponent gravitational potential (supermassive black hole, stars and dark matter halo). With the aim of having a good agreement between the kinematics obtained from the Jeans equations, and the observed σ and rotation velocity V of MASSIVE (Veale et al. 2016, 2018), I derived constraints on the dark matter distribution and orbital anisotropy. This work suggests a trend of the dark matter amount and distribution with the shape of the velocity dispersion profiles in the outer regions: the models of galaxies with flat or rising velocity dispersion profiles show higher dark matter fractions fDM both within 1 Re and 5 Re. Orbital anisotropy alone cannot account for the different observed trends of σ(R) and has a minor effect compared to variations of the mass profile. Galaxies with similar stellar mass M∗ that show different velocity dispersion profiles (from falling to rising) are successfully modelled with a variation of the halo mass Mh.

Die Induktion von Toleranz spielt bei der Inhibition allergischer Immunreaktionen eine wichtige Rolle. Hierbei ist die Induktion regulatorischer T Zellen (Treg) von großer Bedeutung. Da zu einer erfolgreichen Behandlung von allergischen Erkrankungen bisher nur wenige Therapiemöglichkeiten zur Verfügung stehen wie die spezifische Immuntherapie (SIT), die allerdings nicht immer zum Erfolg führt, ist es wichtig neue Therapieformen zu entwickeln. rnIn dieser Arbeit wurde daher die biolistische DNA-Immunisierung mit Kombinations-Vakzinen bestehend aus einem allergenkodierenden Plasmid (βGalaktosidase (βGal)) in Kombination mit einem Plasmid, welches für ein immunmodulatorisches Molekül kodiert (Indolamin-2,3-Dioxygenase (IDO), Transforming Growth Factor beta (TGF-β) oder Interleukin-10 (IL-10)), durchgeführt und im Mausmodell der allergeninduzierten IgE-vermittelten Atemwegsinflammation auf ihre Wirksamkeit untersucht. Die Expression des Allergens zusammen mit dem immunregulatorischen Molekül in transfizierten Dendritischen Zellen (DCs) sollte zu einer Induktion von Treg führen und somit eine Suppression der Immunantwort bewirken. rnIn den Versuchen wurde zunächst der Effekt einer Transgenexpression unter der Kontrolle des ubiquitären CMV-Promotors mit dem der Transgenexpression unter der Kontrolle des Fascin-Promotors, der eine Genxpression spezifisch in DCs erlaubt, verglichen. Hierbei stellte sich heraus, dass es wichtig ist die Expression des Antigens mit Hilfe des Fascin-Promotors auf DCs zu beschränken. Einzig in diesem Fall konnte nach der Vakzinierung ein inhibitorischer Effekt auf die Entwicklung einer Atemwegshyperreaktivität durch Expression des Immunmodulatoren IDO beobachtet werden. Es zeigte sich auch, dass es von Vorteil ist, wenn das immunregulatorische Molekül unter Verwendung des CMV-Promotors in allen transfizierten Zellen exprimiert wird. Dies bewirkt, dass IDO in ausreichenden Konzentrationen vorhanden ist. rnDie Expression von βGal unter der Kontrolle des Fascin-Promotors (pFascin-βGal) in Kombination mit der Expression der Moleküle IL-10, TGF-β oder IDO unter Kontrolle des CMV-Promotors (pCMV-IL-10, pCMV-TGFβ, pCMV-IDO) bewirkte eine Immunsupprimierung, die sich in einer inhibierten Produktion antigenspezifischer Antikörper, einer verminderten Zytokin-Produktion, einer reduzierten Induktion zytotoxischer T-Zellen und in einer Inhibition der allergeninduzierten Atemwegshyperreaktivität zeigte, im Vergleich zu einer Vakzinierung mit pFascin-βGal in Kombination mit einem Kontroll-Plasmid. Bei nachfolgender Proteinsensibilisierung blieben diese Effekte jedoch nicht bestehen. Einzig durch Vakzinierung mit IL-10-kodierenden Plasmiden konnte eine moderate Verminderung der Atemwegsreaktivität nachgewiesen werden. rnIn einem therapeutischen Modell der Atemwegsinflammation, in dem die Mäuse vor der DNA-Immunisierung mit dem Protein sensibilisiert wurden, wurde demonstriert, dass im Vergleich zu Mäusen, die nur mit dem Protein sensibilisiert wurden, eine DNA-Immunisierung mit pFascin-βGal aber auch mit pCMV-βGal einen inhibierenden Einfluss auf die Entwicklung einer Atemwegsinflammation hat. Eine weitere Reduktion der Atemwegsreaktivität durch eine kombinierte Vakzinierung mit pCMV-IDO wurde nur erreicht, wenn βGal unter der Kontrolle des Fascin-Promotors exprimiert wurde, nicht aber unter Kontrolle des CMV-Promotors.rn

Derzeit stellt die allergenspezifische Immuntherapie die einzige nicht allein antisymptomatische Behandlungsform zur langfristigen Therapie von Typ I-Allergien dar, welche grundlegende Änderungen im immunologischen Geschehen induziert. Sie ist jedoch verbesserungswürdig in Bezug auf Behandlungsdauer, Erfolgschancen und Nebenwirkungen. Daher wurde in dieser Arbeit eine Strategie zur Therapie von Typ I-Allergien entwickelt und evaluiert, welche auf der Inhibition allergenspezifischer T-Zellen durch Dendritische Zellen (DC), die selektiv nach DNA-Immunisierung sowohl das relevante Allergen als auch Indolamin 2,3-dioxygenase (IDO) konstitutiv produzieren, basiert. IDO ist ein Enzym aus dem Tryptophan-Stoffwechsel, dessen Produktion durch DC einen lokalen immunsuppressiven Mechanismus induziert und in verschiedenen Situationen mit der Induktion peripherer Toleranz assoziiert ist. Zunächst wurden Plasmide hergestellt, die entweder IDO alleine oder IDO zusammen mit dem Antigen unter der Kontrolle des ubiquitär aktiven CMV- bzw. des DC-spezifischen Fascin-Promotors kodieren. Die Überprüfung der IDO-Expression durch die monocistronischen Plasmide anhand von Transfektionsexperimenten in vitro ergab, dass die IDO-Expression unter der Kontrolle des CMV-Promotors sehr viel stärker ausfiel als unter der Kontrolle des Fascin-Promotors. Nach Transfektion mit den bicistronischen Vektoren, in denen die Transgene für das Antigen und IDO durch eine IRES-Sequenz verbunden waren, war die IDO-Expression jedoch insgesamt sehr schwach. Im Rahmen der Überprüfung der Funktionalität der IDO-Expressionplasmide in vivo unter Verwendung der Genpistole wurden daher lediglich Plasmide getestet, die alleine IDO unter der Kontrolle des CMV-Promotors bzw. des Fascin-Promotors kodieren. Auch in vivo wurde eine stärkere IDO-Expression nach biolistischer Transfektion mit solchen Vektoren beobachtet, in denen der CMV-Promotor zur Expressionskontrolle verwendet wurde. Die Analyse des Einflusses einer Koexpression von IDO auf die durch biolistische Immunisierung mit einem antigenkodierenden Vektor induzierte systemische Immunantwort offenbarte einen inhibitorischer Effekt für den Fall, dass die Antigenproduktion mittels des Fascin-Promotors auf DC fokussiert war und die Expression des koapplizierten IDO-Transgens unter der Kontrolle des CMV-Promotors stand. In diesem Fall wurde eine Reduktion der antigenspezifischen IgG1- und IgG2a-Produktion, eine verringerte Sekretion von IFN-y durch restimulierte Milz- und Lymphknotenzellen sowie eine Reduktion der Zahl antigenspezifischer CD8+ Effektor-T-Zellen nachgewiesen. Im Mausmodell der IgE-vermittelten Typ I-Allergie wurde weiterhin gezeigt, dass nach prophylaktischer biolistischer Vakzinierung unter Verwendung dieser Vektorkombination eine Inhibition der durch die Vakzinierung bedingten antigenspezifischen Th1-Immunantwort ausgelöst wurde. Die Suppression der Th2-Antwort, welche durch Transfektion mit dem Antigenkodierenden Vektor unter Kontrolle des Fascin-Promotors bewirkt wurde, wurde durch Kotransfektion mit den IDO-kodierenden Vektoren aufrecht erhalten.

The free H(2)xspa ligands [xspa = pspa, Clpspa, tspa or fspa where p = 3-(phenyl), Clp = 3-(2-chlorophenyl), t = 3-(2-thienyl), f = 3-(2-furyl) and spa = 2-sulfanylpropenoato], their Zn(II) complexes of formula [HQ](2)[Zn(xspa)(2)] (HQ=diisopropylammonium) and the Cd(II) equivalents were prepared and characterized by elemental analysis and by IR, Raman and NMR ((1)H, (13)C) spectroscopy. X-Ray studies of the crystal structures of [HQ](2)[Zn(pspa)(2)], [HQ](2)[Zn(Clpspa)2], [HQ](2)[Zn(tspa)(2)] and [HQ](2)[Zn(fspa)(2)] show that the zinc atom is coordinated to two O atoms and two S atoms of the ligands in a distorted tetrahedral ZnO(2)S(2) environment. In the structures of [HQ](2)[Cd(pspa)(2)] and [HQ](2)[Cd(Clpspa)(2)] the cadmium atom is coordinated to three S atoms and two carboxylato O atoms of the ligands in a distorted trigonal bipyramidal environment. The interchange of ligands between Zn( II) and Cd( II) was studied by (113)Cd NMR spectroscopy. The in vitro protective effect of H(2)xspa and their Zn( II) complexes against Cd toxicity was investigated using the human hepatocarcinoma HepG2 cell line and the pig renal proximal tubule LLC-PK1 cell line. The incorporation of Zn( II) was found to be relevant in the case of H(2)pspa, with an increase observed in the cell viability of the LCC-PK1 cells with respect to the value for the free ligand.

The influence of Al(2)O(3) addition and sintering parameters on the mechanical properties and cytotoxicity of tetragonal ZrO(2)-3 mol% Y(2)O(3) ceramics was evaluated. Samples containing 0, 10, 20 and 30 wt.% of Al(2)O(3) particles were prepared by cold uniaxial pressing (80 MPa) and sintered in air at 1500, 1550 and 1600 degrees C for 120 min. The effects of the sintering conditions on the microstructure were analyzed by X-ray diffraction analysis and scanning electron microscopy. Hardness and fracture toughness were determined by the Vickers indentation method and the mechanical resistance by four-point bending tests. As a preliminary biological evaluation, ""in vitro"" cytotoxicity tests were realized to determine the cytotoxic level of the ZrO(2)-Al(2)O(3) composites, using the neutral red uptake method with NCTC clones L929 from the American Type Culture Collection (ATCC) bank. Fully dense ceramic materials were obtained with a hardness ranging between 1340 HV and 1585 HV, depending on the amount of Al(2)O(3) in the ZrO(2) matrix. On the other hand, no significant influence of the Al(2)O(3) addition on fracture toughness was observed, exhibiting values near 8 MPa m(1/2) for all compositions and sintering conditions studied. The non-cytotoxic behavior, the elevated fracture toughness, the good bending strength (sigma(f) = 690 MPa) and the elevated Weibull`s modulus (m = 11) exhibited by the material, show that these ceramic composites are highly suitable biomaterials for dental implant applications.
