Of all laser-based processes, laser machining has received little attention compared with others such as cutting, welding, heat treatment and cleaning. The reasons for this are unclear, although much can be gained from the development of an effcient laser machining process capable of processing diffcult materials such as high-performance steels and aerospace alloys. Existing laser machining processes selectively remove material by melt shearing and evaporation. Removing material by melting and evaporation leads to very low wall plug effciencies, and the process has difficulty competing with conventional mechanical removal methods. Adopting a laser machining solution for some materials offers the best prospects of effcient manufacturing operations. This paper presents a new laser machining process that relies on melt shear removal provided by a vertical high-speed gas vortex. Experimental and theoretical studies of a simple machining geometry have identifed a stable vortex regime that can be used to remove laser-generated melt effectively. The resultant combination of laser and vortex is employed in machining trials on 43A carbon steel. Results have shown that laser slot machining can be performed in a stable regime at speeds up to 150mm/min with slot depths of 4mm at an incident CO2 laser power level of 600 W. Slot forming mechanisms and process variables are discussed for the case of steel. Methods of bulk machining through multislot machining strategies are also presented.

A matriz extracelular (MEC) é capaz de modular a adesão celular, induzindo processos de sinalização celular. No estado de aderência intermediária, induzido por proteínas matricelulares, as células tendem a se diferenciar, migrar e proliferar. A tenascina-C é uma proteína matricelular amplamente secretada em gliomas que está envolvida na proliferação e angiogênese tumoral. A MEC de gliomas, possui elevada incorporação de tenascina-C (TN-C), uma glicoproteína matricelular desadesiva que compete com a glicoproteína adesiva fibronectina (FN), desestabilizando os contatos focais e induzindo proliferação celular em gliomas. Neste trabalho nós nos propusemos a investigar o papel da TN-C tumoral no fenótipo angiogênico de células endoteliais. Recentemente em um trabalho publicado pelo nosso grupo observamos que as células endoteliais semeadas sobre matrizes de glioma (U373 MG) aderem menos e são deficientes na capacidade de formar tubos quando comparadas com àquelas plaqueadas sobre MEC de HUVECs. No entanto, neste trabalho, reproduzimos este fenótipo semeando as células endoteliais em suportes de TN-C /FN miméticos da composição da matriz tumoral nativa. Por western blotting, observamos um aumento na fosforilação em treonina 638 da proteína PKCα, um possível sítio inibitório, e um aumento na ativação de PKCδ. O efeito antagônico na regulação dessas isoformas de PKC foi demonstrado quando usamos inibidores seletivos de PKC α e δ e um ativador de PKCα (PMA). Observamos que quando tratamos as HUVECs plaqueadas sobre MEC de U373 com PMA, resgatamos a capacidade dessas células de formar tubos, o pré-tratamento dessas HUVECs com inibidor de PKC δ (rotlerina) resgatou parcialmente a capacidade tubulogênica dessas células. O pré-tratamento das HUVECs que foram semeadas sobre MEC da HUVEC (que formam tubos normalmente) com um inibidor de PKC α (RO320432) levou a diminuição da capacidade tubulogênica. Além disso, esta matriz também induz ativação de ERK e AKT. Investigamos também se o bloqueio dos diferentes domínios da TN-C na matriz derivada de glioma poderia, de alguma forma, reverter o defeito angiogênico das células, propiciado pela interação com a matriz extracelular de gliomas. O pré-tratamento da matriz extracelular de glioma com anticorpos anti-TN-C (contra os domínios FNIII 1-3, 4-5 FNIII e N-terminal) resgatou parcialmente a capacidade das células endoteliais de formar tubos. Nossos dados sugerem que a indução do fenótipo vascular observado em muitos gliomas, com predomínio de vasos mal formados e sub-funcionais, pode ser parcialmente devido ao comprometido da sinalização mediada por PKCs em células endoteliais, bem como do aumento da ativação das vias de ERK e Akt.

Fisheries models have traditionally focused on patterns of growth, fecundity, and survival of fish. However, reproductive rates are the outcome of a variety of interconnected factors such as life-history strategies, mating patterns, population sex ratio, social interactions, and individual fecundity and fertility. Behaviorally appropriate models are necessary to understand stock dynamics and predict the success of management strategies. Protogynous sex-changing fish present a challenge for management because size-selective fisheries can drastically reduce reproductive rates. We present a general framework using an individual-based simulation model to determine the effect of life-history pattern, sperm production, mating system, and management strategy on stock dynamics. We apply this general approach to the specific question of how size-selective fisheries that remove mainly males will impact the stock dynamics of a protogynous population with fixed sex change compared to an otherwise identical dioecious population. In this dioecious population, we kept all aspects of the stock constant except for the pattern of sex determination (i.e. whether the species changes sex or is dioecious). Protogynous stocks with fixed sex change are predicted to be very sensitive to the size-selective fishing pattern. If all male size classes are fished, protogynous populations are predicted to crash even at relatively low fishing mortality. When some male size classes escape fishing, we predict that the mean population size of sex-changing stocks will decrease proportionally less than the mean population size of dioecious species experiencing the same fishing mortality. For protogynous species, spawning-per-recruit measures that ignore fertilization rates are not good indicators of the impact of fishing on the population. Decreased mating aggregation size is predicted to lead to an increased effect of sperm limitation at constant fishing mortality and effort. Marine protected areas have the potential to mitigate some effects of fishing on sperm limitation in sex-changing populations.

Seeded zone-melt recrystallization using a dual electron beam system has been performed on silicon-on-insulator material, which was prepared with single-crystal silicon filling of the seed windows by selective epitaxial growth. The crystal quality has been assessed by a variety of microscopic techniques, and it is shown that single-crystal films 0.5-1.0 μm thick over 1.0 μm of isolating oxide may be prepared by this method. These films have considerably less lateral variation in thickness than standard material, in which the windows are not so filled. The filling method is suitable for both single- and multiple-layer silicon-on-insulator, and gives the advantages of excellent layer uniformity after recrystallization and improved planarity of the whole chip structure. Experiments using various amounts of seed window filling have shown that the lateral variations of silicon film thickness seen in unplanarized material are due to stress relief in the cap oxide when the silicon film is molten, rather than the effect previously postulated in which they were assumed to be due to the contraction of silicon on melting.

Selectivity studies using cod end and cover to determine the optimum cod end mesh size for commercial size groups of shrimps was carried out at Cochin during 1963-64 fishing season. The normality of the result was checked by trouser cod end method. Although the present investigation was mainly aimed to find out suitable cod end mesh size for commercial varieties of shrimps, five commonly occurring species of fishes were also taken. The 50% escape level, co-efficient of selectivity and selection factor for all the species were worked out. From the findings, the authors stress the necessity of increasing the cod end mesh size from the present condition (25.4 to 31.70 mm) to 41.65 mm fabricated mesh size to avoid depletion of the natural population.

Feeding experiments were conducted for 21 days to study the effect of live food (Tubifex sp.) and three prepared supplemental feeds on the growth and survival of 13 day old magur (C. batrachus) fry. It was observed that the growth of fry varied significantly (p<0.05) with different diets. The best growth was shown by the fry fed with Tubifex sp. followed by those fed with the diet containing yeast (30%), milk powder (30%) and chicken eggs (30%). The poorest growth rate was given by the fry fed on yeast (45%) and fish meal (45%). There was no significant difference in survival rates and condition factors among the fry fed with live food and prepared feeds.

This work demonstrates transmission at 2.5 Gbit/s across two wavelength-division multiplexing (WDM) network nodes, constructed using counter-propagating semiconductor optical amplifier (SOA) wavelength converters and an integrated wavelength-selective router separated by 45 km of fiber, with an overall penalty of 0.6 dB. Minimal degradation of the eye diagram is evident across the whole system. Full utilization of the capacity of the router would allow an aggregate 360-Gbit/s node capacity for a WDM channel of 2.5 Gb/s.
