Terminal sialic acid residues on surface-associated glycoconjugates mediate host cell interactions of many pathogens. Addition of sialic acid-rich fetuin enhanced, and the presence of the sialidiase inhibitor 2-deoxy-2,3-dehydro-N-acetylneuraminic acid reduced, the physical interaction of Neospora caninum tachyzoites and bradyzoites with Vero cell monolayers. Thus, Neospora extracts were subjected to fetuin-agarose affinity chromatography in order to isolate components potentially interacting with sialic acid residues. SDS-PAGE and silver staining of the fetuin binding fraction revealed the presence of a single protein band of approximately 65 kDa, subsequently named NcFBP (Neospora caninum fetuin-binding protein), which was localized at the apical tip of the tachyzoites and was continuously released into the surrounding medium in a temperature-independent manner. NcFBP readily interacted with Vero cells and bound to chondroitin sulfate A and C, and anti-NcFBP antibodies interfered in tachyzoite adhesion to host cell monolayers. In additon, analysis of the fetuin binding fraction by gelatin substrate zymography was performed, and demonstrated the presence of two bands of 96 and 140 kDa exhibiting metalloprotease-activity. The metalloprotease activity readily degraded glycosylated proteins such as fetuin and bovine immunoglobulin G heavy chain, whereas non-glycosylated proteins such as bovine serum albumin and immunoglobulin G light chain were not affected. These findings suggest that the fetuin-binding fraction of Neospora caninum tachyzoites contains components that could be potentially involved in host-parasite interactions.

By applying high pressure freezing and freeze-substitution, we observed large inclusions of homogeneous appearance in the front of locomoting Walker carcinosarcoma cells that have not been described earlier. Live cell imaging revealed that these inclusions were poor in lipids and nucleic acids but had a high lysine (and hence protein) content. Usually one such structure 2-5 mum in size was present at the front of motile Walker cells, predominantly in the immediate vicinity of newly forming blebs. By correlating the lysine-rich areas in fixed and embedded cells with electron microscopic pictures, inclusions could be assigned to confined, faintly stained cytoplasmic areas that lacked a surrounding membrane; they were therefore called pseudovacuoles. After high-pressure freezing and freeze substitution, pseudovacuoles appeared to be filled with 20 nm large electron-transparent patches surrounded by 12 and 15 nm large particles. The heat shock protein Hsp90 was identified by peptide sequencing as a major fluorescent band on SDS-PAGE of lysine-labelled Walker cell extracts. By immunofluorescence, Hsp90 was found to be enriched in pseudovacuoles. Colocalization of the lysine with a potassium-specific dye in living cells revealed that pseudovacuoles act as K+ stores in the vicinity of forming blebs. We propose that pseudovacuoles might support blebbing by locally regulating the intracellular hydrostatic pressure.

Historical stained glass in Calumet and Laurium revealed the complex structures of these industrial communities. Creating an Industrial Archaeology-focused approach, I examined stained glass as material culture. Sacred glass revealed ethnic and religious values of a congregation through the style, iconography, and quality of the glasswork. Residential glass showed how owners represented themselves within cultural settings by meeting social expectations. Commercial glass indicated community status of owners through discreet and artistic shows of wealth and taste. Corporate glass displayed prosperity and belonging through the superior quality and cost of the glasswork. Viewing stained glass as material culture opened new methods of looking at both stained glass and industrial communities. Findings from my research can teach the public about the importance of preserving and conserving stained glass, and that can lead to greater public appreciation for the material culture found within these industrial communities.

Charcoal has been known for a considerable length of time to have the property of recovering gold, silver, and copper from cyanide solutions of these metals. Quantita­tive data that may shed light on the mechanism of the re­moval of these metals is very limited except that char­coal in a form known as activated has the power to abstract gold and silver in considerable quantities from the above solutions.

Between the villages of Rocker and Silver Bow, in south­western Montana, are found an interesting group of placers. Gold occurs in Tertiary gravel beds that are interstratified with beds of rhyolitic volcanic ash. With the aid of a plane table and open-sight alidade, a small portion of the lake-bed area near Rocker was mapped; all distances were paced, but numerous checks assure a fairly accurate map.

A microscopic investigation of the Montana silver minerals was conducted. This study consisted mainly of identifying the silver and silver-bearing minerals and of determining their paragenetic rela­tionships. The increasing amount of research in which the reflecting microscope is employed is evidence of the great value of this method of approach in the solution of problems of paragenesis of the opaque minerals.

The purpose of this study is to detail and analyze the distribution, concentration, and loads of 5 organic compounds along Silver Bow Creek in Butte, Montana from the Municipal Wastewater treatment plant to the Warm Springs Ponds. The chemicals analyzed include Carbamazepine (pharmaceutical), Miconazole (fungicide) and three antibiotics – Sulfamethoxazole, Thiabendazole, and Ciprofloxacin. This project begins a 2 year study to analyze 6 additional compounds (11 compounds total), to develop an effective method to detail and analyze OWCs using Mass Spectrometer/Liquid chromatography system, and to aid in assessment of aquatic health and ongoing restoration work. In many deposits of silver ores the grade of the ore de­creases considerably a few hundred feet below the surface. It is believed that in many cases the better ores owe their richness in part to the process of sulphide enrichment. It is recognized, however, that many rich silver ores are hypogene deposits that have been affected very little, if any, by processes of enrichment.

Secondary Enrichment of Copper at the Madison Gold Skarn Deposit, Silver Star District, Montana. This paper focuses on the chemical reactions responsible for secondary enrichment of copper...we argue that most of the secondary Cu enrichment occurred during a late hydrothermal event that replaced the high temperature skarn mineral assemblage with hematitic jasperoid. Evidence favoring this "hypogene" Cu enrichment hypothesis is presented.
