We have previously shown that the subunit 1 of Leishmania amazonensis RPA (LaRPA-1) alone binds the G-rich telomeric strand and is structurally different from other RPA-1. It is analogous to telomere end-binding proteins described in model eukaryotes whose homologues were not identified in the protozoan's genome. Here we show that LaRPA-1 is involved with damage response and telomere protection although it lacks the RPA1N domain involved with the binding with multiple checkpoint proteins. We induced DNA double-strand breaks (DSBs) in Leishmania using phleomycin. Damage was confirmed by TUNEL-positive nuclei and triggered a G1/S cell cycle arrest that was accompanied by nuclear accumulation of LaRPA-1 and RAD51 in the S phase of hydroxyurea-synchronized parasites. DSBs also increased the levels of RAD51 in non-synchronized parasites and of LaRPA-1 and RAD51 in the S phase of synchronized cells. More LaRPA-1 appeared immunoprecipitating telomeres in vivo and associated in a complex containing RAD51, although this interaction needs more investigation. RAD51 apparently co-localized with few telomeric clusters but it did not immunoprecipitate telomeric DNA. These findings suggest that LaRPA-1 and RAD51 work together in response to DNA DSBs and at telomeres, upon damage, LaRPA-1 works probably to prevent loss of single-stranded DNA and to assume a capping function.

The purpose of this study was to investigate the effect of thermal cycling and disinfection on the microhardness of acrylic resins denture base. Four different brands of acrylic resins were evaluated: Onda Cryl, QC 20, Classico and Lucitone. Each brand of acrylic resin was divided into four groups (n = 7) according to the disinfection method (microwave, Efferdent, 4% chlorhexidine and 1% hypochlorite). Samples were disinfected during 60 days. Before and after disinfection, samples were thermal cycled between 5-55 °C with 30-s dwell times for 1000 cycles. The microhardness was measured using a microhardener, at baseline (B), after first thermal cycling (T1), after disinfection (D) and after second thermal cycling (T2). The microhardness values of all groups reduced over time. QC-20 acrylic resin exhibited the lowest microhardness values. At B and T1 periods, the acrylic resins exhibited statistically greater microhardness values when compared to D and T2 periods. It can be concluded that the microhardness values of the acrylic resins denture base were affected by the thermal cycling and disinfection procedures. However, all microhardness values obtained herein are within acceptable clinical limits for the acrylic resins. © 2013 Informa UK Ltd.

A total of 360 pacus (Piaractus mesopotamicus) were used to study vascular permeability (VP) and inflammatory cell component (CC) in induced aerocystitis in P. mesopotamicus through inoculation of inactivated Aeromonas hydrophila, and the effect of steroidal and nonsteroidal anti-inflammatory drugs. It was observed that after inoculation of A. hydrophila, the maximum VP occurred 180 min post-stimulus (MPS). Pretreatment with anti-inflammatory drugs inhibited VP, and the inhibitory effect of dexamethasone was seen earlier than the effects caused by meloxicam and indomethacin. Inoculation of the bacterium caused a gradual increase in the accumulation of cells, which reached a maximum 24 h post-stimulus (HPS). Pretreatment with dexamethasone, indomethacin and meloxicam reduced the accumulation of lymphocytes, thrombocytes, granulocytes and macrophages. There was no significant difference between the different doses of the drugs tested. The results suggest that eicosanoids and pro-inflammatory cytokines participate in chemical mediation in acute inflammation in pacus. © 2013 Elsevier Ltd.
