We investigated the role of the cdk inhibitor protein p21Cip-1/WAF1/MDA6 (p21) in the ability of MAPK pathway inhibition to enhance radiation-induced apoptosis in A431 squamous carcinoma cells. In carcinoma cells, ionizing radiation (2 Gy) caused both primary (0–10 min) and secondary (90–240 min) activations of the MAPK pathway. Radiation induced p21 protein expression in A431 cells within 6 h via secondary activation of the MAPK pathway. Within 6 h, radiation weakly enhanced the proportion of cells in G1 that were p21 and MAPK dependent, whereas the elevation of cells present in G2/M at this time was independent of either p21 expression or MAPK inhibition. Inhibition of the MAPK pathway increased the proportion of irradiated cells in G2/M phase 24–48 h after irradiation and enhanced radiation-induced apoptosis. This correlated with elevated Cdc2 tyrosine 15 phosphorylation, decreased Cdc2 activity, and decreased Cdc25C protein levels. Caffeine treatment or removal of MEK1/2 inhibitors from cells 6 h after irradiation reduced the proportion of cells present in G2/M phase at 24 h and abolished the ability of MAPK inhibition to potentiate radiation-induced apoptosis. These data argue that MAPK signaling plays an important role in the progression/release of cells through G2/M phase after radiation exposure and that an impairment of this progression/release enhances radiation-induced apoptosis. Surprisingly, the ability of irradiation/MAPK inhibition to increase the proportion of cells in G2/M at 24 h was found to be dependent on basal p21 expression. Transient inhibition of basal p21 expression increased the control level of apoptosis as well as the abilities of both radiation and MEK1/2 inhibitors to cause apoptosis. In addition, loss of basal p21 expression significantly reduced the capacity of MAPK inhibition to potentiate radiation-induced apoptosis. Collectively, our data argue that MAPK signaling and p21 can regulate cell cycle checkpoint control in carcinoma cells at the G1/S transition shortly after exposure to radiation. In contrast, inhibition of MAPK increases the proportion of irradiated cells in G2/M, and basal expression of p21 is required to maintain this effect. Our data suggest that basal and radiation-stimulated p21 may play different roles in regulating cell cycle progression that affect cell survival after radiation exposure.

The crystal structure of human endostatin reveals a zinc-binding site. Atomic absorption spectroscopy indicates that zinc is a constituent of both human and murine endostatin in solution. The human endostatin zinc site is formed by three histidines at the N terminus, residues 1, 3, and, 11, and an aspartic acid at residue 76. The N-terminal loop ordered around the zinc makes a dimeric contact in human endostatin crystals. The location of the zinc site at the amino terminus, immediately adjacent to the precursor cleavage site, suggests the possibility that the zinc may be involved in activation of the antiangiogenic activity following cleavage from the inactive collagen XVIII precursor or in the cleavage process itself.

The European Large Subunit Ribosomal RNA Database compiles all complete or nearly complete large subunit ribosomal RNA sequences available from public sequence databases. These are provided in aligned format and the secondary structure, as derived by comparative sequence analysis, is included. Additional information about the sequences such as literature references and taxonomic information is also included. The database is available from our WWW server at http://rrna.uia.ac.be/lsu/.

The transporter associated with antigen processing (TAP) is essential for intracellular transport of protein fragments into the endoplasmic reticulum for loading of major histocompatibility complex (MHC) class I molecules. On the cell surface, these peptide–MHC complexes are monitored by cytotoxic T lymphocytes. To study the ATP hydrolysis of TAP, we developed an enrichment and reconstitution procedure, by which we fully restored TAP function in proteoliposomes. A TAP-specific ATPase activity was identified that could be stimulated by peptides and blocked by the herpes simplex virus protein ICP47. Strikingly, the peptide-binding motif of TAP directly correlates with the stimulation of the ATPase activity, demonstrating that the initial peptide-binding step is responsible for TAP selectivity. ATP hydrolysis follows Michaelis–Menten kinetics with a maximal velocity Vmax of 2 μmol/min per mg TAP, corresponding to a turnover number of approximately 5 ATP per second. This turnover rate is sufficient to account for the role of TAP in peptide loading of MHC molecules and the overall process of antigen presentation. Interestingly, sterically restricted peptides that bind but are not transported by TAP do not stimulate ATPase activity. These results point to coordinated dialogue between the peptide-binding site, the nucleotide-binding domain, and the translocation site via conformational changes within the TAP complex.

A finales de la década de los años veinte el cine mudo estaba llegando a su fin y comenzaron a producirse masivamente películas habladas. Los grandes estudios de Hollywood temen perder público fuera de Estados Unidos y antes de decidirse por el doblaje, ya que este sistema presentaba en aquella época problemas de sincronización, comienzan filmar versiones múltiples de un mismo guion en diferentes idiomas para exportar a los países de habla no inglesa. Dentro de todas las películas en español producidas por Hollywood, esta tesis doctoral está centrada únicamente en las de género fantástico. Es cierto que en las versiones en español encontramos pocas de dicho género, pero dentro de las existentes hay películas muy variadas, desde películas totalmente de género como las dos versiones de Drácula hasta otras que contienen elementos fantásticos lo suficientemente interesantes como para ser incluidas en esta investigación. La mayoría de las versiones en español que se produjeron en la década de los años treinta eran comedias o melodramas, los dos géneros más populares de la época. Pero de las más de cien películas en español que se filmaron en Hollywood durante la década de los años treinta algunas pueden catalogarse dentro del género fantástico. Estas películas son: ¡Pobre infeliz! (Charles Rogers, 1930), versión en español de The Shrimp (Ídem, Charles Rogers ,1930); Noche de duendes (James Parrott, 1930), versión en español de The Laurel-Hardy Murder Case (Ídem, James Parrot, 1930); La voluntad del muerto (George Melford, 1930), versión en español de The Cat Creeps (Rupert Julian, John Willard, 1930); Wu Li Chang (Nick Grinde, 1930), versión en español de Mr. Wu (William Night, 1927); Drácula (George Melford, 1931), versión en español de Drácula (Dracula, Tod Browning, 1931); Cheri-Bibi (Carlos F. Borcosque, 1931), versión en español de The Phantom of Paris (Ídem, John S. Robertson, 1931) y El último varón sobre la tierra (James Tinling, 1933), versión en español de It's Great to Be Alive (Alfred L. Werker, 1933)...

During the last months, the number of reports on Holstein calves suffering from incurable idiopathic diarrhea dramatically increased. Affected calves showed severe hypocholesterolemia and mostly died within days up to a few months after birth. This new autosomal monogenic recessive inherited fat metabolism disorder, termed cholesterol deficiency (CD), is caused by a loss of function mutation of the bovine gene. The objective of the present study was to investigate specific components of lipid metabolism in 6 homozygous for the mutation (CDS) and 6 normal Holstein calves with different genotypes. Independent of sex, CDS had significantly lower plasma concentrations of total cholesterol (TC), free cholesterol (FC), high-density lipoprotein cholesterol (HDL-C), low-density lipoprotein cholesterol (LDL-C), very-low-density lipoprotein cholesterol (VLDL-C), triacylglycerides (TAG), and phospholipids (PL) compared with homozygous wild-type calves ( < 0.05). Furthermore, we studied the effect of the genotype on cholesterol metabolism in adult Holstein breeding bulls of Swissgenetics. Among a total of 254 adult males, the homozygous mutant genotype was absent, 36 bulls were heterozygous carriers (CDC), and 218 bulls were homozygous wild-type (CDF). In CDC bulls, plasma concentrations of TC, FC, HDL-C, LDL-C, VLDL-C, TAG, and PL were lower compared with CDF bulls ( < 0.05). The ratios of FC:cholesteryl esters (CE) and FC:TC were higher in CDC bulls compared with CDF bulls, whereas the ratio of CE:TC was lower in CDC bulls compared with CDF bulls ( < 0.01). In conclusion, the CD-associated mutation was shown to affect lipid metabolism in affected Holstein calves and adult breeding bulls. Besides cholesterol, the concentrations of PL, TAG, and lipoproteins also were distinctly reduced in homozygous and heterozygous carriers of the mutation. Beyond malabsorption of dietary lipids, deleterious effects of apolipoprotein B deficiency on hepatic lipid metabolism, steroid biosynthesis, and cell membrane function can be expected, which may result in unspecific symptoms of reduced fertility, growth, and health.

During three Antarctic expeditions (2004, ANT XXI-4 and XXII-2; 2006, ANT XXIII-6) with the German research icebreaker R/V Polarstern, six different amphipod species were recorded under the pack ice of the Weddell Sea and the Lazarev Sea. These cruises covered Austral autumn (April), summer (December) and winter (August) situations, respectively. Five of the amphipod species recorded here belong to the family Eusiridae (Eusirus antarcticus, E. laticarpus, E. microps, E. perdentatus and E. tridentatus), while the last belongs to the Lysianassidea, genus Cheirimedon (cf. femoratus). Sampling was performed by a specially designed under-ice trawl in the Lazarev Sea, whereas in the Weddell Sea sampling was done by scuba divers and deployment of baited traps. In the Weddell Sea, individuals of E. antarcticus and E. tridentatus were repeatedly observed in situ during under-ice dives, and single individuals were even found in the infiltration layer. Also in aquarium observations, individuals of E. antarcticus and E. tridentatus attached themselves readily to sea ice. Feeding experiments on E. antarcticus and E. tridentatus indicated a carnivorous diet. Individuals of the Lysianassoid Cheirimedon were only collected in baited traps there. Repeated conventional zooplankton hauls performed in parallel to this study did not record any of these amphipods from the water column. In the Lazarev Sea, E. microps, E. perdentatus and E. laticarpus were regularly found in under-ice trawls. We discuss the origin and possible sympagic life style of these amphipods.
