First published April 9, 2010; doi:10.1152/ajpheart.00620.2009.-We recently observed the enhanced serine and matrix metalloproteinase (MMP) activity in the spontaneously hypertensive rat (SHR) compared with its normotensive Wistar-Kyoto (WKY) rat and the cleavage of membrane receptors in the SHR by MMPs. We demonstrate in vivo that MMP-7 and MMP-9 injection leads to a vasoconstrictor response in microvessels of rats that is blocked by a specific MMP inhibitor (GM-6001, 1 mu M). Multiple pathways may be responsible. Since the beta(2)-adrenergic receptor (beta(2)-AR) is susceptible to the action of endogenous MMPs, we hypothesize that MMPs in the plasma of SHRs are able to cleave the extracellular domain of the beta(2)-AR. SHR arterioles respond in an attenuated fashion to beta(2)-AR agonists and antagonists. Aorta and heart muscle of control Wistar rats were exposed for 24 h (37 C) to fresh plasma of male Wistar and WKY rats and SHRs with and without doxycycline (30 mu M) and EDTA (10 mM) to reduce MMP activity. The density of extracellular and intracellular domains of beta(2)-AR was determined by immunohistochemistry. The density of the extracellular domain of beta(2)-AR is reduced in aortic endothelial cells and cardiac microvessels of SHRs compared with that of WKY or Wistar rats. Treatment of the aorta and the heart of control Wistar rats with plasma from SHRs, but not from WKY rats, reduced the number of extracellular domains, but not intracellular domains, of beta(2)-AR in aortic endothelial cells and cardiac microvessels. MMP inhibitors (EDTA and doxycycline) prevented the cleavage of the extracellular domain. Thus MMPs may contribute to the reduced density of the extracellular domain of beta(2)-AR in blood vessels and to the increased arteriolar tone of SHRs compared with normotensive rats.

In alveolar macrophages, leukotriene (IT) B(4) and cysteinyl LTs (LTC(4), LTD(4) and LTE(4)) both enhance Fc gamma receptor (Fc gamma R)-mediated phagocytosis. In the present study we investigated the role of specific PKC isoforms (PKC-alpha and -delta), the MAP kinases p38 and ERK 1/2, and PI3K in mediating the potentiation of Fc gamma R-mediated phagocytosis induced by addition of leukotrienes to the AMs. It was found that exogenously added LTB(4) and LTD(4) both enhanced PKC-delta and -alpha phosphorylation during Fc gamma R engagement. Studies with isoform-selective inhibitors indicated that exogenous LTB(4) effects were dependent on both PKC-alpha and -delta, while LTD(4) effects were exclusively due to PKC-delta activation. Although both exogenous LTB(4) and LTD(4) enhanced p38 and ERK 1/2 activation, LTB(4) required only ERK 1/2, while LTD(4) required only p38 activation. Activation by both LTs was dependent on PI3K activation. Effects of endogenous LTs on kinase activation were also investigated using selective LT receptor antagonists. Endogenous LTB(4) contributed to Fc gamma R-mediated activation of PKC-alpha, ERK 1/2 and PI3K, while endogenous cysLTs contributes to activation of PKC-delta, p38 and PI3K. Taken together, our data show that the capacities of LTB(4) and LTD(4) to enhance Fc gamma R-mediated phagocytosis reflect their differential activation of specific kinase programs. Background Epidemiological and experimental data suggest that bacteria] lipopolysaccharides (LPS) can either protect from or exacerbate allergic asthma. Lipopolysaccharides trigger immune responses through toll-like receptor 4 (TLR4) that in turn activates two major signalling pathways via either MyD88 or TRIF adaptor proteins. The LPS is a pro-Type 1 T helper cells (Th 1) adjuvant while aluminium hydroxide (alum) is a strong Type 2 T helper cells (Th2) adjuvant, but the effect of the mixing of both adjuvants on the development of lung allergy has not been investigated. Objective We determined whether natural (LPS) or synthetic (ER-803022) TLR4 agonists adsorbed onto alum adjuvant affect allergen sensitization and development of airway allergic disease. To dissect LPS-induced molecular pathways, we used TLR4-, MyD88-, TRIF-, or IL-12/IFN-gamma-deficient mice. Methods Mice were sensitized with subcutaneous injections of ovalbumin (OVA) with or without TLR4 agonists co-adsorbed onto alum and challenged with intranasally with OVA. The development of allergic lung disease was evaluated 24 h after last OVA challenge. Results Sensitization with OVA plus LPS co-adsorbed onto alum impaired in dose-dependent manner OVA-induced Th2-mediated allergic responses such as airway eosinophilia, type-2 cytokines secretion, airway hyper-reactivity, mucus hyper production and serum levels of IgE or IgG1 anaphylactic antibodies. Although the levels of IgG2a, Th1 -affiliated isotype increased, investigation into the lung-specific effects revealed that LPS did not induce a Th1 pattern of inflammation. Lipopolysaccharides impaired the development of Th2 immunity, signaling via TLR4 and MyD88 molecules and via the IL-12/IFN-gamma axis, but not through TRIF pathway. Moreover, the synthetic TLR4 agonists that proved to have a less systemic inflammatory response than LPS also protected against allergic asthma development. Conclusion Toll-like receptor 4 agonists co-adsorbed with allergen onto alum down-modulate allergic lung disease and prevent the development of polarized T cell-mediated airway inflammation.

Focal and segmental glomerulosclerosis (FSGS) is one of the most important causes of end-stage renal failure. The bradykinin B1 receptor has been associated with tissue inflammation and renal fibrosis. To test for a role of the bradykinin B1 receptor in podocyte injury, we pharmacologically modulated its activity at different time points in an adriamycin-induced mouse model of FSGS. Estimated albuminuria and urinary protein to creatinine ratios correlated with podocytopathy. Adriamycin injection led to loss of body weight, proteinuria, and upregulation of B1 receptor mRNA. Early treatment with a B1 antagonist reduced albuminuria and glomerulosclerosis, and inhibited the adriamycin-induced downregulation of podocin, nephrin, and alpha-actinin-4 expression. Moreover, delayed treatment with antagonist also induced podocyte protection. Conversely, a B1 agonist aggravated renal dysfunction and even further suppressed the levels of podocyte-related molecules. Thus, we propose that kinin has a crucial role in the pathogenesis of FSGS operating through bradykinin B1 receptor signaling. Lipopolysaccharides from gram-negative bacteria are amongst the most common causative agents of acute lung injury, which is characterized by an inflammatory response, with cellular infiltration and the release of mediators/cytokines. There is evidence that bradykinin plays a role in lung inflammation in asthma but in other types of lung inflammation its role is less clear. In the present study we evaluated the role of the bradykinin B(1) receptor in acute lung injury caused by lipopolysaccharide inhalation and the mechanisms behind bradykinin actions participating in the inflammatory response. We found that in C57BI/6 mice, the bradykinin B(1) receptor expression was up-regulated 24 h after lipopolysaccharide inhalation. At this time, the number of cells and protein concentration were significantly increased in the bronchoalveolar lavage fluid and the mice developed airway hyperreactivity to methacholine. In addition, there was an increased expression of tumor necrosis factor-alpha, interleukin-1 beta and interferon-gamma and chemokines (monocytes chemotactic protein-1 and KC) in the bronchoalveolar lavage fluid and in the lung tissue. We then treated the mice with a bradykinin B, receptor antagonist, R-954 (Ac-Orn-[Oic(2), alpha-MePhe(5), D-beta Nal(7), Ile(8)]desArg(9)-bradykinin), 30 min after lipopolysaccharide administration. We observed that this treatment prevented the airway hyperreactivity as well as the increased cellular infiltration and protein content in the bronchoalveolar lavage fluid. Moreover, R-954 inhibited the expression of cytokines/chemokines. These results implicate bradykinin, acting through B(1) receptor, in the development of acute lung injury caused by lipopolysaccharide inhalation. (C) 2010 Elsevier B.V. All rights reserved.

Thyroid hormone receptors (TRs) are ligand-gated transcription factors with critical roles in development and metabolism. Although x-ray structures of TR ligand-binding domains (LBDs) with agonists are available, comparable structures without ligand (apo-TR) or with antagonists are not. It remains important to understand apo-LBD conformation and the way that it rearranges with ligands to develop better TR pharmaceuticals. In this study, we conducted hydrogen/deuterium exchange on TR LBDs with or without agonist (T(3)) or antagonist (NH(3)). Both ligands reduce deuterium incorporation into LBD amide hydrogens, implying tighter overall folding of the domain. As predicted, mass spectroscopic analysis of individual proteolytic peptides after hydrogen/deuterium exchange reveals that ligand increases the degree of solvent protection of regions close to the buried ligand-binding pocket. However, there is also extensive ligand protection of other regions, including the dimer surface at H10-H11, providing evidence for allosteric communication between the ligand-binding pocket and distant interaction surfaces. Surprisingly, C-terminal activation helix H12, which is known to alter position with ligand, remains relatively protected from solvent in all conditions suggesting that it is packed against the LBD irrespective of the presence or type of ligand. T(3), but not NH(3), increases accessibility of the upper part of H3-H5 to solvent, and we propose that TR H12 interacts with this region in apo-TR and that this interaction is blocked by T(3) but not NH(3.) We present data from site-directed mutagenesis experiments and molecular dynamics simulations that lend support to this structural model of apo-TR and its ligand-dependent conformational changes. To shed more light on the molecular requirements for recognition of thyroid response elements (TRES) by thyroid receptors (TRs), we compared the specific aspects of DNA TRE recognition by different TR constructs. Using fluorescence anisotropy, we performed a detailed and hierarchical study of TR-TRE binding. This wits done by comparing the binding affinities of three different TR constructs for four different TRE DNA elements, including palindromic sequences and direct repeats (F2, PAL, DR-1, and DR-4) as well as their interactions with nonspecific DNA sequences. The effect of MgCl(2) on suppressing of nonselective DNA binding to TR was also investigated. Furthermore, we determined the dissociation constants of the hTR beta DBD (DNA binding domain) and hTR beta DBD-LBD (DNA binding and ligand binding domains) for specific TRES. We found that a minimum DNA recognition peptide derived from DBD (H1TR) is sufficient for recognition and interaction with TREs, whereas scrambled DNA sequences were unrecognized. Additionally, we determined that the TR DBD binds to F2, PAL, and DR-4 with high affinity and similar K(d) values. The TR DBD-LBD recognizes all the tested TRES but binds preferentially to F2, with even higher affinity. Finally, our results demonstrate the important role played by LBDs in modulating TR-DNA binding.

Angiotensin II (Ang II) and its transmembrane AT(1) receptor were selected in order to test an innovative strategy that might allow the assessment of the agonist binding site in the receptor molecule. With the use of the 2,2,6,6-tetramethylpiperidine-1-oxyl-4-amino-4-carboxylic acid (TOAC) paramagnetic probe, a biologically active agonist (TOAC(1)-Ang II), as well as an inactive control (TOAC(4)-Ang II) analogs were mixed in solution with various synthesized AT(1) fragments. Comparative intermolecular interactions, as estimated by analyzing the EPR spectra of solutions, suggested the existence of an agonist binding site containing a sequence composed of portions of the N-terminal (13-17) and the third extracellular loop (266-278) fragments of the AT(1) molecule. Therefore, this combined EPR-TOAC approach shows promise as an alternative for use also in other applications related to specific intermolecular association processes.

Nicotinic acetylcholine receptors (nAChRs) were studied in detail in the past regarding their interaction with therapeutic and drug addiction related compounds. Using fast kinetic whole-cell recording, we have now studied effects of tacrine, an agent used clinically to treat Alzheimer`s disease, on currents elicited by activation of rat alpha(3)beta(4) nAChR heterologously expressed in KX alpha(3)beta(4)R2 cells. Characterization of receptor activation by nicotine used as agonist revealed a K(d) of 23 +/- 0.2 mu M and 4.3 +/- 1.3 for the channel opening equilibrium constant, Phi(-1). Experiments were performed to investigate whether tacrine is able to activate the alpha(3)beta(4) nAChR. Tacrine did not activate whole-cell currents in KX alpha(3)beta(4)R2 cells but inhibited receptor activity at submicromolar concentration. Dose response curves obtained with increasing agonist or inhibitor concentration revealed competitive inhibition of nAChRs by tacrine, with an apparent inhibition constant, K(I), of 0.8 mu M. The increase of Phi(-1) in the presence of tacrine suggests that the drug stabilizes a nonconducting open channel form of the receptor. Binding studies with TCP and MK-801 ruled out tacrine binding to common allosteric sites of the receptor. Our study suggests a novel mechanism for action of tacrine on nAChRs besides inhibition of acetylcholine esterase.

The mechanism of eupalmerin acetate (EUAC) actions on the embryonic muscle nicotinic acetylcholine receptor (nAChR) in BC3H-1 cells was studied by using whole-cell and single-channel patch-clamp current measurements. With whole-cell currents, EUAC did not act as an agonist on this receptor. Coapplication of 30 mu M EUAC with 50 mu M, 100 N, or 500 mu M carbamoylcholine (CCh) reversibly inhibited the current amplitude, whereas, with 20 mu M CCh, current was increased above control values in the presence of EUAC. EUAC concentration curves (0.01-40 N) obtained with 100 mu M and 500 mu M CCh displayed slope coefficients, n(H), significantly smaller than one, suggesting that EUAC bound to several sites with widely differing affinities on the receptor molecule. The apparent rate of receptor desensitization in the presence of EUAC and CCh was either slower than or equal to that obtained with CCh alone. The major finding from single-channel studies was that EUAC did not affect single-channel conductance or the ability of CCh to interact with the receptor. Instead, EUAC acted by increasing the channel closing rate constant. The results are not consistent with the competitive model for EUAC inhibition, with the sequential open-channel block model, or with inhibition by increased desensitization. The data are best accounted for by a model in which EUAC acts by closed-channel block at low concentrations, by positive modulation at intermediate concentrations, and by negative allosteric modulation of the open channel at high concentrations. As origens das populações e o processo de povoamento das Américas são questões bastante controversas e, por isso, têm sido amplamente estudadas. O gene LDLR (low density lipoprotein receptor), que está localizado no braço curto do cromossomo 19 (p13.1-p13.3) e possui 18 éxons, bem como uma porção 3’UTR onde ocorrem dois elementos Alu completos (chamados de U e D) e um parcial, foi escolhido para esclarecer esses e outros problemas de evolução e variação genética humana. O objetivo geral deste trabalho foi verificar o que esta região hipervariável, especificamente na sua porção U, nos indicaria sobre a história evolutiva das populações ameríndias. Para este trabalho foram analisadas amostras de DNA de indivíduos nativos da Mongólia (n=24), da Sibéria (n=26), das Américas do Norte (n=11), Central (n=26) e do Sul (n=16), totalizando 14 populações e 103 indivíduos. Essas amostras foram amplificadas pela técnica de PCR (polymerase chain reaction), utilizando-se primers específicos para o segmento hipervariável U e, posteriormente, foram elas seqüenciadas automaticamente. Quatorze sítios polimórficos foram encontrados, classificáveis em sete haplótipos, sendo que o sítio 3809 apresentou uma transversão de C para G não descrita na literatura. A estimativa geral de diversidade nucleotídica (π) foi de 0.62%, considerada alta para um marcador autossômico. Verificou-se uma certa uniformidade haplotípica entre a Ásia e a América, mas com a diferenciação maior ocorrendo entre a Mongólia e a América+Sibéria. Não foi detectada diferenciação significativa entre nativos sul e centro-americanos. De um modo geral, o estudo desse marcador confirmou uma provável origem asiática para as populações ameríndias e apoiou a hipótese de uma única onda de migração no processo de povoamento pré-histórico das Américas.

Neste trabalho investigou-se as características do receptor à insulina e a capacidade de captação de glicose nas brânquias do caranguejo Chasmagnathus granulata aclimatado a diferentes tempos (24, 72 e 144 horas) de estresse hiper e hiposmótico. Primeiramente, o cDNA do receptor para insulina foi parcialmente clonado e seqüenciado em brânquias posteriores de Chasmagnathus granulata. A seqüência peptídica mostrou a presença de 39 aminoácidos e foi designada CGIRLTK (C. granulata insulina receptor-like tyrosine kinase). Esta seqüência apresentou significativa homologia com o domínio tirosina quinase da subunidade b dos receptores para insulina de mamíferos (69%) e de Drosophila (74%). Sítios de ligação à insulina foram caracterizados nas membranas plasmáticas das brânquias através do estudo de ligação com 125I-insulina. A atividade tirosina quinase foi determinada pela capacidade do CGIRLTK de fosforilar o substrato sintético poly (Glu; Tyr 4:1). A captação de glicose foi avaliada pela captação de [14C] 2-deoxi-D-glicose pelo tecido branquial. Nas brânquias posteriores a insulina bovina estimulou significativamente a fosforilação do CGIRLTK nos animais aclimatados a 20‰ de salinidade (controle), já nas brânquias anteriores este estímulo não foi observado. O estresse hiperosmótico (34 ‰ de salinidade) levou a uma diminuição do número e da afinidade dos receptores à insulina nas brânquias posteriores, bem como a uma redução na atividade tirosina quinase. A captação de glicose não mudou durante os tempos de estresse osmótico estudados Esses resultados mostram que o estresse hiperosmótico modifica a sinalização da insulina, causando um estado de resistência à insulina nas brânquias posteriores. Nenhuma mudança foi observada na concentração dos receptores à insulina nas brânquias posteriores de caranguejos aclimatados durante 24 horas ao estresse hiposmótico (0‰). Contudo, foi observada uma redução na afinidade dos receptores pela insulina bovina. A fosforilação do CGIRLTK diminui às 24 horas de estresse e retornou aos valores basais às 144horas. A captação de glicose não foi alterada significativamente. Os resultados sugerem que o estresse hiposmótico modifica as características do CGIRLTK nas brânquias posteriores de C. granulata de forma tempo-dependente. Essas mudanças são parte dos ajustes necessários à sobrevivência à baixa salinidade. Nas brânquias anteriores, durante aclimatação ao estresse hiperosmótico, foi observada redução da concentração e da capacidade de fosforilação dos receptores insulínicos. Contudo, a insulina bovina não estimulou a fosforilação nas brânquias anteriores durante o estresse Nenhuma alteração foi observada na concentração e na afinidade de receptores à insulina nas brânquias anteriores após 24 horas de estresse hiposmótico. A fosforilação do receptor à insulina diminuiu após 24 horas de estresse e voltou aos valores basais após 72 horas. A capacidade de captação de glicose, por sua vez, não foi modificada em função de mudanças na osmoliridade do ambiente. Assim como no estresse hiperosmótico, a insulina bovina não estimulou a fosforilação nas brânquias anteriores no estresse hiposmótico. Os resultados deste trabalho demonstram que o estresse osmótico modifica as características do CGILRTK e conseqüentemente a transdução do sinal insulínico nas brânquias. As respostas às alterações de salinidade dependem do tipo de estresse ao qual o animal é submetido e da brânquia estudada (anterior ou posterior). As mudanças observadas no sinal insulínico fazem parte dos ajustes necessários para a regulação osmótica frente às mudanças ambientais de salinidade.

Considerando que as doenças cardiovasculares representam a maior causa de mortalidade e morbidade em países ocidentais, a aterosclerose se destaca pelo fato de predispor os pacientes ao infarto do miocárdio, a acidentes vasculares cerebrais e a doenças vasculares periféricas. Neste contexto, a oxidação de lipoproteínas do plasma, particularmente LDL, é um dos fatores de risco para eventos cardiovasculares, pois é reconhecida e internalizada por macrófagos, ocasionando a sua diferenciação em foam cells. Diversos fatores participam deste processo de diferenciação, como a expressão de receptores de scavenger CD 36, proporcionando aumento na captação de LDL oxidada, aumento na síntese endógena de colesterol e ativação de fatores nucleares que iniciam a transcrição de proteínas específicas e fatores de crescimento que disparam a aterogênese. Os fenômenos celulares relacionados à apoptose também são de especial importância, tanto no desenvolvimento da lesão aterosclerótica como na estabilidade da placa e formação de trombos. As prostaglandinas (PGs) ciclopentenônicas (CP-PGs), em particular a PGA2 e a 15-desóxi-∆12,14-PGJ2 são uma classe especial de PGs que, em diminutas concentrações, disparam a expressão das proteínas de choque térmico (hsp), que são citoprotetoras. Além disso, CP-PGs bloqueiam a ativação do fator nuclear pró-inflamatório NF-κB tornando-as potentes agentes antiinflamatórios. Embora as PGs das famílias A e J guardem uma série de características em comum, a 15-desóxi-∆12,14- PGJ2 é o ligante fisiológico do fator nuclear pró-aterogênico PPAR-γ, enquanto as PGs da família A ativam apenas a via citoprotetora das hsp. Este trabalho teve como objetivo avaliar os efeitos das CP-PGs sobre a expressão gênica de fatores relacionados à diferenciação de macrófagos em foam cells, bem como proteínas reguladoras do processo de apoptose, em células da linhagem pró-monocítica humana U937. Para tal, as células foram tratadas com CPPGs em presença e/ou ausência de LDL nat e LDL ox, o RNA foi extraído para a realização de RT-PCR para PPAR-γ, CD 36, HMG-CoA redutase e proteínas de apoptose Caspase 3, p53 e Bcl-xL. O tratamento estatístico utilizado foi análise de variância (ANOVA one-way) e teste “t” de student, com resultados expressos como médias + desvios-padrão da média, com P<0,05. Os resultados obtidos demontraram que as CP-PGs PGA2 (20µM-24h) e PGJ2 (1,5µM-24h) inibiram a expressão gênica do fator nuclear PPAR- γ (64 % (PGA2), 88 % (15- d-PGJ2)) nas células U937, em presença de LDL oxidada, quando comparado ao controle. PGA2 inibiu a expressão de HMG-CoA redutase (33 %), enzima chave da síntese de colesterol intracelular, e o tratamento com as CP-PGs também inibiu a apoptose nas células tratadas em presença de LDL oxidada. Os dados sugerem que as CP-PGs apresentam grande potencial para o tratamento da aterosclerose, já que, além de apresentarem efeito antiinflamatório, inibem a expressão do fator nuclear pró-aterogênico PPAR-γ, do receptor de scavenger CD36 (apenas a 15-desóxi-∆12,14-PGJ2) e da enzima HMG-CoA redutase. O bloqueio da apoptose nas células estudadas pode estar relacionado à citoproteção oferecida por estas PGs. Embora investigações in vivo deste laboratório tenham mostrado a eficácia do tratamento com CP-PGs em camundongos portadores de aterosclerose, estudos adicionais são necessários para esclarecer-se o efeito antiaterogênico das mesmas.

There is evidence that several fibroblast growth factors (FGFs) are involved in growth and development of the corpus luteum (CL), but many FGFs have not been investigated in this tissue, including FGF10. The objective of this study was to determine if FGF10 and its receptor (FGFR2B) are expressed in the CL. Bovine CL were collected from an abattoir and classed as corpus hemorrhagica (stage 1), developing (stage 11), developed (stage 111), and regressed (stage IV) CL. Expression of FGF10 and FGFR2B mRNA was measured by reverse transcription-polymerase chain reaction (RT-PCR). Both genes were expressed in bovine CL, and FGF10 expression did not differ between stages of CL development. FGF10 protein was localized to large and small luteal cells by immunohistochemistry. FGFR2B expression was approximately threefold higher in regressed compared to developing and developed CL (P < 0.05). To determine if FGF10 and FGFR2B expression is regulated during functional luteolysis, cattle were injected with PGF2 alpha and CL collected at 0, 0.5, 2, 4, 12, 24, 48, and 64 hr thereafter (n = 5 CL/time point), and mRNA abundance was measured by real-time RT-PCR. FGF10 mRNA expression did not change during functional luteolysis, whereas FGFR2B mRNA abundance decreased significantly at 2, 4, and 12 hr after PGF2a, and returned to pretreatment levels for the period 24-64 hr post-PGF2 alpha. These data suggest a potential role for FGFR2B signaling during structural luteolysis in bovine CL.
