To study the influence of different organic amendments on the quality of poultry manure compost, three pilot composting trials were carried out with different mixes: poultrymanure/carcasse meal/ashes/grape pomace (Pile 1), poultry manure/cellulosic sludge (Pile 2) and poultry manure (Pile 3). For all piles, wood chips were applied as bulking agent. The process was monitored, over time, by evaluating standard physical and chemical parameters, such as, pH, electric conductivity, moisture, organic matter and ash content, total carbon and total nitrogen content, carbon/nitrogen ratio (C/N) and content in mineral elements. Piles 1 and 2 reached a thermophilic phase, however having different trends. Pile 1 reached this phase earlier than Pile 2. For both, the pH showed a slight alkaline character and the electric conductivity was lower than 2 mS/cm. Also, the initial C/N value was 22 and reached values lower than 15 at the end of composting process. The total N content of the Pile 1 increased slightly during composting, in contrast with the others piles. At the end of composting process, the phosphorus content ranged between 54 and 236 mg/kg dry matter, for Pile 2 and 3, respectively. Generally, the Piles 1 and 3 exhibited similar heavy metals content. This study showed that organic amendments can be used as carbon source, given that the final composts presented parameters within the range of those recommended in the 2nd Draft of EU regulation proposal (DG Env.A.2 2001) for compost quality.

Biodiesel production from waste cooking oil with methanol was carried out in the presence of poly(vinyl alcohol) with sulfonic acid groups (PVA-SO3H) and polystyrene with sulfonic acid groups (PS-SO3H), at 60°C. The PVA-SO3H catalyst showed higher catalytic activity than the PS-SO3H one. In order to optimize the reaction conditions, different parameters were studied. An increase of waste cooking oil conversion into fatty acid methyl esters with the amount of PVA-SO3H was observed. When the transesterification and esterification of WCO was carried out with ethanol over PVA-SO3H, at 60°C, a decrease of biodiesel production was also observed. The WCO conversion into fatty acid ethyl ester increased when the temperature was increased from 60 to 80°C. When different amounts of free fatty acids were added to the reaction mixture, a slight increase on the conversion was observed. The PVASO3H catalyst was reused and recycled with negligible loss in the activity.

Colibacillosis is a rather important economic problem for poultry production, associated to Avian Pathogenic Escherichia coli (APEC) strains, which may cause several extraintestinal pathologies, such as airsacculitis and cellulitis in broiler chickens, and salpingitis and peritonitis in broiler breeders, leading to septicemic mortality. Control of morbidity and mortality in the outbrakes of colibacillosis may be performed with antibiotics and/or by vaccination. The use antibiotics is frequently inef - fective as Escherichia coli ( E. coli ) is considered the largest reservoir of antimicrobial resistance, characteristic that may even transmit to other bacteria, turning the situation into a serious problem of public health. Vaccination may be the alternative solution but as many different strains arise, flock-specific autovaccines seem to be needed under several possible protocols, with live attenuated and/or inactivated vaccines from different strains that should be identified and characterized according to their virulence factors, within different flocks.

Nel settore alimentare viene utilizzata un’elevata quantità di materie plastiche per conservare i prodotti e facilitarne la distribuzione. L’utilizzo di questi polimeri ha un costo ambientale piuttosto elevato, per questo trovare surrogati ecosostenibili diventa sempre più importante. In questa tesi abbiamo testato l’efficacia del confezionamento di un prodotto altamente deperibile, quale carne di pollo, con un biofilm a base di chitosano. Il chitosano è polisaccaride largamente presente in natura, dotato di caratteristiche chimico-fisiche che permettono l’ottenimento di un film con proprietà meccaniche e di barriera simili ai polimeri tradizionali, oltre a possedere attività antibatterica. Abbiamo realizzato film contenenti chitosano e altri biocomposti, quali montmorillonite, nanoparticelle di ossido di zinco e olio essenziale di rosmarino, per un totale di 6 film con diversa composizione. Tramite analisi microbiologiche e chimico-fisiche abbiamo confrontato l’efficacia dei diversi film prodotti rispetto ad un controllo (carne conservata in un contenitore asettico). Le analisi sono state svolte in doppio, a 0, 3, 7, 10, 15 giorni di conservazione ad una temperatura di 4°C. In diversi film abbiamo ottenuto una riduzione significativa rispetto al controllo (p<0,05) della conta totale dei microrganismi mesofili aerobici (TMAM) e delle Enterobacteriaceae. La rilevazione del pH e dell’acidità titolabile ha fornito risultati in linea a quelli microbiologici. I campioni nel biofilm hanno spesso subito una variazione significativa (p<0,05) dell’umidità rispetto al controllo, a causa dell’elevata permeabilità al vapore acqueo. L’analisi dei TBARS non ha spesso riportato differenze significative rispetto al controllo (p>0,05), e quando presenti, è perché il campione era più ossidato del controllo (p<0,05). Invece, è stato ottenuto un miglioramento significativo (p<0,05) dello Hue angle tra i film e il controllo. I risultati ottenuti forniscono le basi per studi aggiuntivi.

The better understanding of mechanisms at the basis of host-pathogen interaction can represent a valid tool to increase productivity and contain economic losses in animal production through the maintenance of intestinal homeostasis. With this project, three preliminary in vitro studies were conducted with the aim of investigating how bioactive compounds could influence mechanisms of host-pathogen interaction in poultry and swine. Different panels of nature identical compounds, medium chain fatty acids, and plant extracts were employed against strains of Salmonella Typhimurium, Brachyspira hyodysenteriae, and Salmonella Enteritidis, respectively. When bacterial field strains were tested, the comparison between natural compounds and antibiotics was examined, with the aim of evaluating the role of the substances in the antibiotic-resistance context. Results demonstrate that bioactive compounds have positive effects on the host, the pathogen, or both in different experimental conditions. Additionally, when compared to antibiotics, bioactive compounds have proven to be valid alternatives to address the phenomenon of antibiotic resistance.

Within this master thesis, various aspects related to the issue of sustainability in the food sector were addressed, focusing on the greenhouse gas emissions derived from livestock production. The increment in population number and wealth is directly related to the growing demand for meat products, which is, in turn, related to an increase in greenhouse gas emissions. Consumers are becoming more and more aware of these environmental issues and, therefore, sustainability factors are becoming even more relevant also from the environmental point of view. A very useful tool in this field is Response-Inducing Sustainability Evaluation (RISE), a software that allows you to determine the sustainability of a farm under many aspects, like energy consumption, livestock management and soil use. The RISE software processes the information obtained through a questionnaire submitted by the farmer, in which 10 different areas of sustainability in the farm are covered. For each theme, the results are expressed clearly with a score that goes from 0 to 100. The experimentation discussed in this work included two different projects, one regarding a dairy farm and the other regarding a poultry farm. The first one was conducted on a dairy farm in Germany and the results allowed to highlight the weakest areas of the farm on which recommendations were given for ecological improvement. The second project was conducted on a chicken broiler farm in Italy, on an experimental basis since it was the first time that the software was applied to poultry. The results pointed out the aspects that can be improved in the RISE software in order to make it more suitable for future poultry studies.

Il concept alla base di Onecook è stato ispirato dalle esigenze dei giovani adulti che vivono da soli in piccole case. Infatti, molte persone in questa fascia di età hanno appena iniziato a vivere da sole e spesso hanno uno spazio limitato in cucina. Il mio obiettivo era creare un set che includesse tutti gli elementi essenziali, per cucinare in modo più efficiente risparmiando anche energia, con un design compatto e salvaspazio. Per raggiungere questo obiettivo, ho progettato una piastra ad induzione con un’area di riscaldamento flessibile, affinché potesse permettere l’utilizzo simultaneo di pentole e padelle di diverse dimensioni. Grazie a questa flessibilità, la piastra si scalda solamente nell’area utilizzata, aumentando, quindi, l’efficienza energetica e contribuendo a ridurre il consumo energetico complessivo. Inoltre, è stata progettata una cappa di aspirazione filtrante portatile, che può essere facilmente spostata e riposta quando non viene utilizzata. Onecook include anche una varietà di altri strumenti essenziali come un coltello, un tagliere, un mestolo e una spatola. Una delle caratteristiche uniche di questo set è che le padelle e le pentole sono progettate per essere impilabili e modulari, massimizzando lo spazio di archiviazione e facilitando la ricerca della pentola o padella giusta per una ricetta particolare. Oltre alla funzionalità, ho considerato anche l’aspetto estetico del set e scelto colori e materiali moderni e senza tempo. Il prodotto finale è un set funzionale, poco ingombrante e visivamente gradevole. Nel complesso, Onecook è progettato per soddisfare le esigenze dei giovani adulti che cercano un modo compatto ed efficiente per cucinare per loro stessi. Credo che questo set li aiuterà a creare pasti deliziosi e salutari nelle loro piccole case.

Avian pathogenic Escherichia coli (APEC) strains belong to a category that is associated with colibacillosis, a serious illness in the poultry industry worldwide. Additionally, some APEC groups have recently been described as potential zoonotic agents. In this work, we compared APEC strains with extraintestinal pathogenic E. coli (ExPEC) strains isolated from clinical cases of humans with extra-intestinal diseases such as urinary tract infections (UTI) and bacteremia. PCR results showed that genes usually found in the ColV plasmid (tsh, iucA, iss, and hlyF) were associated with APEC strains while fyuA, irp-2, fepC sitDchrom, fimH, crl, csgA, afa, iha, sat, hlyA, hra, cnf1, kpsMTII, clpVSakai and malX were associated with human ExPEC. Both categories shared nine serogroups (O2, O6, O7, O8, O11, O19, O25, O73 and O153) and seven sequence types (ST10, ST88, ST93, ST117, ST131, ST155, ST359, ST648 and ST1011). Interestingly, ST95, which is associated with the zoonotic potential of APEC and is spread in avian E. coli of North America and Europe, was not detected among 76 APEC strains. When the strains were clustered based on the presence of virulence genes, most ExPEC strains (71.7%) were contained in one cluster while most APEC strains (63.2%) segregated to another. In general, the strains showed distinct genetic and fingerprint patterns, but avian and human strains of ST359, or ST23 clonal complex (CC), presented more than 70% of similarity by PFGE. The results demonstrate that some zoonotic-related STs (ST117, ST131, ST10CC, ST23CC) are present in Brazil. Also, the presence of moderate fingerprint similarities between ST359 E. coli of avian and human origin indicates that strains of this ST are candidates for having zoonotic potential.

Background and aims: Recent findings have highlighted enhanced fish consumption as a potential measure to increase intake of healthy fatty acids, particularly omega-3. The generalizability of this recommendation, however, may fall short by differences in fish species and cooking techniques. Hence, we investigated how these 2 variables affect the lipid content in fish flesh. Methods and Results: Nine species of freshwater, deep sea or shore fish were grilled, steamed or fried with or without the addition of soybean oil, olive oil or butter. The lipid composition was analysed and a significant difference was observed in cholesterol, saturated fatty acids, polyunsaturated fatty acids, omega-3 fatty acids, and omega-6 fatty acids contents between species (p<0.05). The use of soybean or olive oil was associated with a significant change in flesh concentration of polyunsaturated, omega-3 and omega-6 fatty acids (p<0.05). Conclusion: This study calls attention to the specific lipid content that must be expected from different fish species and cooking techniques.

Bologna-type sausages were produced with 50% of their pork back-fat content replaced with gels elaborated with different ratios of pork skin, water, and amorphous cellulose (1:1:0, 1:1:0.1, 1:1:0.2, 1:1:0.3, and 1:1:0.4). The impact of such replacement on the physico-chemical characteristics and the consumer sensory profiling was evaluated. The modified treatments had 42% less fat, 18% more protein, and 8% more moisture than the control group. Treatments with amorphous cellulose had a lower cooking loss and higher emulsion stability. High amorphous cellulose content (1:1:0.3 and 1:1:0.4) increased hardness, gumminess, and chewiness. The gel formulated with the ratio of 1:1:0.2 (pork skin: water: amorphous cellulose gel) provided a sensory sensation similar to that provided by fat and allowed products of good acceptance to be obtained. Therefore, a combination of pork skin and amorphous cellulose is useful in improving technological quality and producing healthier and sensory acceptable bologna-type sausages.

Avian Pathogenic Escherichia coli (APEC) strains are extra-intestinal E. coli that infect poultry and cause diseases. Nitrite is a central branch-point in bacterial nitrogen metabolism and is used as a cytotoxin by macrophages. Unlike nitric oxide (NO), nitrite cannot diffuse across bacterial membrane cells. The NirC protein acts as a specific channel to facilitate the transport of nitrite into Salmonella and E. coli cells for nitrogen metabolism and cytoplasmic detoxification. NirC is also required for the pathogenicity of Salmonella by downregulating the production of NO by the host macrophages. Based on an in vitro microarray that revealed the overexpression of the nirC gene in APEC strain SCI-07, we constructed a nirC-deficient SCI-07 strain (ΔnirC) and evaluated its virulence potential using in vivo and in vitro assays. The final cumulative mortalities caused by mutant and wild-type (WT) were similar; while the ΔnirC caused a gradual increase in the mortality rate during the seven days recorded, the WT caused mortality up to 24h post-infection (hpi). Counts of the ΔnirC cells in the spleen, lung and liver were higher than those of the WT after 48 hpi but similar at 24 hpi. Although similar number of ΔnirC and WT cells was observed in macrophages at 3 hpi, there was higher number of ΔnirC cells at 16 hpi. The cell adhesion ability of the ΔnirC strain was about half the WT level in the presence and absence of alpha-D-mannopyranoside. These results indicate that the nirC gene influences the pathogenicity of SCI-07 strain.

The biodegradability of animal wastes production was evaluated through a simplified methodology that allowed the verification of the applicability of anaerobic processes. The experiments were performed in bath reactors, with granular sludge of three origins: UASB reactor treating dairy effluent, UASB reactor treating swine effluent and UASB reactor treating effluent of slaughterhouse of poultry. The experiments (1) - dairy effluent and poultry slaughterhouse non-adapted sludge; (2) -swine effluent and poultry slaughterhouse non-adapted sludge; (3) - dairy effluent and poultry slaughterhouse adapted sludge; (4) - swine effluent and poultry slaughterhouse adapted sludge; (5) - dairy effluent and dairy sludge, and (6) - swine effluent and swine sludge were performed in Incubator Shaker, at a temperature of 35 °C, under agitation at a 150 rpm, for 5 minutes, every 1 hour. A substrat:biomass relationship of 0.5 was used. Kinetic models of Monod, Zero Order, First and Second Order were tested and it was verified that the First Order model provided the best adjustment. The apparent First Order kinetic parameter (k1) was estimated for the experiments 1; 2; 3; 4; 5, and 6, as 2.51 x 10-2; 2.49 x 10-2; 1.90 x 10-2; 3.09 x 10-2; 2.54 x 10-2; 4.09 x 10-2 h-1, respectively.

Knowing the importance that the poultry industry represents for the Brazilian economy, this work, searched to understand and to identify new welfare pointers inherent to the animal that contributed for the increase of the productive effectiveness, studying different behavior reactions in broiler breeders, in climatic chamber. The experiment was delineated as a Latin Square 3x3x3, where the variable: temperature of air, birds ration and birds age had been controlled. The birds of different ages had been lodged in distinct boxes. Observations of the behavior of the birds in two schedules of the day had been made, being one in the morning and the other one in the afternoon, during a period of 15 minutes each through video cameras, installed in the ceiling of the climatic chamber, having no interference of human being in the register of the data. It was verified the influence of the controlled variables in diverse observed behaviors where it was concluded that the presence of food resulted in bigger occurrences of aggressiveness reactions.
