We hypothesized that a function of sleep is to replenish brain glycogen stores that become depleted while awake. We have previously tested this hypothesis in three inbred strains of mice by measuring brain glycogen after a 6h sleep deprivation (SD). Unexpectedly, glycogen content in the cerebral cortex did not decrease with SD in two of the strains and was even found to increase in mice of the C57BL/6J (B6) strain. Manipulations that initially induce glycogenolysis can also induce subsequent glycogen synthesis thereby elevating glycogen content beyond baseline. It is thus possible that in B6 mice, cortical glycogen content decreased early during SD and became elevated later in SD. In the present study, we therefore measured changes in brain glycogen over the course of a 6 h SD and during recovery sleep in B6 mice. We found no evidence of a decrease at any time during the SD, instead, cortical glycogen content monotonically increased with time-spent-awake and, when sleep was allowed, started to revert to control levels. Such a time-course is opposite to the one predicted by our initial hypothesis. These results demonstrate that glycogen synthesis can be achieved during prolonged wakefulness to the extent that it outweighs glycogenolysis. Maintaining this energy store seems thus not to be functionally related to sleep in this strain.

Soluble MHC-peptide (pMHC) complexes induce intracellular calcium mobilization, diverse phosphorylation events, and death of CD8+ CTL, given that they are at least dimeric and co-engage CD8. By testing dimeric, tetrameric, and octameric pMHC complexes containing spacers of different lengths, we show that their ability to activate CTL decreases as the distance between their subunit MHC complexes increases. Remarkably, pMHC complexes containing long rigid polyproline spacers (> or =80 A) inhibit target cell killing by cloned S14 CTL in a dose- and valence-dependent manner. Long octameric pMHC complexes abolished target cell lysis, even very strong lysis, at nanomolar concentrations. By contrast, an altered peptide ligand antagonist was only weakly inhibitory and only at high concentrations. Long D(b)-gp33 complexes strongly and specifically inhibited the D(b)-restricted lymphocytic choriomeningitis virus CTL response in vitro and in vivo. We show that complications related to transfer of peptide from soluble to cell-associated MHC molecules can be circumvented by using covalent pMHC complexes. Long pMHC complexes efficiently inhibited CTL target cell conjugate formation by interfering with TCR-mediated activation of LFA-1. Such reagents provide a new and powerful means to inhibit Ag-specific CTL responses and hence should be useful to blunt autoimmune disorders such as diabetes type I.

Motherhood and reproduction have been at the core of the feminist discourse about women's rights ever since its onset. For the first and second feminist movements, the right to abortion and the public recognition of motherhood have been main issues in the discourse on reproduction. Since the last two dec- ades of the 20th century, the potentials of assisted reproductive technologies (ART) have opened up new venues of feminist discourse.In this paper we sketch the main feminist lines of argumentation regarding motherhood and reproduction since the 1970s, and we identify specific shifts in their recurrent issues. We argue that an essential contribution of feminism to the understanding of motherhood as a structuring category has been its insis- tence on the distinction between biological and social motherhood. Feminist discourse shows how ART has further decomposed biological motherhood and has altered the meaning of motherhood and reproduction. Feminist analysis maintains that despite the rhetoric of choice surrounding ART, these technolo- gies have not increased women's reproductive freedom. The decomposition of biological motherhood, the medical, legal, and commercial development of re- production, and the change in the social perception of motherhood have rather established new forms of control over female reproduction.

PURPOSE OF REVIEW: One of the seven key scientific priorities identified in the road map on HIV cure research is to 'determine the host mechanisms that control HIV replication in the absence of therapy'. This review summarizes the recent work in genomics and in epigenetic control of viral replication that is relevant for this mission. RECENT FINDINGS: New technologies allow the joint analysis of host and viral transcripts. They identify the patterns of antisense transcription of the viral genome and its role in gene regulation. High-throughput studies facilitate the assessment of integration at the genome scale. Integration site, orientation and host genomic context modulate the transcription and should also be assessed at the level of single cells. The various models of latency in primary cells can be followed using dynamic study designs to acquire transcriptome and proteome data of the process of entry, maintenance and reactivation of latency. Dynamic studies can be applied to the study of transcription factors and chromatin modifications in latency and upon reactivation. SUMMARY: The convergence of primary cell models of latency, new high-throughput quantitative technologies applied to the study of time series and the identification of compounds that reactivate viral transcription bring unprecedented precision to the study of viral latency.

The trabecular bone score (TBS) is a new parameter that is determined from gray-level analysis of dual-energy X-ray absorptiometry (DXA) images. It relies on the mean thickness and volume fraction of trabecular bone microarchitecture. This was a preliminary case-control study to evaluate the potential diagnostic value of TBS as a complement to bone mineral density (BMD), by comparing postmenopausal women with and without fractures. The sample consisted of 45 women with osteoporotic fractures (5 hip fractures, 20 vertebral fractures, and 20 other types of fracture) and 155 women without a fracture. Stratification was performed, taking into account each type of fracture (except hip), and women with and without fractures were matched for age and spine BMD. BMD and TBS were measured at the total spine. TBS measured at the total spine revealed a significant difference between the fracture and age- and spine BMD-matched nonfracture group, when considering all types of fractures and vertebral fractures. In these cases, the diagnostic value of the combination of BMD and TBS likely will be higher compared with that of BMD alone. TBS, as evaluated from standard DXA scans directly, potentially complements BMD in the detection of osteoporotic fractures. Prospective studies are necessary to fully evaluate the potential role of TBS as a complementary risk factor for fracture.

O fracionamento físico densimétrico é uma técnica que permite estudar a proteção física da matéria orgânica (MO) no solo, sendo utilizadas, de forma indistinta, soluções densas de iodeto de sódio (NaI) e de politungstato de sódio (PTS). O objetivo deste estudo foi avaliar (a) o efeito de soluções de NaI (1,8 g cm-3) e de PTS (1,8, 2,0 e 2,2 g cm-3) no rendimento de carbono (C) nas frações leve-livre (FLL) e leve-oclusa (FLO) da MO na camada de 0-5 cm de um Argissolo Vermelho [PVd, Eldorado do Sul (RS)] e de um Latossolo Vermelho [LVdf, Dourados (MS)] e (b) a magnitude da proteção física da MO em agregados no acúmulo de C orgânico nesses solos submetidos, respectivamente, ao sistema plantio direto por 18 anos (PD) e pastagem (Brachiaria decumbens) por nove anos, em comparação à sua condição em preparo convencional (PC). Em ambos os locais, blocos indeformados de solo foram coletados em experimentos de longa duração; os agregados foram separados nos planos de fraqueza e passados em peneira de 9,51 mm; esse material foi submetido ao fracionamento densimétrico da MO do solo. O uso da solução de PTS 1,8 g cm-3 aumentou de 176 a 727 % o rendimento de C na FLO em comparação à solução de NaI na mesma densidade, mas não teve efeito no rendimento de C da FLL da MO do solo. O aumento da densidade da solução de PTS de 1,8 g cm-3 para 2,0 g cm-3 incrementou de 30 a 99 % o rendimento de C da FLO; contudo, o aumento da densidade para 2,2 g cm-3 resultou em aumento pouco expressivo (< 18 %) no rendimento de C na FLO nos diferentes solos e sistemas de manejo. O uso de NaI no fracionamento densimétrico leva à conclusão errônea de que a proteção física não é um mecanismo importante de estabilização da MO no solo. Por outro lado, a utilização da solução de PTS 2,0 g cm-3, a qual é recomendada para o fracionamento densimétrico devido à sua alta eficiência na recuperação do C da FLO, permitiu constatar que a proteção física da MO em agregados de solo foi responsável por 54 e 23 % do acúmulo de C orgânico no PVd e LVdf, respectivamente, sob plantio direto e pastagem.

The case of a man exposed during 25 years to vibration while maneuvering a heavy earth moving tractor is reported. The first clinical manifestation of hand-arm vibration syndrome was a bilateral Raynaud's phenomenon followed five years later by digital necrosis. The arteriography revealed a proximal and bilateral ulnar artery occlusion. Bilateral median nerve conduction abnormalities were also present. Vibration exposure level was much higher than the threshold level proposed by the European Commission. Laboratory examinations for vasculitis and other vascular diseases were all negative. The purpose of this report is to show that vibration can be responsible for proximal occlusion of a medium sized artery and severe neurovascular abnormalities which must be distinguished from the usual vasospastic Raynaud's phenomenon and the classical hypothenar hammer syndrome. An early and correct diagnosis is crucial because continued repetitive trauma can result in irreversible ischemic injury and loss of digits.

During the period from January 1990 to December 1993, 558 new patients (250 female and 308 male, mean age 44 years; range 5-92) were seen at the Uveitis Clinic of the Hopital Jules Gonin. These 558 patients (740 eyes) were subdivided into anterior uveitis (343 patients-61%), intermediate uveitis (57 patients-10%), posterior uveitis (118 patients-21 %) and panuveitis (40 patients-7%). The incidence of uveitis for the referral area considered was calculated to be 17.5 per 100,000 inhabitants per year. A specific diagnosis was found in 386 cases (69%). The most frequently diagnosed entities were HLA-B27-associated acute anterior uveitis (89 cases-15.9%), uveitis associated with acute herpes zoster ophthalmicus (54 cases-9.7%), toxoplasmosis (53 cases-9.5%), sarcoidosis (33 cases-5.9%), typical pars planitis (31 cases-5.6%), Fuchs' heterochromic cyclitis (30 cases-5.4%), herpetic anterior uveitis (23 cases-4.1 %) and acute retinal necrosis (13 cases-2.3%). Incidence and distribution of most disease entities correspond to those of other European and American series.
