Gene therapy is one of the major challenges of the post-genomic research and it is based on the transfer of genetic material into a cell, tissue or organ in order to cure or improve the patient s clinical status. In general, gene therapy consists in the insertion of functional genes aiming substitute, complement or inhibit defective genes. The achievement of a foreigner DNA expression into a population of cells requires its transfer to the target. Therefore, a key issue is to create systems, vectors, able to transfer and protect the DNA until it reaches the target. The disadvantages related to the use of viral vectors have encouraged efforts to develop emulsions as non-viral vectors. In fact, they are easy to produce, present suitable stability and enable transfection. The aim of this work was to evaluate two different non-viral vectors, cationic liposomes and nanoemulsions, and the possibility of their use in gene therapy. For the two systems, cationic lipids and helper lipids were used. Nanoemulsions were prepared using sonication method and were composed of Captex® 355; Tween® 80; Spam® 80; cationic lipid, Stearylamine (SA) or 1,2-dioleoyl-3-trimethylammoniumpropane (DOTAP) and water (Milli-Q®). These systems were characterized by average droplet size, Polidispersion Index (PI) and Zeta Potential. The stability of the systems; as well as the DNA compaction capacity; their cytotoxicity and the cytotoxicity of the isolated components; and their transfection capacity; were also evaluated. Liposomes were made by hydration film method and were composed of DOTAP; 1,2-dioleoyl-sn-glycero-3-phosphoethanolamine (DOPE), containing or not Rhodaminephosphatidylethanolamine (PE- Rhodamine) and the conjugate Hyaluronic Acid DOPE (HA-DOPE). These systems were also characterized as nanoemulsions. Stability of the systems and the influence of time, size of plasmid and presence or absence of endotoxin in the formation of lipoplexes were also analyzed. Besides, the ophthalmic biodistribution of PE-Rhodamine containing liposomes was studied after intravitreal injection. The obtained results show that these systems are promising non-viral vector for further utilization in gene therapy and that this field seems to be very important in the clinical practice in this century. It seeks to find an alternative to the current tantalum electrolytic capacitors in the market due to its high cost. Niobium is a potential substitute, since both belong to the same group of the periodic table and because of this have many similar physical and chemical properties. Niobium has several technologically important applications, and Brazil has the largest reserves, around 96%. There are including niobium in reserves of tantalite and columbite in Rio Grande do Norte. These electrolytic capacitors have high capacitance specifies, ie they can store high energy in small volumes compared to other types of capacitors. This is the main attraction of this type of capacitor because is growing demand in the production of capacitors with capacitance specifies increasingly high, this because of the miniaturization of various devices such as GPS devices, televisions, computers, phones and many others. The production route of the capacitor was made by powder metallurgy. The initial niobium powder supplied by EEL-USP was first characterized by XRD, SEM, XRF and laser particle size, to then be sieved into three particle size, 200, 400 e 635mesh. The powders were then compacted and sintered at 1350, 1450 and 1550°C using two sintering time 30 and 60min. Sintering is one of the most important parts of the process as it affects properties as porosity and surface cleaning of the samples, which greatly affected the quality of the capacitor. The sintered samples then underwent a process of anodic oxidation, which created a thin film of niobium pentóxido over the whole porous surface of the sample, this film is the dielectric capacitor. The oxidation process variables influence the performance of the film and therefore the capacitor. The samples were characterized by electrical measurements of capacitance, loss factor, ESR, relative density, porosity and surface area. After the characterizations was made an annealing in air ate 260ºC for 60min. After this treatment were made again the electrical measurements. The particle size of powders and sintering affected the porosity and in turn the specific area of the samples. The larger de area of the capacitor, greater is the capacitance. The powder showed the highest capacitance was with the smallest particle size. Higher temperatures and times of sintering caused samples with smaller surface area, but on the other hand the cleaning surface impurities was higher for this cases. So a balance must be made between the gain that is achieved with the cleaning of impurities and the loss with the decreased in specific area. The best results were obtained for the temperature of 1450ºC/60min. The influence of annealing on the loss factor and ESR did not follow a well-defined pattern, because their values increased in some cases and decreased in others. The most interesting results due to heat treatment were with respect to capacitance, which showed an increase for all samples after treatment

The use of raw materials from renewable sources for production of materials has been the subject of several studies and researches, because of its potential to substitute petrochemical-based materials. The addition of natural fibers to polymers represents an alternative in the partial or total replacement of glass fibers in composites. In this work, carnauba leaf fibers were used in the production of biodegradable composites with polyhydroxybutyrate (PHB) matrix. To improve the interfacial properties fiber / matrix were studied four chemical treatments to the fibers..The effect of the different chemical treatments on the morphological, physical, chemical and mechanical properties of the fibers and composites were investigated by scanning electron microscopy (SEM), infrared spectroscopy, X-ray diffraction, tensile and flexural tests, dynamic mechanical analysis (DMA), thermogravimetry (TGA) and diferential scanning calorimetry (DSC). The results of tensile tests indicated an increase in tensile strength of the composites after the chemical treatment of the fibers, with best results for the hydrogen peroxide treated fibers, even though the tensile strength of fibers was slightly reduced. This suggests a better interaction fiber/matrix which was also observed by SEM fractographs. The glass transition temperature (Tg) was reduced for all composites compared to the pure polymer which can be attributed to the absorption of solvents, moisture and other low molecular weight molecules by the fibers

The ionic nitriding process presents some limitations related with the control of the thickness of the layer and its uniformity. Those limitations that happen during the process, are produced due to edge effects, damage caused by arcing arc and hollow cathode, mainly in pieces with complex geometry and under pressures in excess of 1 mbar. A new technique, denominated ASPN (active screen shapes nitriding) it has been used as alternative, for offering many advantages with respect to dc plasma conventional. The developed system presents a configuration in that the samples treated are surrounded by a large metal screen at high voltage cathodic potencials, (varying between 0 and 1200V) and currents up to 1 A. The sample is placed in floting potential or polarized at relatively lower bias voltages by an auxiliary source. As the plasma is not formed directly in the sample surface but in the metal screen, the mentioned effects are eliminated. This mechanism allows investigate ion of the transfer of nitrogen to the substrate. Optical and electronic microscopy are used to exam morphology and structure at the layer. X-ray difration for phase identification and microhardness to evaluate the efficiency of this process with respect to dc conventional nitriding

With the advances in medicine, life expectancy of the world population has grown considerably in recent decades. Studies have been performed in order to maintain the quality of life through the development of new drugs and new surgical procedures. Biomaterials is an example of the researches to improve quality of life, and its use goes from the reconstruction of tissues and organs affected by diseases or other types of failure, to use in drug delivery system able to prolong the drug in the body and increase its bioavailability. Biopolymers are a class of biomaterials widely targeted by researchers since they have ideal properties for biomedical applications, such as high biocompatibility and biodegradability. Poly (lactic acid) (PLA) is a biopolymer used as a biomaterial and its monomer, lactic acid, is eliminated by the Krebs Cycle (citric acid cycle). It is possible to synthesize PLA through various synthesis routes, however, the direct polycondensation is cheaper due the use of few steps of polymerization. In this work we used experimental design (DOE) to produce PLAs with different molecular weight from the direct polycondensation of lactic acid, with characteristics suitable for use in drug delivery system (DDS). Through the experimental design it was noted that the time of esterification, in the direct polycondensation, is the most important stage to obtain a higher molecular weight. The Fourier Transform Infrared (FTIR) spectrograms obtained were equivalent to the PLAs available in the literature. Results of Differential Scanning Calorimetry (DSC) showed that all PLAs produced are semicrystalline with glass transition temperatures (Tgs) ranging between 36 - 48 °C, and melting temperatures (Tm) ranging from 117 to 130 °C. The PLAs molecular weight characterized from Size Exclusion Chromatography (SEC), varied from 1000 to 11,000 g/mol. PLAs obtained showed a fibrous morphology characterized by Scanning Electron Microscopy (SEM)

Brazil has vast amounts of hydric resources, whose quality has been deteriorating due to pollutant dumping. Household waste disposal is one of the main sources of water pollution, stimulating bacteria proliferation and introducing microorganisms, including those from fecal matter. Conventional water disinfection methods are a solution, but on the downside, they lead to the formation byproducts hazardous to human health. In this study, aiming to develop bactericidal filters for the disinfection of drinking water; silver nanoparticles were deposited on alumina foams through three routes: sputtering DC, dip coating and in situ chemical reduction of silver nitrate. The depositions were characterized through X-ray diffraction, scanning electron microscopy and EDS element mapping. The influence of the depositions on permeability and mechanical properties of the ceramic foams was assessed and, in sequence, a preliminary antibacterial efficiency analysis was carried out. Characterization results indicate that the chemical reduction routes were efficient in depositing homogeneously distributed silver particles and that the concentration of the metallic precursor salt affects size and morphology of the particles. The antibacterial efficiency analysis indicates that the chemical reduction filters have potential for water disinfection

O presente trabalho teve como objetivo compartimentar a sub-bacia do Baixo Rio Piracicaba - SP, em unidades homogêneas quanto à potencialidade à erosão visando a subsidiar o gerenciamento ambiental. A importância desta pesquisa concentra-se no intenso desenvolvimento dos processos erosivos na área em foco e na sua importância socioeconômica em níveis estadual e nacional. O procedimento adotado para atingir o objetivo foi a análise da rede de drenagem e dos lineamentos obtidos pelas imagens TM/Landsat-5. O resultado obtido foi a divisão da área em quatro compartimentos quanto à potencialidade à erosão: muito alta, alta, média e baixa. Concluiu-se que a área estudada é heterogênea, com regiões sujeitas a diferentes intensidades de processos erosivos e que a sistemática adotada se mostrou eficiente para caracterizar as compartimentações fisiográficas da potencialidade erosiva, e essas compartimentações podem e devem ser utilizadas como ponto de partida para estudos ambientais e de utilização do território, em consonância com o desenvolvimento sustentável.

Bactérias endofíticas são microrganismos que habitam o interior dos tecidos vegetais e são conhecidas promotoras do crescimento de plantas, favorecendo sua nutrição, pela solubilização de fosfatos, além de produzirem fito-hormônio, enzimas e sideróforos, apresentando potencial para uso como bioinoculantes. O objetivo deste trabalho foi isolar e caracterizar bactérias endofíticas de milho eficientes na solubilização de fosfatos de cálcio e de ferro e na produção de sideróforos, substâncias que atuam na captação de ferro.Para o isolamento das bactérias, foram coletadas plantas de milho no florescimento em área de cerrado, sendo estas posteriormente desinfestadas e maceradas para o isolamento dos endófitos. Foram isoladas 113 bactérias provenientes de raízes (54,9%), folhas (20,4%) e seiva de milho (24,8%). Dessas bactérias, 49 foram caracterizadas geneticamente com base na sequência do 16S rDNA e avaliadas quanto à capacidade de solubilização de fosfatos em meio de cultura líquido contendo fosfato de cálcio e de ferro e produção de sideróforos. Os isolados mais eficientes na solubilização de fosfato de cálcio e fosfato de ferro produziram entre 179,4 a 193,7 mgP. L-1 e 62,7 a 68,7 mg P. L-1, respectivamente. Foi observada uma correlação negativa de -0,58 e -0,48 entre a solubilização de fósforo nos dois fosfatos testados, P-Ca e P-Fe, e os valores de pH, respectivamente, indicando a produção de ácidos orgânicos como o principal mecanismo de solubilização de P. O sideróforo produzido por 69% dos microrganismos avaliados foi do tipo carboxilato. As bactérias endofíticas eficientes na solubilização de P foram identificadas principalmente como pertencentes ao gênero Bacillus e Pantoea. Foi possível concluir que as estirpes bacterianas provenientes do microbioma interno de milho cultivado em solo de cerrado possuem características promissoras de biossolubilização de fosfatos e promoção de crescimento de plantas, sendo possível neste estudo a seleção de seis isolados do gênero Bacillus para futuros testes de inoculação em plantas.

Os produtos obtidos da fermentação de pescados marinhos frescos registrados como fertilizantes orgânicos são ricos em nutrientes, possuem em sua composição quitina e quitosana, podendo-se constituir em um produto alternativo adequado para o controle de fitopatógenos. O objetivo deste trabalho foi estudar o potencial de um hidrolisado de peixe em controlar o oídio em abobrinha, F. oxysporum f. sp. lycopersici raça 3 em tomate e Pythium spp. em pepino. Nesse sentido, um fertilizante orgânico obtido da fermentação de resíduos de pescados marinhos frescos foi pulverizado semanalmente nas plantas de abobrinha, com o auxílio de um compressor de pintura 10 1b/pol2 m a 0%, 0,5%, 1%, 2%, 3%, 4%, 5% e 10% (v/v) para o controle do oídio. Este mesmo fertilizante foi incorporado ao substrato nas concentrações de 0%, 5%, 10%, 20%, 30%, 40%, 50% e 100% (por volume de água necessária para atingir a capacidade de campo), em experimento realizado dentro e fora de casa de vegetação. Outros experimentos realizados avaliaram a eficiência do hidrolisado de peixe no controle do tombamento causado por Pythium spp., em pepino e a murcha-de-fusário causada por Fusarium oxysporum f. sp. lycopersici raça 3, em tomate. Os experimentos foram realizados em casa de vegetação e o delineamento experimental foi inteiramente casualizado, com dez repetições por tratamento. Para o pepino, foi utilizada a técnica de estimular a população original do solo com aveia. Assim, o hidrolisado de peixe foi incorporado ao solo dez dias após a mistura com aveia, em concentrações de 0%, 5%, 10%, 20%, 30%, 40%, 50% e 100% do volume de água, para atingir a capacidade de campo do solo e com incubação aberta e fechada. Após dez dias de incubação, 200 ml da mistura foram adicionados ao colo das plantas de pepino no estádio de 2° folhas verdadeiras. A avaliação foi realizada após cinco dias, determinando-se o número de plântulas tombadas. O hidrolisado de peixe, tanto pulverizado na folhas quanto incorporado no substrato, não controlou o oídio da abobrinha. Por outro lado, pode ser observado o efeito do hidrolisado de peixe no desenvolvimento das plantas e no desenvolvimento de Trichoderma no substrato. A partir da concentração de 30%, não houve tombamento de plantas. Por outro lado, o tombamento foi de 100% para os tratamentos com 0 e 5% do fertilizante. Para o experimento do Fusarium, foram utilizados três isolados da raça 3 de Fusarium oxysporum f. sp. lycopersici (isolados 145, 146 e 149). Após a infestação, o substrato foi incubado por quinze dias com o hidrolisado de peixe e foi incorporado ao substrato nas seguintes concentrações: 0%, 5%, 10%, 20%, 30%, 40% e 50% volume de água necessária para atingir a capacidade de campo. Uma muda de tomate cultivar Santa Clara suscetível à raça 3 com 30 dias de idade, foi transferida para cada vaso. A severidade da doença foi avaliada após 40 dias, por meio de escala de notas para escurecimento vascular e sintomas externos. De modo geral, todas as doses do hidrolisado de peixe reduziram, significativamente, a severidade da doença.
