The long-term variability of the Siberian High, the dominant Northern Hemisphere anticyclone during winter, is largely unknown. To investigate how this feature varied prior to the instrumental record, we present a reconstruction of a Dec-Feb Siberian High (SH) index based on Eurasian and North American tree rings. Spanning 1599-1980, it provides information on SH variability over the past four centuries. A decline in the instrumental SH index since the late 1970s, related to Eurasian warming, is the most striking feature over the past four hundred years. It is associated with a highly significant (p < 0.0001) step change in 1989. Significant similar to 3-4 yr spectral peaks in the reconstruction fall within the range of variability of the East Asian winter monsoon (which has also declined recently) and lend further support to proposed relationships between these largescale features of the climate system.

Asymmetric hydrogenation of C=C bonds is of the highest importance in organic synthesis, and such reactions are currently carried out with organometallic homogeneous catalysts. Achieving heterogeneous metal-catalyzed hydrogenation, a highly desirable goal, necessitates forcing the crucial enantiodifferentiating step to take place at the metal surface. By synthesis and application of six chiral sulfide ligands that anchor robustly to Pd nanoparticles and resist displacement, we have for the first time accomplished heterogeneous enantioselective catalytic hydrogenation of isophorone. High resolution XPS data established that ligand adsorption from solution occurred exclusively on the Pd nanoparticles and not on the carbon support. All ligands contained a pyrrolidine nitrogen to enable their interaction with the isophorone substrate while the sulfide functionality provided the required interaction with the Pd surface. Enantioselective turnover numbers of up to similar to 100 product molecules per ligand molecule were found with a very large variation in asymmetric induction between ligands: observed enantiomeric excesses increased with increasing size of the alkyl group in the sulfide. This likely reflects varying degrees of ligand dispersion on the surface: bulky substituent groups hinder close approach of ligand molecules to each other, inhibiting close-packed island formation, favoring dispersion as separate molecules, and leading to effective asymmetric induction. Conversely, small substituents favor island formation leading to very low asymmetric induction. Enantioselective reaction most likely involves initial formation of an enamine or iminium species, confirmed by use of an analogous tertiary amine, which leads to racemic product. Ligand rigidity and resistance to self-assembled monolayer formation are important attributes that should be designed into improved chiral modifiers.

Physiological evidence using Infrared Video Microscopy during the uncaging of glutamate has proven the existence of excitable calcium ion channels in spine heads, highlighting the need for reliable models of spines. In this study we compare the three main methods of simulating excitable spines: Baer & Rinzel's Continuum (B&R) model, Coombes' Spike-Diffuse-Spike (SDS) model and paired cable and ion channel equations (Cable model). Tests are done to determine how well the models approximate each other in terms of speed and heights of travelling waves. Significant quantitative differences are found between the models: travelling waves in the SDS model in particular are found to travel at much lower speeds and sometimes much higher voltages than in the Cable or B&R models. Meanwhile qualitative differences are found between the B&R and SDS models over realistic parameter ranges. The cause of these differences is investigated and potential solutions proposed.

The associative sequence learning model proposes that the development of the mirror system depends on the same mechanisms of associative learning that mediate Pavlovian and instrumental conditioning. To test this model, two experiments used the reduction of automatic imitation through incompatible sensorimotor training to assess whether mirror system plasticity is sensitive to contingency (i.e., the extent to which activation of one representation predicts activation of another). In Experiment 1, residual automatic imitation was measured following incompatible training in which the action stimulus was a perfect predictor of the response (contingent) or not at all predictive of the response (noncontingent). A contingency effect was observed: There was less automatic imitation indicative of more learning in the contingent group. Experiment 2 replicated this contingency effect and showed that, as predicted by associative learning theory, it can be abolished by signaling trials in which the response occurs in the absence of an action stimulus. These findings support the view that mirror system development depends on associative learning and indicate that this learning is not purely Hebbian. If this is correct, associative learning theory could be used to explain, predict, and intervene in mirror system development.

As an obligatory parasite of humans, the body louse (Pediculus humanus humanus) is an important vector for human diseases, including epidemic typhus, relapsing fever, and trench fever. Here, we present genome sequences of the body louse and its primary bacterial endosymbiont Candidatus Riesia pediculicola. The body louse has the smallest known insect genome, spanning 108 Mb. Despite its status as an obligate parasite, it retains a remarkably complete basal insect repertoire of 10,773 protein-coding genes and 57 microRNAs. Representing hemimetabolous insects, the genome of the body louse thus provides a reference for studies of holometabolous insects. Compared with other insect genomes, the body louse genome contains significantly fewer genes associated with environmental sensing and response, including odorant and gustatory receptors and detoxifying enzymes. The unique architecture of the 18 minicircular mitochondrial chromosomes of the body louse may be linked to the loss of the gene encoding the mitochondrial single-stranded DNA binding protein. The genome of the obligatory louse endosymbiont Candidatus Riesia pediculicola encodes less than 600 genes on a short, linear chromosome and a circular plasmid. The plasmid harbors a unique arrangement of genes required for the synthesis of pantothenate, an essential vitamin deficient in the louse diet. The human body louse, its primary endosymbiont, and the bacterial pathogens that it vectors all possess genomes reduced in size compared with their free-living close relatives. Thus, the body louse genome project offers unique information and tools to use in advancing understanding of coevolution among vectors, symbionts, and pathogens.

A Biossegurança constitui um campo do conhecimento com limites amplos e em constante construção. Entretanto, no Brasil, sua evolução teve fraco acompanhamento, especialmente em relação aos conhecimentos e procedimentos dos profissionais e infraestrutura dos laboratórios nível de Biossegurança 3, com poucos dados que possam indicar o grau de desenvolvimento e aplicabilidade de suas normas nas diversas instituições de saúde. Contudo, não bastam a adequação e funcionabilidade da infraestrutura predial, mas sim o conhecimento das necessidades dos usuários no sentido de propiciar aceitação das normas de Biossegurança por meio da conscientização dos riscos levando à mudança na conduta e no comportamento. Para isto, torna-se imprescindível a elaboração de treinamentos teóricos e práticos com atualizações constantes dos profissionais da área de saúde que deverão ser avaliados periodicamente para que haja melhorias nos programas de treinamentos oferecidos, uma vez que esses programas visam a garantir a qualidade e a segurança dos profissionais, dos ambientes internos e externos, à população em geral e a qualidade dos resultados pretendidos. Neste contexto, este trabalho teve, como objetivo principal, avaliar, a partir da pesquisa descritiva com abordagem qualitativa e exploratória, os dados coletados por questionários e entrevistas estruturadas, aplicadas aos 37 profissionais e nove gestores dos seis laboratórios NB3 participantes. A análise dos dados foi feita pela abordagem da multirreferencialidade, considerando os conhecimentos e procedimentos em Biossegurança e os possíveis impactos das estratégias dos últimos treinamentos oferecidos a esses profissionais, assim como as condições das instalações laboratoriais no país Dentre as respostas obtidas, há constatações importantes: alguns profissionais relataram não ter recebido treinamento específico para o início dos trabalhos, demonstrando falta de organização e critérios, por parte das instituições e gestores, para o ingresso de novos trabalhadores em laboratórios NB3; não houve padronização de conceitos e percepções relacionados à Biossegurança dentre os profissionais avaliados; mais da metade dos laboratoristas, incluindo gestores, alegaram trabalhar sozinhos em ambientes nível 3; a maior parte dos profissionais relatou que os laboratórios foram bem construídos mas todos, incluindo usuários e gestores, afirmaram que gostariam que a situação atual fosse melhor. Em relação à gestão dos laboratórios, foram relatadas várias falhas, tais como falta ou não atualização de POP específico para o trabalho em NB3, ausência de documentos de notificação de acidentes, falta de acompanhamento dos profissionais destinados à limpeza do laboratório NB3, ausência de realização de exames clínicos, imunizações e armazenamento de soro base como controle. De acordo com as respostas apresentadas, observamos que nem todos os indivíduos, incluídos neste estudo, trabalham da maneira correta ou seguem uma metodologia de trabalho padronizada, deficiência relatada, inclusive, pela maioria dos próprios profissionais A realização dos cursos de Biossegurança e treinamentos somente não garantem preparo pleno dos profissionais para o exercício de suas obrigações em laboratórios NB3. Esta atividade e o monitoramento das instalações NB3 devem ser connuos e de responsabilidade dos profissionais e das instituições, garantindo que as rotinas de trabalho estejam de acordo com as normas de Biossegurança
