In the plant-beneficial, root-colonizing strain Pseudomonas fluorescens CHA0, the Gac/Rsm signal transduction pathway positively regulates the synthesis of biocontrol factors (mostly antifungal secondary metabolites) and contributes to oxidative stress response via the stress sigma factor RpoS. The backbone of this pathway consists of the GacS/GacA two-component system, which activates the expression of three small regulatory RNAs (RsmX, RsmY, RsmZ) and thereby counters translational repression exerted by the RsmA and RsmE proteins on target mRNAs encoding biocontrol factors. We found that the expression of typical biocontrol factors, that is, antibiotic compounds and hydrogen cyanide (involving the phlA and hcnA genes), was significantly lower at 35 degrees C than at 30 degrees C. The expression of the rpoS gene was affected in parallel. This temperature control depended on RetS, a sensor kinase acting as an antagonist of the GacS/GacA system. An additional sensor kinase, LadS, which activated the GacS/GacA system, apparently did not contribute to thermosensitivity. Mutations in gacS or gacA were epistatic to (that is, they overruled) mutations in retS or ladS for expression of the small RNAs RsmXYZ. These data are consistent with a model according to which RetS-GacS and LadS-GacS interactions shape the output of the Gac/Rsm pathway and the environmental temperature influences the RetS-GacS interaction in P. fluorescens CHA0.

Human immunodeficiency virus type 1 uses ribosomal frameshifting for translation of the Gag-Pol polyprotein. Frameshift activities are thought to be tightly regulated. Analysis of gag p1 sequences from 270 plasma virions identified in 64% of the samples the occurrence of polymorphism that could lead to changes in thermodynamic stability of the stem-loop. Expression in Saccharomyces cerevisiae of p1-beta-galactosidase fusion proteins from 10 representative natural stem-loop variants and three laboratory mutant constructs (predicted the thermodynamic stability [Delta G degrees] ranging from -23.0 to -4.3 kcal/mol) identified a reduction in frameshift activity of 13 to 67% compared with constructs with the wild-type stem-loop (Delta G degrees, -23.5 kcal/mol). Viruses carrying stem-loops associated with greater than 60% reductions in frameshift activity presented profound defects in viral replication. In contrast, viruses with stem-loop structures associated with 16 to 42% reductions in frameshift efficiency displayed no significant viral replication deficit.

As soluções analíticas de distribuição de água para ponto fonte e regime de fluxo não- permanente são dependentes de parâmetros de solos considerados constantes em suas deduções. Erros na determinação desses parâmetros implicam insucesso dessas soluções. Este trabalho teve por objetivo avaliar o comportamento dos parâmetros alfada equação de Gardner k(h) = Ks ealfah e o parâmetro de linearização k = dK(teta)/dteta em diferentes posições do bulbo molhado, considerando os processos de infiltração isolado e seguido de redistribuição de água, à semelhança de um ciclo de irrigação. Dados de potencial matricial foram coletados em diversas posições do bulbo molhado em duas situações:(i) início da irrigação até atingir regime permanente em todo o bulbo molhado (infiltração); e (ii) durante dois ciclos de irrigação envolvendo infiltração e redistribuição de água. Os resultados mostraram que os parâmetros alfa e k variaram nas posições do bulbo molhado em relação ao gotejador, de acordo com o regime de umidade a que tais posições estiveram sujeitas. A obtenção desses parâmetros pelo método inverso requer testes que considerem as fases de infiltração e redistribuição em pelo menos dois ciclos de irrigação.

This study aimed to assess application of ultrasound (US) combined with microbubbles (MB) to transfect the ciliary muscle of rat eyes. Reporter DNA plasmids encoding for Gaussia luciferase, β-galactosidase or the green fluorescent protein (GFP), alone or mixed with 50% Artison MB, were injected into the ciliary muscle, with or without US exposure (US set at 1 MHz, 2 W/cm(2), 50% duty cycle for 2 min). Luciferase activity was measured in ocular fluids at 7 and 30 days after sonoporation. At 1 week, the US+MB treatment showed a significant increase in luminescence compared with control eyes, injected with plasmid only, with or without MB (×2.6), and, reporter proteins were localized in the ciliary muscle by histochemical analysis. At 1 month, a significant decrease in luciferase activity was observed in all groups. A rise in lens and ciliary muscle temperature was measured during the procedure but did not result in any observable or microscopic damages at 1 and 8 days. The feasibility to transfer gene into the ciliary muscle by US and MB suggests that sonoporation may allow intraocular production of proteins for the treatment of inflammatory, angiogenic and/or degenerative retinal diseases.

O objetivo deste trabalho foi estimar o tamanho amostral mínimo (n) para comparar tratamentos em experimentos de consumo e de digestibilidade com bovinos, envolvendo múltiplos caracteres. Foram utilizados dados de digestibilidade de 72 novilhas com média de 18 meses de idade e 250 kg de peso. O experimento foi realizado na Embrapa-Centro de Pesquisa de Pecuária do Sudeste, São Carlos, SP, de 1988 a 1989, em delineamento inteiramente casualizado, com nove tratamentos organizados em esquema fatorial 3 x 3 (três grupos genéticos: Canchim, ½ Canchim + ½ Nelore e Nelore, e três níveis de proteína bruta: 6, 10 e 13%, com oito repetições cada, sendo a unidade experimental a novilha). Foram analisados consumo de ração (g/kg0,75) por quilograma de peso metabólico, energia digestível, nitrogênio retido (NR), NR (mg/kg0,75) e digestibilidades da matéria seca, proteína bruta, fibra em detergente neutro e fibra em detergente ácido. O valor mínimo de n, que permite detectar diferenças significativas (delta) entre vetores de médias de tratamentos, foi obtido por meio de um programa SAS (Statistical Analysis System), considerando modelo de distribuição normal t-variada, média zero e matriz de covariância sigma, estatística T² de Hotelling, distribuição F com parâmetro de não centralidade (ACE=Symbol>d²D), erros do tipo I (alfa), poder do teste (1 beta) e delta. O valor de n variou de 6 a 47, sendo mais influenciado por alteração nos valores de delta, do que nos valores de alfa e poder do teste.

OBJECTIVE: Fabry disease is an X-linked disorder resulting from alpha-galactosidase A deficiency. The cardiovascular findings include left ventricular hypertrophy (LVH) and increased intima-media thickness of the common carotid artery (CCA IMT). The current study examined the possible correlation between these parameters. To corroborate these clinical findings in vitro, plasma from Fabry patients was tested for possible proliferative effect on rat vascular smooth muscle cells (vascular smooth muscle cell [VSMC]) and mouse neonatal cardiomyocytes. METHODS AND RESULTS: Thirty male and 38 female patients were enrolled. LVH was found in 60% of men and 39% of women. Increased CCA IMT was equally present in males and females. There was a strong positive correlation between LV mass and CCA IMT (r2=0.27; P<0.0001). VSMC and neonatal cardiomyocyte proliferative response in vitro correlated with CCA IMT (r2=0.39; P<0.0004) and LV mass index (r2=0.19; P=0.028), respectively. CONCLUSIONS: LVH and CCA IMT occur concomitantly in Fabry suggesting common pathogenesis. The underlying cause may be a circulating growth-promoting factor whose presence has been confirmed in vitro.

O objetivo do trabalho foi avaliar um método para estimar o número de indivíduos (n) a ser utilizado em experimentos que envolvam análises multivariadas de medidas repetidas no tempo, avaliadas sobre a mesma unidade experimental. O método foi testado com dados de produção de leite com 10 controles mensais (t = 1, 2, ... , 10) ou condições de avaliação de vacas da raça Holandesa. As estimativas de n foram obtidas por meio de um programa desenvolvido no Statistical Analysis System (SAS), considerando distribuição normal t variada, vetor de média zero e matriz de covariância sigma, estatística T² de Hotelling e distribuição F com parâmetro de não-centralidade delta²delta. A ligação dos dados observados com o método é feita por meio da matriz de variância-covariância. Para t > 2 condições de avaliação, o método estima o valor de n que permite detectar diferença mínima significativa (delta) entre médias de condições de avaliação, considerando diferentes níveis de erros do tipo I (alfa), poder do teste F (1-beta) e delta. Para as 10 condições de avaliação consideradas, as estimativas de n variaram de 11 a 89, sendo mais influenciadas por variações na delta, seguidas de alfa e beta.

O objetivo deste trabalho foi estudar o processo germinativo dos grãos na espiga, o rendimento de grãos, a adaptabilidade e a estabilidade em genótipos de trigo (Triticum aestivum L.) sensíveis às variações ambientais. Os experimentos foram instalados no Núcleo Experimental do Instituto Agronômico, em Campinas, SP, no período de 1996 a 1998. A germinação na espiga foi avaliada pelo método do "Falling Number", que consiste em determinar o nível de atividade da alfa-amilase nos grãos. A segunda e a terceira colheitas foram realizadas ao sete e quatorze dias após a primeira colheita. O atraso da colheita determinou redução no rendimento, notadamente na terceira colheita; os teores de alfa-amilase foram maiores também na terceira colheita, em virtude da degradação dos grãos expostos ao tempo. O genótipo IAC 289 apresentou adaptabilidade a ambientes favoráveis, mas apresentou suscetibilidade à ocorrência de germinação dos grãos na espiga. Os genótipos IAC 24, Mochis, IAC 370 e IAC 351 foram os mais resistentes à germinação dos grãos na espiga, apresentando baixa atividade de alfa-amilase.

BACKGROUND: We have developed a nonviral gene therapy method based on the electrotransfer of plasmid in the ciliary muscle. These easily accessible smooth muscle cells could be turned into a biofactory for any therapeutic proteins to be secreted in a sustained manner in the ocular media. METHODS: Electrical conditions, design of electrodes, plasmid formulation, method and number of injections were optimized in vivo in the rat by localizing β-galactosidase expression and quantifying reporter (luciferase) and therapeutic (anti-tumor necrosis factor) proteins secretion in the ocular media. Anatomical measurements were performed via human magnetic resonance imaging to design a human eye-sized prototype that was tested in the rabbit. RESULTS: In the rat, transscleral injection of 30 µg of plasmid diluted in half saline (77 mM NaCl) followed by application of eight square-wave electrical pulses (15 V, 10 ms, 5.3 Hz) using two platinum/iridium electrodes, an internal wire and an external sheet, delivered plasmid efficiently to the ciliary muscle fibers. Gene transfer resulted in a long-lasting (at least 5 months) and plasmid dose-/injection number- dependent secretion of different molecular weight proteins mainly in the vitreous, without any systemic exposure. Because ciliary muscle anatomical measurements remained constant among ages in adult humans, an integrated device comprising needle-electrodes was designed and manufactured. Its usefulness was validated in the rabbit. CONCLUSIONS: Plasmid electrotransfer to the ciliary muscle with a suitable medical device represents a promising local and sustained protein delivery system for treating posterior segment diseases, avoiding repeated intraocular injections.

The serine protease thrombin plays a role in signalling ischemic neuronal death in the brain. Paradoxically, endogenous neuroprotective mechanisms can be triggered by preconditioning with thrombin (thrombin preconditioning, TPC), leading to tolerance to cerebral ischemia. Here we studied the role of thrombin's endogenous potent inhibitor, protease nexin-1 (PN-1), in ischemia and in tolerance to cerebral ischemia induced by TPC. Cerebral ischemia was modelled in vitro in organotypic hippocampal slice cultures from rats or genetically engineered mice lacking PN-1 or with the reporter gene lacZ knocked into the PN-1 locus PN-1HAPN-1-lacZ/HAPN-1-lacZ (PN-1 KI) exposed to oxygen and glucose deprivation (OGD). We observed increased thrombin enzyme activity in culture homogenates 24 h after OGD. Lack of PN-1 increased neuronal death in the CA1, suggesting that endogenous PN-1 inhibits thrombin-induced neuronal damage after ischemia. OGD enhanced β-galactosidase activity, reflecting PN-1 expression, at one and 24 h, most strikingly in the stratum radiatum, a glial cell layer adjacent to the CA1 layer of ischemia sensitive neurons. TPC, 24 h before OGD, additionally increased PN-1 expression 1 h after OGD, compared to OGD alone. TPC failed to induce tolerance in cultures from PN-1(-/-) mice confirming PN-1 as an important TPC target. PN-1 upregulation after TPC was blocked by the c-Jun N-terminal kinase (JNK) inhibitor, L-JNKI1, known to block TPC. This work suggests that PN-1 is an endogenous neuroprotectant in cerebral ischemia and a potential target for neuroprotection.

BACKGROUND: In Fabry nephropathy, alpha-galactosidase deficiency leads to accumulation of glycosphingolipids in all kidney cell types, proteinuria and progressive loss of kidney function. METHODS: An international working group of nephrologists from 11 Fabry centres identified adult Fabry patients, and pathologists scored histologic changes on renal biopsies. A standardized scoring system was developed with a modified Delphi technique assessing 59 Fabry nephropathy cases. Each case was scored independently of clinical information by at least three pathologists with an average final score reported. RESULTS: We assessed 35 males (mean age 36.4 years) and 24 females (43.9 years) who mostly had clinically mild Fabry nephropathy. The average serum creatinine was 1.3 mg/dl (114.9 micromol/l); estimated glomerular filtration rate was 81.7 ml/min/1.73 m(2) and urine protein to creatinine ratio was 1.08 g/g (122.0 mg/mmol). Males had greater podocyte vacuolization on light microscopy (mean score) and glycosphingolipid inclusions on semi-thin sections than females. Males also had significantly more proximal tubule, peritubular capillary and vascular intimal inclusions. Arteriolar hyalinosis was similar, but females had significantly more arterial hyalinosis. Chronic kidney disease stage correlated with arterial and glomerular sclerosis scores. Significant changes, including segmental and global sclerosis, and interstitial fibrosis were seen even in patients with stage 1-2 chronic kidney disease with minimal proteinuria. CONCLUSIONS: The development of a standardized scoring system of both disease-specific lesions, i.e. lipid deposition related, and general lesions of progression, i.e. fibrosis and sclerosis, showed a spectrum of histologic appearances even in early clinical stage of Fabry nephropathy. These findings support the role of kidney biopsy in the baseline evaluation of Fabry nephropathy, even with mild clinical disease. The scoring system will be useful for longitudinal assessment of prognosis and responses to therapy for Fabry nephropathy.
