Accurate diagnosis of orthopedic device-associated infections can be challenging. Culture of tissue biopsy specimens is often considered the gold standard; however, there is currently no consensus on the ideal incubation time for specimens. The aim of our study was to assess the yield of a 14-day incubation protocol for tissue biopsy specimens from revision surgery (joint replacements and internal fixation devices) in a general orthopedic and trauma surgery setting. Medical records were reviewed retrospectively in order to identify cases of infection according to predefined diagnostic criteria. From August 2009 to March 2012, 499 tissue biopsy specimens were sampled from 117 cases. In 70 cases (59.8%), at least one sample showed microbiological growth. Among them, 58 cases (82.9%) were considered infections and 12 cases (17.1%) were classified as contaminations. The median time to positivity in the cases of infection was 1 day (range, 1 to 10 days), compared to 6 days (range, 1 to 11 days) in the cases of contamination (P < 0.001). Fifty-six (96.6%) of the infection cases were diagnosed within 7 days of incubation. In conclusion, the results of our study show that the incubation of tissue biopsy specimens beyond 7 days is not productive in a general orthopedic and trauma surgery setting. Prolonged 14-day incubation might be of interest in particular situations, however, in which the prevalence of slow-growing microorganisms and anaerobes is higher.

Résumé : Dans le modèle murin d'infection avec le parasite protozoaire Leishmania major (L. major), la souche de souris C57BL/6 est résistante a |'infection et développe une réponse protectrice Thelper (Th) 1. Inversement, les souris de la souche BALB/c développent une réponse Th2 et sont sensibles a cette infection. A la suite d'une infection avec ce parasite, les neutrophiles sont les premières cellules présentes au site d'infection et sont recrutées de manière égale dans les souches résistantes et sensibles à L. major, Néanmoins, trois jours après l'infection, la majorité des neutrophiles disparaissent du site d'infection chez les souris C57BL/6, tandis que ils restent jusqu'a dix jours chez les souris BALB/c. Un rôle crucial des neutrophiles a été démontré durant l'infection avec L. major. En effet, la déplétion de ces cellules avant |'infection dans les souris BALB/c, conduit a une réduction du développement des lésions, associée à une baisse de la charge parasitaire et a une modification de la réponse immunitaire vers une réponse Th1 dans des souris normalement sensibles a |'infection, suggérant un rôle immunorégulateur de ces neutrophiles durant les premiers jours de l'infection. Dans la première partie de cette thèse, nous avons étudié le rôle des neutrophiles suite à l'infection avec L. major. Nous avons démontré que le parasite induisait des phénotypes de neutrophiles distincts chez les souris résistantes ou sensibles à L. major. Suite à l'exposition au parasite, les neutrophiles de souris C57BL/6 ont montré une expression élevée des récepteurs Toll-like 2, 7 et 9 ainsi que la sécrétion d'lL-12p7O et d'lL-10, alors que ceux de souris BALB/c sécrétaient de l'IL-12p40 et du TGFB. Nous avons ensuite démontré qu'en réponse à L. major, au contraire des neutrophiles de BALB/c, les neutrophiles de souris résistantes C57BL/6, libéraient la chimiokine CCL3 attirant les cellules dendritiques. Le rôle crucial de cette chimiokine dans la migration de la première de vague de cellules dendritiques au site d'infection ainsi que son rôle dans le développement de la réponse immunitaire subséquente a été établi. Ces résultats démontrent que les neutrophiles, suite a |'infection avec le parasite L. major, créent un microenvironnement capable de déterminer le développement d'une réponse immunitaire spécifique a un antigène. Dans un second temps, nous nous sommes intéressés au rôle des neutrophiles suite a l'infection avec d'autres espèces de Leishmania: L, doriovani et L. mexicaria, agents responsables de leishmaniose viscérale et cutanée chronique respectivement. Un rôle crucial des neutrophiles a été démontré dans la réponse protectrice suite a l'infection avec L. donovani, l'absence de ces cellules amenant à une susceptibilité au parasite accrue, associée avec une induction préférentielle d'une réponse Th2. Inversement, la déplétion des neutrophiles lors de l'infection avec L. mexicaria aboutit a une résistance accrue, comme constaté par la baisse dela charge parasitaire, la hausse de la réponse Th1 ainsi la baisse de la réponse Th2 dans les souris déplétées en neutrophiles. Néanmoins, malgré le rôle délétère des neutrophiles sur le développement d'une réponse protectrice suite à |'infection avec L. mexicana, ces cellules sont nécessaires pour une résolution correcte dela réponse inflammatoire. En résumé, cette étude révèle un rôle majeur des neutrophiles lors de |'infection avec plusieurs especes de Leishmania. Résumé pour un large public : Les neutrophiles font partie de la famille des globules blancs. A la suite d'une infection, ces cellules sont les premières a être recrutées au site d'infection et sont impliquées dans |'élimination des pathogènes. Dans cette thèse, nous nous somme donc intéressés au rôle que pouvaient jouer ces neutrophiles durant l'infection avec le parasite protozoaire Leishmania major (L. major). Dans le modèle murin d'infection avec L. major, la majorité des souches de souris utilisées dans la recherche, dont les souris de la souche C57BL/6, développent de petites lésions qui guérissent spontanément après quelques semaines (souris résistantes). ll existe néanmoins, quelques souches de souris, dont la souche de souris BALB/c, qui développent des lésions qui ne guérissent pas (souris sensibles). Il a été observé que lors de l'lnfection avec ce parasites les neutrophiles étaient les premières cellules recrutées au site de l'lnfection dans toutes les souches de souris, toutefois trois jours après le début dela réaction immunitaire, la majorité des neutrophiles disparaissent chez les souris C57BL/6, tandis qu'ils restent jusqu'à dix jours chez les souris BALB/c. De plus, un rôle crucial des neutrophiles a été démontré durant l'infection avec L. major. En effet, l'absence de neutrophiles durant les trois premiers jours de l'infection chez les souris sensibles à |'infection, rend ces souris résistantes. Ces résultats suggèrent donc un rôle régulateur de la réponse immunitaire des neutrophiles durant les premiers jours de l'infection. Dans la première partie de cette thèse, nous avons étudié le rôle des neutrophiles suite à l'infection avec L. major. Nous avons donc analysé la sécrétion des cytokines, molécules essentielles qui déterminent la réponse immunitaire, par les neutrophiles. Nous avons démontré que le parasite induisait une sécrétion de cytokines différente entre les souris résistantes ou sensibles a L. major. Nous avons ensuite démontré que seule la souche de souris résistante sécrétait la chimiokine CCL3, connue pour être impliquée dans le recrutement de différentes cellules au site d'infecti0n, dont les cellules dendritiques. Les cellules dendritiques sont un élément fondamental pour un bon déroulement d'une réponse immunitaire, de par leur rôle décisif de liaison entre une réponse précoce non-spécifique au pathogène et une réponse plus tardive spécifique au pathogène et nécessaire pour |'élimination de dernier. Nous avons démontré que les neutrophiles de souris résistantes sécrétaient CCL3 et recrutaient les cellules dendritiques au site d'infecti0n, jouant de ce fait un rôle essentiel dans le développement de la réponse immunitaire. Ces résultats démontrent que les neutrophiles, suite à l'infection avec le parasite L. major, créent un microenvironnement capable de déterminer le développement d'une réponse immunitaire. Dans un second temps, nous nous sommes intéressés au rôle des neutrophiles suite à l'lnfection avec d'autres espèces de Leishmania, L. donovani et L. mexicana. Nous avons pu montrer un rôle crucial de ces cellules dans la réponse à ces deux parasites. En effet, suite à |'infection avec L. donovani, un rôle protecteur des neutrophiles a été observé, leur absence menant à une susceptibilité accrue aux parasites. Dans le cas de l'infection avec L. mexicana, une réduction de |'infection a été observée en absence de neutrophiles, avec néanmoins une augmentation de la lésion, suggérant un rôle important de ces cellules dans le développement de la réponse immunitaire ainsi que dans le contrôle de la réponse inflammatoire. En résumé, cette étude révèle un rôle majeur des neutrophiles lors de l'lnfection avec plusieurs membres de la famille Leishrnania. Summary : Upon infection with the protozoan parasite Leishmania major (L. major), C57BL/6 mice show a resistant phenotype, developing a protective Thelper (Th) 1 response. ln contrast, BALB/c mice develop a Th2 response and are susceptible to infection. Following inoculation with the parasite, neutrophils are the first cells migrating at the site of infection and are equally recruited in both L. major- resistant and susceptible mouse strains. However, after three days of infection, almost all neutrophils disappear from the site of infection in C57BL/6 mice, while they persist until ten days in BALB/c mice. Neutrophils were shown to play a crucial role during infection with L. major. indeed, depletion of these cells in BALB/c mice prior to infection with the parasite led to a lower Iesion development, associated with a lower parasite burden and a modification in the immune response towards a Th1 response in these otherwise susceptible mice, suggesting an immunomodulatory role for neutrophils during the first days of infection. ln the first part of this thesis, we were interested in better understanding the role of neutrophils in infection with L. major. \/\/e found that this parasite was inducing distinct neutrophil phenotypes in L. major-resistant and susceptible mice. Upon exposition with L. major, C57BL/6 neutrophils were reported to express high level of Toll-like receptors 2, 7, 9 mRNA and secrete IL-12p70 and IL-10, while BALB/c neutrophils secreted homodimers of IL-12p40, and TGFB. We then demonstrated that in response to L. major, neutrophils from L. major-resistant C57BL/6 mice release the CCL3 dendritic cell attracting chemokine, which is critical for the first wave of dendritic cell migration to the site of infection and in the development of the subsequent immune response. Altogether, these results demonstrated that upon infection with L. major, neutrophils create a microenvironment that can determine the development of an antigen-specific immune response. ln the second part of the thesis we were interested in understanding the role of neutrophils upon infection with of other species of Leishmania: L. donovani causing visceral leishmaniasis and L. mexicana, agent of chronic cutaneous leishmaniasis. Upon infection with L. donovani, neutrophils were found to play a crucial role in the early protective response, their absence leading to an increased susceptibility to the parasite, associated with the preferential induction of a Th2 response. ln contrast, depletion of these cells early in infection with L. mexicana was leading to an increased resistance, as observed by a decreased parasite burden, increased Th1 and decreased Th2 response in neutrophil-depleted mice. However, despite the deleterious role of neutrophils on the development of a protective immune response upon L. mexicana infection, these cells were required for the proper resolution of the inflammatory response. Altogether, these results highlight a major immunomodulatory role for neutrophils in infection with several species of Leishmania.

Les neutrophiles constituent la première ligne de défense contre un grand nombre de pathogènes. Après infection avec Leishmania major, les neutrophiles migrent rapidement et massivement au site d'infection par le parasite. Les neutrophiles sont d'importants acteurs dans l'orchestration de la réponse anti-Leishmania, via la sécrétion de nombreuses cytokines, chimiokines et composés stockés dans leurs granules. De plus, les neutrophiles interagissent avec les cellules présentatrices d'antigènes, telles que les cellules dendritiques et en conséquences contribuent au développement de la réponse adaptative. A ce jour, l'impact des neutrophiles sur 1'activation des cellules dendritiques et les possibles conséquences de l'interaction neutrophiles avec ces dernières sur l'évolution de la maladie suite à l'infection avec L. major reste peu connu. Ainsi, nous avons dans un premier temps investigué l'influence des neutrophiles sur 1'activation des cellules dendritiques in vitro. Suite à cela, nous avons analysé le rôle des neutrophiles sur 1'activation des cellules dendritiques présentes au site d'infection et dans les ganglions drainants in vivo, après inoculation intra-dermale de L. major dans le pavillon auriculaire de souris de souche C57BL/6 et BALB/c, ainsi que les conséquences de la déplétion des neutrophiles sur l'évolution de la maladie. Nous avons pu démontrer que les neutrophiles ont un impact négatif sur l'activation des cellules dendritiques exposées à L. major in vitro, via des mécanismes impliquant la sécrétion de Prostaglandines par les neutrophiles et la séquestration des parasites. La déplétion des neutrophiles dans les souris BALB/c durant les premiers jours après infection avec L. major dans le derme de l'oreille résulte en une augmentation de l'expression de marqueurs d'activation des cellules dendritiques présentes dans les ganglions drainants, mais pas au site d'infection. De plus, les souris BALB/c transitoirement déplétées en neutrophiles développent des lésions significativement plus petites, une réponse de type Th2 diminuée et une charge parasitaire plus faible au site d'infection que les souris non déplétées. La déplétion des neutrophiles dans les souris C57BL/6 n'a pas d'influence détectable sur l'activation des cellules dendritiques que ce soit au niveau des ganglions drainants ou au site d'infection. La progression de la lésion et la charge parasitaire ne sont pas affectées par la déplétion des neutrophiles, malgré le développement d'une réponse Th2 diminuée par rapport aux souris non déplétées. En résumé, dans les premières heures après infection, les neutrophiles ont un effet négatif sur l'activation des cellules dendritiques et sur la réponse anti-Leishmania dans les souris BALB/c tandis que leur rôle paraît moins important dans les souris C57BL/6. Ces résultats peuvent avoir d'importantes implications en terme de développement de nouveaux vaccins contre Leishmania. - Neutrophils constitute the first line of defense against a variety of pathogens. Following Leihmania major infection neutrophils migrate rapidly and massively to the site of parasite inoculation. They are important players in the orchestration of the anti-leishmania response through the release of a plethora of cytokines, chemokines and granular components. In addition, neutrophils interact with antigen-presenting cells such as dendritic cells (DCs) and thereby contribute to the development of the adaptive immune response. However, the impact of neutrophils on the activation of DCs and possible consequences on disease progression following L. major infection are poorly understood. Therefore, we first investigated the influence of neutrophils on DC activation in vitro. Next we analyzed the role of neutrophils on the activation of DCs present at the site of infection and in the draining lymph node (dLN) following inoculation of L. major in the ear pinna of C57BL/6 and BALB/c mice and analyzed the consequences of early neutrophil depletion on disease progression. We could demonstrate that neutrophils had a negative impact on the activation of DCs exposed to L. major in vitro through mechanisms involving neutrophil-derived prostaglandins and sequestration of parasites. In BALB/c mice, depletion of neutrophils during the first days of infection with L. major in the ear dermis increased the expression of activation markers on dLN DCs but not on DCs present at the site of infection. In addition, BALB/c mice transiently depleted of neutrophils developed significantly smaller lesions, a decreased Th2 immune response and harbored fewer parasites at the site of infection compared to non-depleted littermates. In C57BL/6, mice early depletion of neutrophils had no detectable impact on the activation of DCs both at the site of infection and in the dLN following L. major inoculation. Furthermore, depletion of neutrophils had no major effect on lesion evolution and parasite loads despite the development of a decreased Th2 immune response compared to non- depleted littermates. In summary, at the onset of infection, neutrophils are detrimental for DC activation and the anti-leishmania response in BALB/c mice while their role appears to be less important in C57BL/6 mice. These findings could have important implications for the design of new vaccination strategies.

The objective of this work was to assess the parasitism potential of Acarophenax lacunatus (Cross & Krantz) on Tribolium castaneum (Herbst). The experimental units encompassed Petri dishes containing 25 g of wheat infested with 20 adults of this beetle species. After seven days of infestation, different mite densities (0, 2, 4, 6, 8 and 10 females) were inoculated into the Petri dishes. The increase in mite density led to a linear increase in parasitism, and a consequent reduction in populations of this insect. It is possible to use this mite species in the biological control of T. castaneum.

OBJECTIVE: The major objective of this study was to investigate the effects of several days of intense exercise on growth hormone (hGH) testing using the World Anti-Doping Agencies hGH isoform differential immunoassays. Additionally the effects of circadian variation and exercise type on the isoform ratios were also investigated. STUDY DESIGN: 15 male athletes performed a simulated nine day cycling stage race. Blood samples were collected twice daily over a period of 15days (stage race+three days before and after). hGH isoforms were analysed by the official WADA immunoassays (CMZ Assay GmbH). RESULTS: All measured isoform ratios were far below the WADA decision limits for an adverse analytical finding. Changes in the isoform ratios could not be clearly connected to circadian variation, exercise duration or intensity. CONCLUSIONS: The present study demonstrates that the hGH isoform ratios are not significantly affected by exercise or circadian variation. We demonstrated that heavy, long term exercise does not interfere with the decision limits for an adverse analytical finding.

The Lane-Wells Road Logger was utilized primarily to determine the feasibility of employing such a device for moisture and density control in Iowa highway construction. A secondary objective was the use of the Road Logger to obtain information concerning moisture content and density during and after construction. Correlation studies with conventional test results required a small portion of the lease period. Practically all phases of construction and most materials utilized in base and surface courses were surveyed. Results of this study were good, in general, with the Road Logger indicating dry density slightly higher and the moisture content slightly lower than conventional results in most instances. Economic feasibility seemed to pose the greatest problem for the acceptance of the Road Logger as a standard compaction control device. It would appear from the findings of this study that probably only large projects, or several smaller contracts tested simultaneously, could justify the expense of the Logger. A total of about 128 miles were surveyed with the Logger during the lease period. Approximately 16 days of downtime due to minor breakdowns were recorded. Inclement weather forcing construction delays resulted in several idle days in which the Logger's full capabilities were not realized.

BACKGROUND: Findings from randomised trials have shown a higher early risk of stroke after carotid artery stenting than after carotid endarterectomy. We assessed whether white-matter lesions affect the perioperative risk of stroke in patients treated with carotid artery stenting versus carotid endarterectomy. METHODS: Patients with symptomatic carotid artery stenosis included in the International Carotid Stenting Study (ICSS) were randomly allocated to receive carotid artery stenting or carotid endarterectomy. Copies of baseline brain imaging were analysed by two investigators, who were masked to treatment, for the severity of white-matter lesions using the age-related white-matter changes (ARWMC) score. Randomisation was done with a computer-generated sequence (1:1). Patients were divided into two groups using the median ARWMC. We analysed the risk of stroke within 30 days of revascularisation using a per-protocol analysis. ICSS is registered with controlled-trials.com, number ISRCTN 25337470. FINDINGS: 1036 patients (536 randomly allocated to carotid artery stenting, 500 to carotid endarterectomy) had baseline imaging available. Median ARWMC score was 7, and patients were dichotomised into those with a score of 7 or more and those with a score of less than 7. In patients treated with carotid artery stenting, those with an ARWMC score of 7 or more had an increased risk of stroke compared with those with a score of less than 7 (HR for any stroke 2·76, 95% CI 1·17-6·51; p=0·021; HR for non-disabling stroke 3·00, 1·10-8·36; p=0·031), but we did not see a similar association in patients treated with carotid endarterectomy (HR for any stroke 1·18, 0·40-3·55; p=0·76; HR for disabling or fatal stroke 1·41, 0·38-5·26; p=0·607). Carotid artery stenting was associated with a higher risk of stroke compared with carotid endarterectomy in patients with an ARWMC score of 7 or more (HR for any stroke 2·98, 1·29-6·93; p=0·011; HR for non-disabling stroke 6·34, 1·45-27·71; p=0·014), but there was no risk difference in patients with an ARWMC score of less than 7. INTERPRETATION: The presence of white-matter lesions on brain imaging should be taken into account when selecting patients for carotid revascularisation. Carotid artery stenting should be avoided in patients with more extensive white-matter lesions, but might be an acceptable alternative to carotid endarterectomy in patients with less extensive lesions. Over three years the postharvest quality of 'Marli' peaches harvested from the integrated (IFP) and conventional production (CFP) systems was evaluated. The peaches were harvested from commercial orchards of Prunus persica at two locations close to the city of São Jerônimo, RS, Brazil, and stored at 0.5°C for 10, 20 or 30 days. The peaches were evaluated at harvest, at retrieval from storage and after ripening periods at 20°C. No differences in fruit weight losses were determined. Decay incidence was low, and no differences were detected amongst systems in both 2001 and 2002 seasons, but in the 2000 season CFP peaches were more decayed. Flesh firmness of peaches from the IFP system were greater than CFP fruits in the years 2000 and 2001. In 2002, firmness changed little during storage and ripening. Peaches from the IFP in 2000 had higher titratable acidity and lower soluble solids. In the 2000 season, flesh browning was observed in decayed fruits, always at ripening after 20 or 30 days of cold storage Chilling injuries such as browning, woolliness and leatheriness ocurred in 2002. There were no differences amongst systems related to peach quality.

The objective of this work was to evaluate the effect of somatic cell counts (SCC) in casein fractions of ultra high temperature (UHT) milk. Raw milks were categorized in SCC groups of low (200,000-320,000 cells mL-1), intermediate (380,000-560,000 cells mL-1) and high cells (600,000-800,000 cells mL-1). Five replicates of UHT milks within each SCC category were analyzed for casein fractions after 8, 30, 60, 90 and 120 days of storage through high performance liquid chromatography. SCC showed effect only on beta-casein reduction. SCC in raw milk increases the proteolysis of UHT milk, as a consequence of beta-casein degradation.

Exposure to PM10 and PM2.5 (particulate matter with aerodynamic diameter smaller than 10 μm and 2.5 μm, respectively) is associated with a range of adverse health effects, including cancer, pulmonary and cardiovascular diseases. Surface characteristics (chemical reactivity, surface area) are considered of prime importance to understand the mechanisms which lead to harmful effects. A hypothetical mechanism to explain these adverse effects is the ability of components (organics, metal ions) adsorbed on these particles to generate Reactive Oxygen Species (ROS), and thereby to cause oxidative stress in biological systems (Donaldson et al., 2003). ROS can attack almost any cellular structure, like DNA or cellular membrane, leading to the formation of a wide variety of degradation products which can be used as a biomarker of oxidative stress. The aim of the present research project is to test whether there is a correlation between the exposure to Diesel Exhaust Particulate (DEP) and the oxidative stress status. For that purpose, a survey has been conducted in real occupational situations where workers were exposed to DEP (bus depots). Different exposure variables have been considered: - particulate number, size distribution and surface area (SMPS); - particulate mass - PM2.5 and PM4 (gravimetry); - elemental and organic carbon (coulometry); - total adsorbed heavy metals - iron, copper, manganese (atomic adsorption); - surface functional groups present on aerosols (Knudsen flow reactor). Several biomarkers of oxidative stress (8-hydroxy-2'-deoxyguanosine and several aldehydes) have been determined either in urine or serum of volunteers. Results obtained during the sampling campaign in several bus depots indicated that the occupational exposure to particulates in these places was rather low (40-50 μg/m3 for PM4). Bimodal size distributions were generally observed (5 μm and <1 μm). Surface characteristics of PM4 varied strongly, depending on the bus depot. They were usually characterized by high carbonyl and low acidic sites content. Among the different biomarkers which have been analyzed within the framework of this study, mean urinary levels of 8-hydroxy-2'-deoxyguanosine increased significantly (p<0.05) during two consecutive days of exposure for non-smoker workers. On the other hand, no statistically significant differences were observed for serum levels of hexanal, nonanal and 4- hydroxy-nonenal (p>0.05). Biomarkers levels will be compared to exposure variables to gain a better understanding of the relation between the particulate characteristics and the formation of ROS by-products. The objective of this work was to determine the effects of postharvest application of 1-methylcyclopropene (1-MCP) and two calcium salts, applied individually or combined, on firmness and visual quality of fresh-cut muskmelon stored in air, for 18 days. Two sets of fruits, one of them exposed to 1-MCP at 300 nL L-1, were cut into cubes, dipped in deionized water, or in 1% Ca solutions as CaCl2, or in calcium amino acid chelate (Ca-chelate), placed in clamshell containers, and stored in air at 5±1ºC and 90±5% RH, for 18 days. The assay was conducted using an entirely randomized design, with three replications, in a split plot array. Evaluation of visual appearance, color, flesh firmness, total soluble solids, titratable acidity, and pH was performed right after treatments, and every period of three days, up to eighteen days. Application of 1-MCP at 300 nL L-1, calcium chloride or Ca-chelate, or the combination 1-MCP and calcium, preserved initial freshness and reduced softening of the samples. Ca-chelate synergistically enhanced the effect of 1-MCP on firmness after nine days of storage, while calcium chloride improved firmness of the samples throughout storage. Ca-chelate may serve as an alternative for shelf life extension of cantaloupe fresh-cut muskmelon.

The objective of this study was to evaluate the mycorrhizal dependency of mangaba tree (Hancornia speciosa) plantlets, under increasing levels of phosphorus fertilization. The experimental design was completely randomized in a 4×5 factorial arrangement with three mycorrhizal fungi inocula - Gigaspora margarita, Glomus etunicatum, or a pool of native mycorrhizal fungi (Acaulospora longula, Glomus clarum, Gigaspora albida, Paraglomus sp.) -, and a nonmycorrhizal control, in combination with five levels of phosphorus applied to the substrate: 0, 25, 50, 75, and 100 mg kg-1. After 180 days of growth, plantlets with inoculation of native mycorrhizal pool produced more shoot and root dry biomass and had higher shoot phosphorus content and accumulation. The noninoculated control showed the lowest values, independently of the phosphorus level. The highest relative mycorrhizal dependency occurred with the inoculation of native mycorrhizal fungi. Plants with mycorrhizal fungi did not respond to phosphorus addition above 50 mg kg-1. Mangaba tree is highly dependent on mycorrhiza, but the degree of dependency varies according to phosphorus levels and fungal inocula. In general, mangaba tree is more responsive to mycorrhizal fungi inoculation than to phosphorus addition.

OBJECTIVE: To assess whether vasopressin V1a receptor blockade reduces the abnormal vasoactive response to cold in patients suffering from Raynaud's phenomenon (RP). METHODS: SR 49059, an orally active, non-peptidic vasopressin V1a receptor antagonist, was given orally (300 mg once daily) to 20 patients with RP in a single-centre, double-blind, placebo-controlled, randomized cross-over study with two 7-day periods of treatment separated by 21 days of washout. Bilateral finger systolic blood pressure and skin temperature were assessed before and after immersion of the hand in cold water for 3 min (15 degrees C) during the screening phase and three times (before and 2 and 4 h after drug intake) on days 1 and 7 of each of the two treatment periods. Recovery of digital pressure and skin temperature was measured 0, 10, 20 and 32 min after the end of the cold immersion test. RESULTS: SR 49059 significantly attenuated the cold-induced fall in systolic pressure by 14.5% (95% confidence interval 0-29; P = 0.045) on the most affected hand on day 7 compared with placebo. Temperature recovery after the end of the cold test showed a trend to enhancement 2 and 4 h after SR 49059 on day 7 (P = 0.060 and P = 0.062 respectively). The beneficial effects on finger pressure and temperature recovery were obtained without changes in supine blood pressure or in heart rate. CONCLUSION: SR 49059 given orally once a day for 7 days to patients with RP showed favourable effects compared with placebo on finger systolic pressure and temperature recovery after cold immersion, without inducing side-effects.

Objectives: To compare the clinical characteristics, species distribution and antifungal susceptibility of Candida bloodstream isolates (BSI) in breakthrough (BTC) vs. non-breakthrough candidemia (NBTC) and to study the effect of prolonged vs. short fluconazole (F) exposure in BTC.Methods: Candida BSI were prospectively collected during 2004- 2006 from 27 hospitals (seven university, 20 affiliated) of the FUNGINOS network. Susceptibility to F, voriconazole (V) and caspofungin (C) was tested in the FUNGINOS mycology reference laboratory by microtitre broth dilution method with the Sensititre YeastOneTM test panel. Clinical data were collected using standardized CRFs. BTC was defined as occurring during antifungal treatment/prophylaxis of at least three days duration prior to the candidemia. Susceptibility of BSI was defined according to 2010/2011 CLSI clinical breakpoints.Results: Out of 567 candidemia episodes, 550 Candida BSI were available. Of these, 43 (7.6%) were from BTC (37/43, 86% were isolated after F exposure). 38 BTC (88.4%) and 315 NBTC (55.6%) occurred in university hospitals (P < 0.001). The majority of patients developing BTC were immunocompromised: higher proportions of haematological malignancies (62.8% in BTC vs. 47.1% in NBTC, P < 0.001), neutropenia (37.2% vs. 11.8%, P < 0.001), acute GvHD (14% vs. 0.2%, P < 0.001), immunosuppressive drugs (74.4% vs. 7.8%, P < 0.001), and mucositis (32.6% vs. 2.3%, P < 0.001) were observed. Other differences between BTC and NBTC were higher proportions of patients with central venous catheters in the 2 weeks preceding candidemia (95.3% vs. 83.4%, P = 0.047) and receiving total parenteral nutrition (62.8% vs. 35.9%, P < 0.001), but a lower proportion of patients treated with gastric proton pump inhibitors (23.3% vs. 72.1%, P < 0.001). Overall mortality of BTC and NBTC was not different (34.9% vs. 31.7%, P = 0.73), while a trend to higher attributable mortality in BTC was found (13.9% vs. 6.9%, P = 0.12). Species identification showed a majority of C. albicans in both groups (51.2% in BTC vs. 62.9% in NBTC, P = 0.26), followed by C. glabrata (18.6% vs. 18.5%), C. tropicalis (2.3% vs. 6.3%) and C. parapsilosis (7.0% vs. 4.7%). Significantly more C. krusei were detected in BTC versus NBTC (11.6% vs. 1.6%, P = 0.002). The geometric mean MIC for F, V and C between BTC and NBTC isolates was not significantly different. However, in BTC there was a significant association between duration of F exposure and the Candida spp.: >10 days of F was associated with a significant shift from susceptible Candida spp. (C. albicans, C. parapsilosis, C. tropicalis, C. famata) to non-susceptible species (C. glabrata, C. krusei, C. norvegensis). Among 21 BTC episodes occurring after £10 days of F, 19% of the isolates were non-susceptible, in contrast to 68.7% in 16 BTC episodes occurring after >10 days of F (P = 0.003).Conclusions: Breakthrough candidemia occurred more often in immunocompromised hosts. Fluconazole administered for >10 days was associated with a shift to non-susceptible Candida spp.. Length of fluconazole exposure should be taken into consideration for the choice of empirical antifungal treatment.

The objective of this work was to determine the best conditions of use of the synthetic sex pheromone of Phyllocnistis citrella Stainton for monitoring this species in citrus groves in northeastern Brazil. Pheromone doses (0.0, 0.1, 1, 10 and 100 μg) and longevity (1, 15, 29, 43 and 57-day-old lures) and trap height (0.5, 1.5 and 2.5 m), color (green, red, and white) and model influence on P. citrella males capture were evaluated. The doses of 10 and 100 μg of the synthetic sex pheromone - a 3:1 blend of (Z,Z,E)-7,11,13-hexadecatrienal and (Z,Z)-7,11-hexadecadienal - attracted the greatest number of P. citrella males. Traps baited with these two both dosages continued to capture P. citrella males at a comparable rate for over eight weeks in citrus groves. Although there was no significant decrease in activity of both dosages until 57 days of exposure to the environment, the higher dose, as time passed, attracted significantly more P. citrella males than the lower dose. There were no significant differences in male capture in traps with synthetic sex pheromone placed at 1.5 and 2.5 m height, wich had the better results. Trap color and model did not affect male capture.
