The efficacy of 1-methylcyclopropene (1-MCP) gas to prevent the adverse effects of ethylene is limited by its short-term residual activity in some plants. Development of a simple 1-MCP sustained release device that prolongs 1-MCP exposure is reported herein. Sustained release devices comprised of polyvinylchloride tubes containing 0.1 g SmartFresh(TM) powder (a.i. 3.3% 1-MCP) and 1.25 ml deionised water were used to release 1-MCP into fibreboard cartons containing cut Geraldton waxflower (Chamelaucium uncinatum Schauer) cv. CWA Pink bunches during export shipment by air (107 h) from Australia to the UK. The devices protected flowers against abscission induced by subsequent test exposures to ethylene (1011,mul l(-1), 12 h, 20 degreesC) for 3-5 days after arrival. In contrast, pre-shipment treatments with either a single application of 790 nl l(-1) 1-MCP for 14 h at 2 degreesC or a 0.2 mM Ag+ (as silver thiosulphate; STS) pulse for 14 h at 2 degreesC protected flowers against exogenous ethylene for only 1-2 days of post-export life. However, pre-shipment 1-MCP fumigation was up to about three-fold more effective than either sustained 1-MCP release or pre-shipment STS treatments in reducing floral organ and leaf abscission from bunches during export. Thus, it is suggested that a combination of pre-shipment 1-MCP fumigation before export with sustained 1-MCP release during shipment should maximise efficacy against ethylene-induced waxflower flower abscission. Background: The purpose of the present study was to compare the effectiveness of three burns dressings (TransCyte, a bio-engineered skin substitute; Biobrane; and Silvazine cream (silver sulphadiazine and 0.2% chlorhexidine)), in treating children with partial-thickness burns. The primary objective was to determine the days until greater than or equal to90% re-epithelialization. The secondary objectives were to evaluate the number of wounds requiring autografting and the number of dressing changes/local wound care required. Methods: Study wounds were identified on each patient and the patients were randomized to receive TransCyte or Biobrane or Silvazine. Assessment of study wound closure began at 2 days after treatment and continued at least every other day thereafter until the wounds re-epithelialized or were autografted. A laser Doppler imaging system was used as an adjunct to assessing the depth of the burn. Results: Thirty-three patients with 58 wound sites enrolled in the study (TransCyte, n = 20, Biobrane, n = 17; Silvazine, n = 21). Mean time to re-epithelialization was 7.5 days for TransCyte, 9.5 days for Biobrane, and 11.2 days for Silvazine. The number of wounds requiring autografting were 5/21 (24%) for Silvazine, 3/17 (17%) for Biobrane, and 1/20 (5%) for TransCyte. Conclusions: When used in partial-thickness burns in children, TransCyte promotes fastest re-epithelialization and required less overall dressings then Biobrane or Silvazine. Patients who received Silvazine or Biobrane require more autografting than those treated with TransCyte.

The aim of the study presented was to implement a process model to simulate the dynamic behaviour of a pilot-scale process for anaerobic two-stage digestion of sewage sludge. The model implemented was initiated to support experimental investigations of the anaerobic two-stage digestion process. The model concept implemented in the simulation software package MATLAB(TM)/Simulink(R) is a derivative of the IWA Anaerobic Digestion Model No.1 (ADM1) that has been developed by the IWA task group for mathematical modelling of anaerobic processes. In the present study the original model concept has been adapted and applied to replicate a two-stage digestion process. Testing procedures, including balance checks and 'benchmarking' tests were carried out to verify the accuracy of the implementation. These combined measures ensured a faultless model implementation without numerical inconsistencies. Parameters for both, the thermophilic and the mesophilic process stage, have been estimated successfully using data from lab-scale experiments described in literature. Due to the high number of parameters in the structured model, it was necessary to develop a customised procedure that limited the range of parameters to be estimated. The accuracy of the optimised parameter sets has been assessed against experimental data from pilot-scale experiments. Under these conditions, the model predicted reasonably well the dynamic behaviour of a two-stage digestion process in pilot scale. (C) 2004 Elsevier Ltd. All rights reserved.

Protective roles for protease-activated receptor-2 (PAR2) in the airways including activation of epithelial chloride (Cl-) secretion are based on the use of presumably PAR(2)-selective peptide agonists. To determine whether PAR(2) peptide-activated Cl- secretion from mouse tracheal epithelium is dependent on PAR(2), changes in ion conductance across the epithelium [short-circuit current (I-SC)] to PAR(2) peptides were measured in Ussing chambers under voltage clamp. In addition, epithelium and endothelium-dependent relaxations to these peptides were measured in two established PAR(2) bioassays, isolated ring segments of mouse trachea and rat thoracic aorta, respectively. Apical application of the PAR(2) peptide SLIGRL caused increases in I-SC, which were inhibited by three structurally different neurokinin receptor-1 (NK1R) antagonists and inhibitors of Cl- channels but not by capsaicin, the calcitonin gene-related peptide (CGRP) receptor antagonist CGRP(8-37), or the nonselective cyclooxygenase inhibitor indomethacin. Only high concentrations of trypsin caused an increase in I-SC but did not affect the responses to SLIGRL. Relaxations to SLIGRL in the trachea and aorta were unaffected by the NK1R antagonist nolpitantium (SR 140333) but were abolished by trypsin desensitization. The rank order of potency for a range of peptides in the trachea I-SC assay was 2-furoyl-LIGRL > SLCGRL > SLIGRL > SLIGRT > LSIGRL compared with 2-furoyl-LIGRL > SLIGRL > SLIGRT > SLCGRL (LSIGRL inactive) in the aorta relaxation assay. In the mouse trachea, PAR(2) peptides activate both epithelial NK1R coupled to Cl- secretion and PAR(2) coupled to prostaglandin E-2-mediated smooth muscle relaxation. Such a potential lack of specificity of these commonly used peptides needs to be considered when roles for PAR(2) in airway function in health and disease are determined.

As dinâmicas proféticas no prólogo de Dêutero-Isaías que delineiam grande parte das linhas da mensagem apresentada em todo o bloco de Isaías 40-55 como uma introdução, é via desta pesquisa. Estas dinâmicas contêm conceitos teológicos renovados, pertinentes, e que fortaleceram o ânimo dos exilados. Para isto, o profeta usou como modelo antigas memórias fundamentais do povo hebreu. Em especial no prólogo, destacam-se o êxodo do Egito e a peregrinação no deserto, que têm seu propósito em chamar os exilados à resistência ao assédio e incorporação babilônica. Tendo em vista que, entre sucumbir aos encantos da religião e cultura babilônica, ou continuar a firmar a fé em Javé esperando a mudança da sorte, estava o drama vivido pelos exilados. O povo precisava de respostas pertinentes às suas dúvidas e é neste sentido que surge a profecia de Dêutero-Isaías combatendo a crise com esperança e movimentando à fé antes resignada. Neste sentido, a história da salvação serve como apoio e garantia para o futuro de libertação e restauração. Quando o presente não era capaz de suscitar esperança de mudanças no futuro, o profeta buscou no passado, memórias capazes de suscitarem aos exilados o desejo de servir a Javé ao invés de Marduque. Por trás desta necessidade surge o conceito de que a história do povo é o caminho pelo qual Javé escolheu para manifestar-se. No prólogo é possível perceber, como, e, o que, o profeta utilizou para levar os judaítas exilados a sentir-se verdadeiramente o povo de Deus. Foi importante fazer com que buscassem sua identidade em suas experiências de libertação do passado para que pudessem ter esperança.

Pertence ao senso comum atribuir-se à forças malévolas os dissabores que ocorrem ao ser humano, desde intentos pessoais que não dão certo até fenômenos naturais, tais como terremotos, enchentes, etc. A Deus, credita-se a paz entre os povos e as bem-aventuranças da vida. A imaginação do demoníaco é sempre muito forte, evocando imagens de seres horrendos que aparecem com asas de morcego, chifres, rabo e em cores flamejantes e tenebrosas que, apesar de assombrar o dia-a-dia das pessoas, acabam sendo nada mais do que tentativas de explicar e personalizar a realidade abstrata do mal. Este estudo tem por objetivo fazer uma análise literária do texto de Marcos 1.21-28, procurando investigar uma possível influência de tradições populares de povos e culturas antigas sobre a redação do Evangelho segundo Marcos em sua forma de descrever os relatos de exorcismo (Para isto, examinaremos as concepções do mal em várias correntes de tradição numa seqüência histórica relativa para vislumbrar como ao longo do tempo os povos têm imaginado o mal). A hostilidade, o dualismo, a luta cósmica parecem constituir elementos comuns percebidos nas fontes literárias da antiguidade. Todavia, o Evangelho de Marcos tende a fazer um uso particular da figura de demônios e espíritos imundos contrapondo-os a Jesus. Na Bíblia Hebraica, Satanás e os demônios fazem parte do cenário, mas são quase que ignorados e servem aos propósitos soberanos de Iahweh, enquanto que nos relatos de exorcismo tais figuras adquirem uma importância fundamental. Tudo o que ameaça a ordem estabelecida por aquele grupo social onde a narrativa se concentra acaba sendo identificado como manifestações de demônios e espíritos malignos.(AU)

Tetraspanins are thought to exert their biological function(s) by co-ordinating the lateral movement and trafficking of associated molecules into tetraspanin-enriched microdomains. A second four-TM (transmembrane) domain protein family, the Claudin superfamily, is the major structural component of cellular TJs (tight junctions). Although the Claudin family displays low sequence homology and appears to be evolutionarily distinct from the tetraspanins, CD81 and Claudin-1 are critical molecules defining HCV (hepatitis C virus) entry; we recently demonstrated that CD81-Claudin-1 complexes have an essential role in this process. To understand the molecular basis of CD81-Claudin-1 complex formation, we produced and purified milligram quantities of full-length CD81 and Claudin-1, alone and in complex, in both detergent and lipid contexts. Structural characterization of these purified proteins will allow us to define the mechanism(s) underlying virus-cell interactions and aid the design of therapeutic agents targeting early steps in the viral life cycle.

The first and third extracellular loops (ECL) of G protein-coupled receptors (GPCRs) have been implicated in ligand binding and receptor function. This study describes the results of an alanine/leucine scan of ECLs 1 and 3 and loop-associated transmembrane (TM) domains of the secretin-like GPCR calcitonin receptor-like receptor which associates with receptor activity modifying protein 1 to form the CGRP receptor. Leu195Ala, Val198Ala and Ala199Leu at the top of TM2 all reduced aCGRP-mediated cAMP production and internalization; Leu195Ala and Ala199Leu also reduced aCGRP binding. These residues form a hydrophobic cluster within an area defined as the "minor groove" of rhodopsin-like GPCRs. Within ECL1, Ala203Leu and Ala206Leu influenced the ability of aCGRP to stimulate adenylate cyclase. In TM3, His219Ala, Leu220Ala and Leu222Ala have influences on aCGRP binding and cAMP production; they are likely to indirectly influence the binding site for aCGRP as well as having an involvement in signal transduction. On the exofacial surfaces of TMs 6 and 7, a number of residues were identified that reduced cell surface receptor expression, most noticeably Leu351Ala and Glu357Ala in TM6. The residues may contribute to the RAMP1 binding interface. Ile360Ala impaired aCGRP-mediated cAMP production. Ile360 is predicted to be located close to ECL2 and may facilitate receptor activation. Identification of several crucial functional loci gives further insight into the activation mechanism of this complex receptor system and may aid rational drug design.

In this work we present extensive comparisons between numerical modelling and experimental measurements of the transmission performance of either CSRZ-ASK or CSRZ-DPSK modulation formats for 40-Gb/s WDM ULH systems on UltraWave (TM) fiber spans with all-Raman amplification. We numerically optimised the amplification and the signal format parameters for both CSRZ-DPSK and CSRZ-ASK formats. Numerical and experimental results show that, in a properly optimized transmission link, the DPSK format permits to double the transmission distance ( for a given BER level) with respect to the ASK format, while keeping a substantial OSNR margin ( on ASK modulation) after the propagation in the fiber line. Our comparison between numerical and experimental results permits to identify what is the most suitable BER estimator in assessing the transmission performance when using the DPSK format. A self-starting all-fiber passively mode-locked Tm-doped fiber laser based on nonlinear loop mirror (NOLM) is demonstrated. Stable soliton pulses centered at 2017.33 nm with 1.56 nm FWHM were produced at a repetition rate of 1.514 MHz with pulse duration of 2.8 ps and pulse energy of 83.8 pJ. As increased pump power, the oscillator can also operate at noise-like (NL) regime. Stable NL pulses with coherence spike width of 341 fs and pulse energy of up to 249.32 nJ was achieved at a center wavelength of 2017.24 nm with 21.33 nm FWHM. To the best of our knowledge, this is the first 2 μm region NOLM-based mode-locked fiber laser operating at two regimes with the highest single pulse energy for NL pulses. © 2014 Optical Society of America.
